  • This study was made to objectify the principle of oriental medical prescriptions so that we could obtain the principle and the formulation of them For that purpose, we analysed the formula and rule of 205 prescriptions recorded in Gum-goe-yo-rak, which have a few of components, and are widely used in clinical from ancient times to these days. At first we classified those prescriptions by their effect. Then we re-classified the herbs of the effect groups into four natures (cold, hot, warm and cool) and five kinds of flavors (sour, bitter, sweet, acrid and salty). And we classified the herbs into three grades - superior, medium, and inferior - which were used in Shen Nong's Herbal Classic. By these means, we statistically evaluated the prescriptions recorded in Gum-goe-yo-rak based on the Four Regular Components(Monarch, Minister, Assistant, and Laborer). As a result we could obtain some facts about the prescriptions recorded in Gum-goe-yo-rak, those are what kind of herbal material was used frequently and distribution of the natures flavors and grades according to thier effects. And we are sure that these results can be great help for establishment of fundamental formulation of theory of traditional herbal medicinal polyprescription.

  • The spasmogenic activities and acute toxicity study of Yumijangquebo (YMJQB), a Korean herbal laxative formulation, were subjected to pharmacological evaluation. When tested in the contractile extent of guinea pig ileum, YMJQB displayed a spasmogenic effects in a concentration up to 0.05mg/ml and then decreased contractility of ileum; exhibiting the similar pattern with acetylcholine, a physiological regulator for peristaltic movement of the gut.(omitted)

방풍통성산의 동시분석 및 항산화 효능 연구 (Simultaneous Analysis of Bangpungtongseong-san and Its Antioxidant Effect)

  • Objectives : We carry out the simultaneous quantification for quality control of four components in Bangpungtongseong-san (BPTSS) sample. In addition, we assessed the antioxidant effects of BPTSS sample. Methods : The used column for separation and analysis of four compounds was Luna C18 column and column oven temperature was maintained at $40^{\circ}C$. The mobile phase for simultaneous determination consisted of two solvent systems, 1.0% acetic acid in water and 1.0% acetic acid in acetonitrile. High performance liquid chromatography-photodiode array (HPLC-PDA) method for analysis was performed at a flow rate of 1.0 mL/min with PDA detection at 254 and 280 nm. The injection volume was 10 ${\mu}L$. The antioxidant activities of BPTSS were evaluated by measuring free radical scavenging activities on 2,2'-Azinobis-3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) and 1-1-diphenyl-2-picrylhydrazyl (DPPH). The inhibitory effects on low-density lipoprotein (LDL) oxidation were evaluated by the formation of thiobarbituric acid relative substances (TBARS) and relative electrophoretic mobility (REM). Results : Calibration curves were acquired with $r^2{\geq}0.9999$. The values of limit of detection (LOD) and quantification (LOQ) were 0.06-0.29 ${\mu}g/mL$ and 0.20-0.98 ${\mu}g/mL$, respectively. The amounts of geniposide, liquiritin, baicalin, and glycyrrhizin in BPTSS were 5.06, 7.33, 27.56, and 7.81 mg/g, respectively. The BPTSS showed the radical scavenging activity in a dose-dependent manner. The concentration required for 50% reduction (RC50) against ABTS and DPPH radicals were 72.51 ${\mu}g/mL$ and 128.49 ${\mu}g/mL$. Furthermore, GMGHT reduced the oxidation properties of LDL induced by CuSO4. Conclusions : The established HPLC-PDA method will be helpful to improve quality control of BPTSS. In addition, BPTSS has potentials as therapeutic agent on anti-atherosclerosis.

Evidence-based herbal medicine in efficacy and safety assessments

  • Herbal medicine is the use of medicinal plants for prevention and treatment of diseases: it ranges from traditional and popular medicines of every country to the use of standardized and tritated herbal extracts. Generally cultural rootedness enduring and widespread use in a traditional medical system may indicate safety, but also efficacy of treatments, especially in herbal medicine where tradition is almost completely based on remedies containing active principles at very low and ultra low concentrations or relying on magical-energetic principles. The efficacy and safety assessments of medicines, whether modern or herbal, invariably encounter challenges or problems during the course of pre-clinical and clinical research. Some of the challenges in evidence-based herbal medicinal research are unique, and the researcher must be cognizant of them in order to safeguard the quality of the data obtained. Key challenges are: the quality of raw materials; appropriateness of biological/pharmacological activity assessment methodology, and data interpretation; standardization methodology; pharmacokinetics and bioavailability of active constituents and metabolites; clinical dosage formulation/production; and clinical study designs and outcome measures.

무증상성 단백뇨에 대한 단기 한약 투약 경과 : 증례보고 (Progress of Short-Term Herbal Medicine Administration for Asymptomatic Proteinuria: Case Report)

  • Objectives: This case reports a certain level of improvement for asymptomatic proteinuria with short-term administration of herbal medicine. Methods: In the first treatment phase, the patient was treated with Dangguisu-san (DGSS) for his rib fracture. In the second treatment phase, the patient was treated with a herbal formulation of Astragali and Angelicae Sinensis (A&As particle) for asymptomatic proteinuria. Results: Treatment with herbal medicine resulted in a decrease in the patient's complaints regarding symptoms. Also, proteinuria-related items in hematology and urinalysis were improved. Conclusions: Herbal medicine therapy may be effective for proteinuria treatment.

황련과 Pelargonium sidoides 복합제제의 호흡기 감염 세균에 대한 항균 효과 (Anti-bacterial effect of fusion formulation of Coptis rhizoma and Pelargonium sidoides on the growth of bronchial diseases bacteria)

  • Objective : Coptis rhizoma is traditional herb in Korean medicine, and Pelargonium sidoides extract has been used for relief of acute bronchitis in Western medicine. The present study examined the antibacterial effect of fusion formulation of Coptis rhizoma extract and Pelargonium sidoides extract against bronchial diseases bacteria. Methods : Test sample, fusion formulation of Korean and Western medicine, was prepared by mixing Coptis rhizoma extract and Pelargonium sidoides extract at a ratio of 1:2 (w/w). Antimicrobial properties of test sample were determined by agar diffusion assay and minimum inhibitory concentration (MIC) against bronchus diseases bacteria. Results : In agar diffusion assay, the highest amount of test sample (4 mg/disk) exhibited antibacterial activity against all microorganisms tested. Test sample showed the high activity for S. aureus (19.5 mm), C. diptheriae (16.5 mm), A. fumigatus (19.3 mm), F. nucleatum (22.7 mm) and Mycobactrium sp. (17.3 mm), whereas it showed a low activity for K. pneumonia (9.7 mm). The MIC value was determined as $250{\mu}g/m{\ell}$ against C. diptheriae. Test sample showed better growth inhibitory effects against S. aureus and A. fumigatus with the MIC valus of $125{\mu}g/m{\ell}$. Conclusion : These results suggest the possibility of application to chronic respiratory diseases of fusion formulation of Korean and Western medicine, which was prepared with Coptis rhizoma extract and Pelargonium sidoides extract.

Effect of an herbal formulation on DOCA-salt and fructose induced models of hypertension in rats

  • The present study was carried out to investigate the antihypertensive effect of a folklore herbal formulation (HF) (300mg/kg/day; p.o.) in deoxycorticosterone acetate (DOCA)-salt induced and fructose induced hypertensive rats. In DOCA model, DOCA (15 mg/kg, s.c., twice a week) was administered to unilateral nephrectomized rats for 4 weeks. In fructose model, drinking water was replaced with 10% fructose solution for 6 weeks to induce hypertension. Systolic blood pressure (BP) was measured once every week during the treatment schedule. After completion of treatment schedule, BP and vascular reactivity to various agonists like Noradrenaline, Adrenaline, Phenylephrine and Serotonin (5-hydroxytrptamine; 5-HT) were recorded in rats of both models. A cumulative concentration response curve of 5-HT was carried out in isolated rat fundus strip of the DOCA-salt induced and fructose induced hypertensive rats. The results tend to suggest that HF possesses antihypertensive activity.

LC-MS/MS를 이용한 곽향정기산(藿香正氣散) 추출물 중 20종 성분의 함량분석 (Quantitative Analysis of the Twenty Marker Components in Gwakhyangjeonggi-san using Ultra-Performance Liquid Chromatography with Mass Spectrometer)

  • Generally, Gwakhyangjeonggi-san has been used for treatment of diarrhea-predominant irritable bowel syndrome. In this study, an ultra-performance liquid chromatography-electrospray ionization-mass spectrometer method was established for the simultaneous quantification of marker compounds 1-20 in Gwakhyangjeonggi-san water extract. All analytes were separated by gradient elution using two mobile phases on a UPLC BEH $C_{18}$ ($100{\times}2.1mm$, $1.7{\mu}m$) column and maintained at $45^{\circ}C$. The injection volume was $2.0{\mu}L$ and the flow rate was 0.3 mL/min with detection at mass spectrometer. Regression equations of the compounds 1-20 were acquired with $r^2$ values ${\geq}0.9950$. The values of limit of detection and quantification of all analytes were 0.01-2.79 ng/mL and 0.03-8.37 ng/mL, respectively. The amounts of the compounds 1-20 in Gwakhyangjeonggi-san water extract were not detected $-3,236.67{\mu}g/g$. The established LC-MS/MS methods will be valuable to improve quality control of traditional herbal formula, Gwakhyangjeonggi-san.

Chemical Composition and Antimicrobial Activity of Cone Volatile oil of Cupressus macrocarpa Hartwig from Nilgiris, India

  • The aim of the present study was to investigate the various chemical components present in the cone volatile oil of Cupressus macrocarpa and also determine its antimicrobial activity. Totally 13 compounds were identified with 99.99% by GC-MS analysis. The major compounds identified were terpinene-4-ol (19.42%), dinopol (15.63%), ${\alpha}$-pinene (13.58%), and ${\beta}$-pinene (12.16%). The antimicrobial activity was carried out for the oil and a 2% cream formulation using cup plate method by measuring the zone of inhibition. The gram positive organisms used were Bacillus subtilis, Staphylococcus aureus, Bacillus megaterium, and Bacillus cogulans. The gram negative organisms used were Escherichia coli, Kleibseilla pneumonia, Pseudomonas aeruginosa and Salmonella typhi. In vitro antifungal studies were also carried out by using organisms, Candida albicans, Aspergillus flavus, Trichoderma lignorum and Cryptococcus neoformans. The standard drugs used were penicillin ($100{\mu}g/mL$), gentamycin ($100{\mu}g/mL$) and griseofulvin ($100{\mu}g/mL$) for gram positive bacteria, gram negative bacteria and fungi respectively. Both oil and cream formulation showed good activity against fungi than bacteria. This study is being reported for the first time on cone volatile oil of this plant.

  • Dangguisu-san is a well-known traditional Korean herbal medicine prescription and has been widely used to treat ecchymosis, blood stagnation, and pain resulting from physical shock in Korea. In this study, an ultra-performance liquid chromatography-electrospray ionization-mass spectrometer method was established for the simultaneous determination of the 17 biomarker components in Dangguisu-san. All analytes were separated on an UPLC BEH $C_{18}$ ($100{\times}2.1$ mm, $1.7{\mu}m$) column and maintained at $45^{\circ}C$. The mobile phase consisted of two solvent systems, 0.1% (v/v) formic acid in water (A) and acetonitrile (B) by gradient flow. The injection volume was $2.0{\mu}l$ and the flow rate was 0.3 ml/min with detection at mass spectrometer. Calibration curves of the 17 biomarker components were acquired with $r^2$ values ${\geq}0.9951$. The values of limit of detection and quantification of all analytes were 0.02~6.32 ng/ml and 0.05~18.95 ng/ml, respectively. The amounts of the 17 components in Dangguisu-san sample were $3.17{\sim}13,224.50{\mu}g/g$.
