Effect of Beverage from Inonotus Obliquus on Serum Lipid Profile Improvement (체내 지질 농도 개선에 미치는 차가버섯 음료의 효과)

  • The purpose of this study was to investigate the possibility of Inonotus obiquus as an functional resources. We carried out to develop a beverage by using Inonotus obliquus, and we examined the effects of beverage on the reduction of serum lipid and improvement of blood parameters in rats fed high fat diet 5 weeks. Sprague-Dawley rat weigh $150\;g{\pm}15\;g$, were randomly assigned to 4 groups, basal diet only(BDG), high fat diet without beverage(FDCG), high fat diet and 10% beverage(FD10M), high fat diet and 20% beverage(FD20M). The result of this study were as follow. Hematological datas of 4 groups were same level, which were not significant. Serum GOT activity were reasonable levels in FD10M and FD20M groups compared to FDCG group. Total cholesterol, LDL-cholesterol, triglyceride in serum and atherogenic index were remarkably reduced in beverage supplemented groups as compared control group. These results imply that beverage from Inonotus obliquus could be used as possible for decrease of serum lipid concentration.

The Antiobese Effects of Gamikwakhyangjungkisan and Fermented GamiKwakhyangjungkisan in Preadipocytes and Mice Fed High Fat Diet (지방전구세포와 고지방식이비만마우스에서 가미곽향정기산의 전탕액과 발효액의 항비만효과)

  • Objectives This experimental study was designed to investigate the antiobese effects of Gamikwakhyangjungkisan and Fermented GamiKwakhyangjungkisan. Methods The cellular lipid contents were assessed by Oil-Red-O staining. The expression of $PPAR{\gamma}$ and $C/EBP{\alpha}$ were determined by real time RT-PCR and western blotting. In addition, body weight gain and serum lipid levels were measured in the mice with obesity induced by the high fat-diet for four weeks. Results Gamikwakhyangjungkisan and Fermented GamiKwakhyangjungkisan is reduced 3T3-L1 cells' differentiation and the expressions of $PPAR{\gamma}$ and $C/EBP{\alpha}$ in high concentration group. High-fat diet + Fermented GamiKwakhyangjungkisan group significantly reduced body weight gain. High-fat diet + Fermented GamiKwakhyangjungkisan group significantly increased HDL-cholesterol contents and reduced LDL-cholesterol contents. Furthermore, Fermented GamiKwakhyangjungkisan is excellent antiobese effects than Gamikwakhyangjungkisan. Conclusions These results demonstrate that Gamikwakhyangjungkisan and Fermented GamiKwakhyangjungkisan exerts antiobese effect in 3T3-L1 cells and mice fed high fat diet. Furthermore, Fermented GamiKwakhyangjungkisan is excellent antiobese effects than Gamikwakhyangjungkisan.

  • The study was performed by examining the effects of fermented liquid of Crataegi Fructus on the lipid profile improvement in rats fed high fat diets. Sprague-Dawley rats of weighting $180.0{\pm}30g$ were randomly divided into five groups : basal diet (Normal control group, NCG), only high fat diet (High fat control group, HFC), high fat diet and supplemented with 1.69 mg/100 g body weight, 3.38 mg/100 g body weight, 6.76 mg/100 g body weight by fermented liquid of Crataegi Fructus - HFL, HFM, HFH group). These experimental diets were fed for 6 weeks. The fermented liquid of Crataegi Fructus fed groups had more significantly decreased in the levels of serum total cholesterol, triglyceride, LDL-cholesterol and atherogenic index than the high fat control group, while the HDL-cholesterol was higher when compared to the normal control group. Total lipid, total cholesterol, triglyceride, LDL-cholesterol contents in liver were decreased in high fat experimental groups. But the degree of increment was reduced by administration of fermented liquid of Crataegi Fructus. while the fermented liquid of Crataegi Fructus fed group had ore significantly increased in the level of HDL-cholesterol than the high fat control group. The singularity of the unsaturated fatty acid contents attracted our attention. Especially, the polyunsaturated fatty acid compositions were 36.36%, 34.70%, 20.31%in serum, liver and fecal of fermented liquid of Crataegi Fructus fed groups, respectively. These results imply that the fermented liquid of Crataegi Fructus can be used as possible food resources and medicinal food materials.

Effect of Dietary Fibers on Retinol Binding Protein (RBP, cRBP I, cRBP II) Gene Expression in Rats Fed High Fat Diet (고지방 식이내 식이섬유질이 흰쥐의 레티놀 결합 단백질 (RBP, cRBP I, cRBP II) 유전자 발현에 미치는 영향)

  • Recently it has been reported that vitamin A and retinol binding proteins (RBPs) in blood and urine were changed in the condition of diabetes mellitus or hyperlipidemia. Fruits and vegetables are recommended to consume for the people suffered from these chronic degenerative diseases. The main components of fruits and vegetables are dietary fibers, for example cellulose and pectin, of which function to affect the absorption and excretion of dietary fat and fat-soluble substances. This study was conducted to investigate the effect of dietary fibers on RBPs mRNA expression in liver, small intestine and serum of rat fed high fat diet during 4 weeks. Sprague-Dawley rats, weighing 121g on average, were divided into four groups; (Control; $17\%$ fat & cellulose supplement diet, HF0: $25\%$ fat & fiber free diet, B:.Uc: $25\%$ fat & cellulose supplement diet and HF0: $25\%$ fat & pectin supplement diet) . The rats fed high fat diet groups (HF0, HFC, HFP) tended to consume the food less than the control group, but FER of HF0 groups was significantly higher than the control (p < 0.05) . The weight of adrenal gland in high fat diet groups (HF0, HFC, HFP) was significantly less than the control. Total lipid in feces daily excreted and in liver did not show any significant differences among the groups. Total cholesterol in HFP group was significantly different from that of HFC group. Serum total cholesterol and triglyceride in other group tended to lower than other groups and HDL cholesterol higher. Consequently, AI (atherogenic index) was the lowest in HFP group. Vit A contents in feces daily excreted tended to lower in high fat diet groups (HF0, HFP) compared to the control group. That content in adrenal gland was the lowest in HF0 group, but not in liver. In HFP group were down-regulated cRBPI mRNA in liver and cRBPII mRNA in small intestine and up-regulated RBP and transthyretin expression in serum compared to the other groups. In conclusion, dietary fibers, especially pectin, in high fat diet might down-regulate the expression of CRBP I, CRBP II mRNA in liver and small intestine, but increase the secretion of RBP into serum and therefore inhance the bioavailability of Vit A through the body.   • 1.Purpose This study was researched in order to investigate the effect of Hyungbangdojucksan-Gami and Kamsuchunilhwan on the obesity in rats. 2. Methods The rats were divided into 10 groups - The normal group fed with normal diet. One of the sample groups fed with high fat diet, and since fifth week had fed with Hyungbangdojucksan-Gami. The other sample groups fed with high fat diet, and since fifth week had fed with Kamsuchunilhwan. The control group fed with high fat diet, and since fifth week had fed sodium chloride. 3. Results 1. The rate of increasing weight was significantly decreased after oral administration of Hyungbangdojucksan-Gami and Kamsuchunilhwan. 2. The rate of feeding effect was not significantly decreased after oral administration of Hyungbangdojucksan-Gami and Kamsuchunilhwan. 3. After oral administration of Hyungbangdojucksan-Gami, the quantities of total cholesterol, HDL cholesterol, LDL cholesterol and triglyceride on the serum were significantly decreased but that of lipid peroxide on the serum was significantly increased. 4. After oral administration of Kamsuchunilhwan., the quantities of glucose, total cholesterol, LDL cholesterol and triglyceride on the serum were significantly decreased but those of HDL cholesterol, lipid peroxide on the serum were significantly increased. 4.Conclusion Hyungbangdojucksan-Gami and Kamsuchunilhwan have significant effects on the obesity in the rats.

Body-fat Suppressive Effects of Capsaicin through $\beta$-adrenergic Stimulation in Rats Fed a High-fat Diet (고지방식이를 섭취시킨 흰쥐에서 $\beta$-adrenergic 활성의 증가에 의한 Capsaicin의 체지방 감소효과)

  • The effects of capsaicin, a pungent principle of hot red pepper, on body fat gain, balance serum lipid values were investigated in rats fed a high-fat(30%) diet. Administration of capsaicin by dietary administration caused a complete cessation of increased in body weight and fat gain induced by the high-fat diet. However, energy intake and body protein gain were not affected by capsaicin. Therefore, the suppression of body fat gain by capsaicn was believed due to an increased in energy expenditure. Simultaneous administration of capsaicin and a $\beta$-adrenergic blocker, propranolo, resulted in the inhibition of changes in body fat gain by capsaicin without remained unchanged, indicating an increase in the number of mitochondria in brown adipose tissue. Therefore, it appears that capsaicin possesses potent body fat suppressive effects mediated by $\beta$-adrenergic stimulation in which brown adipose tissue may be involved. On the other hand, capsaicin had no effects on serum triglyceride, total cholesterol or HDL-cholesterol levels. These results are in contrast to those reported by other investigators. Perhaps expression of the effects of capsaicin on plasma lipids is a rather complicated process, dependent on the type of diet administered, fat content of the diet, period and route of capsaicin administration, and species and strain of animals used.

Effects of herbal acupuncture (Atratylodes japonica Koizumi and Coix lachrymajobi var. mayeun Stapf) at Pungnyung(ST40) and Umnungchon(SP9) in obese Rats induced by high fat diet (풍륭(風隆)·음릉천(陰陵泉)에 대한 약침(藥鍼)(창출(蒼朮), 의이인(薏苡仁))이 고지방식이(高脂肪食餌)로 유발(誘發)된 비만백서(肥滿白鼠)에 미치는 영향(影響))

  • Objectives : This research was performed to investigate the effect of herbal acupuncture (Atratylodes japonica Koizumi and Coix lachrymajobi var. mayeun Stapf) at Pungnyung(ST40) and Umnungchon(SP9) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL to total cholesterol ratio of rats fed high fat diet for 5weeks. Methods : Experimental groups were divided into normal group(Sham), high fat diet group(Control), high fat diet and Atractylodes japonica-herbal acupuncture in ST40 SP9 treated group (AJ), high fat diet and Coix lachrymajobi var. mayeun Stapf-herbal acupuncture in ST40 SP9 treated group(CL), high fat diet and Atractylodes japonica+Coix lachrymajobi var. mayeun Stapf-herbal acupuncture in ST40 SP9 treated group(AJ+CL). Herbal acupuncture was bilaterally treated at the level of 132.5mg/kg body weight per day. Results : Body weight and food efficiency were decreased in AJ, AJ+CL. Feed intake was decreased in AJ, CL. The level of serum HDL-cholesterol was increased in AJ, CL. The level of serum total cholesterol, LDL-cholesterol and phospholipid were not significant. The level of serum triglyceride, free fatty acid were increased in AJ. The level of serum AST was decreased in AJ, AJ+CL. The level of serum ALT was decreased in AJ, CL, AJ+CL. The level of serum ALP was decreased in AJ+CL. The HDL to Total cholesterol ratio was increased in AJ. Conclusions : Atractylodes japonica-herbal acupuncture in ST40 SP9 can control the body weight, feed intake and feed efficiency ratio, HDL-cholesterol and herbal acupuncture combined Atractylodes japonica with Coix lachrymajobi var. mayeun Stapf is effective on Body weight, feed intake, feed efficiency ratio, protection of liver function and prevention cardiovascular risk by obesity.

Gene Expression Profile in the Liver Tissue of High Fat Diet-Induced Obese Mice

  • The purpose of this study was to investigate the gene profiles that were up- or down-regulated in the livers of high-fat diet-induced obese mice and $db_-/db_-$ mice with deficient leptin receptor. C57/BL6 normal mice and $db_-/db_-$ mice, respectively, were divided into two groups and fed a standard or high-fat diet for four weeks. Liver weight was unchanged in the normal mice but the high-fat diet led to a 10% weight increase in the $db_-/db_-$mice. Adipose tissue mass increased by about 88% in the normal mice that were fed a high-fat diet and by about 17% in the $db_-/db_-$mice on the high-fat diet. In terms of serum lipids, total cholesterol significantly increased in mice on the high-fat diet. Microarray analysis was carried out using total RNA isolated from the livers of standard or high-fat diet-fed mice of the normal and $db_-/db_-$ strains. The change of gene expression was confirmed by RT-PCR. About 1.6% and 6.8% of total genes, respectively, showed different expression patterns in the normal mice fed the high-fat diet and $db_-/db_-$ mice. As a result of microarray, many genes involved in metabolism and signal pathways were shown to have different expression patterns. Expression of Mgst3 gene increased in the livers of normal and $db_-/db_-$ mice that were fed a high-fat diet. Wnt7b and Ptk9l were down-regulated in the livers of the normal mice and $db_-/db_-$ mice that were fed a high-fat diet. In conclusion, a high-fat diet induced obesity and affected gene expression involved in metabolism and signal pathway.

Effects of Bambusae Caulis in liquamen on inhibition of angiotensin-converting enzyme in vitro and blood flow in mouse model (죽력의 in vitro에서의 angiotensin-converting enzyme 저해효과와 마우스모델에서 혈행개선효과)

  • The purpose of this study was to investigate the effects of the Bambusae Caulis in liquamen (BCL) on blood circulation in animal models. Previous studies on BCL have shown effects on thrombolytic activity and angiotensin-converting enzyme (ACE) inhibitory activity. In the mouse model, the triglyceride content were 301.5 mg/dL in the high fat diet+BCL II 0.01% group, 289.2 mg/dL in the high fat diet+BCL II 0.05% group, which was significantly lower than the high fat diet group. The total cholesterol content was 311.9 mg/dL in high fat diet+BCL II 0.01% and 293.7 mg/dL in high fat diet+BCL II 0.01% 0.05%, respectively, which was significantly lower than the high fat diet group. The HDL-cholesterol level was 206.0 mg/dL for the high fat diet, 196.6 mg/dL for the high fat diet+BCL II, and 189.2 mg/dL for the high fat diet+BCL II. There was no significant difference between the 0.01% and 0.05% groups. The high-fat diet+0.05% group was significantly improved in the blood flow compare to the high fat diet and the high fat diet+0.01% group. Platelet aggregation inhibition ability was inhibited in the high fat diet+0.01% and 0.05% groups compared to the high fat diet group.

Red beet(Beta vulgaris L.) leaf supplementation improves antioxidant status in C57BL/6J mice fed high fat high cholesterol diet

  • The effect of diet supplemented with red beet (Beta vulgaris L.) leaf on antioxidant status of plasma and tissue was investigated in C57BL/6J mice. The mice were randomly divided into two groups after one-week acclimation, and fed a high fat (20%) and high cholesterol (1%) diet without (control group) or with 8% freeze-dried red beet leaf (RBL group) for 4 weeks. In RBL mice, lipid peroxidation determined as 2-thiobarbituric acid-reactive substances (TBARS value) was significantly reduced in the plasma and selected organs (liver, heart, and kidney). Levels of antioxidants (glutathione and $\beta$-carotene) and the activities of antioxidant enzyme (glutathione peroxidase) in plasma and liver were considerably increased, suggesting that antioxidant defenses were improved by RBL diet. Comet parameters such as tail DNA (%), tail extent moment, olive tail moment and tail length were significantly reduced by 25.1%, 49.4%, 35.4%, and 23.7%, respectively, in plasma lymphocyte DNA of RBL mice compared with control mice, and indicated the increased resistance of lymphocyte DNA to oxidative damage. In addition, the RBL diet controlled body weight together with a significant reduction of fat pad (retroperitoneal, epididymal, inguinal fat, and total fat). Therefore, the present study suggested that the supplementation of 8% red beet leaf in high fat high cholesterol diet could prevent lipid peroxidation and improve antioxidant defense system in the plasma and tissue of C57BL/6J mice.
