Effect of the Fractionated Extracts of Mugwort on the in vitro Growth of Some Intestinal Microorganisms (쑥의 분획추출물들이 주요 장내세균의 in vitro 생육에 미치는 영향)

  • Mugwort was successively fractionated with n-hexane, chloroform, ethyl acetate, n-butanol and water and the fractions were evaluated by their growth-promoting activites for Bifidobacterium sp. in vitro experiments. The growths of Bifidobacterium adolescentis, B. bifidum, B. infantis and B.longum were enhanced with the addition of the water fraction, while the fractions of chloroform and ethylacetate inhibited Clostridium perfringens. When the wate fraction was added to media at a concentration of 0.01-0.5%(w/v), the growhts of Bifidobacterium sp. were increased according to the concentration of water fraction used. The water fraction stimulated also the growth of lactobacillus acidophillus, whereas those of E. coli and Enterococcus faecalis were not affected. The growth-promoting activity of water fraction was stable at the range of pH 2 to pH 10 and kept in thermal treatment at 10$0^{\circ}C$ for 30 minutes.

  • In a preliminary screening of the plant extracts for the antigastritic action in rats, the extract of Aralia elata(Araliaceae) showed positive activity in HCI plus ethanol induced gastric lesion. Systematic fractions with hexane, chloroform, ethyl acetate and butanol resulted in the most patent activity with the butanol fraction: This butanol fraction at the oral dose of 200 mg/kg exhibited significant inhibition of absolute alcohol induced gastric lesion which was more potent than 100 mg/kg of cimetidine and had significant stimulation of mucus secretion. The butanol fraction showed significant decreases in the ulcer indices of Shay ulcers and inhibition of gastric juice secretion with acid output in pylorus-ligated stomachs of rats. It also suppressed the acetic acid induced gastric ulcer. These results might suggest that the butanol fraction had inhibitory action in gastric lesion and ulceration through inhibition of gastric acid secretion and stimulation of mucin secretion in the stomachs of rats.

Phenolic Glycosides from Cercidiphyllum japonicum Leaves

  • Cercidiphyllum japonicum leaves were collected, air-dried and extracted with 70% aqueous acetone, then concentrated and sequentially fractionated using n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc), and $H_2O$. A portion of EtOAc fraction (10 g) was chromatographed on a Sephadex LH-20 column, by the successively elution with various aqueous $MeOH-H_2O$ (1:9, fraction $1-2{\rightarrow}3:7$, fraction $3-5{\rightarrow}1:1$, fraction $6-9{\rightarrow}7:3$, fraction $10-13{\rightarrow}9:1$, fraction 14-16). Compound 2 was isolated from fraction 6 and compound 1 was separated from fraction 11 and 12. Compound 3 and 4 were purified from fraction 13. The isolated compounds were elucidated as quercetin-3-O-${\alpha}$-L-rhamnopyranoside (1), chlorogenic acid (2), quercetin-3-O-${\alpha}$-L-arabinofuranoside (3) and quercetin-3-O-${\beta}$-D-xylopyranoside (4) by the spectral and literature data, and by comparison with the authentic samples. These compounds were reported, for the first time, from the extracts of C. japonicum leaves. Also chlorogenic acid (2) has never been reported before in domestic tree species and can be used as an index compound for C. japonicum.

  • A new aromatic amide isolated from the hexane soluble fraction of the root of Aconitum pseudolaeve var. erectum has been characterized as methyl-N-(2-acetaminobenzoyl) anthranilate on the basis of spectroscopic data, and a monoglyceride was also isolated and identified as glycerol-1-hexadecanoate from the fraction.

Antioxidative Effect of Persimmon Leaves (감잎의 항산화 효과)

  • The production of malondiadehyde(MDA) was significantly decreased when the methoanol extract of persimmon leaves was added to the system. The methanol extract of persimmon leaves was fractionated by using various solvents such as hexane, chloroform, ethylacetate and butanol. Among the above fractions especially the chloroform fraction, ethylacetate fraction revealed the strong antioxidative activities. The hot water extract of the persimmon leaves was less effective than tannin which was extracted from persimmon leaves in antioxidative activity.

  • An hydrocarbon alcohol and steroids were isolated from the hexane fraction of the extract from the seeds of Phaseolus radiatus L. And also a flavonoid was identified from the ethyl acetate and butanol fraction. The structure of the obtained compound were elucidated by spectroscopic methods (UV, IR, mass and NMR) and their physicochemical properties. As the results, triacontanol, stigmasterol, ${\beta}-sitosterol$ and vitexin were identified.

  • The root of Polygonum cuspidatum (Polygonaceae) has been used as treatments of hyperlipidemia, dermatitis, gonorrhea, favus athlete's foot, inflammation in traditional medicine. In order to examine anti-lipidperoxidation activity, hexane, EtOAc, BuOH and water fractions of its methanol extract were administered to carbon tetrachloride intoxicated rats. Ethylacetate fraction exhibited antilipidperoxidative effect on liver lipid homogenate and the radical scavenging effect on DPPH. Serum transaminase, AlP, triglyceride and total cholesterol contents significantly decreased by administrations of ethylacetate fraction.

Antioxidant Activity of Rubus crataegifolius Bge. Fruit Extracts (Rubus crataegifolius Bge. 열매 추출물의 항산화 활성)

  • We investigated the fruits of Rubus crataegifolius Bge, a plant which has been traditionally used in Korea in phytotherapy, to describe antioxidant materials from plant sources. R. crataegifolius fruits were extracted with methanol and further fractionated into n-hexane, diethyl ether, and ethyl acetate. The antioxidant activity of each fraction and the residue was assessed using a 1,1-diphenyl-2-picrylhydrazyl (DPPH), $H_2O_2$ radical scavenging method, and their cytotoxicity on human primary kerationcyte (HK) was determined by an MTS assay. The R. crataegifolius fruit methanol extract showed strong antioxidant activity (75.04%, 50%) compared with vitamin C (79.9%, 54.1%) by the DPPH, and $H_2O_2$ method, respectively. The measured activity from the subsequent extracts of the methanol extract were 20.3% for n-hexane fraction (HF), 68.8% for diethyl ether fraction (DF), 67.1% for ethyl acetate fraction (EF), and 67.1% for the residue fraction (RE) by DPPH and 2.2% for HF, 1.6% for DF, 10% for EF, and 50% for the RE by $H_2O_2$ assay. An oxidative stress model of HK was established under a suitable concentration (1 mM). The cell viability of the RE treated group increased and the percentage of apoptotic cells decreased at concentrations of 0.005-0.02% RE compared with the $H_2O_2$ treated group. Fruit extracts of the medicinal plant R. crataegifolius showed potent antioxidant activity and the ability to relieve cell damage from $H_2O_2$ induced injury to HK.

  • Achillea sibirica Ledeb. is widely distributed in Korea and has been often used as folk medicine in peptic and tonic. As one of our searches for bioactive (anti oxidation) compounds from medicinal plants, we studied Achillea sibirica Ledeb. (Compositae). Antioxidant activity of Achillea sibirica was determined by measuring lipid peroxide produced when a mouse liver homogenate was exposed at $90^{\circ}C$ using 2-thiobarbituric acid (TBA) and by evaluation the radical scavenging activity on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical. Whole parts of Achillea sibirica was extracted with methanol and its extracts was fractionated with organic solvent; n-Hexane, methylene chloride, ethyl acetate, n-Butanol. EtOAc fraction exhibited antioxidant activity and From its, two flavonoid glycosides were isolated by silica gel and gel filtration colume chromatography and identified to kampferol 3-O-glucoside and luteolin 7-O-neo-hesperidoside, respectively, by physico-chemical and spectroscopical method. At antioxidant activity test for two compounds isolated, antioxidant activity was showed too. And from hexane fraction sterol was is isolated and identificated to mixture of campesterol, stigmasterol, and ${\beta}-sitosterol$.

Study on the Effects of Jiaweicitaowan (加味慈桃丸) Ingredients on Angiogenic Inhibition (가미자도환(加味慈桃丸) 구성약물(構成藥物)의 혈관신생(血管新生) 억제효과(抑制效果)에 관한 연구(硏究))

  • Jiaweicitaowan (JWCTW) has been used to inhibit recurrence and metastasis of cancer in clinical practice. Further study has shown its anti-metastatic and anti-angiogenic effects. By applying in vitro and in vivo anti-angiogenic evaluation model, the author assayed the each ingredients of JWCTW. The author performed the following experiments and the results are listed as below: Cell viability assay showed that the viability was almost identical between that of the control and that of the ingredients extract 40 ${\mu}g/m{\ell}$ treated, except of hexane fraction of Curcumae Radix (40 ${\mu}g/m{\ell}$, 2.0% of control), ethylacetate fraction (40 ${\mu}g/m{\ell}$, 26.7%), butanol fraction (20 ${\mu}g/m{\ell}$, 87.2%; 40 ${\mu}g/m{\ell}$, 12.5%) of Cremastrae appendiculatae Tuber, water fraction of Persicae Semen (40 ${\mu}g/m{\ell}$, 82.7%), ethylacetate fraction of Hippocampus (40 ${\mu}g/m{\ell}$, 85.3%). The results of gelatin zymogram assay showed that the ingredients of JWCTW decreases the gelatinolytic activity of MMP-9 from ECV304, at the concentration of 10 ${\mu}g/m{\ell}$. In in vitro invasion assay, the ingredients of JWCTW effectively inhibited the invasion of cancer cells as compared with the control (+PMA) groups. In capillary-like tube formation assay, the hexane and ethylacetate fractions of Curcumae Radix, Cremastrae appendiculatae Tuber and Persicae Semen showed the dramatic inhibition effects on tube formation of ECV304 at the concentration of 40 ${\mu}g/m{\ell}$. In ex vivo rat aortic ring assay, the hexane and ethylacetate fractions of Curcumae Radix, Cremastrae appendiculatae Tuber and Persicae Semen showed the inhibition effects on angiogenesis of rat aorta at the concentration of 40 ${\mu}g/m{\ell}$. According to the above research, the anti-angiogenic effects of the ingredients of JWCTW was proved and it suggested that the more effective prescription for anti angiogenesis could be developed.
