성장정지효과에 의한 InGaAs/InP 양자우물구조의 Photoluminescence 특성 변화 (Effects of growth interruption on the photoluminescence characteristics of InGaAs/InP quantum wells)

  • 저압 MOCVD 방법을 이용하여 InGaAs/InP 양자우물구조를 성장하였다. 성장 정지 시간에 따른 photoluminescence특성의 변화를 통하여 계면구조를 분석하였다. InP표면을 $PH_3$ 분위기로, InGaAs표면을 $AsH_3$분위기로 유지하며 성장을 정지하는 경우에는 성장 정지 시간이 길어짐에 따라 불순물 유입에 의한 것으로 생각되는 PL반가폭의 증가를 관찰하였다. InP표면에 AsH3을 공급하는 경우에는 As-P교환에 의해 우물층 두께가 증가하여 PL피크가 저에너지로 이동하였고, 반가폭의 변화는 크지 않았다. 계면 양자우물구조를 형성하여 As-P 교환작용에 대해 조사하였고, 1-2monolayer가 InAs유효두께로 계산되었다. InGaAs 표면에 $PH_3$을 공급한 결과, PL피크가 고에너지로 이동하는 것을 관찰하였고 동시에 반가폭도 증가 하였다. 이는 메모리 효과에 의해 InP층으로 As침투를 억제하고, InGaAs표면에서의 국부적 인 As-P교환에 의한 것으로 생각된다.

배추김치의 숙성에 미치는 Mono Sodium Glutamate의 영향 (Effect of Mono Sodium Glutamate on the Fermentation of Korean Cabbage Kimchi)

  • Effects of various monosodium glutamate(MSG_ concentration (0-400mg%) on the quality of Korean cabbage kimchi during fermentation at 2$0^{\circ}C$ were investigated. The growth of lactic acid bacteria titratable acidity pH content of thiamine ascorbic acid $\beta$-carotene and sensory evaluation were observed in the presencve of MSG. Titratable acidity was increased in the kimchi contained MSG during the fermentation.l Wheeas the changes of pH in the kimchi contained MSG were similar to those in control, Maximum growth of lactic acid bacteria was observed after 49 hour of fermentation in kimchi contained with 200mg MSG. The might be due to the fact that MSG was given optimum pH for acid bacteria growth because of its buffering action. Therefore the periods of kimchi fermentation were reduced by addition of MSG, MSG also affected on the stability of ascorbic acid thiamine and $\beta$-carotene. According of MSG. MSG also affected on the stability of ascorbic acid thiamine and $\beta$-carotene. According to sensory evaluation MSG reduced sour taste in kimchi which increased edible periods.

생물반응기에서 형질전환된 식물조직 배양을 위한 기초연구 (Basic Studies on Cultivation of Transformed Plant Tissue in Bioreactor)

  • Growth properties of carrot hairy roots transformed by Agrobacterium rhizogenes were compared in flask and bioreactor. Oxygen transfer coefficient KLa was measured during the cultivation in bioreactor. In flask cultures, initially sucrose 30g/l was nearly exhausted after 20days. pH was dropped from initially 5.8 to 4.79 after 4 days, but it is stable after that time. Finally, after 28 days, hairy roots were grown about twelve times. In view of the results studied optimum conditions, hairy roots were maintained high growth rates in sucrose 50g/l, pH 5.8, total nitrogen 60mM. Also in bioreactor cultures, fixed stainless sieve in bottom and aerated 0.31 vvm, the results of cultivation by the use of sucrose 50g/l had grown about twenty-eight times and pH variations were liked in flask. As a results, growth rate of 1.756g fresh weight/day/g inoculum in bioreactor were higher about three times than 0.57g fresh weight/day/g inoculum in flask culture. KLa values were showed a tendency to decrease from 0.209 min-1 to 0.068 min-1.

감자를 첨가한 김치의 발효 특성 및 항암효과 (Fermentation Properties and In vitro Anticancer Effect of Kimchi Prepared with Potato)

  • Potato kimchi, fermentation was carried out at $10^{\circ}$C for 15 days using various ratios of potato to kimchi (2.5%, 5%, 10%). The samples were determined according to the fermentation time, pH, acidity and growth of lactic acid bacteria in potato kimchi. The addition ratio of potato to kimchi had little effect on the pH, acidity or growth of lactic acid bacteria in potato kimchi. In comparison to baechu kimchi and mul-kimchi, the pH, acidity and growth of lactic acid bacteria was better in potato kimchi than in the other kimchi samples. The in vitro anticancer effect of potato kimchi was investigated using human cancer cells, AGS human gastric adenocarcinoma cells and HT-29 human colon adenocarcinoma cells. MTT assay revealed that the methanol extract of potato kimchi showed the highest anticarcinogenic effects.

홍국으로부터 불리한 Monascus 균주의 향균효과 (Antimicrobial Effect of Monascus Strains Isolated from Ang-Khak)

  • Total 29 Monascus strains were isolated from Ang-Khak and 4 of them were selecte based on the relative intensity of soluble red pigment and growth rate. The optimum growth temperature of the selected isolates was 32.5$^{\circ}C$ on malt extract agar(MEZ) plate. The optimum growth pH was 5.0 on czapek yeast extract agar plate, while it was pH 6.2 or 6.5 on MEA plate. Isolate No. 116, especially, showed the strongest animicrobial activity aganist Bacillus subtilis and Staphylococcus aureus but much less aganist Escherichia coli and Enterobacter aerogenes. The maximun antimicrobial activity of isolate No. 116 against St. aureus was achieved at initial pH 5.3 on rice extract broth. The acitivity was increased with increasing amount of culture broth concentrate of isolate No. 116.

Effect of Temperature, pH, and Media on the Mycelial Growth of Tuber koreanum

  • Members of the genus Tuber are ectomycorrhizal fungi; this genus includes more than 180 species worldwide. In the present study, the optimal pH, temperature, and medium suitable for the mycelial growth of the Korean truffle, Tuber koreanum, were determined. Mycelium of T. koreanum, isolated from fruiting bodies collected in Korea, was used to investigate the effects of these environmental factors. The results showed that malt extract agar and potato dextrose agar were the most suitable for the mycelial growth of T. koreanum when cultured at a pH of 6.0 at 25 ℃ for 30 days.

  • 고추 역병균(疫病菌)의 유주자낭(游走子囊) 발아억제(發芽抑制) 및 균사생장(菌絲生長)에 미치는 효과(效果)를 구명(究明)하기 위하여 $NH_4^+/ NH_3,NO_2^-/ NHO_2$$Ca^{+2}$을 in vitro에서$NH_4OH,KNO_2$$CaCl_2$에서 공시(供試)하여 수행한 결과는 다음과 같다. 1. $NH_4OH,KNO_2$$CaCl_2$로 조제(調劑)된 mol 용액(溶液)으로 처리(處理)한 결과(結果) 비(非)이온화(化)인$NH_3, NHO_2$와 이온화형(化型)인 $Ca^{+2}$ 농도(濃度)에 의해 고추 역병균(疫病菌)의 유주자낭(游走子囊) 발아(發芽)가 무처리구(無處理區)에 비(比)하여 $15{\sim}50%$의 억제효과(抑制效果)를 보였다. 2. pH와 mol농도(濃度)가 증가(增加)함에 따라서 $NH_3$의 농도(濃度)가 증가(增加)되었으며 반면에 $HNO_2$농도(濃度)는 pH 감소(減少)와 mol농도(濃度) 증가(增加)에 의하여 증가(增加)되었는데 동일(同一)한 농도(濃度)에서 $NH_3$는 pH6보다 pH8에서 유주자낭(游走子囊) 발아(發芽)에 더 저해적(沮害的)이였으나 반대로$HNO_2$는 pH8보다 pH6에서 더 억제(抑制)되었다. 3. $NH_3$$HNO_2$은 저농도(低濃度)에서 고농도(高濃度)로 될수록 유주자낭(游走子囊) 발아(發芽)가 더욱 억제(抑制)되는 경향을 보였다. 4. $Ca^{+2}$은 pH와 mol농도(濃度)가 증가(增加)됨에 따라 유주자낭(游走子囊) 발아(發芽)가 억제(抑制)되는 경향이었다. 5. 균사생장(菌絲生長)은 pH에 의하여 영향(影響)을 받았는데 특히 높은 PH에서 다소 억제(抑制)되었으나 $NH_3, HNO_2$$Ca^{+2}$의 농도(濃度)에는 아무런 영향(影響)이 미치지 많았다. 6. 이상(以上)의 결과(結果)로 보아 고추 재배(栽培)에 있어서 토양개량제(土壤改良劑)인 계분(鷄糞)같은 실소(室素)를 ,함유(含有)한 유기물(有機物)의 $NH_4^+/,NH_3,\;NO_2^-/HNO_2$$Ca^{+2}$은 고추의 역병균(疫炳菌)의 유주자낭(游走子囊) 발아(發芽)에 저해적(沮害的)으로 작용(作用)함으로 이는 실제로 토양(土壤) 개량제(改良劑) 처리(處理) 토양(土壤)에서 고추 역병균(疫病菌)의 발생억제현상(發生抑制現象)을 부분적(部分的)으로 나마 뒷받침하여 준다.

Biochemical Characteristics of Micrococcus varians, Staphylococcus carnosus and Staphylococcus xylosus and Their Growth on Chinese-Style Beaker Sausage

  • This study was conducted to investigate protein and carbohydrate utilization of Micrococcus varians, Staphylococcus carnosus and Staphylococcus xylosus. Sensitivity to pH, sodium chloride, potassium sorbate and sodium nitrite of these strains was also determined. In Chinese-style beaker sausage manufacturing, the growth rate of these strains during the curing period ($20^{\circ}C$ and $30^{\circ}C$) was evaluated. The results indicated that no strains could hydrolyze azo-casein and sarcoplasmic protein and only S. xylosus could hydrolyze gelatin at $30^{\circ}C$. All of these strains could oxidize and ferment fructose and mannitol. S. carnosus and S. xylosus could slightly oxidize lactose and utilize citrate. Arabinose was oxidized by S. xylosus and sorbitol was oxidized by S. carnosus. Growth of M. varians was restricted at pH 5.0 and S. carnosus and S. xylosus were restricted at pH 4.5. S. xylosus and S. carnosus were able to grow with 0.1~0.5% potassium sorbate, 50~200 ppm sodium nitrite or 1~15% sodium chloride. S. xylosus had a higher growth rate than the other strains. Staphylococcus species grew well during curing period of Chinese-style beaker sausage then followed by Micrococcaceae.

차가버섯의 균사체 성장에 대한 배지성분의 영향 (Effect of Medium Compositions on the Mycelial Growth of Inonotus obliquus)

  • 차가버섯(Inonotus obliquus) 균사체의 액체배양에서 온도($22-32^{\circ}C$)와 pH(5-7) 그리고 배지성분의 변화가 차가버섯 균사체의 성장에 미치는 연구를 수행하였다. Glucose, starch, peptone, yeast extract, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$ 그리고 $CaCl_2$를 각각 30-120 g/L, 0-10 g/L, 0-20 g/L, 0-15 g/L, 0-2 g/L, 0-1.5 g/L, 0-0.5 g/L의 범위로 변화시키면서 실험하였다. 차가버섯 균사체의 성장속도는 $26-27^{\circ}C$, pH 6, glucose 70 g/L, yeast extract 5 g/L 그리고 $CaCl_2$ 0.1 g/L일 때에 최대값을 나타났다. Starch와 peptone 그리고 $K_2HPO_4$의 농도가 증가할수록 차가버섯 균사체의 성장속도는 증가하였으나, $MgSO_4{\cdot}7H_2O$의 농도가 증가할수록 차가버섯 균사체의 성장속도는 감소하였다. 차가버섯 균사체의 최대 성장 속도를 위한 액체배지는 glucose 70 g/L, peptone 5-20 g/L, starch 10 g/L, yeast extract 5 g/L, $K_2HPO_4$ 2 g/L 그리고 $CaCl_2$ 0.1 g/L로 구성된다.

  • 예산근교의 하우스재배농가에서 상치, 쪽파, 고추에 대한 작항과 식물양분 및 토양화학성을 조사하였다. 토양중 Ca와 Mg가 낮고 K가 높은경우 상치에서 Ca결핍과 Mg부족증상을 보였다. 이 경우 pH는 낮고 EC는 상당히 낮었다. 쪽파는 출아가 안되거나 엽선단이 백색으로 고사하고 생육이 불량한데(50%) 높은 EC(0.5mmhos/cm 이상)와 낮은 pH(6.2이하)에 기인하는 것으로 나타났다. 고추는 시드름병을 보였는데 pH가 낮은것이(5.9이하) 원인으로 보였다.
