Histopathological Study and Expression of Beta-Catenin in Congenital Choledochal Cyst in a Tertiary Care Pediatric Referral Center in South India

  • Purpose: Choledochal cysts are congenital anomalies that occur as localized cystic or fusiform dilatations of the biliary tree. Reflux and stasis of pancreatic enzymes in the biliary duct may relate to the development of intestinal metaplasia which might be an important factor related to the carcinogenesis of choledochal cyst, thus the expression of beta-catenin in the metaplastic epithelium might be associated with malignant transformation of choledochal cyst epithelium. Methods: This study was conducted at a tertiary care pediatric center between October 2014 and March 2017. Forty patients were evaluated for epithelial lining, mural ulceration, fibrosis, inflammation, and metaplasia. Results: Out of 40, 12 cases (30.0%) were the infantile age group and 28 cases (70.0%) were in the classic pediatric group. Ulceration was classified as grade 0 (14 cases, 35.0%), grade 1 (17 cases, 42.5%), or grade 2 (nine cases, 22.5%). Inflammation was classified as grade 0 (2 cases, 5.0%), grade 1 (26 cases, 65.0%), or grade 2 (12 cases, 30.0%). Fibrosis was classified as grade 0 (five cases, 12.5%), grade 1 (11 cases, 27.5%), grade 2 (17 cases, 42.5%), or grade 3 (seven cases, 17.5%). Metaplasia was noted in five (12.5%) out of 40 cases. All choledochal cysts with metaplasia showed beta-catenin nuclear positivity on immunohistochemistry and were followed up. Conclusion: This study emphasizes the importance of detailed histopathological examination and documentation of metaplastic changes. Metaplasia was associated with beta-catenin nuclear positivity. These findings suggest a potential role for beta-catenin as a marker of metaplastic changes in choledochal cysts.

The recovery effect of polyacetylene constituents from Acanthopanax senticosus extracts on hepatic injury in dogs

  • The present study was performed to clarify the recovery effect of polyacetylene constituents from Acanthopanax senticosus ethanol extracts on $CCl_4$-induced acute hepatic injury in dogs. Sixteen clinically healthy dogs were divided into the control group (six dogs), and experimental group I (three dogs), II (three dogs) and III (four dogs). Acute hepatic injury was induced by single intraperitoneal injection of 1 ml/kg $CCl_4$ solution ($CCl_4$ : olive oil = 1 : 1) after sterilization. The control group was orally administered with 10 ml of corn oil, and experimental groups I, II and III were orally administered with extracts of A. senticosus at the dose of 5, 10 and 30 mg/kg in 10 ml corn oil for seven consecutive days at 24 h after $CCl_4$ injection. We assessed changes occurring in serum ALT, AST and GGT activities on the pre, day 0, day 1, day 2, day 3, day 5 and day 7, together with the histopathological findings. In experimental group I, the significant changes in serum ALT activities were found on day 5 (p<0.05) and day 7 (p<0.05), compared with those in the control group. However, significant differences were not found in the changes of serum AST and GGT activities. In experimental group II, significant changes were found on day 2 (p<0.05), day 3(p<0.05), day 5 (p<0.05) and day 7 (p<0.05) in serum ALT activities, on day 1 (p<0.05) in serum AST, and on day 2 (p<0.05) and day 3 (p<0.05) in serum GGr activities, respectively. In experimental group III, the changes in serum ALT, AST and GGT activities were not significant. In the histopathological evaluation, experimental group I and II showed much improvement, while experimental group III became worse when compared with the control group. Based on the results of the present study, it was concluded that low dose (below 10 mg/kg) administration of polyacetylene constituents from A. senticosus ethanol extracts was effective for recovery of acute hepatic injury induced by $CCl_4$ in dogs.

Single-Dose Toxicity and Four Week Repeated-Dose Toxicity Study on Tensolin-F® (3,9-diferuloyl-6-oxopterocarpen) (Tensolin-F® (3,9-diferuloyl-6-oxopterocarpen)의 단회 독성시험 및 4주 반복투여 독성시험)

  • This study was to investigate single and repeated-dose toxicities of Tensolin-$F^{(R)}$, an anti-wrinkle agent, in Sprague-Dawley (SD) rats or ICR mice. In single-dose oral toxicity study, the test materials were administered once by gavage to male and female SD rats at dose levels of 0 and 2,000 mg/kg. No dead animals and abnormal necropsy findings were found in control and Tensolin-$F^{(R)}$ treated group. Therefore, the approximate lethal dose of Tensolin-$F^{(R)}$ was considered to be higher than 2,000 mg/kg in rats. In the 4-week repeated oral toxicity study, the test material was administered once daily by gavage to male and female ICR mice at dose levels of 0, 25, 50 and 100 mg/kg/day for 4-weeks. In the results, no abnormality was observed in mortality, clinical findings, body weight changes, food and water consumptions, opthalmoscopic findings, necropsy findings, histopathological findings. In hematological analysis, there was a trend of increase in reticulocyte at male 25 mg/kg, although such changes were in normal ranges. On the other hand, there was a trend of decrease in hemoglobin at female 50, 100 mg/kg, such changes were in normal ranges. In addition, serum biochemical parameters including sodium, BUN and chloride increased at 25, 50 and 100 mg/kg. Relative organ weights of right testis, brain, lung and left epididymis were increased in 100 mg/kg groups of male rats in contrast to not change in female groups. However, these changes of relative organ weights, hematological and serum biochemical parameters were not accompanied with related signs such as histopathological changes or clinical findings. In conclusion, 4-week repeated oral dose of Tensolin-$F^{(R)}$ to ICR mice did not cause apparent toxicological change at the dose of 25, 50, 100 mg/kg body weight. Consequently the no-observed-adverse-effect level (NOAEL) for Tensolin-$F^{(R)}$ in ICR mice following gavage for at least 4-week is higher than 100 mg/kg/day.

Effects of Ligature of Mandibular Gland and Sublingual Gland on the Mandibular Gland in Dogs (개에서 턱밑샘관 및 혀밑샘관의 결찰이 턱밑샘에 미치는 영향)

  • It was performed to evaulate the effects of occlusion of mandibular gland and sublingual gland on the mandibular gland in dogs. The occlusion of mandibular gland and sublingual gland was induced by ligature in clinically normal dogs, and ultrasonograpic appearance, hematological changes and histological changes of the mandibular gland were observed daily during 6 days. The results were as follows. 1. The length, width, circumference and area of the mandibular gland in clinically normal dogs were 25.1 mm, 8.4 mm, 56.4 mm and 163.0 $\textrm{mm}^2$ in the left, and 23.4 mm, 8.0 mm, 53.3 mm and 141.6 $\textrm{mm}^2$in the right. 2. The length, width, circumference and area of the mandibular gland were gradually increased after ligature of the salivary duct, and revealed maximum values, as 33.4 mm, 10.9 mm, 73.0 mm and 287.2 $\textrm{mm}^2$, respectively, at 24 hours after ligature, and returned to normal size at 3 days after ligature. 3. The size of the opposite mandibular gland without ligature was also gradually increased until 24 hours, and returned to normal size at 3 days, similar to the changes of the mandibular gland with ligature. 4. There were no significant changes after ligature of the salivary duct in hematological findings. 5. In histopathological findings, the major of glandular alveoli was enlarged at 24 hours after ligature of the salivary duct, and neutrophils and monocytes were infiltrated at 6 days after ligature.

Effects of Cryosurgery of the Third Eyelid Gland on Tear Production and Histological Changes in Dogs (개에서 제3안검선 냉동수술이 눈물생산량 및 조직학적 변화에 미치는 영향)

  • The effects of cryosurgery on tear production and histological changes of the third eyelid were studied in dogs. Clinically normal 12 mixed breeds weighing 2∼6 kg were divided into three groups and treated as follows; 45 seconds double freeze-thaw treated group, 60 seconds double freeze-thaw treated group and 90 seconds double freeze-thaw treated group. The significant decrease of the tear production after cryosurgery was shown in all groups throughout the observed periods(p<0.05). However, there was no difference among groups. The main complications after cryosurgery were chemosis and conjunctival injection. Other complications such as eyelid edema, eyelid depigmentation and keratitis were more preominent in group III compared to those of groups I and II. On histopathological examination, chronic inflammatory changes and regeneration of the third eyelid glands were noted in group I and predominated loss of the third eyelid glands and necrosis of hyaline cartilage were observed in group III However, such changes were less appeared in group II. The results of this study suggested that double freeze-thaw cryosurgery for 60 seconds on the third eyelid glands would be the most effective method for treating tear staining syndrome.

Histopathological changes in lymphoid organs of chickens inoculated with IBDV (SBV92) (IBDV (SH/92)의 인공감염에 의한 닭 면역장기의 병리조직학적 연구)

  • Sequential morphologic changes in the lymphoid organs were examined after ocular and cloacal inoculation in 3weekold chicks with a highly virulent strain (SH/92) of infectious bursal disease virus (IBDV). The infected chickens were sacrificed at 6, 12, 24, 48, 72, and 96 hrs post inoculation (Pl), and thymus, harderian gland, ceacal tonsil, and spleen were observed. Histologically, the significant lesions were characterized by lymphocyte depletion and the earliest detectable changes were evident at 12 hrs Pl. In thymic cortex, lymphoid depletion with apoptosis and prominent "tingible body macrophages" were observed. As the infection advanced, the lesions showed more severe changes. Dying cells were characterized either by capping of nuclear chromatin (apoptosis) or by cytoplasmic swelling (necrosis). In situ staining for apoptosis, some lymphoid cells revealed typical positive reaction, even the stainability was variable depend on every lymphoid organs. These results suggest that IBBV (SH/92) cause severe damage both primary and secondary lymphoid organs, and both T and B lymphocytes. Also the lymphoid depletion of these organs is caused by necrosis and apoptosis induced by IBDV.d by IBDV.

  • This study was concerned with assessment of dietynitrosamine (DEN) induced liver cell carcinogenesis by measurement of changes preceding the development of neoplasms. The changes of hepatic morphology in rats(Sprague-Dawley) were detected by hematoxylin-eosin stain and immunohistochemistry(PCNA). The results were as follows ; 1. Minor behavioral change, brittleness of hair and decreased amount of water and diet intake. were observed in rats 7 weeks after DEN administration. 2. Variable size of liver tumor and hepatomegaly were observed in rats treated with DEN after 10 weeks. 3. Numerous vacuoles were showed on the midzonal and or peripheral areas of hepatic lobules. The large and polymorphological hepatocytes with eosinophilic cytoplasm or densely basophilic mitotic nucleoli were showed. 4. Several proliferative small round cells were shown on vacuolated and necrotic areas in peripheral hepatic lobules or portal areas. 5. PCNA-positive cells were showed on the vacuolated portal areas and peripheral areas of hepatic lobules. Maximal positivity was 23.6% in the areas of small round cells. In conclusion, this result confirmed that the DEN was one of the potent hepatocarcinogens. In histopathological analysis, the altered foci, hyperplastic nodules, neoplastic nodules, adenomas and carcinomas were observed in liver tumors induced by administration of DEN in rats.

Subacute Toxicity Test of Guh Sung Y.L.S.-95 (Guh Sung Y.L.S.-95의 아급성 독성시험)

  • Guh Sung Y.L.S.-95 is one of the polyacidic solution of which main component is acetic acid. We investigated the subchronic toxicity of the Guh Sung Y.L.S.-95 using SPF ICR mouse for 4 weeks. The Guh Sung Y.L.S.-95 was administered by gastric intubation, 1.0, 2.5, 5.0 g/kg body weight. The results are as follows: 1. There are no adverse effects on the clinical obserbation and body weight changes. Also, there are some significant changes in organ weight, but it was meaningless because of the absence of dose-response relationships. 2. In the hematological patterns of administered mouse, there are no significant changes between the treated groups. Also, there are no serological enzymatic changes in the treated mouse. In the 1.0 g/kg treated group, ASP activity was increased significnatly compared with control group. But, this level of activity was fall under the normal physiological range of control mouse. 3. Histopathological findings of the brain, liver, heart, spleen, kidneys, stomach, lung, testis, ovary, uterus and thymus were not observed in the treated mouse. From the above results, the Guh Sung Y.L.S.-95 has no toxicity upto the 5.0 g/kg/day of oral dose for 4 weeks.

  • This study was designed to evaluate a repeated intravenous dose toxicity of a new anticancer agent, SB extracted from Pulsatilla korean Nakai in Beagle dogs. Animals were intravenously injected with dosages of 0, 0.062, 0.25, and 1 mg/kg of SB everyday for 13 weeks, respectively. There were no dose-related changes in clinical signs, body weight changes, food and water consumptions, opthalmoscopy, and urine analysis. There were somewhat significant differences compared with control group in organ weight, biochemical examination, and hematology findings of animals treated with SB. However, these changes were not dose-related changes. Gross and histopathological findings revealed no evidence of specific toxicity related to SB. These indicate that intraveous maximum tolerated dose value of SB may be over 1mg/kg in rats.

Experimental Studies of Kamidaebotang on the Hypothyroidism (가감대보탕(加減大補湯)이 백서(白鼠)의 갑상선기능저하증(甲狀腺機能低下症)에 미치는 영향(影響))

  • To study the effect of Kamidaebotang on the hypothyroidism induced by thiourea in SD(Splague Dawley) rats, the weight of thyroid gland, the level of serum $T_3\;and\;T_4$. total cholesterol, hematological changes and histopathological findings were measured. The following results were obtained. 1. Thyroid gland weight of the Kamidaebotang B treated group was significantly increased as compared with that of control group(p<0.05). 2. The level of serum $T_3$ of the Kamidaebotang B treated group was slightly increased as compared with that of control group. But no significant changes appeared between two groups. 3. The level of serum $T_4$ of the Kamidaebotang A, B treated groups were significantly increased as compared with that of control group(p<0.05). 4. Total cholesterol of the Kamidaebotang A, B treated groups were not significant changes as compared with that of control group. 5. Hematological data of the Kamidaebotang A, B treated groups were not significant changes as compared with that of control group. 6. Histopathologically, Kamidaebotang B administration recovered effectly the dystropy, degeneration of thyroid follicle induced by thiourea. From above results, it is suggested that Kamidaebotang B is available to the hypothyroidism induced by thiourea in rats. Therefore, Kamidabotang B seems to be applicable to hypothyroidism in clinical practice.
