Persistence of Organophosphorus Insecticides in/on Mulberry Leaves with Reference to Silkworm Mortality (뽕잎중(中) 유기인계농약(有燐燐系農藥)의 잔류성(殘留性)과 잠독성(蠶毒性)에 관(關)한 연구(硏究))

  • The present study was aimed to investigate persistence of dichlorvos EC, fenitrothion EC, fenthion EC, and phenthoate EC in and on mulberry leaves with special reference to silkworm mortality and cocoon production under greenhouse condition. The halflives of fenitrothion, fenthion, and phenthoate in and on mulberry leaves were ranged from two to three days, while that of dichlorvos was less than 9 hours. The insecticide residues in and on mulberry leaves persisted longer in spring cropping season than in autumn cropping season. Elapsed periods from last application of each insecticide to leaf harvest for silkworm feed and maximum residue limits for safe cocoon production as well as relations between insecticide residues in and on mulberry leaves and silkworm mortality were produced.

Effects of dietary forage-to-concentrate ratio on nutrient digestibility and enteric methane production in growing goats (Capra hircus hircus) and Sika deer (Cervus nippon hortulorum)

  • Objective: Two experiments were conducted to determine the effects of forage-to-concentrate (F:C) ratio on the nutrient digestibility and enteric methane ($CH_4$) emission in growing goats and Sika deer. Methods: Three male growing goats (body weight $[BW]=19.0{\pm}0.7kg$) and three male growing deer ($BW=19.3{\pm}1.2kg$) were respectively allotted to a $3{\times}3$ Latin square design with an adaptation period of 7 d and a data collection period of 3 d. Respiration-metabolism chambers were used for measuring the enteric $CH_4$ emission. Treatments of low (25:75), moderate (50:50), and high (73:27) F:C ratios were given to both goats and Sika deer. Results: Dry matter (DM) and organic matter (OM) digestibility decreased linearly with increasing F:C ratio in both goats and Sika deer. In both goats and Sika deer, the $CH_4$ emissions expressed as g/d, g/kg $BW^{0.75}$, % of gross energy intake, g/kg DM intake (DMI), and g/kg OM intake (OMI) decreased linearly as the F:C ratio increased, however, the $CH_4$ emissions expressed as g/kg digested DMI and OMI were not affected by the F:C ratio. Eight equations were derived for predicting the enteric $CH_4$ emission from goats and Sika deer. For goat, equation 1 was found to be of the highest accuracy: $CH_4(g/d)=3.36+4.71{\times}DMI(kg/d)-0.0036{\times}neutral$ detergent fiber concentrate (NDFC,g/kg)+$0.01563{\times}dry$ matter digestibility (DMD,g/kg)-$0.0108{\times}neutral$ detergent fiber digestibility (NDFD, g/kg). For Sika deer, equation 5 was found to be of the highest accuracy: $CH_4(g/d)=66.3+27.7{\times}DMI(kg/d)-5.91{\times}NDFC(g/kg)-7.11{\times}DMD(g/kg)+0.0809{\times}NDFD(g/kg)$. Conclusion: Digested nutrient intake could be considered when determining the $CH_4$ generation factor in goats and Sika deer. Finally, the enteric $CH_4$ prediction model for goats and Sika deer were estimated.

An efficient protocol for the production of transgenic Alstroemeria plants via particle bombardment

  • Alstroemeria plants were transformed by using an improved particle-gun-mediated transformation system. Friable embryogenic callus (FEC) induced from the leaves with axil tissues of Alstroemeria plant was used as the target tissue. Also, FEC was transformed with the bar gene was used as a selectable marker. In the case of plasmid pAHC25, 7.5% of the twice-bombarded FEC clumps showed blue foci, whereas the clumps with single bombardment showed only 2.3%. Additionally, a 90° rotation with double bombardment led to a higher frequency (6 times) of luciferase gene expression in PBL9780 than the control treatment. After 8 weeks of bombardment, more than 60 independent transgenic lines were obtained for pAHC25 and nearly 150 independent transgenic lines were obtained for PBL9780, all of which were resistant to PPT and demonstrated either GUS or luciferase activity. Regarding effect of osmotic treatment (0.2 M mannitol) with 7 different periods, the highest transient gene expression was obtained in 8 h before and 16 h after transformation in both pAHC25 and PBL9780. Compared with the control, at least three times more GUS foci and photons were observed in this treatment. With respect to different combinations of mannitol and sorbitol with 8 h before and 16 h after transformation, high numbers of transient and stable transgene expressions were observed in both 0.2 M mannitol and 0.2 M sorbitol used in the osmotic pre-culture. This combination showed the highest transformation efficiency in both pAHC25 (8.5%) and PBL9780 (14.5%). In the control treatment, only 10% of the FEC clumps produced somatic embryos. However, by using 0.2 M mannitol and 0.2 M sorbitol, the frequency of somatic embryos increased to 36.5% (pAHC25) and 22.9% (PBL9780). Of the somatic embryos produced, at least 60% germinated. Approximately 100 somatic embryos from these 210 independent transgenic lines from 2 plasmids developed into shoots, which were then transferred to the greenhouse. PCR analysis confirmed the presence of the bar gene. This is the report on the production of transgenic Alstroemeria plants by using particle bombardment with a high efficiency, thereby providing a new alternative for the transferring of gene of interests in Alstroemeria in the breeding program in the future.

Use of Pellet or Cube-type Phenolic Foam as an Artificial Medium for Production of Tomato Plug Seedlings (토마토 플러그 묘 생산을 위한 펠릿 및 큐브형 phenolic foam 인공배지의 이용)

  • Growers in plug seedling production think that root media in which rockwool is a component has given rise to several environmental problems. Therefore, the demand for new materials as a substitute for rockwool has been increased. This study examined the possibility of cultivation of tomato plug seedlings using a newly developed growing medium with phenolic foam. Plug seeds of tomato cultivar 'Madison' were sown in four pellet-type growing media: Grodan rockwool (GRW), UR rockwool (URW), phenolic foam LC (LC) or phenolic foam LC-lite (LC-lite). Then, the seedlings were transplanted to the four cube-type growing media 19 days after sowing. Seeds were germinated in a growth chamber ($25{\pm}2^{\circ}C$, 80% relative humidity, and dark) for 4 days and then the seedlings were grown with a nutrient solution supplied by an overhead irrigation system in a greenhouse. Plant height, number of leaves, leaf area, and fresh or dry weight of tomato seedlings were the greatest for the seedlings transplanted to URW cube media after being grown on LC-lite pellets. Root grade was the greatest for the seedlings transplanted on LC or LC-lite cube media after being grown on LC pellets. Chemical properties of all media tested for tomato growth were maintained within a stable range, while physical properties of URW showed high values in container capacity, air space, and total porosity. These results demonstrated that the phenolic foam media were effective for seedling growth and can substitute for rockwool as a root medium.

Development of a Trial Product for Irrigation Management in Substrate Culture (고형배지경 급액관리 시작기 개발)

  • This experiment was carried out to commercialize an irrigation control system by finding out problems and solving them in application of a nutrient supply system through this experiment. Its efficiency had been tested through hydroponic cultivation of tomato and cucumber using this system in the farmer's plastic house (1-2W, 20a : Yanggyo-ri, Oseong-myeon Gyeonggi-do) from November. 2006, too. In the first cultivation, tomato seeds (cultivar Coco, Takii Seed Co. Japan) were sowed on November 8, 2006, and transplanted on January 8, 2007. and then, in the second, cucumber (Chuichong, Nongwoo Seed Co.) were cultivated in the same plastic house (sowing date : June 27, transplanting date : July 13). In the third, another cucumber cultivar (Jo-woon, Dongbu-hannong Seed Co.) were cultivated (sowing date : September 5, transplanting date : September 23). All of seedlings were transplanted on perlite bag ($W340{\times}L1,200{\times}H150mm$, 40L). By using this system, 971 boxes (5 kg/box) of tomato were produced and sold, and then total income was 5,466 thousand won per 10a. On the second cultivation, total amount of cucumber production was 489 boxes (50 ea/box), and total income was 7,380 thousand won. On the third cultivation, total amount of production was 67 boxes (100 ea/box), and total income was 1,854 thousand won. On the other hand, this system saved irrigated water by 50% ($4,000{\rightarrow}2,000L/10a/day$) in tomato cultivation, and by 44%($4,500{\rightarrow}2,500L/10a/day$) in cucumber cultivation. It also saved cost of nutrients by 50% in tomato ($1,648{\rightarrow}824thousand\;won/10a$), and 44% in cucumber ($1,648{\rightarrow}725thousand\;won/10a$). Furthermore this irrigation system maintained moisture content in perlite bag stable during cultivation period. Therefore, this system was successfully applied on farmer's greenhouse without a problem and can be commercialized for farmers.

  • Sustaining future wheat production is challenged by anthropogenically forced climate warming and drying led by increased concentration of greenhouse gases all around the globe. Warming stresses, originating from the elevated $CO_2$ concentration, are continuously reported to have negative impacts on wheat growth and yield. Yet, elevated $CO_2$ concentration, despite being disparagingly blamed for promoting warming, is also associated with a phenomenon called $CO_2$ enrichment; in which wheat yield can improve due to the enhanced photosynthesis rates and less water loss through transpiration. The conflicting nature of climate warming and $CO_2$ enrichment and their interplay can have specific implications under different environments. It is established form the field and simulation studies that the two contrasting phenomena would act severely in their own respect under arid and semi-arid environments. Wheat is a dietary staple for masses in Pakistan. The country's wheat production system is under constant stress to produce more from irrigated agricultural lands, primarily lying under arid to semi-arid environments, to meet the rapidly growing domestic needs. This work comprehensively examines the warming impacts over wheat yield and water productivity (WP), with and without the inclusion of $CO_2$ enrichment, under semi-arid environment of Punjab which is the largest agricultural province of Pakistan. Future wheat yields and WPs were simulated by FAO developed AquaCrop model v 5.0. The model was run using the bias-correction climate change projections up to 2080 under two representative concentration pathways (RCP) scenarios: 4.5 and 8.5. Wheat yield and WPs decreased without considering the $CO_2$ enrichment effects owing to the elevated irrigation demands and accelerated evapotranspiration rates. The results suggested that $CO_2$ enrichment could help maintain the current yield and WPs levels during the 2030s (2021-2050); however, it might not withhold the negative climate warming impacts during the 2060s (2051-2080). Furthermore, 10 - 20 day backward shift in sowing dates could also help ease the constraints imposed by climate warming over wheat yields and WPs. Although, $CO_2$ enrichment showed promises to counteract the adverse climate warming impacts but the interactions between climate warming and $CO_2$ concentrations were quite uncertain and required further examination.

A New Spray Chrysanthemum Cultivar, "Water Fog" with Single Type and White Petals for Cut Flower (절화용 백색 흩꽃 스프레이국화 "워트포그" 육성)

  • A New spary chrysanthemum (Dendranthema grandiflorum) cultivar 'Water Fog' was bred by open pollination of 'Angaesoguk'. Its characteristics were investigated three times from 2004 to 2006 under forcing culture in spring and retarding culture in autumn. The natural flowering time of 'Water Fog' was 24 October, but year-round production was possible by shading and lighting. It had flower diameter of 2.3 cm, 46.5 petals and 34.4 flowers per stem in autumn. The petal was narrow(0.4cm) and the color of petal was white with presence of green at flower center. Days to flower from short-day start for 'Water Fog' was about 48 in spring. 'Water Fog' showed the vase life of 22.6 days in autumn. This cultivar was registered for a commercialization in 2007. 'Water Fog' would be cultured under greenhouse and rain shelter condition in Korea.

A New Flowering, Spray Chrysanthemum Cultivar for Cut Flower, "Yellow Eye" with Single Type and Light Yellow Petals (황색 홑꽃인 조기개화성의 절화용 스프레이국화 "옐로우아이" 육성)

  • A new spary chrysanthemum (Dendranthema grandiflorum) cultivar 'Yellow Eye' was developed from a cross between 'Angela' and 'Angaesoguk' followed by selections of seedlings and lines at the Flower Research Institute, Gyeongnam Agricultural Research and Extension Services (ARES) from 2003 to 2007. Its characteristics were investigated three times from 2005 to 2007 under forcing culture in spring and retarding culture in autumn. The natural flowering time of 'Yellow Eye' was October 24th, but year-round production was possible by shading and lighting. Its flower was 3.3 cm in diameter, and had 31.7 petals and 27.9 flowers per stem in autumn. It has pollenless, single type flowers with yellow petals and green flower center. To flower in under the short day condition, for 'Yellow Eye' was about 41 days in spring, and 'Yellow Eye' showed a vase life of 23.1 days in autumn. This cultivar was registered for a commercialization in 2007. 'Yellow Eye' would be cultured under the greenhouse and rain shelter condition in Korea.

  • The proportion of farm households in the total population is decreasing every year. The aging of rural areas is expected to deepen. The aging of agriculture is continuing due to the aging of the aged population and the decline of the young population, and agricultural manpower shortage is emerging as a threat to agriculture and rural areas. The existing facility cultivation was concentrated on the production / yield per unit area. However, nowadays, not only production but also crop quality should be good so that the quality of crops must be improved because they can secure competitiveness in the market. Therefore, the government plans to increase the productivity by hi-techization of ICT infrastructure horticulture and to plan the dissemination of energy saving smart greenhouse. Therefore, it is necessary to develop a Smart Farm convergence service system based on a hybrid algorithm to enhance diversity and connectivity. Therefore, this study aims to develop smart farm convergence service system which collects data of growth environment of the rhizosphere environment of crops by wireless and monitor smartphone.

Enhanced Tolerance of Chinese Cabbage Seedlings Mediated by Bacillus aryabhattai H26-2 and B. siamensis H30-3 against High Temperature Stress and Fungal Infections

  • Two rhizobacteria Bacillus aryabhattai H26-2 and B. siamensis H30-3 were evaluated whether they are involved in stress tolerance against drought and high temperature as well as fungal infections in Chinese cabbage plants. Chinese cabbage seedlings cv. Ryeokgwang (spring cultivar) has shown better growth compared to cv. Buram-3-ho (autumn cultivar) under high temperature conditions in a greenhouse, whilst there was no difference in drought stress tolerance of the two cultivars. In vitro growth of B. aryabhattai H26-2 and B. siamensis H30-3 were differentially regulated under PEG 6000-induced drought stress at different growing temperatures (30, 40 and $50^{\circ}C$). Pretreatment with B. aryabhattai H26-2 and B. siamensis H30-3 enhanced the tolerance of Chinese cabbage seedlings to high temperature, but not to drought stress. It turns out that only B. siamensis H30-3 showed in vitro antifungal activities and in planta crop protection against two fungal pathogens Alternaria brassicicola and Colletotrichum higginsianum causing black spots and anthracnose on Chinese cabbage plants cv. Ryeokgwang, respectively. B. siamensis H30-3 brings several genes involved in production of cyclic lipopeptides in its genome and secreted hydrolytic enzymes like chitinase, protease and cellulase. B. siamensis H30-3 was found to produce siderophore, a high affinity iron-chelating compound. Expressions of BrChi1 and BrGST1 genes were up-regulated in Chinese cabbage leaves by B. siamensis H30-3. These findings suggest that integration of B. aryabhattai H26-2 and B. siamensis H30-3 in Chinese cabbage production system may increase productivity through improved plant growth under high temperature and crop protection against fungal pathogens.
