본 연구에서는 대나무, 계피, 생강 등 8종의 한약재를 포함한 한방목초액의 소재특성을 시험하여 화장품 및 피부질 환용 의약품에의 응용가능성을 조사하였다. 목초액의 항산화력은 DPPH 자유라디칼 소거법으로 측정되었으며 $50{\mu}g/ml$의 농축목초액 농도에서 97%의 매우 높은 항산화능을 보여주었다. 목초액의 항균력은 원판확산법으로 시험되었으며, 피부상재균인 Staphylococcus aureus에 대해 우수한 항균력을 보여주었다. 목초액의 미백효과는 tyrosinase 활성억제시험법으로 측정되었으며 대조군인 비타민 C에 비해 매우 낮아서 미백효과는 미미하였다. 목초액의 안전성 시험은 MTT assay에 의해 측정되었으며, 세포 독성이 비교적 높은 것으로 나타났다. 목초액의 화장품소재 안정성 시험 결과 색, 향, 외관 및 pH에서 큰 변화가 없어서 매우 안정하였다. 무모생쥐를 이용한 항 아토피 시험결과, 목초액은 DNCB에 의해 유도된 피부염증을 9일 째 거의 정상상태로 회복시켰으며, 피부내 IgE의 농도도 대조군에 비해 30% 감소시켜 항 아토피 효과가 우수한 것으로 나타났다. 따라서 목초액은 항산화력, 항균력 및 항 아토피 효과가 우수하여 화장품소재나 피부질환치료제로서의 응용 가능성이 큼을 알 수 있었다.
Exposure of the skin to ultraviolet radiation can cause skin damage with various pathological changes including inflammation. In the present study, we identified the skin-protective activity of 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucose (pentagalloyl glucose, PGG) in ultraviolet B (UVB) radiation-induced human dermal fibroblasts and mouse skin. PGG exhibited antioxidant activity with regard to intracellular reactive oxygen species (ROS) generation as well as ROS and reactive nitrogen species (RNS) scavenging. Furthermore, PGG exhibited anti-inflammatory activity, inhibiting the activation of nuclear factor-kappaB (NF-${\kappa}B$) and mitogen-activated protein kinase (MAPK) signaling, resulting in inhibition of the expression of pro-inflammatory mediators. Topical application of PGG followed by chronic exposure to UVB radiation in the dorsal skin of hairless mice resulted in a significant decrease in the progression of inflammatory skin damages, leading to inhibited activation of NF-${\kappa}B$ signaling and expression of pro-inflammatory mediators. The present study demonstrated that PGG protected from skin damage induced by UVB radiation, and thus, may be a potential candidate for the prevention of environmental stimuli-induced inflammatory skin damage.
Background and Objectives: The skin barrier protects skin against a harmful environment. Its function includes an antimicrobial barrier as well as a physical barrier. Stress is harmful to the skin barrier and there are many studies on this, but there are few about the effect of stress during pregnancy on the skin barrier of offspring. The aim of this study was to investigate the effect of stress during pregnancy on the skin barrier of offspring by examination with the naked eye, cortisol, weight, TEWL and histologic tests. Materials and Methods: Male hairless mice ten weeks old were coupled with females for three days. After twelve days the females were divided into two groups. We stressed one group and not the other group. The offspring from the non-stressed (N group) and stressed (S group) grew up until four weeks. The S group was exposed to chronic mild stress using the chronic stress model protocol modified method of Wilner and Towell for a week. We made examinations with the naked eye, assessment of cortisol, weight, TEWL, and histologic test to contrast the S group with the N group. Statistical analysis was performed by using Mann-Whitney and Friedman test. Statistical significance was achieved if the probability was less than 5% (p<0.05). Results: 1. By making an examination with the naked eye, S group showed papules, creases and dryness on their skin, but N showed no change compared with normal skin. 2. There was no statistically significant difference between S group and N group in cortisol (p>0.05). 3. There was no statistically significant difference between S group and N group in weight. N group had less weight than S group (p<0.05). 4. There was no statistically significant difference between S group and N group in TEWL (p>0.05). 5. There was no statistically significant difference between S group and N group in TEWL recovery (p>0.05). 6. In TEWL and TEWL recovery, there was statistically significant difference by time between S group and N group (p<0.005). Histologically, S group showed a thicker epidermis and epidermal crest than N group. Conclusions: We found that stress during pregnancy has negative effects on the skin barrier of offspring. Though there were no statistically significant differences between S group and N group in TEWL recovery, we found evidence that the recovery of skin barrier function is interrupted by stress during pregnancy by examination with both the naked eye and histology.
In order to develop optimal formulation and iontophoresis condition for the transdermal delivery of levodopa, we have evaluated the effect of two permeation enhancers, ethanol and oleic acid in microemulsion, on transdermal delivery of levodopa. In vitro flux studies were performed at $33^{\circ}C$, using side-by-side diffusion cell and full thickness hairless mouse skin. Current density applied was $0.4\;mA/cm^2$ and current was off after 6 hours application. Levodopa was analysed by HPLC at 280 nm. The o/w microemulsions of oleic acid in buffer solution (pH 2.5 & 4.5) were prepared using oleic acid, Tween 80 and ethanol. The existence of microemulsion regions were investigated in pseudo-ternary phase diagrams. Contrary to our expectation, cumulative amount of levodopa transported from microemulsion (pH 2.5) for 10 hours was similar to that from aqueous solution in all delivery methods (passive, anodal and cathodal). When pH of the micro-emulsion was pH 4.5, cumulative amount of levodopa transported for 10 hours increased about 40% (anodal) to 50% (cathodal), when compared to that from aqueous solution. Flux from pH 4.5 microemulsion showed higher value than that from pH 2.5 in all delivery methods. These results seem to indicate that electroosmosis plays more dominant role than electrorepulsion in the flux of levodopa at pH 2.5. The effect of ethanol on iontophoretic flux was studied using pH 2.5 phosphate buffer solution containing 3% or 5% (v/v) ethanol. Flux enhancement was observed in passive and anodal delivery as the concentration of the ethanol increased. Without ethanol, cathodal delivery showed higher flux than anodal delivery. Anodal delivery increased the cumulative amount of levodopa transported 1.6 fold by 5% ethanol after 10 hours. However, in cathodal delivery, no flux enhancement of levodopa was observed during current application and only marginal increase in cumulative amount transported after 10 hours was observed by 5% ethanol. These results seem to be related to the decrease in dielectric constant of the medium and the lipid extraction of the ethanol, which decrease the electroosmotic flow, and thus decrease the flux. Overall, the results provide important insights into the role of electroosmosis and electrorepulsion in the transport of levodopa through skin, and provide some useful informations for optimal formulation for levodopa.
본 연구에서 프로모터-리포터 분석방법을 통해 인슐린 유사 성장인자-1의 프로모터를 자극하는 천연물을 선별한 결과 사인 추출물이 가장 좋은 프로모터 자극효과를 나타냈으며, 무모생쥐에서 패쇄첩포 후 RT-PCR로 실험한 결과 IGF-1 mRNA를 $35\%$ 증가시키는 것으로 나타났다. 사인 추출물의 피부 주름개선 효과를 알아보기 위하여 인체 섬유아세포에서는 type-I collagen과 MMP-1 합성 변화를 관찰하였으며, 무모생쥐에서는 콜라젠의 증가와 진피 두께를 관찰하였다. 그 결과, 동위원소를 이용한 콜라젠 증가실험에서 type-I collagen은 $38\%$ 증가하였으며 무모생쥐에서 실시한 RT-PCR 결과에서는 mRNA가 $21\%$ 증가하는 것으로 관찰되었다. MMP-1 효소발현의 경우 ELISA 분석을 통해서 $63\%$의 높은 발현저해능을 확인하였고 Western blot에서도 발현이 저해되는 것을 확인하였다 추출물을 무모생쥐에 패쇄첩포 하였을 경우 대조군에 비해 진피 두께가 두꺼워지고 콜라젠 양도 증가되는 것으로 조직염색 관찰을 통해 확인하였다. 이상의 결과를 통해 사인추출물은 피부 주름개선에 좋은 효과를 나타내는 것으로 보이며, 여기에는 인슐린 유사성장 인자-I의 발현증가와 관련된 기전이 관여하는 것으로 판단된다.
지금까지 (-)-epigallocatechin-3-gallate(EGCG)는 인간의 피부에 유용한 녹차 카테킨 중에서 가장 강력한 항산화 성분으로 알려져 왔다. 본 연구팀은 용매, 온도, 압력 등 다양한 조건을 변화시키며, 멸균과정(autoclaving) 중에 발생하는 이성질체화(epimerization) 과정을 연구하여, gallocatechin-3-gallate(GCG) 함량이 크게 증가된 열전환-EGCG-복합체(heat-epimerzied-EGCG-mixture, HE-EGCG-mix)를 순수한 EGCG로부터 조제 하였다. 이러한 열전환-EGCG-복합체는 무모쥐 SKH-1을 이용한 실험에서, 손상된 피부 장벽의 회복 시에 인보루크린 7(involucrin 7) 단백질의 발현량을 EGCG 처리 시보다 증가시킴을 확인하였다. 또한, in vitro 실험을 통하여 GCG는 $PPAR-{\alpha}$에 대한 전이활성(transactivation) 효과가 EGCG보다 뛰어남을 확인하였다. 이러한 결과는 열전환-EGCG-복합체에 함유된 고함량의 GCG 성분에 의해서, 피부 장벽 손상 회복 시 PPAR에 의해 매개된 각질형성세포(keratinocyte)의 분화가 더욱 촉진될 수 있음을 암시한다. 따라서, EGCG의 C-2 에피머(epimer)인 GCG는 녹차 카테킨을 이용한 피부 장벽 개선 용도의 화장품과 건강식품 개발 시 주요 소재로 활용될 수 있다.
These studies were designed to determine the effect of hydroalcoholic gel system (lower alkanol concentration: 40-60%) compared to general hydrogel system (lower alkanol concentration: 10-35%) on transdermal delivery of piroxicam and its anti-inflammatory activity. Piroxicam was incorporated into a hydroalcoholic gel and a hydrogel containing polymers, solvents, and cosolvents. The pH of gel was about 6.3-7.3 and the solvent mixtures were composed of water and various concentrations of ethanol (35, 40, 50, and 60%). For the in vitro study, the skin permeation of piroxicam from the gel formulations was investigated using Franz modified diffusion cells fitted with hairless mouse skin. For the in vivo study, the anti-inflammatory activity of hydroalcoholic gel was compared to other commercial products (piroxicam hydrogel and ketoprofen hydrogel) in rat and human. The anti-inflammatory activity was determined using carrageenan induced foot edema model in rat. For the clinical study, it was evaluated from determining efficacy and acceptability with 98 patients suffering from musculoskeletal pain. A novel piroxicam hydroalcoholic gel was successfully formulated in the range of 40-50% of ethanol as solvent, more than 10% of propylene glycol, 5% of $Transcutol^{\circledR}$ and 1 % of benzyl alcohol. The skin permeation of piroxicam using hydroalcoholic gel system was greater than that of general hydrogel system $(flux\;:\;139.1-148.2\;{\mu}g/cm^2/hr\;vs.43.0-84.5 {\mu}g/cm^2/hr)$ in vitro. Background and Objectives: Surgical ablation of tumors in the oral cavity and the oropharynx results in a three dimensional defect because of the needs to resect the adjacent area for the surgical margin. Although a variety of techniques are available, radial forearm free flap has been known as an effective method for this defect, which offers a thin, pliable, and relatively hairless skin and a long vascular pedicle. We report the clinical results of our 54 consecutive radial forearm free flaps used for oral cavity and oropharynx cancers. Materials and Methods: We reviewed the medical records of patients who were offered intraoral reconstruction with a radial forearm free flap after ablative surgery for oral cavity and oropharyngeal cancers from August 1994 to February 2003 and analyzed surgical methods, flap survival rate, complication, and functional results. Among these, 20 cases were examined with modified barium swallow to evaluate postoperative swallowing function and other 8 cases with articulation and resonance test for speech. We examined recovery of sensation with two-point discrimination test in 15 cases who were offered sensate flaps. Results: The primary sites were as follows : mobile tongue (18), tonsil (17), floor of mouth (4), base of tongue (2), soft palate (2), retromolar trigone (3), buccal mucosa (1), oro-hypopharynx (6), and lower lip (1). The paddles of flaps were tailored in multilobed designs from oval shape to tetralobed design and in variable size according to the defects after ablation. This procedures resulted in satisfactory flap success rate (96.3%) and showed good swallowing function and social speech. Eight of 15 cases (53.3%) who had offered sensate flap showed recovery of sensation between 1 and 6 postoperative months (average 2.6 month). Conclusion: The reconstruction with radial forearm free flap might be an excellent method for the maximal functional results after ablative surgery of oral cavity and oropharyngeal cancers that results in multidimensional defect.
본 연구는 PFEG(글루코실세라마이드 4.0% 함유)가 저 미네랄 특수사료(HR-AD)를 섭취시킨 헤어리스 마우스에서 발생하는 피부장벽 붕괴에 대한 억제작용이 있는지를 확인하고자 하였다. 4주간의 투여를 통해 정상군(Rabo MR Stock)과 비교하여 대조군(HR-AD)에서는 경피 수분 손실량(TEWL)이 서서히 상승하여 피부장벽기능의 붕괴가 발생함과 동시에 표피 수분량 및 각층 수분량도 서서히 저하되는 것이 확인되었다. 육안적으로는 깊은 주름 형성이 확인되었으며 조직학적 소견에서는 표피의 비후와 각화 항진이 확인되었다. 그러나 PFEG를 섭취시킨 시험군에서는 대조군에서 유발되는 피부장벽기능 붕괴에 대한 억제 효과가 확인되었다. 즉, 글루코실세라마이드 0.01% 배합군 및 0.1% 배합군에서는 섭취 시작 2주일째 이후 야기되는 TEWL 상승을 완전히 억제하였다. 또한, 저하되는 표피 수분량, 각층 수분량에 대해서도 유의하게 억제하여 피부 보습 기능을 유지하고 있었다. 육안적으로도 깊은 주름 형성은 억제되었으며, 조직학적인 검토에서도 표피의 비후나 각화 항진은 확인되지 않았다. 이러한 결과는 HR-1 마우스의 피부 세라마이드 생산 개선에 기인한 결과라고 예상된다. 즉, HR-AD 섭취에 의해 HR-1 마우스는 세라마이드를 포함한 피부 지질의 함량 및 구성에 이상이 생기고, 피부 수분량이 급격히 줄어들어 피부 보습 능력이 저하된다. 그런데 PFEG의 혼합 투여는 손상된 피부의 세라마이드 함량 및 구성을 정상과 가깝게 만들어, 대조군에서 나타나는 증상의 발현을 억제하는 것으로 판단된다. 이상의 결과로부터 PFEG의 섭취는 피부장벽기능의 손상으로 발생하는 다양한 피부 수분 손실을 억제함으로써 피부 보습 개선에 우수한 효능이 있음을 알 수 있었다. 따라서 PFEG가 피부 보습에 도움을 주는 건강기능성 원료로서 이용 가능성이 높음을 확인할 수 있다.
본 연구는 포도수확 후 버려지는 가지를 이용한 포도전정가지 추출물(Grape Pruning Stem Extracts, GPSE)에 함유된 polyphenol류 중 항산화, 항염증, 항암 등의 효과가 있다고 알려진 rutin, procyanidin B3, quercetin, kaempferol의 함량을 분석하고, UVB로 유도된 HR-1 mice의 손상된 피부에 대한 교원섬유, 탄력섬유 파괴억제 효과를 측정하여 기능성 화장품 소재로서의 응용 가능성을 확인하고자 하였다. 포도전정가지에서 polyphenol 성분을 80% EtOH로 추출하고 여과하여 농축한 후, 동결건조하여 $-20^{\circ}C$에 보관하면서 사용하였다. GPSE의 유효성분 함량은 HPLC를 사용하여 분석하였다. 피부손상을 유도하기 위해 UVB를 HR-1 hairless mice에 조사하였고, 손상된 피부에 교원섬유 파괴억제 효과를 보기 위해 Masson's trichrome stain, 탄력섬유 파괴 억제 효과를 보기 위해 Verhoeff's elastic fiber stain, MMP-9 단백 발현을 보기 위해 면역조직화학적 염색을 실시하였다. 본 연구에 사용된 포도전정가지 시료 53 kg에서 EtOH 분획 추출물 2.34 kg을 추출하여 4.42%의 수율 결과를 얻었다. 유효성분 분석결과, procyanidin B3, 0.28 mg/g, rutin 12.81 mg/g, quercetin 0.51 mg/g 및 kaempferol 8.24 mg/g로 나타났다. GPSE와 serum base를 혼합 도포한 그룹에서 UVB에 의한 교원섬유와 탄력섬유를 분해하는 MMP-9 단백발현 결과, 대조군 대비 평균 약 90% 감소 억제하는 효과를 볼 수 있었다. 교원섬유 파괴억제 효과는 모든 농도에서 평균 24%의 억제율과, GPSE와 serum base의 혼합물 농도 2,000 mg/kg 에서 43%로 가장 우수한 교원섬유 파괴억제효과가 나타났으며, 탄력섬유파괴 억제 효과는 모든 농도에서 평균 12%의 억제율과 GPSE와 serum base의 혼합물 농도 1,000 mg/kg에서 35%로 가장 우수한 탄력섬유 파괴 억제 효과가 있는 것으로 나타났다. 이 결과는 GPSE가 진피층의 세포 외 기질에 UVB 조사에 의해 파괴되는 교원섬유, 탄력섬유의 파괴를 억제하는 작용을 한다는 것을 알 수 있었다. 이상의 결과로 포도전정가지로부터 rutin, kaempferol, quercetin, procyanidin B3 등의 폴리페놀성 화합물이 화장품 기초 원료인 serum base에 함유토록 제조된 GPSE 는 교원섬유 파괴 억제, 탄력섬유파괴 억제효과 및 피부손상 기능개선 효과의 결과를 얻을 수 있었다. 본 연구의 의의는 지역포도농가에서 수확 후 버려지는 포도전정가지에서 폴리페놀류와 같은 유효성분을 확보하여 화장품 원료로서의 활용 가치가 높을 것으로 기대된다.

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
