Alcaligenes sp. GB-77 에 의한 Polyhydroxyalkanoic Acid의 생산 (Polyhydroxyalkanoic Acid Production by Alcaligenes sp. GB-77)

  • For polyhydroxyalkanoic acid (PHA) production, several microorganisms were isolated from sewage sludge. One of them, GB-77 strain, was chosen from its PHB/HV copolymer production on only fructose without cosubstrate. The isolated strain GB-77 was identified as the genus Alcaligenes. Optimal temperature and pH for cell growth were 36C and 6.8. Optimal medium composition was 10 g/l of fructose and 5 g/l of polypeptone, 1 $\times$ 10$^{-2}$M Na$^{2}$HP0$^{4}$, 1.3 $\times$ 10$^{-2}$M KH$^{2}$PO$^{4}$. To investigate the optimal condition for polyhydroxyalkanoic acid production two-stage culture technique was used; first stage for cell growth and second stage for PHA production on unbalanced growth conditions. Optimal conditions for high PHA production were C/N ratio 50, temperature 36$\circ$C and pH 6.8. To overcome fructose inhibition on cell growth, intermittent feeding fed-batch culture technique was used. Total cell concentration was 17.4 g/l with 9.1 g/l of PHA. The purified PHA was identified PHB/HV copolymer by NMR analysis.

한국산 도꼬마리 추출물의 항균효과 및 분리 정제

  • Antimicrobial activity of various extracts of Xanthium strumarium L. was tested against 25 strains of bacteria, yeast and fungus. The crude ethylacetate extract exhibited strong growth inhibition to the tested strains with the exception of partial Gram-negative bacteria. The property of antimicrobial compound was very stable under heat treatment at $120^{\circ}C$, but it was unstable in acid (pH 3.0) and alkali (pH 10.0) treatment. The antimicrobial compounds were purified by boiling water extraction, ethylacetate extraction, charcoal column chromatography, silica gel column chro- matography and reverse phase HPLC. The purified compound A and B were detected in a single peak (each above 98% purity) through the HPLC analysis. The compound A and B showed a strong growth inhibition against Gram-negative and positive bacteria in the agar diffusion method. When tested by the FDA method using the esterase, compound A mainly inhibited the growth of bacteria and compound B showed the growth inhibition of both bacteria and yeasts.

쑥의 분획추출물들이 주요 장내세균의 in vitro 생육에 미치는 영향 (Effect of the Fractionated Extracts of Mugwort on the in vitro Growth of Some Intestinal Microorganisms)

  • Mugwort was successively fractionated with n-hexane, chloroform, ethyl acetate, n-butanol and water and the fractions were evaluated by their growth-promoting activites for Bifidobacterium sp. in vitro experiments. The growths of Bifidobacterium adolescentis, B. bifidum, B. infantis and B.longum were enhanced with the addition of the water fraction, while the fractions of chloroform and ethylacetate inhibited Clostridium perfringens. When the wate fraction was added to media at a concentration of 0.01-0.5%(w/v), the growhts of Bifidobacterium sp. were increased according to the concentration of water fraction used. The water fraction stimulated also the growth of lactobacillus acidophillus, whereas those of E. coli and Enterococcus faecalis were not affected. The growth-promoting activity of water fraction was stable at the range of pH 2 to pH 10 and kept in thermal treatment at 10$0^{\circ}C$ for 30 minutes.

산패유를 이용한 느타리버섯 균사체의 생산 (Production of Pleurotus spp. Mycelium Using Rancid Frying Oils)

  • Conditions for the culture of Pleurotus spp. mycelium using rancid frying oils were investigated. Among the six strains tested, Pleurotus ostreatus CBS 03 showed the greatest mycelial growth on fish paste and ramyon frying oil, and was used in this study. The optimum temperature and pH for mycelial growth were from 25 to $30^{\circ}C$ and pH 5.5 to 6.0, respectively. Tryptone for mycelial growth was better than any other nitrogen sources. The addition of $KH_2PO_4 and MgSO_4$ enhanced mycelial growth at 0.2 and 0.01% on fish frying oil, and at 0.1 and 0.03% on ramyon frying oil. Among the vitamins used, thiamine and nicotinic acid were the most effective ones.

Culture Conditions and Growth Characteristics of Bifidobacterium longum

  • A simple and low-cost medium was developed for the growth of Bifidobacterium longum KFRI 977. Of three bifidobacterial strains, B. longum KFRI 977 (ATCC 15707) showed the best growth in MRSC broth containing 0.3% oxgall, grew well in partially anaerobic condition, exhibited highest $\beta$-galactosidase activity, and was inhibitory against Clostridium perfringens KFRI 434. Of three developed media, the population of B. longum KFRI 977 was highest (1.9$\times$$10^9$/ml) in ISP based medium. The composition of ISP based medium is ISP (5%), glucose (1%), L-cysteine HCI (0.05%), Trypticase peptone (0.5%), yeast extract (0.5%), $MgSO_4$ (0.05%), Tween-80 (0.1%), and phosphate buffer (pH 7.0). Hydrolysis of ISP by Protease A was unnecessary, and the use of phosphate buffer (pH 7.0) prevented the formation of protein precipitate. Associative culture of B. longum KFRI 977 with Lactobacillus acidophilus KFRI 233 was proven to be deleterous to the growth of B. longum KFRI 977.

Effect of Glycine on the Growth of Leuconostoc mesenteroides and Lactobacillus plantarum in Kimchi Fermentation

  • This study was aimed to investigate the effect of glycine supplementation on the growth of Leuconostoc mesenteroides and Lactobacillus plantarum during kimchi fermentation. As preliminary experiment, the effect of supplementation of glycine (0-2.0%, w/v) to MRS medium on the growth of Leuc. mesenteroides and L. plantarum was evaluated. At 2.0% glycine in the MRS medium, cell growth rate was inhibited by 79% for L. plantarum and 27% for Leuc. mesenteroides. Subsequently, different concentrations of glycine (0, 0.5, and 2.0%, w/v) were applied for kimchi fermentation for 21 days, at $5^{\circ}C$. At day 14, the pH and titratable acidity (TA) of kimchi supplemented with 2.0% glycine were 4.83 and 0.38%, respectively, whilst the control kimchi had a pH of 4.49 and TA of 0.44%, respectively. The ratio of Leuconostoc/ Lactobacillus in kimchi increased as the concentration of glycine increased. The results show that the presence of glycine affected the growth of the 2 lactic acid bacteria, particularly of L. plantarum.

환경인자와 중금속이 Alcaligenes sp.의 생장과 인 제거에 대한 영향 (Effects of Environmental Factors and Heavy Metals on the Growth and Phosphorus Removal of Alcaligenes sp.)

  • Alcaligenes sp.의 인 제거에 대한 환경인자로 각각 온도, pH, 탄소원의 효과를 알아본 결과 $25^{\circ}C$, pH 7, glucose+acetate이 탄소원일 때 인 제거효율이 가장 좋은 것으로 확인 할 수 있었다. 5가지 중금속의 독성효과에 따른 Alcaligenes sp.의 생장은 중금속의 농도가 높아질수록 억제되었으며, $IC_{50}$값은 구리 5.03 mg/L, 카드뮴 0.08 mg/L, 아연 0.73 mg/L, 비소 282.20 mg/L, 니켈 4.74 mg/L로 나타났다. 모든 중금속의 농도가 높아질수록 생장이 억제되어 인 제거 역시 억제되는 것을 확인 할 수 있었다.

  • Temperature and pH conditions were studied for an effective biosurfactant production by Pseudomonas aeruginosa YPJ-80. Efficient methods of biosurfactant separation were also investigated. pH-uncontrolled experiments at 35$^{\circ}C$ and an initial pH of 8 resulted in the best cell growth (3.6 g/L) and biosurfactant production (0.073 g biosurfactant/g cell). Biosurfactant separation was most efficient using solvent extraction with chloroform/methanol (2:1 vol%) followed by acidification using 1N HCl.

  • To increase the level of $CoQ_{10}$ production in mass culture, the effects of pH and light irradiation on $CoQ_{10}$ production by Rhodobacter sphaeroides were investigated in a 1-L bioreactor. $CoQ_{10}$ production was growth-associated, and the highest production of $CoQ_{10}$ (1.69 mg/g dry cell) was obtained under uncontrolled pH: this production was 1.7 times higher than that obtained at controlled pH 7. Therefore, pH was a key factor affecting $CoQ_{10}$ production. The effect of light irradiation on $CoQ_{10}$ production was negligible. This result offers an advantage for mass production of $CoQ_{10}$.

Effects of Light on Production of Toxins by Helminthosporium victoriae and H. carbonum

  • Helminthosporium vicotoriae and H. carbonum were grown under fluorescent plus incandescent lights, or in darkness, with several different temperature regimes. There was little or no effect of light on toxin production by H. victoriae. Light-grown cultures of H. carbonum had significantly higher titres of host-specific toxin than did dark-grown cultures. Light had no significant effect on growth of either fungus. Maximum titres of host-specific toxins from both fungi were evident at the time maximum growth was reached. Minimum pH values in Fries modified medium occurred at the time of, or slightly before maximum level of toxin was reached.
