Schizophyllum commune is an edible and medicinal mushroom widely distributed in the world. The optimal growth conditions for the mycelia of 10 strains of the fungus were investigated. The temperature suitable for the mycelial growth and density was obtained at $30{\sim}35^{\circ}C$. Among the tested conditions, the minimum mycelial growth was found at $15^{\circ}C$. In case of pH, the most favorable growth was found at pH 5. The results indicated that this mushroom well adapted to high temperature and low pH for its mycelial growth. Considering growth phenotype of mycelia, Hamada, Hennerberg, PDA and YM were the most suitable and Lilly, Glucose triptone, Glucose peptone and Hoppkins were the most unfavorable among tested media for the mycelial growth of S. commune. Out of tested carbon sources, dextrin and fructose were the most suitable and lactose, mannose and sorbitol were the unsuitable for the fungus. Compact mycelial density was obtained from most of the carbon sources. Among used nitrogen sources, calcium nitrate, potassium nitrate and alanine were the most appropriate and the most incompatible were ammonium phosphate, histidine, urea and arginine for mycelial growth of S. commune on the culture media. Calcium nitrate, histidine and potassium nitrate showed moderately thin or thin, and rest of nitrogen sources showed compact or moderately compact mycelial density.
The influences of glucose concentration, initial medium acidity (pH), and temperature on the growth and ethanol production of Saccharomyces cerevisiae NK28, which was isolated from kiwi fruit, were examined in shake flask cultures. The optimal glucose concentration, initial medium pH, and temperature for ethanol production were 200 g/l, pH 6.0, and $35^{\circ}C$, respectively. Under this growth condition, S. cerevisiae NK28 produced $98.9{\pm}5.67$ g/l ethanol in 24 h with a volumetric ethanol production rate of $4.12{\pm}0.24g/l{\cdot}h$. S. cerevisiae NK28 was more tolerant to heat and ethanol than laboratory strain S. cerevisiae BY4742, and its tolerance to ethanol and fermentation inhibitors was comparable to that of an ethanologen, S. cerevisiae D5A.
We investigated the effect of fish oil (FOS) on growth performance, ruminal metabolism and fatty acid composition and physical characteristics of longissimus muscle in 10 steers and 10 bulls of Korean cattle. Concentrates diet was supplemented with FOS at 5% of the diet. FOS contained 3.34% eicosapentaenoic acid (EPA) and 24.87% docosahexaenoic acid (DHA) of total fatty acids by weight. Average daily weight gain and feed efficiency were not affected (p>0.871) by FOS, but feed intake was decreased. FOS had lower (p<0.003) pH and higher (p<0.001) $NH_3$-N than that of control. There was a treatment effect (p<0.001) for serum cholesterol concentrations. FOS increased (p<0.009) concentrations of n-3 fatty acids, including linolenic, EPA and DHA in longissimus muscle. Physical traits were significantly (p<0.015) changed by feeding FOS except for pH and lightness (L). We concluded that the fatty acid composition and physical properties of the muscle in fattening Korean cattle can be altered by feeding 5% FOS.
This study pointed at isolation of bunker-C oil degrading bacteria and then estimation of it's degrading capability in environmental conditions. Degradation ratio of the excellent isolate was appeared to 40.5% and 44.7% when the oil was treated to 1% and 5%, respectively. The isolate was identified to Acinetobater calcoaceticus SEBCM. In pH test, high degrading effect was appeared to about 73% at pH 6 and pH 7, and low degrading ratio was 37% at pH 4. Its growth condition at temperature has not large variation in $15^{\circ}C\;{\sim}30^{\circ}C$, Quantity of nitrogen for it's good growth was ranged of $0.5\;g/L{\sim}2\;g/L$. As these results, we realized that this isolate have good activity when treated to $15\;{\sim}30^{\circ}C$ of temperature and $6{\sim}7$ of pH.
The effects of pH, sodium chloride and potassium sorbate on the germination of Bacillus cereus spores in the medium of cooked rice homogenate were studied. At the range of pH $4.5{\sim}10.0$, the germination of spores were observed. Germinated spores were reached to the number of $10^7/ml$ within 5 hours at $32^{\circ}C$ under the condition of pH 7.0, which was found as optimum pH of germination. In the range of sodium chloride $2{\sim}10\%$, the maximum growth were exhibited under $2\%$ concentration, while it proportionally decreased under the salinity condition higher than $5\%$. The growth of Bacillus cereus were inversely related to the concentration of potassium sorbate within the range of $0{\sim}0.2\%$. Maximum sporulation ratio was observed under the culturing condition: $10\%$ NaCl and $0.2\%$ potassium sorbate in the medium of cooked rice homogenate.
The measurement of pH in the rhizosphere soil was conducted to clarify whether the growth of pokeberry plants affect the acidity of rhizosphere soil in two environmentally contrasting area Ulsan and Chongju city. The rhizosphere pH between 5.25 and 5.33 was shown in the pokeberry stand at Mt. Boomo located at Chongju. The rhizosphere pH of pokeberry stands at Mt. Bongdae, Mt. Sinsun and Mt. Totchil was below 5.0, and did not differ with depth and distance from the main axis of root. At Mt. Bongdae, however, the pH in the rhizosphere soil was significantly changed with soil depths though that was not changed horizontally. The rhizosphere pH at top soil was lower than that at subsoil, which indicates the fact that soil acidification at Mt. Bongdae was not caused by pokeberry plants. Furthermore, the rhizosphere pH did not change with the growth of pokeberry plants. These results indicate that the hypothesis that pokeberry plants acidify local soil environment should be reconsidered.
To investigate clonal variations of recombinant Chinese hamster ovary(rCHO) clones in response to culture pH and temperature, serum-free suspension cultures of two antibody-producing CHO clones(clones A and B), which were isolated from the same parental clone by the limiting dilution method, were performed in a bioreactor at pH values in the range of 6.8-7.6, and two different temperatures, $33^{\circ}C\;and\;37^{\circ}C$. In regard to cell growth, clone A and clone B displayed similar responses to temperature, although their degree of response differed. In contrast, clones A and B displayed different responses to temperature in regard to antibody production. In the case of clone A, no significant increase in maximum antibody concentration was achieved by lowering the culture temperature. The maximum antibody concentration obtained at $33^{\circ}C$(pH 7.4) and $37^{\circ}C$(pH 7.0) were $82.0{\pm}2.6$ and $73.2{\pm}4.1{\mu}g/ml$, respectively. On the other hand, in the case of clone B, an approximately 2.5-fold increase in maximum antibody concentration was achieved by lowering the culture temperature. The enhanced maximum antibody concentration of clone B at $33^{\circ}C$($132.6{\pm}14.9{\mu}g/ml$ at pH 7.2) was due to not only enhanced specific antibody productivity but also to prolonged culture longevity. At $33^{\circ}C$, the culture longevity of clone A also improved, but not as much as that of clone B. Taken together, CHO clones derived from the same parental clone displayed quite different responses to culture temperature and pH with regards antibody production, suggesting that environmental parameters such as temperature and pH should be optimized for each CHO clone.
Pleurotus has increased rapidly production and consumption because of highly nutritional value, natural healthy food and so on. The basic studies for Pleurotus need for development of mushroom industry. This study was to investigate the cultural characteristics among 15 strains of 6 species and to analyze their phylogenetic relationships. The cultural characteristics were investigated by mycelial growth activity at different media, temperature and pH. The optimum media for mycelial growth were YM and MCM in most species. The optimum temperature for mycelial growth were $25^{\circ}C$ and $30^{\circ}C$. The optimum pH for mycelial growth were widly range from pH 5.1 to 7.4. Through the RFLP (restriction fragment length polymorphism) of IGS (intergenic spacer) I region in ribosomal DNA, it was analyzed phylogeny of interspecies and intraspecies. Each species was discriminated well as isolates within each species formed clade to be distinguished other species. P. florida was highly similar to P. floridanus, and P. flabellatus was P. cornucopiae. P. fuscus var. ferulae was highly similar to P. eryngii but discriminated different species in analysis of RFLP of IGS I region and showed different characteristics in mycelial culture. RFLP of IGS I region was useful of studying phylogenetic relationships of species and population.
This study was carried out to survey the effect of Jujube extract on the growth of 3 strains of lactic acid bacteria(LBA) starter cultures in the MRS broth by the addition of 0, 0.1, 0.5, 1 and 2% extract The pH, titratable acidity and OD of LAB were investigated in order to get fundamental knowledge for the development a new product. The effects of Jujube extract on the growth of LAB were variable depending upon the LAB species and concentrations of Jujube extract significantly (p<0.05). In the results, Jujube extract enhanced the acid production and propagation by the 3 LAB strains with increasing concentration of Jujube extracts until 2.0% added was to the MRS broth medium (p<0.05). Addition of Jujube extract markedly stimulated the acid production and propagation of Lactobacillus acidophilus, Streptococcus thermophilus and Bifidobacterium longum.
