Cecropin A-Magainin 2 유도체 펩티드의 Trichosporon beigelii에 대한 항진균 활성 및 인간 적혈구 세포에 대한 용혈활성 (Fungicidal and Hemolytic Activity of Cecropin A-Magainin 2 Analogue Peptides against Tri-chospoon beigelii and Human Red Blood Cells)

  • 강한 항진균 활성을 나타내며, 낮은 적혈구 용혈활성을 갖는 새로운 합성 펩티드를 얻기 위하여 cecropin A(1-8)-magainin 2(1-12) 및 이들의 유사체들을 설계하고 이들을 고상법에 의하여 합성하였다. 합성 펩티드의 항진균 활성은 Trichosporon beigelii에 대한 성장억제능에 의하여 측정하였으며, 세포독성은 인간의 적혈구 세포에 대한 용혈활성에 의하여 측정하였다. 펩티드의 양쪽 친매성의 증가는 항진균활성보다는 적혈구 용혈황성에 큰 영향을 미쳤다. Cecropin A(1-8)-magainin 2(1-12)의 12번 아미노산이 Lys을 Ala으로 치환시킨 유사체 펩티드(A2)는 가장 강한 항진균활성(minimum inhibitory concentration : 2.5.$\mu$g/ml)을 나타내며 비교적 낮은 적혈구 용혈활성(200.$\mu$g/ml의 펩티드 농도에서 0.5% hemolysis를 나타냄)을 나타내었다. 따라서 A2의 펩티드는 세포독성을 갖지 않으며 강한 항진균 활성을 가지는 모델로서 유용하게 사용될 것이다.

학슬 및 관동화 에탄올 추출물의 항산화 및 항용혈 효과 (Antioxidant and Antihemolytic Activities of Ethanol Extracts of Carpesii Fructus and Farfarae Flos)

  • Objectives : To develop a natural antioxidant and anti-hemolytic agents, we investigated the effects of ethanol extracts of Carpesii Fructus and Farfarae Flos. Methods : Aerial parts of Carpesii Fructus and Farfarae Flos were extracted with 80% ethanol. Antioxidant activity of Carpesii Fructus or Farfarae Flos extract was evaluated by employing three different assays, i.e., 1,1-diphenyl-2-picryl hydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzthiazoline-6-suphonic acid) diammonium (ABTS) scavenging and reducing power activities. Also, anti-hemolytic activity of Carpesii Fructus or Farfarae Flos extract was determined using [2,2'-azobis (2-methylpropionamidine) dihydrochloride (AAPH)]-induced hemolysis, glutathione (GSH) depletion and malondialdehyde (MDA) formation in normal rat red blood cells (RBC) or plasma. Results : The extracts obtained from Carpesii Fructus and Farfarae Flos dose-dependently increased the scavenging activity on DPPH- or ABTS-induced radicals and the reducing power activities. Carpesii Fructus and Farfarae Flos were similar to the scavenging activity and the reducing power of butylated hydroxy anisole effect at high concentration ($1,000{\mu}g/mL$). RBC oxidative hemolysis and plasma MDA formation induced by AAPH were significantly suppressed by the extracts of Carpesii Fructus and Farfarae Flos in a dose-dependent manner. Also, Carpesii Fructus and Farfarae Flos extracts prevented the depletion of cystosolic antioxidant GSH in RBCs. Carpesii Fructus generally had better than the free radical scavenging activity, the reducing power and anti-hemolytic effects of Farfarae Flos. Conclusions : These results suggest that Carpesii Fructus and Farfarae Flos may have value as the potential antioxidant and anti-hemolytic medicinal plant.

Structure-Antifungal Activity Relationships of Cecropin A-Magainin 2 and Cecropin A-Melittin Hybrid Peptides on Pathogenic Fungal Cells

  • In order to investigate a relationship of the structure-antifungal and hemolytic activities between cecropin A(1-8)-magainin 2(1-12) and cecropin A(1-8)-melittin(1-12) hybrid peptides, several analogues with amino acid substitution at positions 10 (Ile) and 16 (Ser) were designed and synthesized. The increase of the hydrophobicity by substituting with Leu, Phe, and Trp at position 16 in cecropin A(1-8)-magainin 2(1-12) did not have a significant effect on antifungal activity but caused a remarkable increase in hemolytic activity. These results indicate that the hydrophobic property at position 16 of cecropin A(1-8)-magainin 2(1-12) is more correlated to hemolytic activity than to antifungal activity. Replacement with Pro at position 10 of cecropin A(1-8)- magainin 2(1-12) and cecropin A(1-8)-melittin (1-12) caused a remarkable decrease in a-helical contents in the 50% TFE solution and induced a reduction in lytic activity against Aspergillus flavus, and Aspergillus fumigatus. These results demonstrate that flexibility at the central hinge region is essential for lytic activity against fungal cells and $\alpha$-helicity of the peptides.

대두 사포닌의 활성에 관한 기전 연구

  • Saponins are glycosidic compounds present in many plant foods. They are characterized by their ability to lyse cell membranes due to their surface-active properties. Saponins are believed to interact primarily with cholesterol in the cell membrane. In this study, the interaction of soybean(SS) with cell membrane was investigated using erythrocytes as a model. Mechanisms of interaction was also investigated by measuring their binding capacity with different membrane lipid fractions. Throughout the study, gypsophilla saponin(GS) and quillaja saponin(QS) were used to evaluate the membranolytic activity of soybean saponins. All saponins released hemoglobin in a concentration-dependent manner. SS induced 40% hemolysis at the concentration of 400 ppm, however there was no increase in hemoglobin release above 400ppm concentration. 5ppm of GS and 8 ppm of QS hemolyzed 100% of erythrocytes. Isolation of SS fractions by thin layer chromatography revealed that only one non-polar saponin possesses strong hemolytic activity. When saponins were incubated decreased the release of cholesterol. When the hemolytic activity of saponins was measured in the presence of other major membrane lipid components, sphingomyelin significantly reduced the hemolytic activity of SS, while cholesterol reduced the activity of QS. GS showed high affinity to other component(s) in the incubation media as well as lipids. These results suggest that the membranolytic activity of saponins are related to their specific chemical structure, which determines the interaction behavior between saponins and different membrane components, and thereby influence the biological activity.

Antimicrobial Effects of Ocotillone Isolated from Stem Bark of Ailanthus altisshima

  • Bioassay-directed chromatographic fractionation of a methylene chloride extract of Ailanthus altisshima indicated the presence of 20(S), 24(R), epoxy-25-hydroxydammarane-3-one (compound 1, ocotillone) which was isolated from this plant, for the first time. Antimicrobial activity of compound 1 was measured by inhibition of bacterial and fungal cells growth and by a hemolytic assay with human erythrocytes, respectively. The results revealed that compound 1 had potent antibacterial activity against Cram-negative bacteria, P. aeruginosa and S. typhimurium, that were without hemolytic activity, whereas it had weak antimicrobial activity against Gram-positive bacteria and fungi. These results demonstrated that the compound 1 has more antibacterial activity against 6ram-negative bacteria, which have no hemolytic activity, than Gram-positive bacteria and fungi. This is the first report on the biological activities of the compound 1.

  • It is well-known that onion and garlic of Allium family have been cultivated for major food since ancient times. Especially garlic is known for containing protective materials against cardiovascular disease, neoplasm and cancer, and decreasing cholesterol value. Hemolytic anemia was first reported by Goldsmith in onion-fed cattle in 1909. Subsequently, similar effects have been reported in cattle, horses, sheep, dogs and cats. This toxic effect has been attributed to npropyl disulfide which causes oxidant damage to erythrocyte cell membrane. There are few reports on overconsumption of garlic. Kim et al. reported in 1999 that 45% of Jindo dogs in korea have erythrocytes characterized by high Na-K-ATPase activity with high K and low Na concentrations (HK) which easily results in hemolytic anemia. The purpose of present report is to determine whether garlic-induced hemolytic anemia occurs in normal erythrocytes characterized by complete lack of Na-K-ATPase activity with low K and high Na concentrations (LK) when LK dogs are fed large amounts of garlics. We found in this experiment that Erythrocyte count, PCV and Hemoglobin concentration decreased significantly, and GSH and Met-hemoglobin concentrations increased. Consequently, we determined that overconsumption of garlic induces hemolytic anemia in LK dogs.

Pathogenic Factors of Vibrio spp. Isolated from Seawater of Gwangan Beach in Busan

  • The authors identified 68 Vibrio strains from Gwangan beach seawater from June to October in 2001. We identified them as 19 strains of Vibrio alginolyticus, 15 strains of V. vulnificus, 15 strains of V. parahaemolyticus, 11 strains of V. cholerae non O1, 7 strains of V. fluvialis and just one strain of V. hollisae. They showed their typical biochemical characteristics by API 20E kit (bioMerieux), respectively. It was examined whether their cultural supernatants had enzymatic activities such as hemolysin, protease or urease. The 46 strains showed hemolytic activities and/or protease activities. But we could not find any strain which had urease activity. All isolates of V. cholerae non O1 showed $\beta$ hemolysis. The others showed $\alpha$ hemolysis or did not show clear zones on sheep blood agar plates. These results of Kanagawa phenomenon were not always correspondant with hemolytic activities of cultural supernatants at late log phase. Some strains had higher hemolytic activities despite of showing protease activities on skim milk agar plates and in litmus milk media. On the other hand, some strains showed protease activities but did not show hemolytic activities. Therefore we could guess that there were the relationships between hemolysins and proteases produced by pathogenic vibrios.

Enhanced Expression and Functional Characterization of the Recombinant Putative Lysozyme-PMAP36 Fusion Protein

  • The porcine myeloid antimicrobial peptide (PMAP), one of the cathelicidin family members, contains small cationic peptides with amphipathic properties. We used a putative lysozyme originated from the bacteriophage P22 (P22 lysozyme) as a fusion partner, which was connected to the N-terminus of the PMAP36 peptide, to markedly increase the expression levels of recombinant PMAP36. The PMAP36-P22 lysozyme fusion protein with high solubility was produced in Escherichia coli. The final purified yield was approximately 1.8 mg/L. The purified PMAP36-P22 lysozyme fusion protein exhibited antimicrobial activity against both Gram-negative and Grampositive bacteria (Staphylococcus aureus, Salmonella enterica serovar Typhimurium, Pseudomonas aeruginosa, and Bacillus subtilis). Furthermore, we estimated its hemolytic activity against pig erythrocytes as 6% at the high concentration ($128{\mu}M$) of the PMAP36-P22 lysozyme fusion protein. Compared with the PMAP36 peptide (12%), our fusion protein exhibited half of the hemolytic activity. Overall, our recombinant PMAP36-P22 lysozyme fusion protein sustained the antimicrobial activity with the lower hemolytic activity associated with the synthetic PMAP36 peptide. This study suggests that the PMAP36-P22 lysozyme fusion system could be a crucial addition to the plethora of novel antimicrobials.

Antimicrobial, Antioxidant and Hemolytic Activity of Water-soluble Extract of Mottled Anemone Urticina crassicornis

  • We evaluated the biological activities of five water extracts of tissue of the mottled anemone Urticina crassicornis. Most extracts exhibited broad-spectrum antimicrobial activity as determined by ultrasensitive radial diffusion assay (URDA) against gram-positive and -negative bacteria, including a fish pathogen, Aeromonas hydrophila, but no activity against fungi. The activity of the extracts was abolished by tryptic digestion, indicating that protein compounds were responsible for the antimicrobial activity. Furthermore, in a 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging activity assay, only the visceral tissue extract showed activity. However, no extract had hemolytic activity against human red blood cells. Consequently, this study suggests the water-soluble extract of mottled anemone to be a promising source of proteinaceous antimicrobial compounds that can be utilized for development of novel antibiotics.
