Characteristics and Health Benefit of Highly Marbled Wagyu and Hanwoo Beef

  • This review addresses the characteristics and health benefit of highly marbled Wagyu and Hanwoo beef. Marbling of Wagyu and Hanwoo beef has been increased in Japan and Korea to meet domestic consumer preferences. Wagyu and Hanwoo cattle have high potential of accumulating intramuscular fat (IMF) and producing highly marbled beef. The IMF content varies depending on the feeding of time, finishing diet, and breed type. IMF increases when feeding time is increased. The rate of IMF increase in grain-fed cattle is faster than that in pasture-fed cattle. Fatty acid composition are also different depending on breeds. Highly marbled Wagyu and Hanwoo beef have higher proportions of monounsaturated fatty acid (MUFA) due to higher concentrations of oleic acid. MUFAs have little effect on total cholesterol. They are heart-healthy dietary fat because they can lower low-density lipoprotein (LDL)-cholesterol while increasing high-density lipoprotein (HDL)-cholesterol. Clinical trials have indicated that highly marbled beef does not increase LDL-cholesterol. This review also emphasizes that high oleic acid beef such as Wagyu and Hanwoo beef might be able to reduce risk factors for cardiovascular disease.

Anti-Glycemic and Anti-Lipid Effect of Ethanol Extract of Supungsunkihwan-gagambang in High Fat Diet-induced Obese Mice (수풍순기환가감방(搜風順氣丸加減方) 투여가 비만형 제2형 당뇨병 동물모델의 당대사 및 지질대사에 미치는 영향)

  • Objectives: This study was undertaken to evaluate anti-glycemic and anti-lipid effects of Supungsunkihwan-gagambang, which is composed of Cornus officinalis, Dioscorea Batatas Decaisne, Auantii fructus and Platicodon grandiflorum. Methods: Diabetes was induced in ICR male mice ($24{\pm}1g$) with Surwit's high fat and high sucrose diet. Mice were divided into 3 groups (n=10) of normal, control and Supungsunkihwan-gagambang. The Supungsunkihwan-gagambang group was given 5% herbal medicine in their diet. The animals were fed on each experimental diet for 8 weeks. Body weight was assessed every week. At the 7th week, the fasting blood sugar (FBS) and oral glucose tolerance test (OGTT) were conducted in all experimental groups. After 8 weeks, fructosamine, lipid profile, epididymal fat weight, liver weight and white adipose tissue (WAT) size were measured. Results: Supungsunkihwan-gagambang significantly reduced FBS, OGTT and fructosamine. It also increased high density lipoprotein (HDL) cholesterol and significantly reduced triglyceride/HDL cholesterol ratio, total cholesterol/HDL cholesterol ratio and WAT size. Conclusions: These results show that Supungsunkihwan-gagambang improves anti-glycemic and anti-lipid effect in high fat diet-induced obese mice. Therefore we suggest that Supungsunkihwan-gagambang could be an effective treatment for patients with type 2 diabetes.

Effects of Laminariae Thallus Extracts on Body Weight and Lipid Metabolism in High Fat Diet Rats (곤포(昆布)추출물이 고지방 식이에 의한 체중변화와 체내 지질대사에 미치는 영향)

  • Laminariae Thallus has been known to be effective for the treatment of obesity. Te show the effectiveness of Laminariae Thallus in a more scientific way, Laminariae Thallus extract was prepared and evaluated in high fat diet rats by measuring the changes of body weight and lipid metabolism as described briefly below. 200 g of crushed Laminariae Thallus was extracted with methyl alcohol. The extract was evaporated under reduced pressure to give 14.4g. For 10 weeks, control group rat were ingested high fat diet, while the test group rat were fed with a fat diet pius Laminariae Thallus extract. Normal group was fed with normal diet. 150 mg of Laminariae Thallus extract per 1 kg of body weight was added to the diet in a test group rats. Control group rats on a high fat diet gained weight significantly, whereas the test group rats on a high fat diet plus Laminariae Thallus extract gained weight less. Significant increase of liver weight caused in a high fat diet was also inhibited by the Laminariae Thallus extract treatment. Total lipid, triglyceride, phospholipid and total cholesterol levels of serum in the high fat diet rats were remarkably increased, whereas their levels on a high fat diet plus Laminariae Thallus extract were less increased. While serum HDL-cholesterol levels was remarkably decreased in a high fat diet, its level was less decreased in a high fat diet plus Laminariae Thallus extract. Furthermore, we observed that the activities of hepatic acetyl-CoA carboxylase and fatty acid synthetase increased in a high fat diet, while their activities in a high fat diet plus Laminariae Thallus extract were getting back nearly to the normal levels of normal diet rats. These result showed that the obesity caused by a high fat diet was effectively inhibited by a Laminariae Thallus extract. Our results also showed that the abnormal lipid metabolism caused by a high fat diet was effectively cured by adding Laminariae Thallus extract.

Antiobesity Effect of Baek-Kimchi (Whitish Baechu Kimchl) in Rats Fed High Fat Diet

  • Baek-kimchi (whitish baechu kimchi) was evaluated for anti-obesity properties and effects on triglyceride (TG) and cholesterol in blood and adipose tissues in rats fed a high fat (20 %) diet, and compared to the similar effects of baechu kimchi. Baek-kimchi does not use red pepper powder but contains higher levels of sliced radish and pear than baechu kimchi. SD rats were raised for four weeks on either a normal diet (ND, based on the AIN-93M diet), high fat diet (HFD, supplemented with 16% lard oil in the ND), or HFD containing 5 % baek-kimchi or 5 % baechu kimchi. Feed consumption was not different among the groups, but weight gains were significantly lower in the groups fed either the normal diet or HFD with baek-kimchi or baechu kimchi diets than the group fed HFD alone. The weights of liver and epididymal and perirenal fat pads in baek-kimchi and baechu kimchi diet groups were lower than those of the HFD groups, but the baek-kimchi diet group had lower epididymal and perirenal fat pad weights than the baechu kimchi diet group (p<0.05). The baechu kimchi dietary group also had significantly lower triglyceride and cholesterol contents in liver and epididymal and perirenal fat, reversing the higher levels seen in HFD. Baek-kimchi and baechu kimchi diets were also effective in lowering serum triglyceride and cholesterol levels (p<0.05). These results suggest that baek-kimchi and baechu kimchi consumption can reverse the effects of HFD on weight gain and blood and tissue lipids, and that baek-kimchi is more effective than baechu kimchi. The greater effect is probably due to the higher content of radish and pear used in baek-kimchi.

Reciprocal Effect of DHEA and Rietary Fat on Glutathione Utilizing Detoxifying System in Rat Liver Tissue

  • This study was intended to examine whether dehydroepiandrosterone (DHEA) and dietary fat level or source could modulate glutathione utilizing detoxifying system activity and the cytosolic NADPH generation in rat liver. Male Sprague-Dawley rats were fed semipurifed diet containing either 2%(w/w) corn oil (low level of corn oil diet: 5 ca% of fat) 15% corn oil (high level of corn oil diet: 31 cal% of fat) or 13% sardine oil plus 2% corn oil(high level of fish oil diet: 31 cal% of fat) for 9 weeks. Half of the rats in each diet group were fed a diet supplemented with 0.2% DHEA (w/w). DHEA administration increased plasma total cholesterol level in low corn oil diet-fed rats. The high fish oil diet significantly decreased plasma total cholesterol level compared to the high corn oil diet. Plasma triglyceride level was not significantly changed by DHEA administration and dietary fat level and source. Fasting plasma glucose level was increased by DHEA administration and fish oil diet. Glucose 6-phosphate dehydrogenase activity in liver tissue was significantly increased by DHEA administration and high fat diet, especially fish oil diet. Malic enzyme activity in liver tissue was significantly increased by DHEA administration and high fat diet, especially fish oil diet. Malic enzyme activity in liver tissue was significantly increased by DHEA administration. DHEA suppressed the glutathione peroxidase, glutathione-dependent enzymes compared to the low corn oil diet, while fish oil diet elevated the activity of glutathione peroxidase and glutathione reductase compared to corn oil diet. These results suggest that DHEA administration and high level of corn oil diet may suppress the cellular detoxifying system activity through reduction of glutathione utilization, while the fish oil diet did not show these effects.

The Inhibitory Effects of Bimanbang-1(肥滿1號方) on the Obese-mouse Fed High-fat Diet (비만1호방(肥滿1號方)이 고지방식이(高脂肪食餌)로 비만(肥滿) 유도(誘導)된 백서(白鼠)에 미치는 영향)

  • Objective: This study investigated the effects of Bimanbang-1 (肥滿1號方: here in after referred to BMB-1) on the obese gene and obese inhibitory. Methods: C57BL/6 mice were induced by high fat diet. Mice were divided into three groups (normal, high fat diet with control, high fat diet with BMB-1 extract) and fed for 15 weeks. Items of this experimental study are as follows: body weight change, final body weight, the weight change of the adipocytes in body, the level change of LFT, NEFA and creatinine, the expression of ${\beta}3AR$ and leptin gene in primary adipocytes, the production change of leptin in primary adipocytes, the expression of ${\beta}3AR$, leptin and $TNF-{\alpha}$ in adipocytes tissue. Result: 1. All experimental groups showedthat the weight change decreased considerably and the high density group showedthat the final weight decreased considerably. 2. The high density group showed that the amount of the adipocyte in weight decreased considerably. 3. All experimental groups showedthat the amount of ALT decreased considerably, and AST decreased in the high density group. However, the amount of creatinine and glucose did not increase considerably. 4. All experimental groups showed that the amount of total cholesterol, LDL-cholesterol, triglyceride, and NEFA decreased, and HDL-cholesterol increased considerably. 5. The high density groups showedthat the amount of leptin decreased considerably. 6. All experimental groups showed that the revelation of ${\beta}3AR$ in primary adipose cell and 3T3-L1 cell increased considerably, and that the revelation of leptin and $TNF-{\alpha}$ in primary adipose cellsand 3T3-L1 cells decreased considerably. 7. All experimental groups showed that the size of adipocyte in adipocytes tissue decreased. 8. All experimental groups showed that the adipose vacuoles in liver tissue decreased considerably. Conclusion: The findings suggest that Bimanbang-1 causes weight loss and histological change, thus it may be effective to treat obesity.

Beneficial Effect of Anti-obese Herbal Medicine Mixture with Chitosan in High Fat Diet-induced Obese Rats

  • This study was carried out to investigate the dietary effects of chitosan mixture (CM), an herbal medicine mixture with Sukjihwang (HS), and CM containing HS (CHS) on obesity in an induced obese model of rats fed high-fat only (HF), in which supplemented diets of 5% CM (HCM), 5% HS (HHS), or 2.5% CM-2.5% HS (HCH) was tested for 6 weeks. Body weight gains, obesity indexes, and body fat contents in the experimental groups (HCM, HHS, HCH) were decreased compared with HF group. The levels of serum triglyceride, total lipid, total cholesterol and LDL-cholesterol in the experimental groups were markedly decreased, however HDL-cholesterol levels in the experimental groups were slightly increased compared with HF group. In addition, although serum ALT and AST activity, and relative organ weights were lower than those of HF group, serum albumin contents were not significantly different in all experimental groups including the normal control group (NC). In conclusion, there are improved effects on obesity in the obese model of animals with all experimental diets supplementations, and the improvement degrees on obesity depend on the content and compositions of the herbal medicine mixture. Further study is needed on the anti-obesity mechanism of these diets.

Effects of Pomegranate Peel (Granati pericarpium) Extracts on the Antioxidant Biomarkers in C57BL/6J Mice Fed a High Fat and Cholesterol Diet (석류 껍질 추출물이 고지방 고콜레스레롤 식이 급여 C57BL/6J 마우스의 항산화 지표 및 DNA 보호에 미치는 영향)

  • The present study evaluated the effects of pomegranate peel (Granati pericarpium) extract on the lipid profiles and antioxidant biomarkers in mice fed a high fat and cholesterol diet: the measured biomarkers included the TBARS value, GPx, GR, SOD and GST activities. Body fat depositions were significantly decreased in the group that received pomegranate peel. In addition, the activities of GPx, GST and SOD were significantly higher in the liver and plasma of the pomegranate peel group than in the control group. Also, the pomegranate peel diet decreased lipid peroxidation of the liver and kidney. Alkaline single-cell gel electrophoresis (comet assay) showed that the DNA damage in the plasma of the pomegranate peel group was decreased compared to that of control. The present results show that a diet with added pomegranate peel exerts protective effects against oxidative DNA damage and lipid peroxidation possibly via effects on the free radical levels in mice fed a high fat and cholesterol diet.

The Effects Of Chekamuiyiin-tang On Biochemical And Histological Changes Of Rats Fed High Diet (체감의이인탕이 고지방식이로 유발한 비만 흰쥐의 생화학 및 조직화학적 변화에 미치는 영향)

  • Objectives : Chekamuiyiin-tang has effects of promoting diuresis, promoting blood circulation, elimating phlegm and strengthening the body's resistance to disease. We experimented with Chekamuiyiin-tang in order to find effects which might possibly cure obesity. Methods : We fed a control group of rats a high fat diet and administered normal saline for 8 weeks. And we fed an experimental group of rats a high fat diet and administered an extract of Chekamuiyiin-tang for 8weeks. Results : The serum total cholesterol level, triglyceride level, free fatty acid level, total lipid level and phospholipid level in the Chekamuiyiin-tang group showed a significant decrease in comparison with the control group. The serum HDL-cholesterol level in the Chekamuiyiin-tang group showed no change in comparison with the control group. The serum LDL-cholesterol level in the Chekamuiyiin-tang group showed a decrease in comparison with the control group, but this result showed no efficacy. The size of the epididymal fat cell and the fat drops area in the hepatic lobule of rats in the Chekamuiyiin-tang group showed a significant decrease in comparison with the control group. Conclusios : According to the above results, Chekamuiyiin-tang has shown to be capable of curing and preventing obesity.

Study of the Effects of Gugijagami-bang in a Hyperlipidemic Animal Model Induced with a High-Fat Diet (고지방 식이로 유발된 고지혈증 동물 모델에서 구기자가미방(枸杞子加味方)의 효과 연구)

  • Objectives: This study was undertaken to investigate the effects of Gugijagami-bang (GGB) in a hyperlipidemic animal model induced by a high-fat diet using diverse biological methods. Methods: This study was to determine whether fractionated GGB extracts inhibit reactive oxygen species (ROS) and nitric oxide (NO) in RAW 264.7 cells. Hyperlipidemia was induced by a high-fat diet fed for 6 weeks. Total cholesterol, LDL cholesterol, HDL cholesterol, triglyceride, liver function and histologic change of liver were measured after oral administration of GGB. Results: 1. DPPH scavenging bow performance was increased in a concentration-dependent manner by GGB. 2. Compared to the control group, NO production (%) and ROS production (%) were decreased significantly by GGB. 3. Total-cholesterol, LDL-cholesterol, triglyceride were decreased significantly by GGB. 4. HDL cholesterol increased more than the control group, but not significantly. 5. In histopathologic examination, fatty liver (hepatic steatosis) was inhibited, almost no rounds of fat were observed in the liver. Conclusions: GGB would appear effective in the prevention and treatment of atherosclerosis, ischemic heart disease, other cardiovascular diseases caused by hyperlipidemia.
