Biodistribution of $^{99m}Tc$-Lactosylated Serum Albumin in Mice with Diethylnitrosamine or Thiacetamide Induced Liver Injury (Diethylnitrosamine 및 Thioacetamide 유발 간손상 생쥐에서의 $^{99m}Tc$-Lactosylated Serum Albumin의 체내 분포상)

  • Purpose: Tc-99m labeled diethylenetriaminepentaacctic acid (DTPA)-coupled galactosylated human serum albumin (GSA) is a currently used imaging agent for asialoglycoprotein receptor (ASGPR) of the liver, but, it has several shortcomings. Recently a new ASGPR imaging agent, $^{99m}Tc$-lactosylated human serum albumin (LSA), with simple labeling procedure, high labeling efficiency, high stability was developed. In order to assess the feasibility of the $^{99m}Tc$-LSA as a ASGPR imaging radiopharmaceuticals, we performed biodistribution study of the tracer in liver injured mice model and the results were compared with histolgic data. Materals and Methods: To induce hepatic damage in ICR mice, diethylnitrosamine (DEN) ($60mg/kg/week{\times}5time$, low dose or $180mg/kg/week{\times}2times$, high dose) and thioacetamide (TAA) ($50mg/kg{\times}1time$) were administrated intraperitoneally. Degree of liver damage was evaluated by tissue hematoxilin-eosin stain, and expression of asialoglycoprotein receptor (ASGPR) was assessed by immunohistochemistry using ASGPR antibody. $^{99m}Tc$-LSA was intravenously administrated via tail vein in DEN or TAA treated mice, and biodistribution study of the tracer was also performed. Results: DEN treated mice showed ballooning of hepatocyte and inflammatory cell infiltration in low dose group and severe hapatocyte necrosis in high dose group, and low dose group showed higher ASGPR staining than control mice in immunohistochemical staining. TAA treated mice showed severe hepatic necrosis. $^{99m}Tc$-LSA Biodistribution study showed that mice with hepatic necrosis induced by high dose DEN or TAA revealed higher blood activity and lower liver activity than control mice, due to slow clearance of the tracer by the liver. The degree of liver uptake was inversely correlated with the degree of histologic liver damage. But low dose DEN treated mice with mild hepatic injury showed normal blood clearance and hepatic activity, partly due to overexpression of ASGPR in mice with mild degree hepatic injury. Conclusion: Liver uptake of $^{99m}Tc$-LSA was inversely correlated with degree of histologic hepatic injury in DEN and TAA treated mice. These results support that $^{99m}Tc$-LSA can be used to evaluate the liver status in liver disease patients.

Mineralization-inducing potentials of calcium silicate-based pulp capping materials in human dental pulp cells

  • Background: This study was performed to provide a long-term bacterial seal through the formation of reparative dentin bridge, calcium silicate-based pulp capping materials have been used at sites of pulpal exposure. The aim of this study was to evaluate the mineralization-inducing potentials of calcium silicate-based pulp capping materials (ProRoot MTA [PR], Biodentine [BD], and TheraCal LC [TC]) in human dental pulp cells (HDPCs). Methods: Specimens of test materials were placed in deionized water for various incubation times to measure the pH variation and the concentration of calcium released. The morphology of HDPCs cultured on the specimens was examined using a confocal laser scanning microscope (CLSM). Alizarin red S staining and alkaline phosphatase assays were used to evaluate mineralization-inducing potentials of the capping materials. Results: BD showed the highest calcium release in all test periods, followed by PR and TC. (p<0.05). All experimental groups showed high alkalinity after 1 day, except at 14 days. BD showed the highest cell viability compared with PR and TC after 1 and 3 days, while TC showed the lowest value (p<0.05). The CLSM analysis showed that cells were well adhered and expressed actin filaments for all pulp capping materials. Mineralization by PR and BD groups was higher than that by TC group based on alizarin red S staining. BD showed significantly higher alkaline phosphatase activity than PR and TC, while TC showed the lowest value (p<0.05). Conclusion: Within the limitations of the in vitro study, BD had higher mineralization-inducing potential than PR and TC.

Total Coliforms Viability in Effluent of Wastewater Treatment Plant Discharged into the Ocean (해양으로 배출된 하수처리수내 총대장균군의 활성 평가)

  • Difference of total coliforms (TC) survival time of mixed samples (effluent were mixed by seawater or freshwater) were examined in various conditions. The time taken to achieve a 90% reduction in the TC concentration ($T_{90}$) of effluent, high initial TC concentration and low initial TC concentration samples indicated 143.9, 121.9 and 89.6 hours at $25^{\circ}C$, respectively. At $4^{\circ}C$, log removal rate after 336 hours were 0.96, 1.04, and 1.30. TC survival time of effluent-inoculated seawater sample was longer than that of effluent-inoculated freshwater sample in laboratory. At outdoor condition, TC inactivation curves of effluent-inoculated seawater or freshwater samples showed similar patterns. And both of them were greatly influenced by climate condition. There was not enough evidence that TC survived longer in freshwater than sea water. It was unlikely that the salinity of sea water contributed to the inactivation process of TC.

Effects of Stephaniae Tetrandrae Rhizoma and Sinomenii Caulis on Hyperlipidemia in Rat (분방기(粉防己)와 청풍등(淸風藤)이 고지혈증(高脂血症) 흰쥐에 미치는 효과)

  • To compare the efficacy of Stephaniae Tetrandrae Rhizoma and Sinomenii Caulis which are medicinal plants used as Bang Kee(防己), the plants were combined with Astragali Radix and Lycopii Herba into 6 different medicinal decoctions. The specimen decoctions prepared were each decoctions of Stephaniae Tetrandrae Rhizoma(St), Sinomenii Caulis(Si), Stephaniae Tetrandrae Rhizoma-Astragali Radix(StA), Stephaniae Tetrandrae Rhizoma-Lycopii Herba(StL), Sinomenii Caulis-Astragali Radix(SiA) and Sinomenii Caulis-Lycopii Herba(SiL). They were dosed to rats with induced hyperlipidemia. In the groups of rats with hyperlipidemia induced by high cholesterol diet, in the group which St was dosed to, there was no significant effect on the level of Tc, TG, HDLc. And in the group which Si was dosed to, the level of Tc, TG dropped and that of HDLc rose. In the groups of rats with hyperlipidemia induced by high cholesterol diet, in the group which StA was dosed to, there was no significant change in the level of Tc, TG, or HDLc. But in the group which SiA was dosed to, the level of Tc and TG dropped and that of HDLc rose. In the groups of rats with hyperlipidemia induced by high cholesterol diet, in the group which StL was dosed to, the level of TG dropped, but there was no significant change in the level of Tc or HDLc. And in the SiL group, there was no significant change in the level of Tc or TG, but the level of HDLc rose. On diseases such as hyperlipidemia, when using drug couple of Bang Kee-Astragali Radix as a treatment, Sinomenii Caulis is recommended rather than Stephaniae Tetrandrae Rhizoma, and when using drug couple of Bang Kee-Lycopii Herba as a treatment, Sinomenii Caulis and Stephaniae Tetrandrae Rhizoma neither showed significant effect. And as Lycopii Herba interrupts the effect of Sinomenii Caulis, drug couple of Sinomenii Caulis-Lycopii Herba is not recommended.

Biodistribution of 99mTc Tricarbonyl Glycine Oligomers

  • $^{99m}Tc$ tricarbonyl glycine monomers, trimers, and pentamers were synthesized and evaluated for their radiolabeling and in vivo distribution characteristics. We synthesized a $^{99m}Tc$-tricarbonyl precursor with a low oxidation state (I). $^{99m}Tc(CO)_3(H_2O)_3^+$ was then made to react with monomeric and oligomeric glycine for the development of bifunctional chelating sequences for biomolecules. Labeling yields of $^{99m}Tc$-tricarbonyl glycine monomers and oligomers were checked by high-performance liquid chromatography. The labeling yields of $^{99m}Tc$-tricarbonyl glycine and glycine oligomers were more than 95%. We evaluated the characteristics of $^{99m}Tc$-tricarbonyl glycine oligomers by carrying out a lipophilicity test and an imaging study. The octanol-water partition coefficient of $^{99m}Tc$ tricarbonyl glycine oligomers indicated hydrophilic properties. Single-photon emission computed tomography imaging of $^{99m}Tc$-tricarbonyl glycine oligomers showed rapid renal excretion through the kidneys with a low uptake in the liver, especially of $^{99m}Tc$ tricarbonyl triglycine. Furthermore, we verified that the addition of triglycine to prototype biomolecules (AGRGDS and RRPYIL) results in the improvement of radiolabeling yield. From these results, we conclude that triglycine has good characteristics for use as a bifunctional chelating sequence for a $^{99m}Tc$-tricarbonyl-based biomolecular imaging probe.

Synthesis of $^{99m} Tc$-tricarbonyl Precursors for Labeling of Bioactive Molecules

  • To radiolabel bioactive molecules, we synthesized $^{99m}$Tc-tricarbonyl precursor, [$^{99m}$Tc(CO)$_3$($H_2O$)$_3$]$^{+}$ with a low oxidation state ( I ). The [$^{99m}$Tc(CO)$_3$($H_2O$)$_3$]$^{+}$ was prepared by low pressure carbonylation (1 atm of CO) of [$^{99m}$Tc $O_4$)]$^{[-10]}$ in the presence of NaB $H_4$ resulting in higher than 98% of labeling yield and stability up to 8 hrs. We evaluated the characteristics of $^{99m}$Tc- tricarbonyl labeled bioactive molecules by carrying out in vitro and in vitro study. Prepared [$^{99m}$Tc(CO)$_3$($H_2O$)$_3$]$^{+}$ was then reacted with some ligands of significance in modem diagnostic nuclear medicine and some amino acids. Labeling yields were checked by HPLC and found to be usually high, excluding $^{99m}$Tc-tricarbonyl-MDP, -EDTMP and -mIBG. And the biodistribution properties of $^{99m}$Tc-tricarbonyl complexes applied in rabbit showed different appearance comparing with that of the $^{99m}$Tc-labeling by conventional means. From these results, we conclude that [$^{99m}$Tc(CO)$_3$($H_2O$)$_3$]$^{+}$ is a potential precursor for development of radiopharmaceuticals, especially for labeling of biomolecules.

Characterizations of Water-dispersed Biocellulose Nanofibers on the Skin Surface (피부 표면에서의 수분산 바이오셀룰로오스 미세섬유의 특성에 대한 연구)

  • In this study, water-dispersed biocellulose nanofibers (TC) were prepared via an oxidation reaction using 2,2,6,6-tetramethyl-1-piperidine-N-oxy radical (TEMPO) as a catalyst. The TC retained their unique structure in water as well as in emulsion. TC adhered to the skin surface while maintaining nanofibrous structures, providing inherent functions of biocellulose, such as high tensile strength and high water-holding capacity. When gelatin gels as model skin were coated with TC, the hardness representing the elasticity was increased by 20% compared to untreated gelatin gel because TC could tightly hold the gelatin structure. When porcine skin was treated with TC and TC-contained O/W emulsion, the initial water contact angles of TC were lower than other materials, and dramatically decreased over time as water penetrated the fibrous structure of the TC film. Characterization of TC on the skin surface offered insight into the function of nanofibers on the skin, which is important for their applications with respect to fiber-cosmetics.

  • In order to realize the commercial application of HTSC materials, it is necessary to develop the fabrication process of high Tc oxide superconductor materials with desired shape and for practical application and high critical current density as well as good mechanical strength which can withstand high lorenz force generated at high magnetic field. Much studies have been concentrated to develop the fabrication technique for high critical current density but still there are a lot of gap which should be overcome for large scale application of HTSC materials at liquid nitrogen temperature. Recently some new fabrication techniques have been developed for YBaCuO bulk superconductor with high mechanical strength and critical current density. In this project, the establishment of fabrication condition and additive effects of second elements were examined so as to improve the related properties to the practical use of YBaCuO superconductor, and we reported the production of the YBaCuO high Tc superconductor by the pyrolysis method.

Usefulness and Comparison of $^{201}TI$-chloride, $^{99}mTc$-MIBI, $^{99}mTc(V)$-DMSA Single Photon Emission Computed Tomography in Distinguishing Lung Cancer from Benign Lesion (폐암과 양성질환의 감별에 $^{201}TI$-chloride, $^{99}mTc$-MIBI, $^{99}mTc(V)$-DMSA 단일광자전산화단층촬영술의 비교 및 가치)

  • Objectives: $^{201}TI$ - chloride, $^{99m}Tc$ - MIBI, $^{99m}Tc$(V) - DMSA SPECT has been used in distinguishing lung cancer from benign lesion. To compare the diagnostic efficacy of SPECT with these tumor - seeking agents, we perfonned three consecutive SPECT using $^{201}TI$, $^{99m}Tc$ - MIBI, $^{99m}Tc$(V) - DMSA in same subjects with a solitary pulmonary lesion. Methods: SPECT was carried out at 10min and 3hr for $^{201}TI$ after injection of 20mCi, and 2hr for $^{99m}Tc$ - MIBI and $^{99m}Tc$(V) - DMSA after injection of 20mCi, respectively, in 37 patients with a solitary pulmonary lesion(27 lung cancer and 10 benign diseases). In patients showing visual uptake on lesion site, we obtained the lesion - to - background(target lesion/contralateral normal lung) uptake ratio from transverse slice for each radionuclide and also calculated the retention index for $^{201}TI$. Results: The diagnostic sensitivity of $^{201}TI$, $^{99m}Tc$ - MIBI and $^{99m}Tc$(V) - DMSA SPECT to lung cancer was 100%, 96% and 73%, and the specificity was 40%, 70% and 70%, respectively. The low specificities for these agents were mainly due to high positive uptake in patients with active pulmonary tuberculosis. There were no significant differences in uptake ratios and retention index between malignant and benign lesions, and among the histologic types of lung cancer. Conclusion : $^{201}TI$ and $^{99m}Tc$ - MIBI showed higher sensitivity than $^{99m}Tc$(V) - DMSA for detecting lung cancer, but was of limited usefulness in distinguishing lung cancer from benign lesion due to low specificity, especially in area with a high prevalence of active pulmonary tuberculosis.

Antihyperlipidemic Effects of Red Ginseng, Crataegii Fructus and Their Main Constituents Ginsenoside Rg3 and Ursolic Acid in Mice

  • Anti-hyperlipidemic effect of red ginseng (RG; the steamed root of Panax ginseng C.A. Meyer) and Crataegii fructus (CF, the fruit of Crataegus pinnatifida BGE), which are used frequently in China and Korea as herbal medicines to treat arteriosclerosis, were investigated. Treatments of RG and CF significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels in Triton WR-1339-induced hyperlipidemic mice and serum TG levels in corn oil-induced hypertriglyceridemic mice. Ginsenoside Rg3 and ursolic acid, the main constituents of RG and CF, respectively, also reduced TG and TC levels in hyperlipidemic mice. RG and CF significantly lowered the high blood TG and TC levels and body and epididymal mass weights induced by long-term feeding of a high-fat diet and increased the high-fat diet-induced decrease in blood HDL cholesterol levels. RG and Rg3 reduced the blood TC levels more than CF and ursolic acid. However, blood TG level were reduced by CF and ursolic acid more than RG and Rg3. RG, CF, and their constituents also inhibited pancreatic lipase and HMG-CoA reductase activities. The most potent inhibitor was Rg3. These findings suggest that RG and CF may be suitable for the therapies of hypercholesterolemia and triglyceridemia, respectively.
