Substitution of Heavy Complementarity Determining Region 3 (CDR-H3) Residues Can Synergistically Enhance Functional Activity of Antibody and Its Binding Affinity to HER2 Antigen

  • To generate a biobetter that has improved therapeutic activity, we constructed scFv libraries via random mutagenesis of several residues of CDR-H3 and -L3 of hu4D5. The scFv clones were isolated from the phage display libraries by stringent panning, and their antiproliferative activity against HER2-positive cancer cells was evaluated as a primary selection criterion. Consequently, we selected AH06 as a biobetter antibody that had a 7.2-fold increase in anti-proliferative activity ($IC_{50}$: 0.81 nM) against the gastric cancer cell line NCI-N87 and a 7.4-fold increase in binding affinity ($K_D$: 60 pM) to HER2 compared to hu4D5. The binding energy calculation and molecular modeling suggest that the substitution of residues of CDR-H3 to W98, F100c, A101 and L102 could stabilize binding of the antibody to HER2 and there could be direct hydrophobic interactions between the aromatic ring of W98 and the aliphatic group of I613 within HER2 domain IV as well as the heavy and light chain hydrophobic interactions by residues F100c, A101 and L102 of CDR-H3. Therefore, we speculate that two such interactions were exerted by the residues W98 and F100c. A101 and L102 may have a synergistic effect on the increase in the binding affinity to HER2. AH06 specifically binds to domain IV of HER2, and it decreased the phosphorylation level of HER2 and AKT. Above all, it highly increased the overall level of p27 compared to hu4D5 in the gastric cancer cell line NCIN82, suggesting that AH06 could potentially be a more efficient therapeutic agent than hu4D5.

Factors Influencing the Continuity of Volunteer Activities for Disaster Relief (재해구호 자원봉사활동의 지속성에 영향을 미치는 요인)

  • The purposes of this study were to find out the factors affecting the continuous participation of volunteer activities for disaster relief and to provide implications for the efficient management of sustainable disaster relief volunteer activities. In order to achieve these purposes, we investigated how much individual characteristics, the volunteer attitude and motivation, and institutional characteristics, respectively, affect the duration of disaster relief volunteer activities. The research was carried out targeting 261 disaster relief volunteer workers, aged over 20, who affiliated to various volunteer centers. Data were collected by survey methodology from May 7th to May 27th, 2008. A frequency analysis and a multiple regression were utilized for analyzing data. The results of this study were as follows: the continuity of volunteer activities of the subjects are likely to be high when they are older, have a job, are highly motivated, are assigned properly to the placement and task. Based on the results of this study, some practical implications were suggested. First, since placement and task assignment managed by volunteer organizations are very important for the continuity of volunteer activities, those organizations should have developed diverse volunteer activity programs considering volunteers' characteristics and interest. Second, the managers of those organizations need to sufficiently consider the characteristics of volunteers such as age and job when dispatching volunteers in disaster areas. The managers should help their volunteers to immerse in their volunteer work. Finally, it is necessary to induce social concern and support in disaster relief volunteer activities in order to make volunteers especially with jobs be constantly active in disaster relief activities.

Improved Organic Removal Efficiency in Two-phase Anaerobic Reactor with Submerged Microfiltration System (침지형 정밀여과시스템을 결합한 이상 혐기성 시스템에 의한 유기물 제거율의 향상)

  • A two-phase anaerobic reactor with a submerged microfiltration system was tested for its ability to produce methane energy from organic wastewater. A membrane separation system with periodic backwashing with compressed air was submerged in the acidogenic reactor. The cartridge type of microfiltration (MF) membrane with pore size of $0.5{\mu}m$ (mixed esters of cellulose) was tested. An AUBF (Anaerobic Upflow Sludge Bed Filter: 1/2 packed with plastic media) was used for the methanogenic reactor. Soluble starch was used as a substrate. The COD removal was investigated for various organic loading with synthetic wastewater of 5,000 mg starch/L. When the hydraulic retention time (HRT) of the acidogenic reactor was changed from 10 to 4.5 days, the organic loading rate (OLR) varied from 0.5 to $1.0kg\;COD/m^3-day$. When the HRT of the methanogenic reactor was changed from 2.8 to 0.5 days, the OLR varied from 0.8 to $5.8kg\;COD/m^3-day$. The acid conversion rate of the acidogenic reactor was over 80% in the 4~5 days of HRT. The overall COD removal efficiency of the methanogenic reactor showed over 95% (effluent COD was below 300 mg/L) under the highly fluctuating organic loading condition. A two-phase anaerobic reactor showed an excellent acid conversion rate from organic wastewater due to the higher biomass concentration than the conventional system. A methanogenic reactor combined with sludge bed and filter, showed an efficient COD and SS removal.

Biological Dechlorination of Chlorinated Ethylenes by Using Bioelectrochemical System (생물전기화학시스템을 이용한 염화에틸렌의 생물학적 탈염소화)

  • Chlorinated ethylenes such as perchloroethylene (PCE) and trichloroethylene (TCE) are widely used as industrial solvents and degreasing agents. Because of improper handling, these highly toxic chlorinated ethylenes have been often detected from contaminated soils and groundwater. Biological PCE dechlorination activities were tested in bacterial cultures inoculated with 10 different environmental samples from sediments, sludges, soils, and groundwater. Of these, the sediment using culture (SE 2) was selected and used for establishing an efficient PCE dechlorinating enrichment culture since it showed the highest activity of dechlorination. The cathode chamber of bioelectrochemical system (BES) was inoculated with the enrichment culture and the system with a cathode polarized at -500 mV (Vs Ag/AgCl) was operated under fed-batch mode. PCE was dechlorinated to ethylene via TCE, cis-dichloroethylene, and vinyl chloride. Microbial community analysis with polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) showed that the microbial community in the enrichment culture was significantly changed during the bio-electrochemical PCE dechlorination in the BES. The communities of suspended-growth bacteria and attached-growth bacteria on the cathode surface are also quite different from each other, indicating that there were some differences in their mechanisms receiving electrons from electrode for PCE dechlorination. Further detailed research to investigate electron transfer mechanism would make the bioelctrochemical dechlorination technique greatly useful for bioremediation of soil and groundwater contaminated with chlorinated ethylenes.

Discipline Bias of Document Citation Impact Indicators: Analyzing Articles in Korean Citation Index (논문 인용 영향력 측정 지수의 편향성에 대한 연구: KCI 수록 논문을 대상으로)

  • The impact of a journal is commonly used as the impact of an individual paper within that journal. It is problematic to interpret a journal's impact as a single paper's impact of the journal, so there are several researches to measure a single paper's impact with its own citation counts. This study applied 8 impact indicators to Korean Citation Index database and examined discipline bias of each indicator. Analyzed indicators are simple citation counts, PageRank, f-value, CCI, c-index, single publication h-index, single publication hs-index, and cl-index. PageRank has the least discipline bias at highly ranked papers and journal bias in a discipline. On the contrary, simple citation counts showed strongly biased results toward a certain discipline or a journal. KCI database provides only simple citation counts. It needs to show PageRank (global indicator) to discover influential papers in diverse areas. Furthermore it needs to consider to provide the best of local indicators. Local indicators can be calculated only with papers in users' search results because they uses citation counts of citing papers and the number of references. They are more efficient than global indicators which explore the whole database. KCI should also consider to provide Cl-index (local indicator).

Dental IQ and Oral Health Care Status of Elementary School Students (초등학생의 Dental IQ 수준과 구강보건 관리실태)

  • Objectives: Oral health care program for the elementary school children (ESOHCP) should be met the first priority because dental caries are highly sensitive and prevalent among them. This study was performed to enhance the efficiency of ESOHCP, and their oral health promotion. Methods: The Subjects were 346 students (3rd grade 163, 6th grade 185) of one elementary school in Busan. Dental IQ and actual oral health status; the decayed, mixing and filling teeth were checked through questionnaire and oral health examination from April to June 2002. Dental IQ was presented out of one hundred, and data analysis was done using SAS (ver 8.1) program. Results: The students' cognitive level about the causative and preventive factor of dental caries was relatively high. However, the practical aspects of preventive behavior - tooth brushing; three times per day (20%), three minutes per time (22%), oral health examination; one time per three months (10%) left much to be desired. And, only 27% of the subjects had experienced in school oral health education. The mean level of dental IQ was 79 out of 100, and 51 % and 42% of them had the decayed and teeth with filling, respectively, with the rate of DMFT 82%, DT 43%, FT 57%. The level of dental IQ was higher in case of having his (her) own tooth brush (p=0.072), standard tooth brushing (p<0.001) three times per day, post meal, three minutes per time, present experience of oral health examination (p<0.001) and dental clinic visit (p<0.001). The grade of caries was more serious in case of 6th grade (p=0.059), an absence of his (her) own tooth brush (p= 0.090), present experience of oral health examination (p=0.021), and an absence of regular dental clinic visit (p=0.003). The frequency of oral health examination (γ= 0.620), tooth brushing; times per day (γ=0.445), post meal (γ=0.355), expending times per brushing (γ=0.352), right cognition to the treatment of caries (γ=0.401), positive attitude to dental treatment (γ=0.387), the frequency of dental clinic visit for the past one year (γ=0.152) showed significant correlation with dental IQ. In the multivariate analysis, dental IQ was influenced by the frequency of oral health examination, right cognition to the cause of caries, times of teeth brushing per day, right cognition to the treatment of caries, etc., with adjusted R2=0.857. Conclusion: Though the students' cognitive level about the causative and preventive factor of dental caries was high, the practical aspects of preventive behavior left much to be desired. Ant the current ESOHCP considered to be still inefficient. However, frequent dental visits had apparent correlation with hish level of dental IQ. The specialized and practical program should be needed for the efficient ESOHCP. Harmonized effort from educational, health and dental society is essential.

Early Childhood Teachers' Recognition and Actual State on Cooperative Art Activities (협동미술활동에 대한 유아교사의 인식 및 실태)

  • The purpose of this study is to investigate the perception and actual condition of early childhood teachers about the activities of cooperative art in early childhood education institutions. The subjects of the study were 280 teachers of early childhood education institutions in Y city. The research method was a questionnaire survey on the perception and actual condition of the cooperative art activities. For the research problems, frequency, percentage, and average were calculated by using descriptive statistics, and t - test, F - test, and ${\chi}^2$ - test were conducted to examine the differences according to background variables of early childhood teachers. In conclusion, this study showed that the need for cooperative art activities of early childhood education institutions was highly perceived, and there were significant differences according to the background variables of teachers in educational goals, contents and method. There were significant differences according to the teachers' background variables in the actual cooperative art activities in terms of frequency, method, time, evaluation method, teacher training and support. Based on the results of this study, various supporting methods such as activities plan and method, teacher education and training appropriate for the cooperative art activity program and the presentation of young children are sought so that more desirable and efficient cooperative art activities can be operated in the field of early childhood education suggesting that it should be done.

A Design Study of Signal Processor for Small Tracking Radar (소형 추적 레이더를 위한 신호처리기 설계 기술 연구)

  • Recently, the tracking radar has confirmed the necessity of developing a small tracking radar that can be operated without various restrictions in various environments. In addition, the performance of a small tracking radar requires equal to or higher than the existing tracking radar. Such a small tracking radar can be implemented through miniaturization and low power of existing tracking radar. In this paper, the role and function of a signal processor for a small tracking radar are defined and we proposed a method to increase the efficiency of power consumption and miniaturization by minimizing the use of devices required to implement a signal processor for a small tracking radar. Used as a method for miniaturization, a device processor such as DDC and communication controller was implemented in an FPGA to design a signal processor for a small tracking radar. In addition, a low-power signal processor was designed by a power supply using a highly efficient switching regulator. Finally, the signal processor was verified by the performance test of the signal processor for the small tracking radar implemented, the Doppler tracking test using the signal processor on the small tracking radar, and the distance tracking test.

Study on High Degree of Efficiency Chemical Reactor for Air Purification Using the Glow Discharge (글로우 방전을 이용한 고효율 공기 정화용 화학 반응기의 특성관찰에 관한 연구)

  • the basic model of chemical reactor using glow discharge, we used cathode discharge cell with vacant cavity in the middle. Currently glow discharge is widely studied as a radiation source or atomization device in atomic spectroscopy and remarkable technological achievements are made through the graft with other analysis devices such as microanalysis and steel analysis.1 Additionally, as the characteristics of basic glow discharge and radiation have been reviewed many times, those results could be used in this experiment.2-3 In 1993, an article regarding the treatment of poisonous gas in the air using low temperature plasma was published. According to this article, if DC Glow Discharge is used under continuous atmospheric flow, poisonous gases such as SO2 and NO can be removed.4 Based on those findings, we designed highly efficient reactor where stable air plasma is composed and all air flow pass the negative glow area passing through the tube. It was observed that the cathode tube type glow discharge developed in this study would be economical, easy to use and could be used as radiation source as well.

Human Safety Assessment for a 4 × 8 Array Antenna Used for Wireless Power Transfer at 2.4 GHz (2.4 GHz의 무선전력전송에 사용되는 4 × 8 배열 안테나에 대한 인체안전성평가)

  • Wireless Power Transfer(WPT) of array antenna applied to beam-forming techniques enables highly efficient WPT when transmitters and receivers are not contacting and even when they are separated. However, this WPT method is possible to use only when human safety restriction by distance between the transmitters and the receivers is satisfied. In the paper, a $4{\times}8$ array antenna for 2.4 GHz is modeled by simulation, then electric field intensity and 10 gram average head SAR(Specific Absorption Rate) by distance away from the array antenna inputted 1 W of 2.4 GHz sinusoidal wave at each single antenna of the array antenna for 2.4 GHz were obtained. And they were compared with human safety restriction of draft of 2018 ICNIRP(International Commission on Non-Ionizing Radiation Protection) guidelines. As the result, power density of far field derived from the electric field intensity was $33.257W/m^2$, which satisfied with occupational human safety restriction but exceeded public's. In addition, the 10 gram average head SAR exceeded the human safety restriction.
