Effects of Oldenlandiae Diffusae Herba Methanol Extract on Acetaminophen Induced Acute Liver Injury in Mice (백화사설초 메탄올 추출물이 acetaminophen으로 유도된 생쥐의 급성 간손상에 대한 효능 연구)

  • Objectives : Oldenlandiae Diffusae herba has been used as a natural drug for tumor, inflammation and liver disease in traditional medicine. This study was performed in order to investigate the antioxidative effects of Oldenlandiae Diffusae herba methanol extract(ODHM) on acetaminophen induced acute liver injury in mice. Methods : In order to investigate the protective effect of ODHM on acute hepatic injury in vivo, ICR mice were pretreated with ODHM, and then treated with acetaminophen(500mg/kg). And the levels of LPO and glutathione(GSH), antioxidative enzyme activities were measured. The levels of LPO were measured by TBA method. And catalase activity was measured as the decrease in hydrogen peroxide absorbance at 240nm on spectrophotometer using 30mM hydrogen peroxide. Superoxide dismutase(SOD) was assayed by recording the inhibition of nitro blue tetrazolium reduction with xanthine and xanthine oxidase. Glutathione peroxidase(GPX) activity was determined by the modified coupled assay developed by Paglia and Lawrence. The reaction was started by addition of 2.2mM hydrogen peroxide as substrate. The change in absorbance at 340nm was measured for 1min on spectrophotometer. Glutathione-S-transferase(GST) activity was assayed with CDNB as substrate and enzyme activity of GST towards the glutathione conjugation of CDNB. And Total SH and GSH levels were measured. Results : In vivo study, LPO levels of acetaminophen treatment group were significantly higher than other groups. This increased level was significantly reduced by ODHM pretreatment. The acetaminophen treatment resulted in a decrease of catalase, GPX, SOD and GST activities. By contrast, ODHM pretreatment markedly increased compare to those of untreated groups. Total SH and GSH levels were reduced by of acetaminophen treatment, and ODHM pretreatment significantly increased GSH levels.

  • This experiment was carried out to investigate variation of plasma lipid level and liver peroxide level(Thiobarbituric acid, T.B.A) caused by tube feeding of Pteridium sp. extracts for seven weeks in rats. A seven week rat's plasma triacylglyceride (T.G.) level showed remarkably higher than that of the control group and that of the three week rats. On the other hand, no remarkable differences of plasma phospholipid, free fatty acid and thiobarbituric acid level have been recognized. Liver homogenate lipid peroxides value of seven week feeding rats showed remarkably higher level compared with that of the control group and the three weeks rats. So it could not be agreed that short time feeding of Pteridium sp. plant or even it's concentrated extracts influenced to the levels of liver homogenate lipid peroxides, plasma lipids and its peroxides.

  • The purpose of this study is to observe the protective effect of Kojinyumja on serum reaction and hepatic tissue in galactosamine treated rats. In this study, the experimental rats divided four group(Normal group, Control group, Sample A group, and Sample B group) : Under the same condition, normal and control group were administered water, sample A, B group were administered Kojinyumja for 8days. And then, both control group and Sample B group were injected to abdomen with galactosamine for 1day. The rates of of glutathione, lipid peroxide, GOT, GPT, ${\gamma}$-GTP, ALP, LDH, and contents of bile acid level were measured. The results are as follows : The glutathione rate significantly increased in sample group, the others (lipid peroxide, GOT, GPT, ${\gamma}$-GTP, ALP, LDH, bile acid) significantly decreased in sample group.

The Effects of Dietary Proteins on Hepatic Histological Changes and Membrane Stability in Chemically Induced Rat Hepatocarcinogenesis (화학적으로 유도한 랫드 간세포 암화과정에서 단백질 식이가 간의 조직학적 변화와 생체막 안정도에 미치는 영향)

  • The purpose of this study is to determine the effect of dietary proteins and fats on the hepatic histological changes, membrane stability, and drug-metabolizing enzyme activities during chemically induced rat hepatocarcinogenesis. Weanling Sprague-Dawley rats were fed the diet containing 20% casein or soy protein isolate and 15% perilla or corn oil for 10 weeks. Hepatocarcinogensis was initiated with diethylnitrosamine(DEN), and the rats were fed diets containing 0.02% 2-acetylaminofluorene(AAF) followed by 0.05% phenobarbital (PB). The scores of histological changes were decreased in treated rats fed soy protein diet compared to those find casein diet. Liver weights were significantly increased by AAF and PB treatment in rats fed casein diets in both oil groups. Glucose 6-phosphatase(G6Pase) activities, an index of membrane stability, were significantly reduced by AAF and PB treatment in rats find casein diets, and were lower in casein diet compared to soy protein diet groups. Especially, the activities were the highest in the rats fed soy protein-perilla oil diet. Lipid peroxide values also were increased by AAF and PB treatment in rats fed casein diet. Aniline hydroxylase activities were not influenced by protein and fat sources. Glutathione-dependent enzyme activities were increased by AAF and PB treatment. Linoleic and arachidonic acid content were increased in rats fed corn oil diet, and linolenic and eicosapentaenoic acid contents were increased in rats fed perilla oil diet. Our results suggest that soy protein isolate inhibit the abnormal histological changes in liver, possibly by maintaining the membrane stability during chemically induced rat hepatocarcinogenesis. Soy protein may be protective against the hepatocarcinogenesis induced by chemical carcinogen.

  • Objective : This study was designed to investigate the anti-oxidative effects of Oldenlandiae Diffusae Herba Water extract (ODHW) on lipid peroxidation by free radicals oxidative hepatic injury. Methods : In order to evaluate anti-oxidative activities of ODHW in the liver cell, cultured normal rat liver cells(Ac2F) were incubated with or without ODHW. After 16 hours to 18 hours of experiment, cells were placed in DMEM medium without serum, and then incubated with 1mM tert-butyl hydro-peroxide(t-BHP) for two hours. Viable cells were detected by MTT assay. The levels of LPO induced by hydroxyl radical derived from H2O2-Fe2+ system in rat liver homogenate were determined by means of TBA. Inhibitory effect of ODHW on superoxide generation was measured by xanthine-xanthine oxidase system. Results : In the linoleic acid autoxidation system, ODHW exhibited antioxidant activity, which inhibited 85% of linoleic acid peroxidation. These effects were similar to those of dl-a-tocopherol. ODHW showed scavenging effects on DPPH radical, inhibited superoxide generation in xanthine-xanthine oxidase system, and also inhibited lipid peroxidation of rat liver tissue with hydroxyl radical derived from $H_2O_2-Fe^{2+}$ system. In addition, ODHW protected the cell death induced by t-BHP and it significantly increased cell viability in a normal rat liver cell(Ac2F)

Protective effects of alginate-free residue of sea tangle against hyperlipidemic and oxidant activities in rats

  • The antihyperlipidemic and antioxidant activities of dietary supplementation of sea tangle from Goseong and the alginate-free residue of sea tangle were investigated in Sprague Dawley rats treated with a high-fat diet, streptozotocin, poloxamer 407, and bromobenzene. The alginate-free residue of Goseong sea tangle induced a significant reduction in triglycerides and total cholesterol levels, as well as a significant increase in high-density lipoprotein cholesterol levels. Alginate-free Goseong sea tangle residue reduced the activities of the phase I enzymes aminopyrine N-demethylase and aniline hydroxylase, which had been increased by intraperitoneal injection of bromobenzene. Pretreatment with Goseong sea tangle residue prevented a bromobenzene-induced decrease in epoxide hydrolase activity. Bromobenzene reduced hepatic glutathione content and increased hepatic lipid peroxide levels. Pretreatment with alginate-free Goseong sea tangle residue prevented lipid peroxidation induced by bromobenzene, but pretreatment with Goseong sea tangle did not. These results suggest that Goseong sea tangle residue exerted antihyperlipidemic and antioxidant activities that were higher than those induced by alginate-containing sea tangle. Therefore, the alginate-free residue may contain physiologically unknown active components, other than alginic acid, which may potentially be used to prevent hyperlipidemic atherosclerosis.

Effects of Daesihotang and its component groups on diabetes, free radical and antioxidative defense system in alloxan induced diabetic rats (대시호탕(大柴胡湯)이 alloxan으로 유발(誘發)된 고혈당(高血糖) 백서(白鼠)에 미치는 영향(影響))

  • The purpose of this study was to reseach the effect of Daesihotang and its component groups on diabetes, free radical and antioxidative defense system in alloxan-induced diabetic rats. The experimental group was divided into three groups: Daesihotang (DS), Soyangyak (DS1), Yangmyungyak (DS2). The results were obtained as follows: The level of glucose, triglyceride, total cholesterol, creatinine in serum were considerablely reduced by Daesihotang. And the level of HDL cholesterol, albumin, total protein in serum were increased by Daesihotang significantly. And the level of BUN has some decreased, but with no significancy. In the study of Daesihotang on free radical scavenging effect in vitro, it has shown that Daesihotang and its components group have significant suppressing effect on peroxidation of linoleic acid on concentration. And in other experiments as scavenging effect of DPPH radical, inhibitory effect of superoxide in xanthine-xanthine oxidase system and inhibitory effect on lipid peroxidation reaction by hydroxy radical in $H_2O_2-Fe^{2+}$ system, Daesihatang have some activity, but with no significancy. In the study of Daesihotang on antioxidative defense system in vivo, the activity of GOT and GPT in serum were significantly increased; and the amounts of lipid peroxide in serum was reduced significantly but in liver no significancy. In hepatic catalase activity, Daesihotang has showed significant effect. In the level of hepatic glutathione, Daesihotang increased the amount of glutathione significantly. And in the activity of glutathione-s-transferase, Daesihotang has decreasing effect but has no significancy. These result suggest that Daesihotang has strong effect on diabetes and it is useful to prevent diabetes, but has less effect on peroxidative damage by free radical.

Effects of Solvent Fractions of Korean Cabbage Kimchi on Antioxidative Enzyme Activities and Fatty Acid Composition of Phospholipid of Rabbit Fed 1% Cholesterol Diet (배추김치 용매획분이 고 콜레스테롤 식이를 섭취한 토끼의 항산화 효소계 및 인지질 지방산 조성에 미치는 영향)

  • The antioxidant effects of dichloromethane, ethylacetate or water fraction of kimchi added to the 1% cholesterol diet were studied. Six New Zealand white rabbits in each group were fed either control diet (basal diet containing 1% cholesterol) or experimental diet containing dichloromethane (CH$_2$Cl$_2$), ethylacetate (EtOAc) or water ($H_2O$) fraction of kimchi in the control diet for 16 weeks. The amount of each solvent fraction of kimchi added to the experimental diet was equivalent to 5% of freeze-dried kimchi. Levels of hepatic lipid oxidation expressed as TBARS or peroxide value for the experimental groups were lower than that of control (p<0.05). Liver homogenated of the experimental group containing dichloromethane fraction of kimchi inhibited LDL oxidation in the presence of Cu++ by 46% (p<0.05). The activities of catalase, Glutathione peroxidase (GSH-Px), Cu, Zn-superoxide (Cu, Zn-SOD) and Mn-superoxide (Mn-SOD) of experimental groups were lower than those of control group. Low enzyme activities observed from the kimchi solvent fraction groups might be due to the level of lipid oxidation progressed less in these groups. The most significant antioxidant effects were observed from dichloromethane fraction of kimchi among the experimental groups. The major fatty acids of hepatic phospholipid of rabbit were C18:2 and C18:0. But the major fatty acid profile was changed into C16:0, C18:0, C18:1, and C18:2 when rabbit was fed 1% cholesterol diet for 16 weeks, and this profile was almost the same as in rabbit fed diet containing kimchi solvent fraction. The ratio for unsaturated fatty acid to saturated fatty acid decreased by cholesterol induced diet and it was not corrected by kimchi solvent fractions.

The Effect of Angelicae gigantis Radix on the Benzo(a)pyrene-induced Hepatotoxicity in Rats (Benzo(a)pyrene에 의해 유도된 간기능 장해에 미치는 당귀의 효과)

  • This study was performed to investigate the effect of Angelicae gigantis Radix extract oil the hepatic detoxifying enzyme activities and lipids. Male sprague-dawley rats were administrated the extracts with benzo(a)pyrene, a hepatotoxic agent, inducing liver damages. Results obtained from this study were as follows: 1. The markedly increased enzyme activities (AST, ALT, LDH, ALP, ${\gamma}$-GTP, GSH-Px) in B(a)P induced groups tended to be decreased by the treatment of the Angelicae gigantis Radix extract. 2. Liver GSH content and lipid peroxide activity were decreased by the administration of the extracts. 3. It tended that the curative effects were better than the protective effects of the extracts.

Influence of Korean Red Ginseng Water Extract on Recovery of Hepatic Function in Hypercholesterolemic Mice Fed High Cholesterol Diet (홍삼물추출물이 고콜레스테롤 식이로 유도된 고콜레스테롤 혈증에서 간 기능 회복에 미치는 영향)

  • Hypercholesterolemia is associated with many pathological states such as fat deposit in the tendon and skin, hepatomegaly, pancreatitis and heart attack. The present study was focused on the effect of Korean red ginseng water extract (RGWE) on the recovery of hepatic function that was deteriorated in high-cholesterol diet fed mice. Mice were divided into 6 groups ; normal diet group (AIN-2WK), 4 week-high-cholesterol diet group (CHOL-4WK), 2 week-high-cholesterol + 2 week-normal diet group (CHOL-2WK), 2 week-high-cholesterol + 2 week-normal diet + RGWE treated group (R100, R300, R500). RGWE treated group received normal diet and RGWE at daily doses of 100, 300, 500 mg/kg b.w. for 2 weeks after the 2 weeks feeding of high-cholesterol diet. The results are as follows : 1. The hepatic weight and total hepatic lipid weight were markedly lower in RGWE treated group than those in the CHOL-4WK or CHOL-2WK group, indicating that ginseng inhibited hepatic hypertrophy. 2. Total hepatic cholesterol and HDL-cholesterol in normal diet conversion group (CHOL-2WK) were significantly reduced compared with the high cholesterol group (CHOL-4WK). The lowering effect was significantly reinforced by RGWE administration. 3. Hepatic triglyceride level of CHOL-4WK group was markedly lower than that of the normal group (AIN-2WK). However, the level was significantly increased in CHOL-2WK and RGWE treated group. The result indicated that ginseng administration and diet conversion exert normalizing effects of hepatic function. 4. The lipid peroxide levels in RGWE treated groups were markedly lower than CHOL-2WK group. These results suggest that Korean red ginseng water extract can be useful as a dietary supplement for the treatment of obesity or hypercholesterolemia by improving lipid metabolism and hepatic function.
