The Effects of Fig Fermented Product Supplementation on Animal Performance, Serum Profile and Meat Quality in Hanwoo Bulls (무화과 발효물의 급여가 한우의 생산성, 혈액성상 및 육질에 미치는 영향)

  • We investigated the effect of fig fermented product(FFP) supplementation on growth performance, serum profile, carcass performance, meat performance and meat quality in 10 bulls of Korean cattle. Concentrates diet was supplemented with substrate fermented from fig fruit and leaves at 10% of the diet. The feed intake of FFP were slightly higher than the control, but the final weight showed no sifnificant difference between the two. Daily weight gain and feed intake were increased in FFP. The serum profile had no significant difference in the treatment. In carcass performance, the meat quantity grade of the treatment had no significant difference, but in meat quality grade the marbling score of FFP was significantly(P<0.05) increased therefore it showed a positive effect on meat quality grade. Also there was no significant(P<0.05) difference of meat cut performance in the treatment. Due to the proximate characteristics of longissimuss muscles the crude fat content of the FFP was significantly(P<0.05) increased. There was no significant difference(P<0.05) in physical characteristics ; pH level, meat color and heat loss of the treatment, but the shear force value and the cholesterol content of FFP significantly(P<0.05) decreased. Crude fat was increased(P<0.05) and cooking loss, shear force and cholesterol concentration were decreased. In fatty acids composition of the FFP, the linoleic acid from the longissimus increased significantly(P<0.05). In subcutaneous fat of longissimus of the FFP, C16:0(palmitic acid) significantly(P<0.05) decreased, but C18:1 significantly (P<0.05) increased. Therefore in FFP, the concentration of saturated fatty acid significantly decreased (P<0.05), but on the other hand the concentration of unsaturated fatty acids significantly (P<0.05) increased. In sensory evaluation of the FFP, the evaluation of odor increased slightly in a positive manner, also the appearance and the taste increased significantly(P<0.05). In conclusion when annexing additional fig fermented product to Hanwoo bulls, the carcass grade improves and the livestock production increases. Also the shear force, lower cholesterol, improved appearance and taste will open the doors to high quality meat production.

The Effect of Dietary Modified Potato Starch By Chemically Denatured Treatment and Potato Starch on the Weight Loss, Lipid Metabolism and Redox Antioxidant System in High Fat Diet-Induced Obese Rats (화학적 변성 및 생감자 전분이 고지방식이로부터 유도된 비만 흰쥐의 지질대사 및 항산화계에 미치는 영향)

  • For the first 42 days, we made rats obese by feeding them potato starch instead of corn starch and after that we fed them transformed potato starch by 4 groups for 70 days. The 4 groups are GPS group, SPS group, EZ group and H40 groups and each were fed normal potato, small potato, enzyme treated potato, and acid treated potato starches, respectively. We determined body weight and feeding efficiency, lipid profiles in serum, lipid peroxidation in tissues and redox antioxidant system as GSH and GP-x in vivo. As a result, there was no difference in the increment of body weight in the groups of GPS, EZ and H40. Therefore EZ group showed lower body weight increment than other groups. While GPS group and SPS group did not show significant difference in blood glucose, cholesterol level, LDL-cholesterol and TC, and their measured values were lower than those of EZ and H40 groups. No significant difference was found in HDL-cholesterol level except for GPS group. Furthermore, when calculating atherogenic index (AI) by HDL-cholesterol and TC contents, H40 group showed higher measured value than other groups. When measuring the lipid peroxidation in serum, kidney and liver tissues, the serum lipid peroxidation in H40 group was higher than others. In the tissue of liver and kidney, EZ and H40 groups showed significantly lower contents than others. The content of GSH showed different tendency in each tissue, but the measured value of GP-x activity was lower in SPS group.

Alcohol-induced Hyperlipidemia Is Ameliorated by Orally Administered DWP208, a Sodium Succinate Form of ZYM201

  • DWP208 is a sodium succinate form of ZYM-201 which is a triterpenoid glycoside isolated from Sanguisorba officinalis, a medicinal plant prescribed for various diseases, such as duodenal ulcers and bleeding in East Asian counties. We demonstrated that this compound is able to normalize the altered lipid metabolism induced by hyperglycemia and a high fat diet. In this study, we determined whether hyperlipidemic conditions induced with chronically treated alcohol can also be restored by DWP208. Similar to our previous results, orally administered DWP208 (1 to 10 mg/kg) also ameliorated the hyperlipidemia that was induced by alcohol. This compound reversed the alcohol-induced hyperlipidemia including (i) up-regulated hyperlipidemic parameters such as low-density lipoprotein (LDL), very low-density lipoprotein (VLDL), atherosclerotic index (AI), triglyceride, and total cholesterol, and (ii) down-regulated hyperlipidemic parameters such as absolute body weight, superoxide dismutase (SOD) activity, and high-density lipoprotein (HDL) in serum and liver. According to our data, the ameliorative activity of DWP208 is due to its indirect anti-oxidative activity as a result of which lipid peroxide and hydroxyl radical levels were reduced and the activity of SOD was enhanced. Therefore, our data strongly suggest that DWP208 can be used as a remedy against alcohol-induced hyperlipidemia.

Studies on the general analysis and antioxidant component analysis of Chenopodium album var. centrorubrum and biochemical analysis of blood of mice administered C. album (명아주의 일반성분, 항산화활성 분석 및 흰쥐의 혈중 생화학적 분석에 관한 연구)

  • The purpose of this study was to develop new food materials by analyzing nutritional components and antioxidant activities of Chenopodium album var. centrorubrum. The highest amount of mineral obtained in C. album was found to be $1.01{\pm}0.07mg$ per 100 g of iron. The total phenolic content in C. album was found to be 3.77~9.57 GAE (gallic acid equivalent) mg/g. The reducing activities of leaves and roots of C. album determined using FRAP (ferric-reducing antioxidant power) were higher in ethanol extracts than water extracts. The ABTS radical scavenging activities of leaves and roots of C. album were $204.29{\pm}4.98{\mu}mol/g$ and $106.96{\pm}2.81{\mu}mol/g$, respectively. The DPPH radical scavenging activity was high upon extraction using ethanol (roots $20.71{\pm}0.04{\mu}mol/g$, leaves $71.08{\pm}0.33{\mu}mol/g$). HDL-cholesterol was significantly higher in the high fat diet groups supplemented with C. album than the control groups (p<0.05). These results suggest that C. album can be used as a natural antioxidant and a functional dietary supplement.

Effects of Fermented Turmeric (Curcuma longa) by Bacillus natto Supplementation on Liver Function and Serum Lipid Parameters in Mice (낫토균으로 발효한 발효울금의 투여가 마우스의 간 기능 및 혈중 지질 함량에 미치는 영향)

  • The effects of turmeric and fermented turmeric by Bacillus natto on antioxidant activities, liver function recovery of acute hepatotoxicity mice, and serum lipid parameters in high fat diet fed mice were investigated. In the results of antioxidant activity by DPPH method, fermented turmeric had higher antioxidative activity than turmeric. Acute hepatotoxicity was induced by 0.5 mL of carbon tetrachloride ($CCl_4$) per kg of mice. Unlike turmeric, fermented turmeric significantly reduced the levels of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) after 5 days compared to the controls with 0.5% methyl cellulose (p<0.05). In addition, higher recovery of liver damage by $CCl_4$ was observed in mice with fermented turmeric than with turmeric. High fat (20%) diet fed mice were divided into 4 groups to investigate the effects of turmeric and fermented turmeric on serum lipid parameters: C (vehicle), TuL (low dose (80 mg/kg) with turmeric), TuH (high dose (160 mg/kg) with turmeric), FTuL (low dose with fermented turmeric), and FTuH (high dose with fermented turmeric). The levels of LDL-cholesterol and HDL-cholesterol were significantly reduced and increased in FTuL, FTuH and TuH groups compared to the C group, respectively. However, there was no significant change in triglyceride levels by either turmeric or fermented turmeric compared to those by control. Collectively, these results strongly suggest that fermented turmeric by Bacillus natto could be used as a functional food for enhancement of health with better consumer acceptance.

Effect of Spirodela polyrhiza on Antioxidant Activity in Diet-induced Obese Rats (고지방 및 고콜레스테롤 식이로 유도 된 비만 쥐에서 부평초의 간 조직에서의 항산화 활성에 미치는 영향)

  • The aim of this study was to investigate the possible antioxidant effect of Spirodela polyrhiza (SP) on rats fed a high fat and high cholesterol diet supplemented with either 5% (SPA group) or 10% (SPB group) SP for 4 weeks. The hepatic SOD activity of the HF group significantly decreased compared to that of the N group, but that of the SPA and SPB groups significantly increased. The GPx activity of the SPA and SPB groups in the liver was significantly greater than that of the HF group, and the hepatic catalase activity of the SPA and SPB groups significantly increased compared to the HF group. The hepatic superoxide radical content of the mitochondria and microsomes of the HF group significantly increased compared to that of the N group, but the contents were reduced in the group that took SP powder. The hepatic hydrogen peroxide content in the cytosol and mitochondria of the SP powder group was lower than in the HF group. The carbonyl content in the mitochondria and microsomes of the SPA and SPB groups was significantly lower than in the HF group. The TBARS values in the liver significantly decreased in the SPA and SPB groups. Spirodela polyrhiza was thus effective in reducing oxidative stress by regulating the hepatic antioxidant enzymes and the free radicals in rats fed high fat and high cholesterol diets.

Anti-inflammatory Effect of Lactuca sativa L. Extract in Human Umbilical Vein Endothelial Cells and Improvement of Lipid Levels in Mice Fed a High-fat Diet (상추 추출물(Lactuca sativa L.)의 혈관내피세포에서 항염증 작용과 고지방 식이 생쥐에서 혈중 지질농도 개선에 미치는 영향)

  • The objective of this study was to investigate the effects of a lettuce (Lactuca sativa L.) extract on the inflammation of human umbilical vein endothelial cell (HUVEC) and blood lipid improvement in hypercholesterolemic mice fed a high cholesterol diet. The lettuce extract (100% ethanol extract) inhibited the expression of intercellular adhesion molecule-1 and vascular cell adhesion molecule-1 in HUVEC treated with tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$). The lettuce extract suppressed the adhesion of THP-1 to TNF-${\alpha}$-treated HUVEC. The lettuce extract decreased the TNF-${\alpha}$-stimulated production of proinflammatory cytokine interleukin-6, interleukin-8 and chemokine monocyte chemotactic protein 1. In hypercholesterolemic mice, the lettuce extract reduced serum total cholesterol, triglyceride, and low-density lipoprotein-cholesterol level, while the lettuce extract elevated high-density lipoprotein-cholesterol level, resulting in the decrease of atherogenic index and cardiac risk factor level. These results suggested that lettuce extract can be an useful resource to show an anti-inflammatory effect and improve lipid metabolism.

Alteration of Lipid Metabolism Related Proteins in Liver of High-Fat Fed Obese Mice (고지방식이 비만쥐의 지방관련 단백질의 변화)

  • Obesity and being overweight are strongly associated with the development of metabolic disease such as diabetes, hypertension, dyslipidemia. High-fat diet (HFD) is one of the most important factors which cause obesity. In this study, C57BL/6 mice were treated with a HFD for 22 weeks in order to induce obesity and hyperglycemia. Twenty-two weeks later, body weight and plasma glucose level of the HFD group were significantly increased, compared with the normal diet (ND) group. Intra-peritoneal glucose tolerance test (IPGTT) showed glucose intolerance in the HFD group compared with the ND group. These results confirmed that a HFD induced obesity and hyperglycemia in C57BL/6 mice. Plasma levels of triglyceride (TG) and total cholesterol (TC) were increased in the HFD group compared with the ND group. Hepatic levels of TG and TC were also increased by a HFD. To investigate the alteration of lipid metabolism in liver, proteins which are related to lipid metabolism were observed. Among lipid synthesis related enzymes, fatty acid synthase (FAS) and glycerol phosphate acyl transferase (GPAT) were significantly increased in the HFD group. Apolipoprotein B (apoB) and microsomal triglyceride transport protein (MTP), which are related to lipid transport, were significantly increased in the HFD group. Interestingly, protein level and phosphorylation of AMP-activated protein kinase (AMPK), which is known as a metabolic regulator, were significantly increased in the HFD group compared with the ND group. In the present study we suggest that HFD may physiologically increase the proteins which are related with lipid synthesis and lipid transport, but that HFD may paradoxically induce the activation of AMPK.

Effects of Fresh, Red and Black Garlic Powder on Lipid Metabolism of Obese Rats Induced by High Fat Diet (생마늘, 홍마늘 및 흑마늘 분말이 고지방 식이로 유도된 비만 흰쥐의 지질대사에 미치는 영향)

  • This study was aimed to evaluate the effects garlic such as fresh garlic powder (FGP), red garlic powder (RGP) and black garlic powder (BGP) by lyophilized, on serum lipid metabolism in obese rats induced high fat diet. Food efficiency ratio (FER) of the experimental groups was lower than the control group and it was significant difference. Total lipid content of serum decreased from 7.0 to 20.9% compared to the control group. Especially, triglyceride content decreased from 40.8% (BGP) to 42.1% (RGP) group as compared to the control group. There was no significant difference in HDL-C content between groups fed garlic powder and normal group. LDL-C contents of the experimental groups were lower than the control group, but has not showed significant diggerence compared to control group. Total lipid content was significantly increased in liver compared to the control group, but decreased over 30% in RGP and BGP groups to the control group. The lipid level in feces was increased by feeding periods of garlic powder, while total cholesterol and triglyceride were significantly increased in feces of RGP group. TBARS content in serum and liver of RGP and BGP groups was significantly decreased than the control group. Antioxidant activity of serum was 68.05% in the RGP group, which was significantly higher than the control group. From the above result, we suggested that red and black garlic powder were effective in the improvement of lipid level in obese rats induced high fat diet.

Antioxidant Activity of Tea Made from Korean Mountain-Cultivated Ginseng Leaves and its Influence on Lipid Metabolism (장뇌삼 엽차의 항산화활성 및 지질대사에 미치는 영향)

  • This study was conducted to investigate the chemical composition and biological function of tea made from mountain-cultivated ginseng leaves. The antioxidant activities of tea made from mountain-cultivated ginseng leaves were determined by measuring their electron-donating ability based on their DPPH and nitrite-scavenging ability. The electron-donating abilities of tea made from mountain-cultivated ginseng leaves (500 and 1,000 ppm) as determined by DPPH assay were 45.6 and 85.1%, respectively. The nitrite scavenging ability of tea made from mountain-cultivated ginseng leaves (500 and 1,000 ppm) at pH 6.0 were 32.8 and 51.4%, respectively. Furthermore, the nitrite scavenging activity increased in a dose-dependent manner at all pH values. The effects of tea made from mountain-cultivated ginseng leaves on Male Sprague-Dawley rats were also evaluated. To accomplish this, the rats were divided into three groups (A: normal diet group, B: high fat diet group and C: high fat diet supplemented with tea made from mountain-cultivated ginseng leaves group). The anti-obesity effects of tea made from mountain-cultivated ginseng leaves were then evaluated. The serum total lipid, total cholesterol and triglyceride contents in C group were lower than those of B group; however, these differences were not statistically significant. The HDL-cholesterol content was significantly higher in the C group than in the other groups. Taken together the results of this study suggest that tea made from mountain-cultivated ginseng leaves possesses antioxidant activity and improves lipid metabolism.
