Salmonella contamination in chicken meat was studied with 100 chicken meat samples purchased from 55 shops located in various regions. A total of 21 isolates of Salmonella enterica were isolated from 21 chicken meat samples from four shops located at open markets, whereas there were none from supermarkets with well-equipped cold systems. Among these, 18 isolates were identified as Salmonella enterica serotype Haardt (S. Haardt) and three isolates were S. enterica serotype Muenchen. When the minimal inhibitory concentrations of the S. Haardt isolates were assayed with the agar dilution method to determine susceptibility to ampicillin, chloramphenicol, sulfisoxazole, tetracycline, and nalidixic acid, all 18 isolates were resistant to tetracycline and nalidixic acid and nine of these were resistant to ampicillin. These isolates showed reduced susceptibility to eight fluoroquinolones including ciprofloxacin, enrofloxacin, levofloxacin, gatifloxacin, gemifloxacin, moxifloxacin, norfloxacin, and ofloxacin. When quinolone resistance determining regions of gyrA and gyrB were sequenced, every isolate had the same missense mutation Ser83$\rightarrow$Tyr (TCC$\rightarrow$+TAC) in gyrA, whereas no mutation was found in gyrB. Pulsed-field gel electrophoresis with XbaI revealed a close relationship among these isolates, suggesting a contamination of raw chicken meat with clonal spread of nalidixic acid-resistant and quinolone-reduced susceptibility S. Haardt in chickens. Results in this study show the importance of a well-equipped cold system and the prudent use of fluoroquinolone in chickens to prevent the occurrence of quinolone-resistant isolates.
At the present, fish farms are suffering a lot of economic losses due to infectious diseases caused by various pathogens including aeromonad. Aeromonad is ubiquitous bacteria that causes infectious diseases. At least 26 species in the genus Aeromonas have been reported to cause fatal infections not only in salmonid fishes, but also in other freshwater and seawater fishes. Molecular techniques based on nucleic acid sequences of 16S rDNA and housekeeping genes can be used to identify the Aeromonas species. In this study, The genus Aeromonas was isolated from salmonid fishes of sixteen fish farms in Gangwon-Do, Korea and phylogenetically identified based on the sequences of 16S rDNA and housekeeping genes for Aeromonad, i.e. RNA polymerase sigma factor ${\sigma}^{70}$ (rpoD) or DNA gyrase subunit B (gyrB). Consequently, 96 strains were collected from Atlantic salmon (Salmo salar), coho salmon (Oncorhynchus kisutch), masou salmon (Oncorhynchus masou) and rainbow trout (Oncorhynchus mykiss), and 36 isolates were identified as the genus Aeromonas by 16S rDNA analysis. Thirty six Aeromonad isolates were further analysed based on rpoD or gyrB gene sequences and found Aeromonas salmonicida (24 isolates), A. sobria (10 isolates), A. media (1 isolates) and A. popoffii (1 isolates), indicating that A. salmonicida is a main infectious bacteria in Salmonid fishes in Gangwon-Do. It was also proved that the phylogenetic identification of Aeromonas species based on the sequences of housekeeping gene is more precise than the 16S rDNA sequence.
Rice blast, caused by Magnaporthe oryzae, is one of the most destructive rice diseases worldwide. The aim of this study was to screen bacterial isolates to efficiently prevent the occurrence of rice blast. A total of 232 bacterial isolates were extracted from nonrhizospheric rice soil and were screened for antifungal activity against M. oryzae using a leaf segment assay. Strains S170 and S9 showed significant antagonistic activity against M. oryzae in vitro and in leaf disk assays, and controlled M. oryzae infection under greenhouse conditions. The results showed that strains S170 and S9 could effectively control rice leaf blast and panicle neck blast after five spray treatments in field. This suggested that the bacterial strains S170 and S9 were valuable and promising for the biocontrol of rice disease caused by M. oryzae. Based on 16S rDNA, and gyrA and gyrB gene sequence analyses, S170 and S9 were identified as Bacillus amyloliquefaciens and B. pumilus, respectively. The research also demonstrated that B. amyloliquefaciens S170 and B. pumilus S9 could colonize rice plants to prevent pathogenic infection and evidently suppressed plant disease caused by 11 other plant pathogenic fungi. This is the first study to demonstrate that B. amyloliquefaciens and B. pumilus isolated from nonrhizospheric rice soil are capable of recolonizing internal rice stem tissues.

Polarization of the light reflected off the Moon provides information on the size and composition of the particles in the lunar regolith. The mean particle size of the regolith can be estimated from the combination of the albedo and degree of polarization, while the color ratio of the parallel-component polarization (CP) has been suggested to be related to the amount of nanophase metallic iron (npFe^0) inside the regolith particles. Both the mean size and npFe^0 abundance of the particles have been used as maturity indicators of the regolith since sustained impacts of high energy particles and micro-meteoroids cause comminution of particles and production of npFe^0. Based on our multispectral polarimetric observations of the whole near side of the Moon in the U, B, V, R, and I bands, we compare the maps of the mean particle size, CP, and the optical maturity (OM). We find that the mean particle size map is sensitive to the most immature (~0.1 Gyr) soil, the OP map to the intermediate immaturity (a few 0.1 Gyr) soil, and the CP map to the least immature (~1 Gyr) soil.
The enterotoxin production and antimicrobial susceptibility on hemolytic strains from stools of diarrheal pigs was investigated in this study. Through morphological observation, gyrB nucleotide sequence, and API kit analysis, the selected potential pathogenic strain BY06 was identified as Bacillus cereus. Because the characteristic of enterotoxin symptoms were widely caused by Bacillus cereus strains, a PCR test was carried out in order to check the enterotoxin genes (hblA) in this strain. According to the results, this strain was an enterotoxin positive strain containing the hblA gene. The minimum inhibitory concentrations of 10 different antimicrobial agents were screened by the agar dilution test, indicating that this strain was resistant to penicillin G and intermediate to erythromycin; however, it susceptible to cephalothin, vancomycin, clindamycin, fusidic acid, gentamicin, ciprofloxacin, tetracycline and rifampin. These results suggest that the B. cereus BY06 isolated from pig feces has a potential risk of producing enterotoxin and is resistant to penicillin G, but susceptible to various antimicrobial agents.
A bacterial strain capable of hydrolyzing xylan was isolated from fermented soybean paste obtained from a domestic Buddhist temple, using enrichment culture with rice straw as a carbon source. The isolate, named YB-1301, was identified as Bacillus safensis on the basis of its DNA gyrase subunit B gene (gyrB) sequence. The xylanase productivity of strain YB-1301 was drastically increased when it was grown in the presence of wheat bran or various xylans. In particular, the maximum xylanase productivity reached above 340 U/ml in the culture filtrate from LB broth supplemented with only birchwood xylan at shake-flask level. The xylanase production was significantly induced by xylans at the stationary growth phase in LB medium containing xylan, whereas only a small amount of xylanase was constitutively produced from cells grown in LB medium with no addition of xylan. Furthermore, xylanase biosynthesis was induced more rapidly by the enzymatically hydrolyzed products of xylan than by the non-hydrolyzed xylan. In addition, the xylanase in the culture filtrate of B. safensis YB-1301 was found to have optimal activity at 55℃ and pH 6.5–7.0.
Citrus blast caused by bacterium Pseudomonas syringae is a very important disease of citrus occuring in many areas of the world, but with few data about genetic structure of the pathogen involved. Considering the above fact, this study reports genetic characterization of 43 P. syringae isolates obtained from plant tissue displaying citrus blast symptoms on mandarin (Citrus reticulata) in Montenegro, using multilocus sequence analysis of gyrB, rpoD, and gap1 gene sequences. Gene sequences from a collection of 54 reference pathotype strains of P. syringae from the Plant Associated and Environmental Microbes Database (PAMDB) was used to establish a genetic relationship with our isolates obtained from mandarin. Phylogenetic analyses of gyrB, rpoD, and gap1 gene sequences showed that P. syringae pv. syringae causes citrus blast in mandarin in Montenegro, and belongs to genomospecies 1. Genetic homogeneity of isolates suggested that the Montenegrian population might be clonal which indicates a possible common source of infection. These findings may assist in further epidemiological studies of this pathogen and for determining mandarin breeding strategies for P. syringae control.
Bacterial leaf spot of Euphorbia pulcherrima has been reported in many countries. Characterization by polyphasic approaches indicated that the isolates from India, USA and New Zealand could be distinguished based on rep-PCR profiles and gyrB phylogenies, while the Chinese isolates should be ascribed to Xanthomonas axonopodis pv. poinsettiicola.
We present $JHK_S$ near-infrared CCD photometric study for the Galactic open clusters NGC 1641 and NGC 2394. These clusters have never been studied before, and we provide, for the first time the cluster parameters; reddening, distance, metallicity and age. NGC 1641 is an old open cluster with age $1.6{\pm}0.2$ Gyr, metallicity $[Fe/H]=0.0{\pm}0.2$ dex, distance modulus$(m-M)_0=10.4{\pm}0.3\;mag(d=1.2{\pm}02\;kpc)$, and reddening $E(B-V)=0.10{\pm}0.05$ mag. The parameters for the other old open cluster NGC 2394 are estimated to be $age=1.1{\pm}0.2$ Gyr, $[Fe/H]=0.0{\pm}0.2$ dex, $(m-M)_0=9.1{\pm}0.4\;mag(d=660{\pm}120\;pc)$, and $E(B-V)=0.05{\pm}0.10$ mag. The metallicities and distance values for these two old open clusters are consistent with the relation between the metallicities and the Galactocentric distances of other old open clusters. We find the metallicity gradient of 53 old open clusters including NGC 1641 and NGC 2394 to be ${\Delta}[Fe/H]/{\Delta}R_{gc}=-0.067{\pm}0.009\;dex\;kpc^{-1}$.
보현산천문대 1.8 m 망원경과 2K CCD를 이용하여 2002년 4월과 2003년 5월에 중원소 함량이 아주 적은 구상성단 M53(NGC 5024)과 M92(NGC 6341)에 대하여 BVI CCD 측광관측을 수행하였다. 구상성단 M53과 M92의 정밀한 상대 나이 측정을 위하여 M53의 측광관측 자료에 대해 조동환과 이상각이 2007년 출간한 구상성단 M15(NGC 7078)와 M92를 대상으로 수행한 측광연구 논문에서 M92의 측광관측 자료를 분석할 때 적용한 꼭 같은 방식으로 전처리, PSF 측광, 표준화 등의 자료 분석을 수행하였다. 그리고 구상성단 M53의 V 대 BV, V 대 V-I, 그리고 V 대 B-I 색-등급도를 제시하였다. 구상성단 M53과 M92의 상대 나 이는 ${\Delta}$(B-V) 방법을 이용하여 도출하였다. 구상성단 M53과 M92 사이의 상대 나이 비교에서 M92의 절대 나이를 14 Gyr로 취할 경우 M53의 상대 나이가 M92의 상대 나이보다 $1.1-2.6{\pm}0.9$ Gyr 적은 것으로 유도하였다. 구상성단 M53과 M92의 이 상대 나이 차이는 M53과 M92의 약간 다른 수평계열 형태 차이를 유발했을 것으로 추정한다.
