  • Geraniol (1), olivetol (2), cannabinoids (3 and 4) and 5-fluorouracil (5) were tested for their growth inhibitory effects against human oral epitheloid carcinoma cell lines (KB) and NIH 3T3 fibrobalsts using two different 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay and sulforhodamine B protein (SRB) assay. Cannabigerol (3) exhibited the highest growth-inhibitory activity against the cancer cell lines.

  • Polyacetylenes were isolated from Panax ginseng C.A. Meyer (Family Araliaceae), and their inhibitory effects on growth, infection and VacA vacuolation of Helicobacter pylori (HP) were investigated. Ginseng polyacetylenes did not inhibit the infection of HP into KATO cells. However, polyacetylenes inhibited HP growth and vacuolation of Hela by VacA toxin. Panaxytriol showed the most potent inhibition with $IC_{50}$ values of 0.05 and 0.046 mg/ml, respectively.

  • Inhibitory effects of the mushrooms on the growth and urease of Helicobacter pylori (HP), which is associated with human gastroduodenal diseases such as gastritis, peptic ulcer and gastric carcinoma, were investigated. Most of the mushroom extracts did not show inhibitory effect on HP urease except Coriolus versicolar, Auricularia auricular Sarcodon aspratus and Flammulina velutipes. The extract of Ganoderma lucidum, Coriolus versicolar, Gyropora esculenta and Agaricus bisporus var. albidus inhibited the growth of HP. When their extracts were fractionated, the ether fraction of Ganoderma lucidum and Agaricus bisporus var. albidus were the most effective. Among seven components separated from the ether fraction of G. lucidum extract by silica gel column chromatography, P3 was the most potent: MIC was $200{\mu}g/ml$. However, P3 did not inhibit the urease.

  • The recombinant human epidermal growth factor(DWP 401) was investigated on the pharrnacological actions. DWP 401 had no effects on the hexobarbital-induced sleeping time, locomotor activity, rotarod test, body temperature, analgesic action and anticonvulsant action in mice. It also had no influences on the isolated tracheal muscle and ileum of guinea-pig, isolated uterus and fundus strip of rats. Slight hypotensive action with effect on respiration was revealed at a dose of 8 g/kg i.v. of DWP 401 in rabbits. DWP 401 exhibited a weak inhibitory action of glucose tolerance in normal rats, significantly lowered the blood glucose contents in adrenalectomized rats at .a concentration of 160 g,/kg, and produced a significant inhibitory effect on leucocyte migration in CMC-pouch of rats at a concentration of 32 g/rat. Furthermore, DWP 401 showed a significant decrease on gastric juice volume and acidity. However. DWP 401 had no intestinal propulsion rate and influence on urine excretion.

Antifungal Activity of an Endophytic Fungus Aspergillus versicolor DYSJ3 from Aphanamixis grandifolia Blume against Colletotrichum musae

  • An endophytic fungus strain DYSJ3 was isolated from a stem of Aphanamixis grandifolia Blume, which was identified as Aspergillus versicolor based on the morphological characteristics, internal transcribed spacer (ITS) and calmodulin gene sequences analyses. A. versicolor DYSJ3 exhibited strong antagonistic activity against Colletotrichum musae, C. gloeosporioides and Fusarium oxysporum f. sp. cubense with the inhibition rates of 61.9, 51.2 and 55.3% respectively. The antifungal metabolites mainly existed in the mycelium of A. versicolor DYSJ3, and its mycelial crude extract (CE) had broad-spectrum antifungal activities against plant pathogenic fungi. The CE had a good thermal stability, and the inhibition rate of 100 mg/mL CE against C. musae was above 70.0% after disposing at 120 ℃ for 1 h. Five secondary metabolites were isolated from the CE and identified as averufanin, ergosterol peroxide, versicolorin B, averythrin and sterigmatocystin. Activity evaluation showed versicolorin B exhibited inhibitory effects on the mycelial growth and conidial germination of C. musae, and sterigmatocystin had a weak inhibitory effect on the mycelial growth of C. musae.

  • Panax ginseng C.A. Meyer (Family Araliaceae) was treated at low ($60^{\circ}C$, LT), mild ($100^{\circ}C$, MT) and high ($120^{\circ}C$, HT) temperatures, some components (panaxytriol, ginsenosides and polysaccharides) were isolated, and their inhibitory effects on growth, infection and VacA vacuolation of Helicobacter pylori (HP) were investigated. The molecular weights of polysaccharides were decreased according to the increasing processed temperature. Ginseng polysaccharides inhibited the HP infection into KATO III cells, but did not inhibit HP growth and VacA vacuolation of HeLa cells. (omitted)

  • Cannabigerol (1, CBG), methyl 4-[(2E)-3,7-dimethyl-2,6-octad ienyl)oxy]-3-methoxybenzoate (2, DTM), 5-fluorouracil (3, FU) as a reference, and cannabidiol (4, CBD) were tested for their growth inhibitory effects against KB(ATCC NO, OCL 17) cell lines using two different assays, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay and the sulforhod-amine B protein (SRB) assay. These compounds showed inhibitory activity in vitro in the micromolar range against KB cell lines. In general, the antitumor activities of these compounds (1, 2, 3 and 4) were dose-dependent over the micromolar concentration range of 1 to 100 M. The comparison of $IC_{50}$ values of these compounds in tumor cell lines showed that their susceptibility to these compounds decreases in the following order: DTM > CBD > 5-FU > CBG by MTT assay and DTM = CBD > 5-FU > CBG by SRB assay. CBG 1, DTM 2, 5-FU 3, and CBD 4 were tested for their cytotoxic effects on NIH 3T3 fibroblasts using two different assays, the MTT assay and SRB assay. These compounds exhibited potent cytotoxic activities in vitro in the micromolar range against NIH 3T3 fibroblasts. In general, the cytotoxic acivities of these compounds (1, 2, 3 and 4) were dose-dependent over the micromolar concentraion range of 1 to 100 M. The comparison of $CD_{50}$ values of these compounds in NIH 3T3 fibroblasts shows that their susceptibility to these compounds in decreases the following order(:) CBD > 5-FU > DTM > CBG by MTT assay, CBD > 5-FU > CBG > DTM by SRB assay. These results suggest that DTM 2 has the most growth-inhibitory activity against KB cell lines.

Inhibitory Effect of Ponciretin on Helicobacter pylori VacA Toxin-induced Vacuolation in HeLa Cells

  • The inhibitory effects of flavanone derivatives on Helicobacter pylori (HP) growth, infection and VacA toxininduced vacuolation were investigated. Among flavanones tested, ponciretin potently inhibited the growth of HP with a MIC value of 0.01 mg/ml and VacA toxin-induced vacuolation in HeLa cells with $IC_{50}$ value of 0.078 mM. However, other flavanones inhibited neither HP growth nor VacA toxininduced vacuolation. All flavanones tested did not inhibit HP infection to KATO III cells. Ponciretin also inhibited activation of procaspase-3 to caspase-3 in HeLa cell induced by HP VacA toxin, but did not affect Bax and Bcl-2 protein levels. These findings indicate that ponciretin inhibits growth as well as vacuolation by HP VacA toxin, which induces cell death via proteolytic activation of a cascade of caspases.

Inhibitory Effect of Snake Venom Toxin on Colorectal Cancer HCT116 Cells Growth through Induction of Intrinsic or Extrinsic Apoptosis

  • I investigated whether snake venom toxin(SVT) from Vipera lebetina turanica enhances the apoptosis ability of tumor necrosis factor(TNF)-related apoptosis-inducing ligand(TRAIL) in cancer cells. TRAIL inhibited HCT116 cell growth in a dose-dependent manner. Consistent with cell growth inhibition, the expression of TRAIL receptors; DR4 and DR5 was significantly increased as well as apoptosis related proteins such as cleaved caspase-3, 8, 9 and Bax. However, the expression of survival proteins(eg, cFLIP, survivin, XIAP and Bcl2) was suppressed by the combination treatment of SVT and TRAIL. Pretreatment with the reactive oxygen species(ROS) scavenger N-acetylcysteine reduced the SVT and TRAIL-induced upregulation of DR4 and DR5 expression and expression of the apoptosis related protein such as caspase-3 and-9 as well as cell growth inhibitory effects. The collective results suggest that SVT facilitates TRAIL-induced apoptosis in human colorectal cancer HCT116 cells through up-regulation of the TRAIL receptors; DR4 and DR5 via ROS pathway signals.

Effects of herbicide butachlor on Rhodospirillum rubrum KS-301 (Rhodospirillum rubrum KS-301에 미치는 제초제 butachlor의 영향)

  • The biololgical effect of the preemergence rice field herbicide, butachlor(commercial name, Machete) on purple nonsulfur photosynthetic bacterium Rhodospirillum rubrum KS-301 has been studied under cultural conditions. Bacterial growth showed a tendency to decline according to the degree of the concentration of butachlor until $10^{-3}$ M and slmost stopped at $10^{-2}$M. The growth inhibitory action at $10^{-3}$M of butachlor was evident (4.2-18.7% inhibition of growth rate) but had little effect in nitrogen fixation. Conversely, there was a little enhancement effect(1%) in pyruvate, dinitrogen gas growing cultures. At concentration of $10^{-3}$ M, instead of spiral form, rod shapes were observed through phase contrast microscope and instead of vesicular intracytoplasmic membrane, irregular tubular forms were observed through electron microscope. Alkaline pH value slightly reversed tha inhibitory action of butachlor.
