The influence of selenuium deficiency and fish oil on lipid peoxidation status and fatty acid composition of tissues(plasma aorta and liver) was studied. Male Sprague Dawley rats were fed for eight weeks semipurified diets containing 7% corn oil(by weight) or 5, 5% fish oil(MaxEPA) plus 1.5% corn oil with oil with or without selenium status (glutathione peroxidase activity and selenium levels) were significantly lower in the rats given inadequate selenium in plasma aorta (p<0.02 and p<0.001 respectively) gut not that in plasma Selenium supplementation decreased hepatic MDA levels(p<0.02) Increases in the levels of 20:5(n-3) 22:5(n-3), 22:6(n-3) 20:3(n-6) and a decrease in the level of 20:4(n-6) were observed in plasma total lipids and aortic and hepatic phospholipids when fish oil was fed. Though selenium supplementation increased the level of n-3 fatty acids(such as 22:6(n-3)) in plsama and the aorta is overall effect was smaller than the effect of fish oil feeding. These data suggest that selenium may play a significant but minor role in protecting against lipid peroxide-tion even when vitamin E intakes are in excess of current recommendations in both corn oil and fish oil diets.

This study investigated the effects of scopoletin (6-methoxy-7-hydroxycoumarin) supplementation on insulin resistance and the antioxidant defense system in chronic alcohol-fed rats. Rats were fed a Lieber-Decarli liquid diet containing 5% ethanol with or without two doses of scopoletin (0.01 and 0.05 g/L) for 8 weeks. Pair-fed rats received an isocaloric carbohydrate liquid diet. Chronic alcohol did not affect fasting serum glucose levels, although it induced glucose intolerance and hyperinsulinemia compared with the pair-fed group and led to insulin resistance. Both doses of scopoletin similarly improved glucose intolerance, serum insulin level, and insulin resistance. Scopoletin supplementation significantly activated phosphatidyl inositol 3-kinase, which was inhibited by chronic alcohol. Two doses of scopoletin up-regulated hepatic mRNA expression and activity of glucokinase as well as down-regulated mRNA expression and activity of glucose-6-phosphatase compared with the alcohol control group. Both doses of scopoletin significantly reduced cytochrome P450 2E1 activity and elevated aldehyde dehydrogenase 2 activity, resulting in a lower serum acetaldehyde level compared with the alcohol control group. Chronic alcohol suppressed hepatic mRNA expression and activities of antioxidant enzymes such as superoxide dismutase, catalase, and glutathione peroxidase; however, they were reversed by scopoletin supplementation, which reduced hydrogen peroxide and lipid peroxide levels in the liver. These results indicate that dietary scopoletin attenuated chronic alcohol-induced insulin resistance and activated the antioxidant defense system through regulation of hepatic gene expression in glucose and antioxidant metabolism.
To evaluate an effect of growth periods on the bromobenzene-induced liver damage, bromobenzene was administrated to 5-week-old rats and 10-week-old rats pretreated with bromobenzene 5 times every other day for 10 days and then the animals were sacrificed. The results were obtained as follows; The increasing rate of serum levels of alanine aminotransferase, xanthine oxidase activity, hepatic lipid peroxide contents, liver weight per body weight (%) and decreasing rate of hepatic contents of protein to each control group were higher in 10-week-old rats than 5-week-old rats by the pretreatment of bromobenzene. According to the above results, 10-week-old rats indicated more severe liver injury than 5-week-old those in case of bromobenzene pretreatment. On the other hand, hepatic aniline hydroxylase activity was more increased in 10-week-old rats than 5-week-old rats both in control and bromobnezene pretreated rats where as the reverse in hepatic glutathione S-transferase. In case of hepatic GSH determination at the intervals of 2, 4, 8, 24 hours throughout 24 hr after administration of single dose of bromobenzene to 5-week-old and 10-week-old rats both in control and bromobnezene pretreated, the rate of GSH utilization was lower in 10-week-old rats than 5-week-old rats. In conclusion, from the above experimental, it is deduce that the 10-week-old rats showed more severe liver injury than 5-week-old rats by the bromobenzene treatment because the disposal ability of bromobenzene in liver was lower in 10-week-old rats than 5-week-old rats.
This study was conducted to investigate the effect of old antler extracts on galactosamine-induced liver injuries in rats. Male rats of Sprague-Dawley strain with average weight of 110$\pm$10g were fed on diets containing three kinds of old antler extracts(water extract, neutral extract and ether extract) for four weeks. Galactosamine(400mg/kg body weight) was injected intraperitoneally at the same time every week in galactosamine treatment groups. Cytochrome P-450 content was decreased in galactosamine treatment groups and increased by old antler extracts administration. Glutathione-peroxidase activity was increased in water extract group. Hepatic glutathione content was not observed significant differences by the old antler extracts administration. Lipid peroxide content was higher in the galactosamine treatment groups than that of the control group and decreased in galactosamine administerd groups after pretreatment with water extract. Total lipid, triglyceride and total cholesterol contents of liver were decreased in old antler extracts administerd groups and decreased in water extract group.
This study investigated anti-obesity and antioxidant effects of dietary non-fermented soybean crud residue (SCR) and fermented SCR by Monascus pilosus (FSCR) in high-fat induced-obese mice. SCR and FSCR were supplemented with high-fat diet at 2% (wt/wt) dose for 8 weeks. Both SCR and FSCR significantly lowered body weight, epididymal fat weight and weight gain rate compared to high-fat diet control (HC) group and FSCR group showed lowest weight gain rate. In addition, it was observed that serum and hepatic lipid profiles including triglyceride, total cholesterol, LDL-cholesterol and HDL-cholesterol were significantly improved by supplementing SCR or FSCR. Furthermore, SCR and FSCR administration showed increase of glutathione content and decrease of hepatic lipid peroxide content, serum aminotransferase activity, and hepatic xanthine oxidase activity. On the other hand, activities of reactive oxygen species scavenging enzyme such as superoxide dismutase, glutathione S-transferase and glutathione peroxidase in two test groups were higher than those of HC. Lastly, in comparison with SCR, FSCR was more effective in restoring obesity-related biomarkers to normal level in high-diet induced obese mice. In conclusion, the present study indicates that FSCR could have not only anti-obese effects such as inhibition of abdominal fat accumulation, but also protective effects of cardiovascular disease such as atherosclerosis by decreasing serum and hepatic lipid contents. Furthermore, these results suggest that experimental diets in this study could alleviate hepatic damage caused by overproduction of reactive oxygen spices (ROS) due to obesity via inhibition of ROS generating activities and induction of ROS scavenging activities.

Alcohol is well known agent which can damage the human tissues such as liver via stimulating lipid peroxidation. On the other hand, carotenoids in addition to vitamins A, C and I play important roles in protecting these oxidative damages as well as preventing the production of free radicals. This study was carried out to investigate the effect of dietary vitamin A on lipid peroxidation and antioxidants status in ethanol-treated rats. In the experiment, male Sprague-Dawley rats weighing 160~180 g were given a liquid diet containing 36% of total calories as ethanol for 7 weeks. The pair-fed control rats received an isocaloric amount of diet containing sucrose instead of ethanol on the following day Additionally, the liquid diet contained adequate amount of $\beta$-carotene, retinyl acetate or 13-sis-reinoic acid except vitamin A-deficient diet. The results obtained are as follows. The levels of plasma and hepatic lipid peroxide were increased after chronic ethanol feeding in rats. Retinyl acetate supplementation significantly reduced lipid peroxidation induced by ethanol feeding Glucose 6-phosphatase activity was significantly reduced in rats fed vitamin A-deficient diet with ethanol and alkaline phosphatase activity was significantly induced in rats fed 13-cis-reinoic acid diet with ethanol. Catalase and alcohol dehydrogenase activities did not show a consistent tendency in experiment groups. The hepatic antioxidant enzyme activities did not significantly changed by chronic ethanol feeding groups. The striking decrease in conversion of $\beta$-carotene to retinol was observed in rats fed a $\beta$-carotene diet with ethanol feeding The level of retinol and retinoic acid in plasma and liver was decreased after chronic ethanol administration Based on this result, these data suggest that ethanol feeding enhances oxidative stress especially in those fed a vitamin A-deficient diet, and vitamin A supplementation, especially, retinyl acetate intake can prevent enhanced lipid peroxidation and related damage to some extent.

The present work was undertaken to investigate the protective mechanism of Oenanthe iavanicu n-butanol extract on the carbon tetrachloride-induced hepatotoxicity in mice. It was observed that a striking enhancement of serum alanine aminotransferase and hepatic lipid peroxide content after carbon tetrachloride administration were markedly decreased by the presentment of Oenanthe javanica extract for 5 days. It was also observed that the hepatic aniline hydroxylase, catalase, glutathione S-transferase activity and glutathione content were not changed by the injection of Oenanthe javanica extract for 5 days. Whereas, hepatic xanthine oxidase activity was inhibited by the treatment of Oenanthe javanica extract for 5 days. After treatment with Oenanthe javanica extract, xanthine oxidase activity was decreased with dose and time-dependent manner as compared to control group. However, hepatic xanthine oxidase activity was not affected by the addition of Oenanthe javanica extract in vitro. These results suggest that the inhibition of hepatic xanthine oxidase activity by the injection of Oenanthe javanica extract is believed to be a possible protective mechanism for the carbon tetrachloride-indured hepatotoxicity in mice.
This study was designed to test the effect of Korean traditional tea materials on oxygen-free radical metabolism in lead (Pb) -administered rats. Male rats were divided into normal, Pb-control (Pb-Con) and Pb-water extract of green tea (Camellia sinensis; GT) , persimmon leaf (Diospyros kaki; PL) , safflower seed (Carhamus tinctorius: SS) , Du-Zhong (Eucommia ulmoides; EU) groups, respectively. Pb intoxication was induced by administration of lead acetate (25 mg/kg. B.W., oral) weekly. The extract was administered based on 1.26 g of raw material/kg B.W./day for 4 weeks. When the GT, PL, SS and EU were supplemented to the Pb-administered rats, hepatic lipid peroxide levels were significantly lower compared to the Pb-Con group. Hepatic cytochrom P-450 content and aminopyrine N-demethylase activity was lower in the Pb-Con group than in the normal group, whereas xanthine oxidase activity was significantly elevated in Pb-administered rats. The water extract of GT, PL, SS and EU supplementation attenuated changes in enzyme activities generating reactive oxygen species in the liver. Hepatic superoxide dismutase, catalase and glucose 6-phosphate dehydrogenase activities were significantly higher in the Pb-Con group than in the normal group, while monoamine oxidase activity also tended to increase in the Pb-administered rats. However, glutathione peroxidase and glutathione S-transferase activities, and glutathione content significantly decreased through Pb intoxication. The supplementation of GT, PL, SS and EU induced alleviation changes of hepatic antioxidant enzyme activity.

Kimchi contains various antioxidative components, therefore it is expected that kimchi have the inhibitory activity on aging. Among the many factor related to aging, UV radiation is the easiest way to induce skin aging. This experiment was designed to demonstrate the antioxidative activity of dietary kimchi with and without UV radiation in tissues. Lyophilized Korean cabbage kimchi, mustard leaf kimchi and buchu kimchi were used for measurement of the contents of antioxidative components. Chlorophyll contents in buchu and mustard leaf kimchi were higher than cabbage kimchi, and the contents of carotene and ascorbic acid were higher in the mustard leaf kimchi. To clarify the antioxidative activity of various kimchi diets in vivo system, hairless mice were fed the diet containing 10% lyophilized kimchi for 20 weeks. Contents of superoxide anion and hydroxyl radical in liver were not shown significant differences, however hydrogen peroxide were significantly decreased in mustard leaf and buchu kimchi diet groups. Supplementation of all kinds of kimchi diets attenuated the hepatic NDA accumulation, especially in mustard leaf and leek kimchi group. In the skin, the contents of MDA were decreased in kimchi groups compared to control group, especially the buchu kimchi diet significantly inhibited the lipid peroxidation. To elucidate the effect of kimchi diets on photooxidation, the extent of oxidative damage in liver and skin was measured after UVB radiation treatment. The contents of lipid peroxides was decreased in mustard leaf kimchi group and cabbage kimchi group compared to control, while lipid peroxidation was increased in buchu kimchi diet group. It suggests that chlorophylls and other antioxidative nutrients in the various kimchi diets are related to antioxidation or photooxidation and it might be needed further study to certify the exact mechanisms.

Phellinus gilvus (PG) is a widely used mushroom for health promotion. We studied the hepatoprotective effect of the polysaccharide aqueous extract of PG (PGP) against carbon tetrachloride ($CCl_4$)-induced liver injury in rats. Sprague Dawley rats were divided into 5 groups: Normal control, $CCl_4$ control, PGP 50, 100, and 200 mg/kg + $CCl_4$. The levels of serum biochemical parameters, liver lipid peroxide and antioxidant enzymes, and histological appearances were evaluated. The $CCl_4$-induced increments of alanine aminotransferase, asparate aminotransferase, and alkaline phosphatase levels in serum were significantly decreased by PGP-pretreatments. The PGP dose-dependently decreased hepatic malondialdehyde formations in $CCl_4$-treatment groups. Hepatic antioxidant enzymes (superoxide dismutase, catalase, and glutathione peroxidase) were elevated by PGP in $CCl_4$-treatment groups. Histopathological evaluation of liver showed that the loss of hepatocytes, fatty changes, swelling and extensive necrosis of hepatocytes in centrilobular regions of the $CCl_4$-treated rats were ameliorated by PGP pretreatment. The PGP has hepatoprotective and antioxidative effects in $CCl_4$-induced liver injury of rat.
