Lactobacillus acidophilus is known to have an inhibitory activity on growth of Helicobacter pylori and this activity has been attributed to lactic acid and antibacterial agents produced by Lactobacillus. Since every Lactobacilli produces lactic acid, an another factor must exist for L. acidophilus to inhibit H. pylori growth. In this work, the inhibitory activity of L. acidophilus on H. pylori adherence was studied. An immunoabsorbent assay using TLC plate was developed and used for screening the inhibitory activity of various Lactobacilli on H. pylori adherence. Glycolipid, the attachment site for H. pylori, was isolated from blood type O red blood cells and spotted on a TLC plate. The H. pylori adherence increased linearly with increasing amounts of glycolipid spotted on the TLC plate. Various L. acidophilus strains, but not L. casei, appeared to inhibit H. pylori adherence to glycolipid, and the adherence decreased linearly as the concentration of the Lactobacillus increased. The results show that the inhibitory activity of L. acidophilus on H. pylori adherence is an another factor for L. acidophilus to inhibit H. pylori growth.
Purpose : In this study, to prove the antibacterial effect of Disocorea batatas, which is widely used for food, and to confirm the growth inhibitory effect, the antibacterial activity against L. gasseri, S. mutans, and P. gingivalis was verified. Based on this, it is intended to verify the utility as a preventive and therapeutic composition for dental caries and periodontal disease. Methods : RAW 264.7 cells were used to verify the cell survival rate and NO (Nitric Oxide) inhibitory effect on Disocorea batatas ethanol extract (DBEE). In order to verify the antibacterial effect against L. gasseri, S. mutans, and P. gingivalis, concentrations of 125, 250, and 500 ㎎/㎖ of DBEE were used and measured by the disk diffusion method. In order to confirm the growth inhibitory effect, the absorbance was measured at 600 ㎚ at 3, 6, 12, 18, and 24 hours using the liquid medium dilution method, and the growth inhibitory effect was measured compared to the control group. Results : The cell viability for DBEE was 91 % at 50 ㎎/㎖, and there was no cytotoxicity. The NO production inhibitory effect was shown from 10 ㎍/ml, and the higher the concentration, the greater the inhibitory effect. As for the antimicrobial effect using the disk diffusion method, the higher the concentration, the higher the antibacterial effect. At 125 ㎎/㎖ and 250 ㎎/㎖, S. mutans and L. gasseri showed high antimicrobial activity, and at 500 ㎎/㎖, the antibacterial effect was higher in L. gasseri. The growth inhibitory effect in DBEE was concentration-dependent as the higher the concentration, the higher the growth inhibitory effect, and all of them began to show growth inhibitory effects from 6 hours. Conclusion : Considering that it is widely used as an edible and medicinal material, Disocorea batatas has shown the potential to be used as a substance to prevent and alleviate dental caries and periodontal diseases, and it is believed that further research can be applied to oral health care products.
Porcine colostrum and milk were separated into the acid-soluble and casein fractions by acidification followed by centrifuge. The acid-soluble fraction of porcine colostrum was further separated by liquid chromatography and anisotropic membrane filtration. Trypsin and chymotrypsin inhibitory capacity in porcine colostrum, milk and their components was determined by incubating bovine trypsin or chymotrypsin in a medium containing their corresponding substrates with or without addition of various amounts of porcine colostrum, porcine milk or their components. The inhibition of insulin-like growth factor I (IGF-I) and epidermal growth factor (EGF) degradation in pig small intestinal contents by porcine colostrum was measured by incubating iodinated IGF-I or EGF with the intestinal contents with or without addition of porcine colostrum. Degradation of labeled IGF-I or EGF was determined by monitoring the generation of radioactivity soluble in 30% trichloroacetic acid (TCA). The results showed that porcine colostrum had high levels of trypsin and chymotrypsin inhibitory activity and increased the stability of IGF-I and EGF in pig intestinal contents. The inhibitory activity declined rapidly during lactation. It was also found that trypsin and chymotrypsin inhibitory activity and the inhibition on IGF-I and EGF degradation in the acid-soluble fraction were higher than that in the casein fraction. Heat-resistance study indicated that trypsin inhibitors in porcine colostrum survived heat treatments of $100^{\circ}C$ water bath for up to 10 min, but exposure to boiling water bath for 30 min significantly decreased the inhibitory activity. Compared with the trypsin inhibitors, the chymotrypsin inhibitors were more heatsensitive. Separation of the acid-soluble fraction of porcine colostrum by liquid chromatography and anisotropic membrane filtration revealed that the trypsin and chymotrypsin inhibitory capacity was mainly due to a group of small proteins with molecular weight of 10,000-50,000. In conclusion, the present study confirmed the existence of high levels of protease inhibitors in porcine colostrum, and the inhibition of porcine colostrum on degradation of milk-borne growth factors in the pig small intestinal tract was demonstrated for the first time.
Growth inhibitory effects of ethanol extracts of green tea and pine needles on Bacillus stearothermophilus isolated from spoiled red bean paste were detected at concentrations higher than 750 ppm, and antimicrobial activity of pine needle extract was slightly higher than that of green tea exract. Growth inhibitory effect of pine needle extract in nutrient broth adjusted to pH 6.0, water-activity 0.92, and $45\;^{\circ}$Brix was observed at 500 ppm. These results indicated growth of B. stearothermophilus could be inhibited by adding pine needle and green tea extracts.
Methanol extracts of 94 Korean plants were screened for angiogenesis inhibitors using the tube-like formation assay of HUVEC (Human Umbilical Vein Endothelial Cell) and evaluated for growth inhibitory activity on A549 cells, human lung cancer cells. Extracts of Euphorbia sieboldiana, Adonis amurensis, and Anthriscus sylvestris showed antiangiogenic and growth inhibitory activity at $50\;{mu}g/ml$. Aristolochia manshuriens and Styrax obassia expressed antiangiogenic activity without growth inhibitory action.
송이의 공시균(KFR1432)과 실험실에서 자체 분리한 균(YxT, WxT, $\textrm{NK}_3\textrm{T}$)의 생장속도의 차이와 FE SEM으로 확인한 미세구조의 차이로 공시균과 자체 분리균이 서로 다른 균임을 확인할 수 있었다. Tyrosinase 활성 억제 실험을 위해 각각 균사체의 열수 추출물, 에탄올 추출물, 배양액을 hexane, chloroform, ethyl acetate로 분획하였다. 그 결과 모든 균의 ethyl acetate 분획층과 수층에서 tyrosinase 활성을 억제하는 것으로 나타났으며, KFR1432의 ethyl acetate 분획층과 YxT의 수층에서 억제율이 100%를 넘어 큰 저해 활성을 보였다. 또한 항산화제 처리 결과 ascorbic acid와 glutathion은 생장을 다소 증가시키거나 별 영향을 주지 못한 반면, tocopherol 의 경우 농도가 높아질수록 생장을 억제하는 것으로 나타났다. Tyrosinase 활성 억제에 있어서는 모든 항산화제 처리구에서 무처리구에 비해 높은 억제율을 보였다.
Classical dihydropyrrole[3,4-f]quinazoline antifolates 7,8 and 9, in which the tricyclic ring is structurally similar to the pteridine ring of $CH_2-THF(1)$, the cofactor of thymidylate synthase (TS), were synthesized, and their in vitro antitumor activity was evaluated by measuring the cell growth inhibitory activity against cancer cell lines. The target compounds were cytotoxic against CCRF-CEM, human T-cell acute lymphoblastic leukemia, with the cell growth inhibitory activity $(IC_{50})$ of $0.8{\sim}8.3\;{\mu}M$. Among the three compounds, 3-amino analog 7 was 10- and 3.5-fold more cytotoxic compared to the 3-methyl analogs 8 and 9, and its cytotoxicity was similar to that of the reference compound with the $IC_{50}$ value of $0.83\;{\mu}M$. This result was supposed as the consequence of the fact that dihydropyrroloquinazolinone ring with amino group was able to bind well in the active site of TS. In the case of 3-methyl analogs, analog 9, which has two-carbon bridge between the dihydropyrroloquinazolinone ring and benzoyl-L-glutamic acid, was 3-times more potent in cytotoxicity than analog 8 which has one-carbon bridge, and this result indicates that the distance and conformational orientation of the benzoyl-L-glutamic acid moiety with respect to the tricyclic ring may also be a crucial determinant of cell growth inhibitory activity.
We examined the effects of biological activity Phellinus linteus mycelium culture with cassiae semen extract. Firstly, the optimal temperature, initial pH and culture period for mycelial growth in a liquid culture of P. linteus were determined, and they were $30^{\circ}C$, pH 5.0 and 8 days respectively. The five herbal materials were examined against several health functional efficacies, and, as a result, Cassiae semen was chosen, with its superior inhibitory effects in ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities(95.3%, 80.9%, 96.1 and 24.2%, respectively). P. linteus fruit body was investigated on ${\beta}$-glucuronidase inhibitory activity, electron donating activity, ACE inhibitory, and ${\alpha}$-glucosidase inhibitory activities, and they were 54.7%, 81.9%, 30.0% and 20.1%, respectively. Accordingly, C. semen was used in the following experiment, to give an additive functional effect on the P. linteus. As the amount of C. semen in the cultural media increased, mycelial weight and ${\beta}$-glucan contents also increased, but final pH was not influenced. In addition, the ${\beta}$-glucuronidase inhibitory activity, electron donating activity, and ${\alpha}$-glucosidase inhibitory activity increased. P. linteus mycelium culture showed higher activities in the other three tests above, except for electron donating activity, when C. semen was added to the medium before cultivation.
Dandelions have been reported to have medicinal properties and bioactive components that impact human health. However, the precise biological properties of dandelions and the parts of the plants possessing bioactive components remain uncertain. In this study, we evaluated 3 different types of dandelions based on their cultivation origin (Songpa, Uiryung, and native Uiryung types) as well as their 4 different plant parts (leaf, flower, root, skin). Each sample was extracted with $80\%$ methanol and then compared for the biological activities (anti-oxidative, immune cell proliferative and tumor cell growth inhibitory activities). All 3 types of dandelions possessed a degree of biological functions including the hydroxyl radical scavenger activity, immune cell proliferative activity and tumor cell growth inhibitory activity. However, there was no significant difference in these activities between the 3 dandelion types. Leaves of all three dandelion types showed the highest levels of all biological activities. To a lesser degree, the flower and root parts displayed biological activities. In the skin parts, anti-oxidative activity was also detected only at higher doses of dandelion extracts. Heating the dandelion leaf extract did not affect the biological activity, suggesting a heat-stable nature of the biological compounds. Taken together, these collective data suggest that dandelions, in particular their leaves, possess a high concentration of heat-resistant biological compounds, which are responsible for anti-oxidative, immune cell proliferative and tumor cell growth-inhibitory activities.
Periconicin A and B, two new fusicoccane diterpenes originally isolated from the cultures of endophytic fungus Periconia sp. OBW-15, were tested by several biological assays. Periconicin A was consistently more active than periconicin B. In an antifungal activity assay, periconicin A showed potent inhibitory activity against the agents of human mycoses, including Candida albicans, Trichophyton mentagrophytes, and T. rubrum, with minimum inhibitory concentration (MIC) in the range of 3.12- 6.25 $\mug$ /ml. In a plant growth regulatory activity assay, periconicins inhibited hypocotyl elongation and root growth of Brassica campestris L. and Raphanus sativus L. At concentrations below 1 μg/ml, however, both compounds accelerated root growth by 110- 135%. From these results, it is apparent that a methyl group positioned in a cyclopentane ring may play an important role in plant and fungal growth inhibitory activity.
