Gonadal Development and Reproductive Cycle of the North Pacific Seastar, Asterias amurensis(Echinodermata: Asteroidea) (아무르불가사리, Asterias amurensis의 생식소 발달과 생식주기)

  • Gonadal development and reproductive cycle of the North Pacific seastar, Asterias amurensis captured from the Gosung, Gyeongsangnamdo, between November 2003 and February 2005, was investigated monthly changes of gonadosomatic index(GSI), gonadal development and oocyte size-frequency distribution. Monthly changes of GSI values showed similar trends in female and male. GSI values were reached the maximum in March($3.88{\pm}3.04$ in female, $0.87{\pm}0.57$ in male), and then gradually decreased. Based on the monthly changes of GSI, histological observation of gonadal development, reproductive cycle was divided into following successive stage: growing stage(October to January), mature stage(February to March), spent stage(March to April), degeneration and resorption stage(April to May), recovery stage(July to September). based on these result, this species seemed to have a synchronous oocyte development and one spawning season a year.

Experimental Study on Anti-body effects of Anti-BV on the Bee Venom Herbal Acupuncture (Anti-BV의 봉약침 항체 효능에 관한 실험적 연구)

  • Objectives : To observe physiological anti-body effects of anti-BV, acute toxic response, measurement of $LD_{50}$, and the effects of anti-body were evaluated. Methods : $LD_{50}$ of Anti-Bee Venom were measured, and to analyze acute toxic responses, weight, and the anti-body effects various concentrations of Anti-BV were diluted and the survival rate was measured. Cell blood count (CBC), liver, spleen, and kidney pathologies were observed from the histological aspects. Results : Experiment was conducted to observe Anti-BV as the anti-body to the bee venom and the following results were obtained : 1.anti-BV was injected intraperitoneally and no toxic responses were witnessed. All of the experiment subjects stayed alive during the experiment, making $LD_{50}$ analysis impossible. 2.Anti-BV was injected intraperitoneally in mice and no significant weight changes were measured between the control group and the experiment groups. 3. Measuring the concentration dependent survival rate, the highest survival rate was at the concentration of $1.25{\times}10^2mg/kg$(1/2.000) for Anti-BV. 4. No particular results were shown in the CBC test. 5. Observation of changes in the organ tissues, Anti-BV was found to suppress blood stasis in the liver and inhibit necrosis of the cells. Conclusion : Above results suggest that Anti-BV doesn't cause any toxic responses in the body and works as an anti-body to the bee venom. Further studies must be followed to secure the findings.

Effects of Baedokhwan on Immune Modulation in Atopic Dermatitis Model of NC/Nga Mice (배독환(排毒丸)의 아토피피부염 병태모델에서의 면역 억제 효능)

  • Atopic dermatitis induced NC/Nga mice were used to investigate the efficacy of BDH(Baedokhwan) on the recovery of dermatitic symptoms through its influence on the immune related factors and histological changes. First of all, BDH treated group showed improvement of atopic dermatitis with naked eye observation, and significant decrease of clinical index(CI) was observed after 14 weeks. And Infiltration of leukocytes was suppressed in BDH treated group, and the thickness of hypertrophied epidermis and dermis were decreased. In dorsal skin, BDH treated group showed significant decrease of the ratio of CD3+, CD11b+/Gr-1+ immune cells by 52.8%, 25.2, respectively. And also significant decrease the level of IL-5 mRNA and IL-13 mRNA by 44.4%, 28.0, respectively. In PBMC and serum, BDH treated group showed an decrease of CD4+/CD45+, B220+/CD23+, CD4+, CD8+, CD4+/CD25+ immune cells by 35.0%, 12.6%, 42.7%, 31.6% and 55.6%, respectively, and the level of histamine was decreased by 39.0%. The results above indicated that BDH clinically used for atopic dermatitis treatment has objective validity, and therefore can be provided as the basic data for anti-allergic and anti-inflammatory studies.

The Effects of Triamcinolone Acetonide by Iontiophoretic Transdermal Delivery on Inflammation Induced Rat (백서에서 유발된 염증에 대한 Triamcinolone Acetonide 이온도입 경피투과 효과)

  • This study investigated the effects of triamcinolone acetonide by iontophoretic transdermal drug delivery on anti-inflammatory action into the rats and which had carrageenan-induced hyperalgesia and edema in the feet, trauma-induced tissue damage in the thigh. Each group was treated under the fellowing conditions. 1. Group I : Control group 2. Group II : Application of direct current 3. Group III : Application of 0.1$\%$ triamcinolone acetonide solution 4. Group IV : Iontophoresis of 0.1$\%$ triamcinolone acetonide solution The degree of anti-inflammation was evaluated by the paw withdrawal latency, the change in volume of foot the change of paw edema, histological change in rats. 1. In paw withdrawal latency, group IV showed the most significant therapeutic effect than the other groups at 0, 3, 6 and 9 hours(p < 0.001). 2. In paw edema experiment in the foot, group IV showed the most significant effect than group I at 0, 3, 6 and 9 hours. It meant that there was effective anti-inflammatory reaction in group I (p < 0.001). 3. In the light microscopic observation, group IV showed the most significant reduction of haemorrhage, hyperemia and infiltrative inflammation. From the results, the iontophoresis with triamcinolone acetonide is more effective than using each groups. It is one of the effective physical agent which delivered large molecular weight drug into the body. The continuous study is needed for many interesting issues of iontophoretic transdermal drug delivery in new future.

Morphological Development and Histology of Multiple Shoots and Microbulbs of Garlic Cultured in Bioreactors (생물반응기에서 배양된 마늘 신초와 Microbulb의 형태발달 및 조직관찰)

  • Multiple shoots of garlic (Allium sativum L.) were propagated in bioreactors containing MS medium supplemented with 2% sucrose for 3 weeks. Microbulbs were induced on MS medium supplemented with 0.1mg/L NAA and 11% sucrose for 9 weeks. For multiple shoot proliferation, leaves in the shoot must be removed before cultures. When the multiple shoots were cultured without removal of leaves, more than 90% of hyperhydricity and no microbulb formation were observed. Histological observation also indicated irregular size and shape of the cells in hyperhydricity of the shoot. Microbulbs were strarted to form from the shoot after 7 weeks of culture by protuberance of adventitious shoot buds followed by inner periclinal divisions and simultaneous anticlinical division in the epidermis of meristematic bulge. Analysis of ploidy level indicated no phenotypic variations in both multiple shoots and microbulbs induced from the mother plant, suggesting genetic homogeneity among the regenerants.

Effectiveness of freeze-dried bone grafts on the non-union fracture model of dogs (개의 비유합(非癒合) 골절(骨折) model에 있어서 동결건조골이식(凍結乾燥骨移植)의 효과(效果))

  • To investigate the effectiveness of the freeze-dried allografts and fibrin glue in bone grafts, the status of new bone formation and union of the grafted bone were observed in three types of grafting bones; autogenic bone(AT), allogenic bone(AL), and allogenic bone particles mixed with fibrin glue(FG). These were transplanted into non-union fracture model of 7 adult dogs with 2cm defect made in the proximal metaphysis of both fibulae. The autogenic and allogenic grafting bones had been treated by a modified freeze-dried method. The serial radiogram were observed the repair process of grafted bones biweekly until 17 or 21 weeks after transplantation and the observation of histological aspects, tetracycline double labeling and microradiography in the grafted bones were undertaken at 17 or 21 weeks after transplantation. The incorporation of bone minerals to the non-union fracture models were accomplished in 4 of 5 cases grafted with AL and in 2 of 4 cases grafted with AT. None of 5 cases grafted with FG were incorporated. The process of new bone formation and resorption in the grafted bones were observed three types; resorption of the grafted bones after newbone formation(type A) in 4 cases, new bone formation after resorption(type B) in 2 cases and complete or incomplete resorption without new bone formation(type C) in 8 cases. The modified freeze-dried method used in this study contributed to inhibite the rejection in allogenic grafts but the union period of the grafted freeze-dried bone was more prolonged than that of fresh autografts. Fibrin glue did not contribute to induce a new bone formation ofbone grafts.

Immunohistochemical Observation of Plasma Cell Granuloma in Intraoral Chronic Inflammatory Lesions (구강내 만성염증병소에서 보이는 형질세포육아종의 면역조직화학적 관찰)

  • Purpose: Chronic inflammatory gingival lesions occur as pyogenic granulomas or non-specific chronic suppurative lesions. Methods: Of the 59 chronic inflammatory gingival lesions examined, plasma cell granuloma (n=14), which showed an intense antibody-mediated immune reaction with the increased infiltration of plasma cells, was observed as a pseudotumor-like gingival overgrowth and myofibroblastic or fibrohistiocytitc proliferation of stromal cells with a heavy collection of plasma cells. The levels of CD3, CD20, CD31, CD68, RANKL, cathepsin G, cathepsin K, lysozyme, TNF${\alpha}$, MMP-2, and MMP-9 in the 14 cases of gingival plasma cell granuloma with immunohistochemical detection were measured to determine the pathogenetic progresses of the plasma cell granuloma compared to the common pyogenic granuloma (n=45) in the gingiva. Results: The gingival lesions of the plasma cell granuloma could be divided into three histological types, plasma cell predominant type (PPT, n=8), mixed inflammatory cell type (MICT, n=2), and sclerosed fibrosis type (SFT, n=4). The PPT showed a condensed infiltration of plasma cells into the perivascular spaces of the granulomatous lesion with frequent formation of Russel's body in their cytoplasm. The MICT showed the concomitant infiltration of many macrophages together with plasma cells, resulting in the diffuse destruction of stromal fibrous tissue. The SFT showed granulomatous lesions replaced gradually by thick collagenous fibrous tissue, resembling an inflammatory pseudotumor. The SFT expressed strongly the lymphocytic markers, CD3 and CD20, and the macrophage/monocyte markers, CD31 and CD68, but showed reduced expression of common inflammatory markers, TNF${\alpha}$, cathepsin G, lysozyme, MMP-2, and MMP-9, as well as the reduced expression of osteoclastogenic markers, RANKL and cathepsin K. Conclusion: These results suggest that a gingival plasma cell granuloma shows variable gene expression for cell-mediated immunity and stromal tissue degeneration, undergoing sclerotic fibrosis with a persistent inflammatory reaction.

  • A case of Behcet's syndrome found in a 22 years old male was is reported along with the histological and immunohistochemical studiesfindings. He The patient had had received prolonged medical therapy of anti-inflammatory agents for the conjunctival ulcer, genital ulcer, oral ulcer, and intestinal ulcers since the first onset of the Behcet's syndrome about approximately 7 years agoearlier. Recently, he felt the athickening of both corners of his lip causing that caused mouth opening difficultydifficulties. A plastic rReconstructive surgery was performed to enlarge the size of the oral orifice by multiple Z-plasty incisions, and finallywhich resulted in proper enlargement of the circumferential length of the lip. During the operation, a scar-like thick fibrous tissue was obtained and examined pathologically. In theThe microscopic observation the revealed the submucosal lesion was to be diffusely fibrosed with the a distribution of sclerotic collagen bundles. Particularly, sSeveral foci of collagen degeneration were found observed in the deep connective tissue, and the degenerating collagen bundles were gradually lost their fibrillar appearance. In the immunohistochemical observations, the foci of collagen degeneration was were strongly positive for IgK, but almost negative for TNF${\alpha}$ lysozyme, and MMP-3. Taken together, it was presumed that tThe submucosal fibrosis was presumed to have firstly presented in this study was probablybeeninduced by the prolonged anti-inflammatory therapy, which may inhibit the removal of sclerosed collagen bundles by the cell-mediated immunity and proteolytic digestion of macrophages, and that it was secondarily aggravated by the deposition of immunoglobulins derived from an autoimmune origin. Therefore, even after the successful plastic surgery of the lip to ameliorate the mouth opening difficulty, the recurrent submucosal fibrosis of lip should be carefully managed in the follow-up treatment.

Seed Production of Red Sea-Bream, Chrysophrys major (참돔(Chrysophrys major)종묘생산에 관한 연구)

  • A trial on the seed production of red sea-bream (Chrysophrys major) was made in Jejudo island at the southern end of Korea, from June, 1980 to August, 1981. Histological observation of the gonads indicated that the main spawning season was June to early July in Jejudo area. The newly-hatched fry were fed Brachionus sp. in the later stages Artemia nauplii, Tigriopus sp. and the minced fish flesh were given. Though during the winter season the water temperature in rearing tanks was decreased as low as $9.2^{\circ}C$, continuous feeding to the juveniles was carried out, showing body weight increase. This implies that winter rearing of juveniles in the near-shore facilities is feasible in the vicinity of Seogwipo area, in Jejudo. Feed coefficient of the juveniles (>23.2 g) was $4.16\sim4.77$ when fed the minced fish flesh.

Suppressive Effect of GJB-7 on Collagen-Induced Arthritis in Mice (관절7호방이 Type II collagen으로 유발된 흰쥐의 관절염에 미치는 영향)

  • Objectives: This study was carried out to know the effects of Gwan-Jul-Bang-7 (hereafter referred to GJB-7) on the inhibition of arthritis induced by collagen on the mouse. Methods: To assess the effects of GJB-7 on mouse with arthritis induced by collagen II, we conducted several experiments such as analysis of cytotoxicity, hepatotoxicity, arthritis index, total cell number of draining lymph nodes and paw joints, value of immunocyte in PBMC (peripheral blood mononuclear cell), DLN (draining lymph node) and paw joint, measurement of cytokine and anti-collagen II, observation of the histological changes of joint. Results: 1. Cytotoxicity against HFC (human fibroblast cells) was not observed in any concentration and hepatotoxicity was not observed in the GJB-7 treated group. 2. The incidence of arthritis significantly decreased. 3. Total cell number of draining lymph nodes significantly increased and total cell number of paw joints significantly decreased. 4. The percentage of $CD8^+$ cells in PBMC (peripheral blood mononuclear cell) significantly increased. The percentage of $CD3^+/CD69^+$, and $CD3^+/CD49b^+$ cells in PBMC significantly decreased. 5. The percentage of $CD19^+,\;CD3^+$, and $CD4^+/CD25^+$ cells in DLN (draining lymph nodes) significantly increased. The percentage of $B220^+/CD23^+$ cells in DLN significantly decreased. 6. The percentage of $CD3^+,\;CD4^+$, and $CD11b^+/Gr-1^+$ cells in paw joints significantly decreased. 7. The production of TNF-$\alpha$, IL-6, and MCP-1 significantly decreased. 8. Anti-collagen II in serum significantly decreased. 9. With the hematoxylin and eosin stain, inflammation of joint decreased. Under Masson's trichrome stain, the cartilage destruction and synovial cell proliferation and the expression of collagen fibers decreased. Conclusions: Comparison of the results for this study showed that GJB-7 had immunomodulatory effects. So we expect that GJB-7 could be used as an effective drug for not only rheumatoid arthritis but also another auto-immune diseases.
