  • Alpinia katsumadai is used in traditional oriental medicine as an antiemetic and for treatment of stomach disorders. This study was to investigate the effect of the 50% methanol extract of Alpinia katsumadai and Puerariae thunbergiana on ethanol metabolism in rat. The administration of 50% methanol extract of Alpinia katsumadai significantly decrease the ethanol concentration in rat. The AUC and Cmax of Alpinia katsumadai was 3.1 and 2.4 times less than the control group, respectively. It was more effective than Pueraria thunbergiana is known to as ethanol detoxicant. Furthermore, we tested on its ethanol metabolizing effect with the preparation containing this herb (Haeju pill) in beagle. The AUC of Haeju pill was about 1.9 times less than the control group. It was estimated that the extract of Alpinia katsumadai had feasibility as a ethanol detoxication agent.

  • In macrophages, nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions including inflammation. In this study, we have examined the inhibition effects of NO production by 85% methanol extract of the flower of Pueraria thunbergiana (PF) in mouse macrophages. Extract of PF (1, 10, 100 ${\mu}g/ml$) inhibited NO production, inducible NO synthase and cyclooxygenase-2 expression in interferon-g and lipopolysaccharide-stimulated mouse peritoneal macrophages and it had no cytotoxicity. These data suggest that 85% methanol extract of PF might be useful in controlling macrophages mediated inflammatory disease.

  • Scopoliae rhizoma is a perennial herb which has a similar effect with atropine on the cardiovascular system. It is also known to have a seditive and anticonvulsant activity on the central nerve system. In order to evaluate an acute toxicity of Scopoliae rhizoma, the present study was performed after administration the Scopoliae rhizoma prepared by both decoctional and frozen dried extract through three different routes (oral; 5,000 mg/kg, intraperitoneal; 2,000 mg/kg, subcutaneous; 5,000 mg/kg) to the female ICR mice. In the group treated intraperitoneally with a frozen dried extract, abnormal clinical signs such as decreased activity, crouch, potosis and abnormal walking were observed for 40 rain after administration. With regard to WBC, decreased number of lymphocyte and increased number of monocyte and granulocyte were also observed in the animals received intraperitoneally with Scopoliae rhizoma extract. Taken together, what toxicity of Scopoliae rhizoma was shown differently depending on its type for administration may be resulted in the differency of administered dose. The results provided here support a pharmacological and toxicological consideration for its clinical use in the regard of oriental medicine.

  • In order to evaluate the antimicrobial function of natural herb extracts as antimicrobial agent or packaging material for the preservation of foods and greenhouse produce, the water extract of chopi (Zanthoxylum piperitum DC.) was prepared and its antimicobial activity was determined. In the paper disk test its antimicrobial activity was increased in proportion to its concentraion. The growth of microorganisms was completely inhibited above 500ppm of its concentration. It showed wide spectrum of thermal(40 to 180oC) and pH(4 to 10) stabilities. In the electronic microscopic observation(TEM and SEM) of microbial morphological change it showed to decrease the activation of physiological enzymes and to lose the function of cell membranes. Even in the activation test of galactosidase, it seemed to weaken the osmotic function of cell membranes remarkably in comparison with chloroform and its activation corresponded to 40~50% of toluene. Zanthoxylum piperitum DC. extract seemed to be an excellent antimicrobial for the inhibition of food borne microorganisms as well as the pre servation of greenhouse produces.

  • Tripterygium regelii SPRAGUE is distributed in Korea and Northern China. This extract has been used as a herb medicine, especially antiparasitic, anti-inflammatory and detoxifying agent in East asia. During our research to develop new antitumor agents from natural products, Dichlorornethane (CH$_2$Cl$_2$) extract of Tripterygium regelii SPRAGUE (DTR) showed the potent apoptotic effects in A-549 lung cancer, HeLa-3 cervical cancer, SKMEL-2 melanoma cells in a dose-dependent manner. in order to purify major compounds from DTR, column chromatography was carried out gradually. Silica gel and RP-18 column chromatography for active fractions led to the isolation of a compound. The compound determined by 1 H-NMR was turned out to De Celastrol known to have antitumor activity.

  • We previously reported the antimutagenicity, anti-lipid peroxidation and antiinflammtory properties of the three Chinese herb medicine, Kalopanax pictus, Pueraria thunbergiana and Rhus verniciflua, which have been used as therapeutics for diabetes mellitus and inflammation in Korea. Since these drugs have been used for the prescriptions consisted of several crude drugs, we investigated whether the extract of the three drugs could increase the individual drugs or not. The three extracts were independently prepared from K. pictus, P. thunbergiana, and R. verniciflua, which extracts were named the K-1, P-1 and R-1 in this order and the extract (KPR-1) was also prepared from the combined plant materials with each same weight of the three drugs. • Lampranthus spectabilis is an ornamental plant which belongs to one of the Aizoaceae family as an one-year herb. There are no illustrations on the references that Lampranthus spectabilis has been made use for the purpose of the medicinal plant among the traditional herb doctors and the laymen. However, it has the specific characteristics to be analogous to the Cactaceae family in the botanical classification. Besides, mescaline, the main ingredient of Peyote cactus, demonstrates the sympathomimetic effect as well as the psychotomimetic effect. The authors called our attention to the resemblance to the botanical family between Aisoaceae and Cactaceae, and made a survey whether the alcohol extract of Lampranthus spectabilis might possess any pharmacological effect on the isolated heart of clam or not. The results of the experiment were as follows. 1. Administration of LX to the isolated heart of clam produced an inhibitory effect on the cardiac motility in proportion to its concentration $10^{-4}$, $5{\times}10^{-4}$ and $10^{-3}\;g/ml$ respectively. 2. Administration of LX $5{\times}10^{-4$ and $10^{-3}\;g/ml$ produced the blockade effect on the cardiac motility pretreated with serotonin $10^{-7}$ and $10^{-4}\;g/ml$, as well as with methysergide $10^{-6}$ and $10^{-5}\;g/ml$. 3. Administration of LX $5{\times}10^{-4}\;g/ml$ produced no effect on the cardiac motility pretreated with acetylcholine $10^{-7}\;g/ml$. 4. Administration of LX $5{\times}10^{-4}\;g/ml$ produced the blockade effect on the cardiac motility pretreated with barium chloride $10^{-4}\;g/ml$, as well as with prostaglandin $20\;{\mu}g$. Consequently, it is concluded that the alcohol extract of Lampranthus spectabilis has an effect of depression on the motility of the isolated heart of clam. Its mechanism of action might be due to a direct action on the heart.

  • The objective of this study is to investigate the inhibitor effects of an traditional oriental herb, Sophora flavescens on the pancreatic & cardiac side effects of chemotherapy were studied by using Bl6-BL6 melanoma-injected C57BL6 mouse tumor model. In this study, the effects of an traditional oriental herb, Sophora flavescens, on the side effects of chemotherapy were studied using B16 melanoma-injected C57BL6 mouse tumor model. Anticancer drugs widely used in clinical situation. Sophora flavescen has significant effect on the reduction of the side effects of chemotherapy. Water extract of Sophora flavescens significantly inhibited cisplatin-induced increase of amylase (AMY) which is a good indicator of pancreatic toxicity. Sophora flavescens extract does not decrease the anti-tumor activity of cisplatin showing that it can selectively inhibit side effects of anticancer drugs preserving beneficial effect. Theses results suggest a possibility that Sophora flavescens extract can be used for cancer patients for the reduction of the side effects and improving the quality of life during chemotherapy of cancer patients.
