Objectives : The purpose of this study is to evaluate the anti-lipogenic effect of Sobuncheong-eum on non-alcoholic fatty liver disease in free fatty acid induced cellular model. Methods : HepG2 cells were treated with palmitate for 24h to overload intracellular triglyceride (TG) content in the presence or absence of Sobuncheong-eum extract. After palmitate treatment, Intracellular TG content was measured with TG assay kit. Several lipogenesis related markers, including AMP-activated protein kinase (AMPK), sterol regulatory element-binding transcription factor-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), and fatty acid synthase (FAS), were assessed using Western-blot analyses and RT-PCR. Results : Palmitate markedly increased intracellular TG in HepG2 cells, and which were alleviated by coadministered Sobuncheong-eum extract. Sobuncheong-eum extract activated AMPK, which plays a key role in reducing hepatic lipid accumulation, and reduced lipogenic fators, SREBP-1c, ACC, and FAS. Conclusions : Taken together, it is conceivable that Sobuncheong-eum has an potential to alleviate steatosis, and which may be mediated by activating AMPK at least in part.
구기엽 추출물 (LLE)과 클로로필을 제거한 구기엽 추출물 (LLE(Ch-))이 MCD diet로 비알코올성 지방간을 유도한 C57BL/6 mouse와 팔미트산으로 지방 축적을 유도한 HepG2 세포에서 지방축적의 억제에 미치는 영향을 검토하였다. 대조군 대비 LLE(Ch-)는 혈장 TG 농도와 혈장 AST와 ALT의 활성을 유의하게 감소시켰으며 LLE군에서는 혈장 ALT 활성이 유의하게 감소하였다. 두 군 모두 간조직의 TG와 cholesterol 함량을 유의하게 낮추었으며 간조직의 병리학적 변화 결과에서 LLE군에 비해 LLE(Ch-)군에서 지방축적의 억제효과가 더 크게 나타났다. 팔미트산 0.5 mM을 처리한 HepG2 세포에서 LLE와 LLE(Ch-)가 1,000 ㎍/mL 농도까지 독성이 없었으며 대조군에 비하여 각각 200 ㎍/mL과 500 ㎍/mL 농도부터 세포 내 지방 축적량을 유의하게 감소시켰고 pAMPK와 pACC 발현이 농도 의존적으로 증가하였으며 FAS발현은 농도 의존적으로 억제되었다. 결과적으로 구기엽 100% 에탄올 추출물들은 간조직의 지방 축적을 억제할 수 있으며 그 효과는 클로로필 제거 구기엽의 활성이 좀 더 큰 것으로 나타났다. 따라서 이 두 소재 모두 항 지방간 효능이 있는 것으로 판단되며 기능성 소재로서의 개발 가능성을 확인하였다.
The objective of the current study was to investigate the effects of dietary ${\omega}-6/{\omega}-3$ polyunsaturated fatty acid (PUFA) ratios on lipid metabolism in goslings. One hundred and sixty 21-day-old Yangzhou geese of similar weight were randomly divided into 4 groups. They were fed different PUFA-supplemented diets (the 4 diets had ${\omega}-6/{\omega}-3$ PUFA ratios of 12:1, 9:1, 6:1, or 3:1). The geese were slaughtered and samples of liver and muscle were collected at day 70. The activities and the gene expression of enzymes involved in lipid metabolism were measured. The results show that the activities of acetyl coenzyme A carboxylase (ACC), malic enzyme (ME), and fatty acid synthase (FAS) were lower (p<0.05), but the activities of hepatic lipase (HL) and lipoprotein lipase (LPL) were higher (p<0.05), in the liver and the muscle from the 3:1 and 6:1 groups compared with those in the 9:1 and 12:1 groups. Expression of the genes for FAS (p<0.01), ME (p<0.01) and ACC (p<0.05) were higher in the muscle of groups fed diets with higher ${\omega}-6/{\omega}-3$ PUFA ratios. Additionally, in situ hybridization tests showed that the expression intensities of the high density lipoprotein (HDL-R) gene in the 12:1 and 9:1 groups were significantly lower (p<0.01) than that of the 3:1 group in the muscle of goslings. In conclusion, diets containing lower ${\omega}-6/{\omega}-3$ PUFA ratios (3:1 or 6:1) could decrease fat deposition by inhibiting fat synthesis in goslings.
본 연구는 발효시킨 블루베리액이 고지방식이로 비만이 유도된 C57BL/6J mice에 미치는 항비만 효과 및 기전에 대해 평가하였다. 실험동물은 각 7마리씩 3군으로 나누어 고지방식이와 증류수를 섭취하는 HD군, 고지방식이와 블루베리액을 섭취하는 BHD군, 고지방식이와 블루베리발효액을 섭취하는 FBHD군으로 사육하였다. 경구투여량은 10 mg/kg BW/day로 설정하였고, 이는 12%의 블루베리액(2.5 g 블루베리 함유; 블루베리 함유 안토시아닌 함량은 약 3.75 mg, 총 폴리페놀은 약 10.3 mg 추정)을 나타낸다. 본 연구 결과, 블루베리액과 블루베리발효액은 체중 감소, 체지방량 감소, 간의 중성지방과 총 콜레스테롤 함량을 감소시켰다. 그리고 혈중 LDL-콜레스테롤을 낮추고, HDL-콜레스테롤을 증가시켰으며 AST 및 ALT 농도를 감소시켰다. 또한 렙틴 농도가 낮아졌으며 지방 합성 유전자 발현에서는 SREBP-1c, ACC 발현 수준이 유의적으로 낮아졌으며, 지방 산화 유전자 발현에서는 블루베리발효액의 ACOX 발현 수준이 유의적으로 증가하였다. 결론적으로 BHD군과 FBHD군이 항비만 효과를 나타냈었으며, 특히 FBHD군이 BHD군보다 간에서 총 콜레스테롤 20%, AST 9%, ALT 52%, 지방 합성 유전자 SREBP-1c 발현을 감소시켰고, 혈중 HDL-콜레스테롤 16.4%와 지방 산화 유전자 ACOX 발현을 유의적으로 증가시켰다. 따라서 본 실험의 결과, 블루베리는 고지방 섭취로 인한 콜레스테롤 및 지질변화에 감소 효과를 나타내며, 특히 발효를 통해 기능성이 더욱 증진되는 것으로 나타났다. 발효과정은 블루베리의 항비만 효과를 증진시키며 블루베리발효액은 기능성 식품으로서 이용가치가 높을 것으로 사료된다.
본 연구에서는 Sprague-Dawley 종 흰 쥐 수컷을 정상 대조군 (C), 알코올 군 (E), 알코올 + 100 mg/kgBW 곤드레 에탄올 추출물군 (E+LCS), 알코올 + 500 mg/kgBW 곤드레 에탄올 추출물군 (E+HCS)으로 나누어 Lieber-DeCarli control diet 혹은 Lieber-DeCarli ethanol diet를 8주간 공급하였으며, 이때 곤드레 에탄올 추출물은 액상 사료에 직접 섞어 공급하였다. 알코올과 곤드레 에탄올 추출물의 식이 공급 종료 후 간 조직의 지방구 축적 정도를 살펴본 결과, E+LCS군과 E+HSC군은 E군에 비해 지방간 발생이 유의적으로 억제되었으며, 정상 대조군인 C군과 유의적인 차이가 없었다. 이와 비슷하게, 곤드레 에탄올 추출물의 공급은 알코올에 의해 증가된 간 조직과 혈청의 중성지방 농도를 유의적으로 낮추었으며, 혈청 AST와 혈청 ALT 활성도 정상 대조군 수준으로 회복시키는 것으로 나타났다. 한편, 곤드레 에탄올 추출물의 공급은 p-AMPK과 p-ACC 단백질 수준을 농도 의존적으로 증가시켰으며, 두 단백질 모두 E군에 비해 E+HSC군에서 유의적으로 높게 나타났다. 또한 FAS mRNA와 SCD1 mRNA 수준은 E군에 비해 E+HSC군에서 유의적으로 낮게 나타났다. 곤드레 에탄올 추출물은 간 조직에서 알코올 공급에 의해 증가된 $NF{\kappa}B$의 활성을 유의적으로 낮추었으며, $NF{\kappa}B$의 표적 단백질인 $TNF{\alpha}$ 단백질 수준을 농도의존적으로 낮추었다. 본 연구 결과를 통해 곤드레는 알코올에 의한 지방간 발생 및 관련된 간 손상을 유의적으로 억제할 수 있음을 확인하였으며 이 과정에서 AMPK 활성 증가와 $NF{\kappa}B$ 활성 억제가 관여함을 제시하였다.
Objectives : We tried to uncover the anti-lipogenic effect and underlying mechanism of Laminaria japonica on an experimental cellular model of non-alcoholic fatty liver disease. Methods : Ethanol extract of Laminaria japonica (LJ) was prepared. Intracellular lipid content of palmitate-treated HepG2 cells was evaluated with or without LJ treatment. We measured the effects of LJ on liver X receptor ${\alpha}$ ($LXR{\alpha}$) and sterol regulatory element-binding transcription factor-1c (SREBP-1c) expression, transcription level of lipogenic genes, including acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS), stearoyl-CoA desaturase-1 (SCD-1), and nuclear factor erythroid 2-related factor 2 (Nrf2) activation in HepG2 cells. Results : LJ markedly attenuated palmitate-induced intracellular lipid accumulation in HepG2 cells. LJ suppressed $LXR{\alpha}$-dependent SREBP-1c activation, and SREBP-1c mediated induction of ACC, FAS, and SCD-1. Furthermore, LJ activated Nrf2, which plays an important cytoprotective role in non-alcoholic fatty liver disease. Conclusions : Our study suggests that LJ has the potential to alleviate hepatic lipid accumulation, and this effect was mediated by inhibiting the $LXR{\alpha}$-SREBP-1c pathway that leads to hepatic steatosis. In addition, the anti-lipogenic potential may, at least in part, be associated with activation of Nrf2.
The objective of this experiment was to determine the effects of n-3 polyunsaturated fatty acids (n-3 PUFAs)-enriched diet supplemented with different levels of $\alpha$-tocopherol on the activities of hepatic lipogenic-related enzymes and the contents of liver and plasma lipid fractions in laying Tsaiya ducks. A total of 180 30-wk-old laying Tsaiya ducks, at the beginning of peak production, were allotted into 6 treatments with 3 replicates each. Ducks were fed one of the 6 experimental diets, containing 4% tallow (control), and 4% fish oil supplemented with graded levels of $\alpha$-tocopheryl acetate ($\alpha$-tocopherol) at 0, 100, 200, 300 and 400 mg/kg, respectively, for 6 wks. Feed and water were supplied ad libitum throughout the experimental period. The results indicated that the n-3 PUFAsenriched diet supplemented with different levels of $\alpha$-tocopherol did not affect (p>0.05) egg weight, feed intake, body weight change or liver and abdominal fat weights. Egg production, egg mass and feed efficiency significantly (p<0.05) improved as dietary $\alpha$-tocopherol levels increased. The activities of hepatic lipogenic-related enzymes including acetyl-CoA carboxylase (EC 6. 2. 1. 3; ACC), glucose-6-phosphate dehydrogenase (EC 1. 1. 1. 49; G-6-PDH), ATP-citrate cleavage enzyme (EC 4. 1. 3. 8; CCE), NADP-malate dehydrogenase (EC 1.1.1.40; NADP-MDH) and fatty acid synthetase (FAS) were higher (p<0.05) in birds fed with the tallow diet than in those fed with fish oil diets and increased with increasing dietary $\alpha$-tocopherol levels. None of the dietary treatments significantly affected the contents of triglyceride and total cholesterol in the liver, or total cholesterol, phospholipid and total lipid in the plasma. However, the contents of phospholipid and total lipid in the liver, and triglyceride in the plasma increased as dietary $\alpha$-tocopherol levels increased. Increasing dietary $\alpha$-tocopherol levels decreased the non-esterified fatty acid (NEFA) content in the plasma and trended to decrease the cholesterol contents in the egg yolk. The lipid metabolism of laying Tsaiya ducks was influenced not only by the dietary fat but also by the supplementation levels of $\alpha$-tocopherol.
Objective: The present study was conducted to investigate the effects of lycopene on growth performance, abdominal fat deposition, serum lipids levels, activities of hepatic lipid metabolism related enzymes and genes expression in broiler chickens. Methods: A total of 256 healthy one-day-old male Arbor Acres broiler chicks were randomly divided into four groups with eight replicates of eight birds each. Birds were fed basal diet supplemented with 0 (control), 100, 200, and 400 mg/kg lycopene, respectively. Results: Dietary 100 mg/kg lycopene increased the body weight at 21 day of age compared to the control group (p<0.05). Compared to the basal diet, broilers fed diet with 100 mg/kg lycopene had decreased abdominal fat weight, and broilers fed diet with 100 and 200 mg/kg lycopene had decreased abdominal fat percentage (p<0.05). Compared to control, diets with 100, 200, and 400 mg/kg lycopene reduced the levels of total triglyceride and total cholesterol in serum, and diets with 100 and 200 mg/kg lycopene reduced the level of serum low density lipoprotein cholesterol (p<0.05). The activity of fatty acid synthase (FAS) in 400 mg/kg lycopene treated broilers and the activity of acetyl-CoA carboxylase (ACC) in 100, 200, and 400 mg/kg lycopene treated broilers were lower than those fed basal diet (p<0.05). Lycopene increased the mRNA abundance of adenosine monophosphate activated protein kinase α (AMPK-α), whereas decreased the mRNA abundance of sterol regulatory element-binding protein 1, FAS, and ACC compared to the control group (p<0.05). Conclusion: Dietary lycopene supplementation can alleviate abdominal fat deposition and decrease serum lipids levels, possibly through activating the AMPK signaling pathway, thereby regulating lipid metabolism such as lipogenesis. Therefore, lycopene or lycopene-rich plant materials might be added to poultry feed to regulate lipid metabolism.

CD36 및 $PPAR-{\gamma}$ mRNA 유전자 발현 수준 또한 대조군보다 백미군, 혼합잡곡군, 항비만혼합잡곡군에서 감소하였다. 간 내 ${\beta}$산화로 지방축적 억제와 관련된 유전자 인자인 $PPAR-{\alpha}$ 및 CPT-1 mRNA 유전자 발현 수준은 대조군보다 혼합잡곡군, 항비만혼합잡곡군에서 증가하였다. 본 실험 결과를 종합해 볼 때 고당식이로 비알코올성 지방간질환을 유도한 마우스에서 백미군, 혼합잡곡군 및 항비만혼합잡곡군 모두 지질 대사 개선 효과가 나타났으며 항비만혼합잡곡군이 가장 효과적이었다.
Objectives : This study is aimed to compare the changes in Antioxidative capacity of Liriopis Tuber by steaming process and to compare the effects in hyperlipidemia induced rats fed high cholesterol diet between Simvastatin and Liriopis Tuber by steaming process. Methods : The SD rats were divided into six groups: normal diet (Nor), high cholesterol diet (Veh), high cholesterol diet plus Simvastatin 5 mg/kg (Sim), high cholesterol diet plus LT0 extract 200 mg/kg (LT0), high cholesterol diet plus LT6 extract 200 mg/kg (LT6) and high cholesterol diet plus LT9 extract 200 mg/kg (LT9). We compared the total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL), low density lipoprotein cholesterol (LDL) contents and reactive oxygen species (ROS) from each serums. Protein expression in liver tissues related to antioxidant and cholesterol was analyzed. Results : The Antioxidant activity of Liriopis Tuber increased by steaming process. In vivo, TC, TG, LDL-c, atherogenic index (AI) and cardiac risk factor (CRF) decreased and HDL-c increased with increasing steaming frequency. aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatinine, and blood urea nitrogen (BUN) decreased with increasing steaming frequency. ROS decreased only in LT9, and SOD, catalase and glutathione peroxidase (GPx) increased with increasing steaming frequency. phospho-AMP-activated protein kinase (p-AMPK) increased and sterol regulatory element-binding protein 2 (SREBP-2), Phospho-Acetyl-CoA Carboxylase (p-ACC) and HMG-CoA reductase (HMGCR) decreased with increasing steaming frequency. Liver staining showed a decrease in hepatic fat accumulation of LT9. LT9 showed significant results in all experiments. Conclusions : LT9 showed significance of anti-lipid effect and improved fatty liver of hyperlipemia induced rats fed on high cholesterol diet, In conclusion, LP9 can be effectively used for the treatment of hyperlipidemia.

제 5 장 계약 해지 및 이용 제한

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
