Bacterial Diversity in the Initial Fermentation Stage of Korean and Chinese Kimchi (발효 초기 한국산 및 중국산 김치의 Bacteria 다양성 평가)

  • The purpose of this research is to draw the bacterial community difference between Korean and Chinese kimchi for future use in the confirmation of kimchi origin. Initial fermentation stage kimchi samples (above pH 5) were used for the analysis of bacterial diversity. From 26 Korean kimchi samples, 1,017 strains in the 45 genera and from 22 Chinese kimchi samples, 842 strains in the 54 genera were isolated with use of marine medium, nutrient medium, succinate minimal medium (SMM), leuconostocs selective medium (LUSM) agars. In the order of isolated numbers, Bacillus, Weissella, Leuconostoc, Pseudomonas, and Lactobacillus genera and Bacillus, Weissella, Lactobacillus, Pseudomonas, Serratia, and Enterobacter genera were predominated in Korean and Chines kimchi, respectively. Among the isolated lactic acid bacteria, Weissella spp. were isolated most dominantly owing to the biased growth of Weissella spp. on LUSM agar. Species in the genera Leuconostoc and Lactobacillus were the next frequently isolated LAB from Korean and Chinese kimchi, respectively. Weissella confusa was isolated only from Korean kimchi and W. soli and Serratia proteamculans were isolated only from Chinese kimchi. They have a possibility to be used as target bacteria to differentiate Korean kimchi from Chinese kimchi.

Effect of PVA-Encapsulation on Hydrogen Production and Bacterial Community Structure (수소 생산과 세균 군집구조에 미치는 PVA-포괄고정화의 영향)

  • In this study, the performances of PVA-encapsulation and non-encapsulation in a fed-batch bioreactor system were compared for biohydrogen production. Hydrogen production in the PVA-encapsulation bioreactor was not significantly different in comparison to the non-encapsulation bioreactor. However, the hydrogen gas in the encapsulation bioreactor could be stably produced when it was exposed to environmental difficulties such as pH impact by the accumulation of organic acids as fermentative metabolic products. Bacterial communities by DGGE analysis were differently shifted between the PVA-encapsulation and non-encapsulation bioreactors from the initial sludge. The community of hydrogen producing bacteria was stable during the experimental period in the PVA-encapsulation bioreactor compared to the non-encapsulation method. The absolute quantitation of the DNA copy number by a high-throughput droplet digital PCR system for six genera contributed to hydrogen production showing that the numbers of dominant bacteria existed at similar levels in the two bioreactors regardless of encapsulation. In both of two bioreactors, not only Clostridium and Enterobacter, which are known as anaerobic hydrogen producing bacteria, but also Firmicutes, Ruminococcus and Escherichia existed with $1{\times}10^5-1{\times}10^6$ copy numbers of ml-samples exhibiting rapid growth during the initial operation period.

  • A hemolysin producing bacterial strain which belong to Vibrio species was isolated from the Kum River estuary. In the process of identification, the strain did not show characteristics of known Vibrio species; thus, the strain was designated as Vibrio sp, E10 (V. kunsan) tentatively and further identification study was carried out by comparing its bacteriological characteristics. Morphologically Vibrio sp, E10 was comma shaped rod with a polar flagellium. Clear hemolysis zones were observed with the strain against human and sheep blood agar. Hemollytic toxicity was confirmed by strong vascular Permeability and fatal toxicity against mouse was also observed. Therefore the strain was a pathogenic vibrio. Growth conditions for Vibrio sp. E10 were ranged salinity of 0$\~$$4.5\%$, pH of 6.2$\~$9.2, temperature of 14$\~$42$^{\circ}C$, respectively, 16S rDNA partial sequence of Vibrio sp, E10 showed $99\%$ homology with dozens of V. cholerae species including V, cholerae El Tor N16961 and V, snmisnfus ATCC 33653T. This strain belonged to Proteobacteria; gamma subdivision; Vibrionacea: Vibrio. But, among knorn Vibrio species no identical styains were found when using automatic bacteria identification system ($MicroLog^{TM}$system, release 4.0, Biolog Inc., USA) which evaluated the ability of metabolizing 95 kinds of carbon and nitrogen sources. Vibrio sp, E10 showed 18 and 11 different responses as compared to V. mimicus and V, cholerae, respectively.

Case study of good soil management in plastic film-house cultivation (시설하우스 재배농가의 우수토양관리 사례연구)

  • Cultivation area of the plastic film-house has been continuously increased with the increase of consumers' income. Intensive land use without fallowing or crop rotation caused severe problem such as salt accumulation in soils and in turn retarded growth and low productivity. This study was carried out to solve them derived from longterm intensive farming practices. Seven farmers who are practicing plastic film-house cultivation were recommended for case study by municipal government and selected for their excellency of cultivation and soil management. The cultivation periods of these systems were in the range of 5 to 40 years in the regions mainly located in alluvial soil cultivated with cucumber, tomato and red pepper. The soils texture of the excellent farmers' fields were silt loam or sandy loam, ranged from 7 to 15 percents of clay contents. Soil bulk density, depth of plowing layer and soil aggregates contents of the farmers' soils were 0.89, 23.1 cm, 61.6% whereas those in neighboring soils were 1.10, 17.8 cm, 54.2 %, respectively. And pH, OM and $NO_3-N$ of the farmers' soils also were better than those of neighboring soils. There was no difference in population densities of nematode between the good farmers' and neighboring soils, but actinomyces and Fusarium densities of recommended farmers' soils were better than neighboring soils. The major farming practices by the good farmers were characterized by deep plowing with flooding, amendment of crude organic matter, and reduction of chemical fertilizer application before transplanting, and also drip irrigation and liquid manure application after planting. They also conducted solar sterilization with or without flooding, removal of plastic films during rainy days and culturing rice or corn as rotation crops to avoid the problems mentioned above.

Effect of Ammonium Thiosulfate on Biological Activity in a Paddy Soil (Ammonium Thisoulfate 처리가 담수(湛水) 답토양(畓土壤)의 생물활성(生物活性)에 미치는 영향)

  • The objective of this research was to examine the effect of ammonium thiosulfate(ATS) on urease activity and on biological and chemical properties of flooded paddy soil especially having high organic matter content by comparing with the effect of sodium thiosulfate(STS). The results obtained are summarized as follows. 1. The hydrolysis of urea was inhibited at 3 and 5 days after treatment of thiosulfate(ATS and STS) +glucose and thiosulfate only, respectively. The inhibitory effect of ATS on urea hydrolysis was slightly lower than that of STS in glusoce-added soils, but when the glucose was not added, the effects of ATS and STS were not different significantly. 2. The soil pH and Eh was lowered by 0.3~0.5 units and 30~120 mV, respectively, when incubated flooded soil with ATS and glucose at $25^{\circ}C$. 3. Soil respiration rate in/flooded soil was increased by 10~70% with the treatment of ATS during the 20 day experimental period. 4. The contents of acetic and butyric acid in thiosulfate treatment soil was below $10{\mu}g/g$, which was lower than that($220.3{\mu}g/g$) of critical growth inhibition of rice.

Isolation and Optimal Culture Conditions of Brevibacillus sp. KMU-391 against Black Root Pathogens Caused by Didymella bryoniae (덩굴마름병원균인 Didymella bryoniae의 생물학적 방제를 위한 길항세균의 분리 및 특성)

  • We isolated a bacterium which produces antifungal substances from the Sanktpeterburg soils at Russia. The iso-lated strain was identified as Brevibacillus sp. and shown a strong antifungal activity on plant pathogenic fungi. Brevibacillus sp. KMU-391 produced maximum level of antifungal substances under incubation aerobically at $30^{\circ}C$ for 48 hours in trypticase soybean broth containing 1.0% sucrose and 1.0% polypeptone at 180 rpm and initiated pH adjusted to 7.0. Precipitate of culture broth by $30{\sim}60%$ ammonium sulfate precipitation exhibited strong antifungal activity against Didymella bryoniae by dry cell weight. Butanol extract of cultured broth also shown fungal growth inhibitory activity against Botrytis cinerea KACC 40573, Botrytis fabae KACC 40962, Colletotrichum gloeosporioides KACC 40804, Colletotrichum orbiculare KACC 40808, Didymella bryoniae KACC 40669, Fusarium graminearum KACC 41040, Fusarium oxysporum KACC 40037, Fusarium oxysporum KACC 40052, Fusarium oxysporum f, sp. radicis-Iycopersici KACC 40537, Fusarium oxysporum KACC 40902, Monosporascus cannonballus KACC 40940, Phytophthora camvibora KACC 40160, Rhizoctonia solani AG-1(IA) KACC 40101, Rhizoctonia solani AG-4 KACC 40142, and Scleotinia scleotiorum KACC 41065 by agar diffusion method.

Application Potential of Hurdle Technology by Combination of Bacteriocin Produced by Lactobacillus brevis DK25 and Potassium Benzoate (Lactobacillus brevis DK25의 박테리오신과 안식향산칼륨과의 혼용에 의한 Hurdle Technology 적용 가능성)

  • Lactobacillus brevis DK25 isolated from Dongchimi was identified by physiological and biochemical tests and 16S rDNA sequence analysis. Bacteriocin of L. brevis DK25 exhibits inhibitory activity against Enterococcus faecalis and Listeria monocytogenes when using agar well diffusion method. Maximal production of bacteriocin was reached in the beginning of the stationary phase, and inhibitory activity declined after the late stationary phase. This result suggested that bacteriocin was produced in a growth-associated manner. Complete inactivation of bacteriocin activity was observed after treatment with protease, but the activity was stable between pH 4-9 and heat resistant (30 min at $100^{\circ}C$). Bacteriocin showed a concentration-dependent antimicrobial activity against L. monocytogenes KCTC 3569. Moreover, the application experiment showed that combination of bacteriocin (320 AU/ml) with potassium benzoate (0.05%) could significantly reduce the counts of L. monocytogenes KCTC 3569 in mayonnaise during storage at 4 or $25^{\circ}C$ for 10 days. Thus, bacteriocin from L. brevis DK25 may be used for hurdle technology by combination with potassium benzoate in order to increase pathogenic bacteria inactivation in food processing and food safety control.

The Environment-Friendly High Quality Pork Production by feeding the Fermentation Products of Natural Clay Mineral (천연의 점토광물질 발효산물 급여에 의한 친환경 고품질 원료돈육 생산)

  • We investigated the effect of the clay mineral fortified with complex trace elements by specific fermentation microbes on growth performance and a quality of pork in this study. For the declared experimental animal, 80 heads of crossbred with Large White and Yorkshire were randomly splitted into a control group and a test group. Each group were assigned with 4 replicates and 10 heads of each replicate. When 0.3% of the fermentation product of the clay mineral (FCM) was added into feed stuff and fed for 9 weeks of experimental feeding period, the results of the feed intake, weight gains, pH of carcass, lightness of meat color, and brightness of the muscle semimembranosus were similar to those of the control group. However, dressing percentage, reddishness of carcass, and water holding capacity were improved, respectively, as much as 2.7%, 12%, and 10%, and reduced by 6% in cooking loss. In order to investigate the immune modulatory effect of fermented clay mineral, pheripheral blood mononuclear cells (PBMC) were isolated and Immune modulatory parameters were measured. The proliferation activity of PBMC from pigs fed the fermented clay mineral were significantly increased compared to control group pigs, and also those results were more clearly observed as activated with lipopolysaccharide and concanavaline A. The secretion of TNF-${\alpha}$ of the FCM group pigs showed an increasing tendency. Therefore it was suggested that the feeding of FCM which was high in cation metathesis and the value of infrared ray, activated the immune responses, and thus the production of the environment-friendly high quality pork without the use of antibiotics would be possible.

The Effects of Freeze-Dried Potato Flour Addition on the Fermentation Characteristics of Yogurt (감마선 조사된 감자분말 첨가가 요구르트의 발효특성에 미치는 영향)

  • This study was performed to characterize the mechanism of the acceleration of lactic acid fermentation in milk incubated with potato flour irradiated by ${\gamma}$-ray and two types of lactobacillus, Lactobacillus bulgaricus and Streptococcus thermophillus, and to examine its effect on the quality of yogurt by assessing the acidity of lactic acid, viscosity, free amino acid, and sensory. In lactic acid fermented in the presence of 0.5% (w/w) potato flour, its acidity was higher, its pH was lower, and its viscosity was increased than lactic acid fermented in the absence of potato flour. In the sample fermented with the mixture of S. thermophillus and L. bulgaricus, the acid production rate was highest. In the samples fermented with a single type of bacteria, the acid production rate of S. thermophillus was higher than that the L. bulgaricus. Fermented a single type of bacteria with potato flour, the lysine utilization rate of L. bulgaricus and S. thermophillus was high. L. bulgaricus produced a large quantity of glutamic acid whereas S. thermophillus consumed glutamic acid. In sensory evaluation of yogurt fermented with potato flour, the preference of texture was improved while its color, taste, flavor, and overall preference were decreased. The data suggest that L. bulgaricus stimulates the growth of S. thermophillus by providing free glutamic acid that is required by S. thermophillus and thus the addition of potato flour shortens the fermentation period of yogurt.

Optimum Media of Sedum kamtschaticum for Pot Culture in an Ebb & Flow System (Ebb & Flow 시스템에서 기린초 분화재배에 적합한 배지 선발)

  • Sedum kamtschaticum has been used as one of the native valuable plants for ornamental purpose. This experiment was conducted to find out the optimum media for pot cultured Sedum kamtschaticum using peatmoss and coir-based mixtures. Four mixtures such as peatmoss:perlite(5:5, v/v), peatmoss:perlite(7:3, v/v), coir:perlite(5:5, v/v), and coir:perlite(7:3, v/v) were used. Plants were irrigated with a Sonneveld solution in an ebb & flow system. The content of T-N, K, and Ca in the peatmoss-based media was higher than those of the coir-based media. The concentration of T-N, K, and Ca in the shoots of Sedum kamtschaticum increased when plants were cultured with peatmoss-based media. In the peatmoss-based media, plant growth, such as number of branches, leaf length and shoot fresh and dry weights, were significantly increased than those of plant grown in the coir-based media. As the results, the optimum media for pot cultured Sedum kamtschaticum in an ebb & flow system was considered to be the peatmoss-based mixtures.
