Haptoglobin Levels in Turkish Patients with Bladder Cancer and its Association with Clinicopathological Features

  • Although alteration in the haptoglobin phenotype has been reported in patients with bladder cancer, serum haptoglobin levels have not been evaluated. We hypothesized that serum haptoglobin can be used as a biomarker. The aim of this study was to evaluate the expression of haptoglobin in bladder cancer and to determine the relationship with clinicopathological features. A total of 68 serum specimens obtained before surgery were used to investigate haptoglobin expression using the sandwich ELISA technique. Serum haptoglobin levels were higher in the patients with bladder cancer compared to healthy controls (p<0.0001). Additionally, the levels of haptoglobin protein increased with increasing tumor grades (p<0.001) and were significantly higher in patients with metastatic disease and the presence of lymphovascular involvement, lymph node metastases and increasing tumor burden (p<0.0001). This study suggests that elevated haptoglobin levels are associated with a higher stage, grade, and extent of distant metastasis and larger tumor size. Haptoglobin may therefore provide a useful diagnostic and treatment biomarker for patients with bladder cancer.

Haptoglobin Concentration in the Cord Blood of Uninfected Korean Newborns

  • Objective: Highly sensitive haptoglobin measurement should be used in neonates because the haptoglobin concentration in neonates is lower than that of adults. The aim of this study was to establish the reference values of haptoglobin levels in the cord blood of uninfected neonates. Methods: The cord blood of 29 preterm and 51 term babies was collected, and data from the mother and the newborn were recorded. The haptoglobin concentrations of 80 cord blood samples were simultaneously measured by enzyme-linked immunosorbent assay (ELISA; Assaypro, St Charles, MO, USA) and immunoturbidimetry assay (Roche Diagnostics, Basel, Switzerland). C-reactive protein (CRP) was also measured by immunoturbidimetry assay (Roche Diagnostics, Switzerland). Results: Mean values of CRP and ELISA haptoglobin were not significantly different between preterm and term babies. The 2.5 percentile and 97.5 percentile values of ELISA haptoglobin concentration were as follows: 80 neonates, 0.01 mg/dL and 0.59 mg/dL; 29 preterm babies, 0.08 mg/dL and 0.18 mg/dL; and 51 term babies, 0.07 mg/dL and 0.23 mg/dL. There were no differences in ELISA haptoglobin concentration according to maternal underlying diseases, delivery method, usage of antibiotics or steroids before delivery, gestational age, gender of baby, or twin gestation. Conclusion: A highly sensitive haptoglobin method should be used to determine the haptoglobin concentration in Korean newborns because the reference values of cord blood haptoglobin concentration in Korean newborns are less than the lower detection limit for commonly used immunoturbidimetric haptoglobin measurement methods.

  • The purpose this research is to develop QCM (quartz crystal microbalance) biosensor for the determination of haptoglobin. Haptoglobin is an acute-phase protein with a hemoglobin-binding activity and has a potential to be used as a biomarker for infection or cancer. Haptoglobin level in milk has been used for the diagnosis of cow mastitis. In this study, anti-bovine haptoglobin antibody or bovine hemoglobin was chemically immobilized on the surface of the QCM, and the resulting sensor chips were tested for their response to samples containing haptoglobin at different concentrations. Concentration dependent frequency change was observed with both of the sensor chips. Especially, the sensor chip containing anti-bovine haptoglobin antibody showed sufficient sensitivity in the concentrations typically observed in the cows with mastitis.

Haptoglobin as an early indicator of infection in pigs (돼지에서 감염의 조기진단 지표로서의 Haptoglobin 적용에 관한 연구)

  • Sera of pigs with clinically normal and infectious conditions were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) to demonstrate the specific changes in protein profile. In the sera from pigs with infection, haptoglobin with a 40KDa protein was found to be increased as compared to that of sera from normal pigs. As a rapid detection method for monitoring infections at large-scale farms, one of acute phase protein, haptoglobin, was selected to compare the concentrations between infectious and non-infectious conditions. Haptoglobin concentrations were low in pigs with clinically normal conditions but significantly increased in pigs with Aujesky's disease, hog cholera and parvo-virus infection. The studies provide that haptoglobin can be used as an indicator to monitor infections early at farm level.

Haptoglobin as an early indicator of infection in horses (말에서 감염의 조기진단을 위한 haptoglobin 지표 연구)

  • The objective of present study was to assess the diagnostic value of the concentration of haptoglobin, one of a series of acute phase proteins that is found in the blood of both humans and animals. A total of 79 random horse sera(10 dam and 69 foals) with clinically normal were analyzed by ELISA. The mean serum concentrations of haptoglobin of dam and foals were 1.67${\pm}$0.52mg/ml(range 1.08∼2.12 mg/ml), 1.65${\pm}$0.50 mg/ml(range 0.50∼2.66 mg/ml), respectively. These results may provide basic information for developing a useful marker system to monitor infections early in horses.

Changes in the concentrations of serum haptoglobin to infection with Erysipelothrix rhusiopathiae and antibiotics treatment in pigs (돼지 단독균 인공감염 및 항생제 치료에 따른 Haptoglobin치의 변화)

  • The changes of concentration of serum haptoglobin, body temperature, hematological result and antibody titers were compared in pigs given intravenously injection of E rhusiopathiae and control pigs. Pigs administered with E rhusiopathiae injections showed acute septicemia and arthritis. Also, statistically significant changes observed in the total white cell counts and body temperature. An increased haptoglobin concentration in serum was detected after E rhusiopathiae infection, but not during antibiotics treatment. The results indicated that measurement of the concentration of serum haptoglobin may be useful in the early diagnosis of infectious disease, such as Erysipelas.

Acute phase proteins in dairy cows with mastitis

  • In this study, to assess the diagnostic value for mastitis in dairy cows, change of acute phase proteins(haptoglobin and serum amyloid A) concentrations in milk and sera of dairy cows were measured. 50 dairy cows were used in this experiment and divided into two groups. The first group was the healthy dairy cow group whose milk contained less than 2.0${\times}$10$\^$5/ somatic cell counts(n=5). The second group was the mastitis-dairy cow group whose milk counted higher than 5.0${\times}$10$\^$5/ somatic cell counts(n=45). The concentration of haptoglobin and serum amyloid A in milk and sera from these two groups were determined by Tridelta range haptoglobin kit and serum amyloid A kit. The concentration of haptoglobin in the milk from first group was undetectable value and that of the second group was 124.0$\mu\textrm{g}$/$m\ell$. And the concentration of haptoglobin in serum of the first group was 32.0$\mu\textrm{g}$/$m\ell$ and that of the second group was 214.4$\mu\textrm{g}$/$m\ell$. The concentration of serum amyloid A in the milk from first group was 0.32$\mu\textrm{g}$/$m\ell$ and that of the second group was 17.7$\mu\textrm{g}$/$m\ell$. And the concentration of serum amyloid A in serum of the first group was 5.1$\mu\textrm{g}$/$m\ell$ and that of the second group was 25.8$\mu\textrm{g}$/$m\ell$. It was concluded that concentration of haptoglobin and serum amyoid A in milk and serum may be was to discriminate between normal and mastitic milks.

Detection of human and bovine haptoglobin by using quartz crystal microbalance sensor chip containing secondary antibody (이차항체를 포함하는 수정미소저울 센서 칩을 이용한 사람과 소의 헵토글로빈 측정)

  • In this study, secondary antibody-containing quartz crystal microbalance(QCM) sensor chip was prepared and utilized for the detection of human and bovine haptoglobin. Anti-goat immunoglobulin G antibody, which is a secondary antibody capable of capturing primary antibodies raised in goat, was immobilized through the reaction between hydrazide and aldehyde group prepared on the QCM surface and antibody respectively. The resulting sensor chip showed higher stability in the repeated surface regeneration with acidic dissociation solution as well as requiring lower amount of primary antibody when compared to the protein G sensor chip. The secondary antibody sensor chip was applied for the estimation of bovine and human haptoglobin.
