녹차, 오룡차 및 홍차 추출물의 돌연변이원성 억제작용 (Desmutagenicity of Tea Extracts from Green Tea, Oolong Tea and Black Tea)

  • Desmutagenicities against 2-amino-1-methyl-6-phenylimidazo[4, 5-b] pyridine(PhIP) and 2-amino-3, 8-dimethylimidazo[4, 5-f]quinoxaline(MelQx) of tea extracts (steamed green tea, roasted green tea, oolong tea and black tea) were investigated. All the fractions obtained from tea extracts showed strong desmutagenic activity against PhIP and MeIQx toward S. typhimurium TA 98 in the presence of the S-9 mix. The crude catechin fraction exhibited the strongest desmutagenic activity. Among these tea extracts, black tea especially exhibited the strongest desmutagenic activity and the activity was 70.9~91.0% against PhIP and 92.2~98.8% against MelQx at a concentration(0.5~1.0mg/plate) for drinking. The activity of authentic catechins of (-)-EGC, (-)-EGCg, (-)-ECg and (-)-EC were 79.5%, 60.2%, 46.1% and 43.5% against PhIP, and were 52.3%, 11.6%, 8.2% and 22.1% against MelQx by addition of 1.0mg/plate, respectively. The desmutagenic activity was supposedly due to the (-)-EGCg, (-)-EGC and (-)-EC in tea polyphenols, and the browning materials. The desmutagenicity was stronger when mutagens were preincubated with S-9 mix after reaciton with black tea extracts than when preincubated with them after reaction with S-9 mix. The desmutagenicity of tea extracts was rather expressed by reacting directly with mutagens than by deactivating the activated forms of mutagens.

녹차 염색 견포의 카테킨 함량 분석 (HPLC analysis of Catechins in The Residues After Dyeing Silk Fabrics with Korean Green Tea Extracts)

  • We focused our research interests on the characteristic of silk fabrics dyed with green tea extracts and, moreover, revealed the catechin content of silk fabrics after dyeing with Korean tea extract by measuring the residues. Three kinds of green tea samples were taken between April and May, 2003, from Bosung(southern province in South Korea), in different terms as the first corp, the second corp, and the third corp. Before and after dyeing the silk fabrics with these extracts, catechins in the residue were analyzed by HPLC. Four kinds of Catechins, such as (-)-epicatechin(EC), (-)-epicatechin gallate(ECG), (-)-epigallocatechin (EgC), (-)-epigallocatechin gallate(EgCG) were isolated from Korean green tea extracts. Catechine contents in Korean green tea leaves depend on the making process such as heating and steaming, and the younger the richer. The absorbed catechins in sillk fabrics after dyeing followed in a decreasing order of EgCG>EgC>ECG>EC. We have found a 68% uptake of EgCG, and 116.8mg/g of EgCG in the silk fabrics after dyeing with 1% Korean green tea extracts.

Salmonella typhimurium Strain TA98, 100에서 감잎차, 녹차, 우롱차 추출물의 돌연변이 억제 효과 (Antimutagenic Effects of Water Extracts of Persimmon Leaf Tea, Green Tea and Oolong Tea on Reversion and Survival of Selected Salmonella Tester Strains)

  • Water extracts of persimmon leaf tea(PLTE), green tea(GTE) and oolong tea(OTE), at the con centration used for human consumption, were examined for inhibitory effects on the mutagenicity of major classes of dietary and environmental mutagens including indirect acting mutagens, B[ ]P (benzo[ ]pyrene), IQ(2 amino 3 methylimidazo[4,5 f]quinoline), 2 AA(2 aminoanthracene) in the presence of S9 mix and direct acting mutagen, 4 NQO(4 nitroquinoline 1 oxide) without S9 mix, using the modified Ames Salmonella/microsome assay. PLTE, GTE and OTE showed very potent and concentration dependent antimutagenic effects against indirect acting mutagens B[ ]P and IQ. At the maximum concentration(16,200 g/plate) of each tea extract, number of colonies decreased in a dose dependent manner up to 82~100%. Similar inhibition of PLTE, GTE and OTE were seen at higher concentration in the mutagenicity of the 2 AA following an initial increase in the activity at lower concentration. However, the mutagenicity of the direct acting mutagen 4 NQO were not suppressed at lower concentration of the three tea extracts, and higher concentration of the tea extracts enhanced mutagenic activity of the mutagen. There were no differences in the mode of antimutagenesis between PLTE, GTE, and OTE, in both Salmonella typhimurium TA98 and TA100 strains against the same mutagen. In conclusion, the water extracts of persimmon leaf tea, green tea and oolong tea possess marked antimutagenic potential against a variety of important dietary and environmental indirect acting mutagens, but the activity was not observed against the direct acting mutagens. These results suggest that the mode of inhibitory action may not have resulted from direct interaction between tea extracts and the mutagens, but rather from indirect metabolic inactivation of mutagens by tea extracts.

감잎, 녹차의 건분 및 물, 에탄올추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effects of Dried Leaf Powders, Water and Ethanol Extracts of Persimmon and Green Tea Leaves on Lipid Metabolism and Antioxidative Capacity in 12-Month-Old Rats)

  • The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level. 녹차와 보이차 추출물의 항산화 효과 (Antioxidative Effect on the Green Tea and Puer Tea Extracts)

  • 녹차에 대한 보이차의 항산화력을 알아보기 위해 물 추출물과 메탄올 추출물의 항산화 효과를 비교$\cdot$조사하였다. 총 폴리페놀의 함량은 물 추출물의 경우 녹차와 보이차가 비슷하였으며 메탄올 추출물의 경우는 보이차의 함량이 녹차의 약 $58\%$정도 밖에 되지 않았다. 전자공여능은 물 추출물의 경우 녹차와 보이차가 거의 유사하였으며 메탄올추출물의 경우 보이차가 녹차의 $54\%$정도였다. SOD 유사 활성은 용매간에 많은 차이가 있었는데 물 추출물의 활성은 $13.46\~14.07\%$로 녹차와 보이차간에는 거의 차이가 없었으나 메탄올 추출물의 활성은 녹차가 $59.63\%$, 보이차가 $48.93\%$로 물 추출물보다 $3\~4$배 높았으며 또한 녹차가 활성이 높았다. 아질산염 소거능은 물 추출물의 경우 녹차와 보이차간에 거의 차이가 없었으며 메탄올 추출물은 보이차가 녹차보다 다소 효과가 높았다. ACE 저해 활성은 물 추출물의 경우 보이차가 녹차보다 약 $38\%$ 정도 활성이 높았으나 메탄올 추출물은 반대로 녹차가 보이차보다 활성이 높았다. 이상의 연구 결과 보이차의 물추출물 항산화력은 녹차와 비슷하였으나 오히려 ACE 저해활성만은 높은 값을 나타내었고 메탄올 추출물에서는 보이차는 녹차보다 항산화력이 전반적으로 낮았으나 아질산염 소거능만은 높았다.

Effect of Natural Extracts on Oral Care Probiotics Weissella cibaria CMU and Periodontal Pathogens

  • The purpose of this study is to determine if natural extracts could be used as an additive in oral health food made with Weissella cibaria CMU (oraCMU). Natural extracts of green tea, mulberry leaf, licorice, and propolis, which are reported to have antimicrobial activities, were selected and used in this study. The minimum inhibitory concentrations (MIC) of extracts on periodontal pathogens such as Fusobacterium nucleatum and Porphyromonas gingivalis and their synergy effects with oraCMU by the fractional inhibitory concentrations methods were measured. From the results obtained, all the extracts showed no effect on the growth of oraCMU. Green tea extract showed the best antibacterial activity with MIC of 1.8 mg/ml against both F. nucleatum and P. gingivalis. In addition, green tea extract had a synergistic effect with oraCMU against F. nucleatum. Therefore, these results suggested that green tea extract is available as an additive in oral health food made with oraCMU.

감잎, 녹차, 솔잎의 건분 및 에탄올추출물이 흰쥐의 지방대사와 항산화능에 미치는 영향 (Effect of Dried Leaf Powders and Ethanol Extracts of Persimmon, Green Tea and Pine Needle on Lipid Metabolism and Antioxidative Capacity in Rats)

  • The effects of dried leaf powders and ethanol extract of persimmon, green tea and pine needle on lipid metabolism, lipid peroxidation and antioxidative activity were investigated in rats. Forty-nine male Spargue-Dawley rats weighing 107.8$\pm$1.8g were blocked into seven groups according to body weight. Rats were raised for four weeks with diets containing either 5%(w/w) dried leaf powders of one of three different Korea traditional teas, persimmon(Diospyros kaki Thunb), green tea(Camellia Sinensis O.Ktzc)or pine needle(pinus Koreansis Sieb. Et Zucc), or ethanol from equal amounts of each dried tea powder. Food intake, weight gain, food efficiency ratio, and weights of liver, kidney and epididymal fat were significantly higher in the green-tea-powder group, and significantly lower in the pine-needle-powder and pine-needle-extract groups. Persimmon-leaf powder was found to decrease plasma total lipid, triglyceride and cholesterol concentration by increasing fecal total lipid triglyceride and cholesterol excretions. Liver cholesterol concentration was significantly lower in the green-tea and pine-needle-extract groups. Red-blood-cell superoxide dismutase(SOD) and glutathione peroxidase(GSH-px) activities were significantly increased in rats fed green-tea extract. Liver SOD activity was increased in rats fed pine-needle powder or extract, and liver GSH-px activity was increased in rats fed greentea powder. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentration were both decreased in rats fed dried leaf posers or extracts of persimmon or green tea. It is believed that high vitamin E levels in persimmon leaf, and high flavonoid, beta-carotene and vitamin C levels in green tea effectively inhibited lipid peroxidation. In conclusion, persimmon and green tea leaves were effective in lowering lipid levels and inhibiting lipid peroxidation in animal tissue, while pine needles were effective and lowering body weight gain. From these results, persimmon and green tea leaves can be recommended in the treatment and prevention of chronic discorders such as cardiovascular disease, cancer and aging. As ethanol extracts from these teas were also effecitive in lowering tissue lipid levels and inhibiting lipid peroxidation, we recommend the use of discarded tea grounds for this.

Phenolic plant extracts are additive in their effects against in vitro ruminal methane and ammonia formation

  • Objective: The methane mitigating potential of various plant-based polyphenol sources is known, but effects of combinations have rarely been tested. The aim of the present study was to determine whether binary and 3-way combinations of such phenol sources affect ruminal fermentation less, similar or more intensively than separate applications. Methods: The extracts used were from Acacia mearnsii bark (acacia), Vitis vinifera (grape) seed, Camellia sinensis leaves (green tea), Uncaria gambir leaves (gambier), Vaccinium macrocarpon berries (cranberry), Fagopyrum esculentum seed (buckwheat), and Ginkgo biloba leaves (ginkgo). All extracts were tested using the Hohenheim gas test. This was done alone at 5% of dry matter (DM). Acacia was also combined with all other single extracts at 5% of DM each, and with two other phenol sources (all possible combinations) at 2.5%+2.5% of DM. Results: Methane formation was reduced by 7% to 9% by acacia, grape seed and green tea and, in addition, by most extract combinations with acacia. Grape seed and green tea alone and in combination with acacia also reduced methane proportion of total gas to the same degree. The extracts of buckwheat and gingko were poor in phenols and promoted ruminal fermentation. All treatments except green tea alone lowered ammonia concentration by up to 23%, and the binary combinations were more effective as acacia alone. With three extracts, linear effects were found with total gas and methane formation, while with ammonia and other traits linear effects were rare. Conclusion: The study identified methane and ammonia mitigating potential of various phenolic plant extracts and showed a number of additive and some non-linear effects of combinations of extracts. Further studies, especially in live animals, should concentrate on combinations of extracts from grape seed, green tea leaves Land acacia bark and determine the ideal dosages of such combinations for the purpose of methane mitigation.

Medilite 침출수에 의한 녹차 추출물의 항산화 활성연구 (Studies on Antioxidative Activity of Green Tea Extracts in Medilite-Extraction Water)

  • 천연 미네랄 성분을 함유한 medilite 침출수를 이용한 녹차 추출물에 의한 화학적 성분조성과 항산화 활성을 증류수 침출 녹차 추출물과 비교하였다. 항산화 활성은 DPPH법 및 microsone 생체막 지질 과산화물 생성정도를 나타내는 TBARS법으로 측정하였다. 녹차 추출물의 총 폴리페놀성 화합물과 추출 수율은 medilite 325 mesh, 600 mesh, DW 추출물 순으로 나타났다. DPPH법에 의한 0.05%와 0.1%의 녹차 추출물에서 항산화 활성은 60.95% ∼ 64.15% 사이였다. 간 homogenate 및 microsone 생체막 지질 과산화물 생성 억제 정도는 medilite 325 mesh 및 600 mesh 침출 녹차 추출물에 비해 증류수 침출 녹차 추출물에서 현저히 억제되었다. 특히 Fe 함량은 증류수 침출 녹차 추출물에 비해 medilite 325 mesh 및 600 mesh 침출 녹차 추출물에 현저히 높았다 이러한 medilite 침출 녹차 추출물에서 높은 Fe성분 함량은 microsone 생체막 지질 과산화물의 생성 촉진과 관련된 것으로 사료된다. 이상의 결과에서 녹차 추출물 중의 총 폴리페놀성 화합물, 추출 수율 및 미네랄 성분은 증류수에 비해 medilite 침출수를 이용함으로써 대체적으로 증가하는 결과를 얻었다.

솔잎 및 녹차추출물의 항산화성 및 아질산염 소거작용 (Antioxidative and Nitrite Scavenging Activity of Pine Needle and Green Tea Extracts)

  • 솔잎과 녹차의 열수와 에탄올 추출물을 이용하여 free radical 반응 및 지방산화에 미치는 영향과 아질산염 소거작용을 검토하였다. 각종 항산화제와 항산화력 비교에서 추출물 농도별 지방산화 억제능력은 0.1%보다는 0.3%가 낮은 TBARS값을 나타내었고, iron sources에 대한 binding 능력은 열수추출물이 우수하였다. Deoxyribose상에서의 hydroxyl radical 포집능력은 솔잎과 녹차 에탄올추출물이 대조구 보다 낮은 TBARS값을 나타내었다. 추출물의 iron 함량은 솔잎 에탄올 추출물이 가장 높은 함량을 나타내었으며, ascorbic acid 함량은 녹차의 열수와 에탄올 추출물에서 높은 값을 나타내었다. 전자공여능은 열수추출보다는 에탄올 추출물이 높은 활성을 나타내었다. 또한 솔잎과 녹차추출물은 각종 항산화제에 비하여 높은 전자공여능을 나타내었다. 아질산염 소거작용은 pH의 감소에 따라 높게 나타났으며, 0.3%의 솔잎과 녹차 추출물은 pH 1.2와 pH 3.0에서 95% 이상의 소거능을 나타내었으며, ascorbic acid 자체보다도 높은 소거능을 나타내었다. 이상의 결과로 보아 솔잎과 녹차추출물은 각종 육제품 제조시 첨가물로서 산화방지 효과 및 아질산염 소거제로서의 사용을 시사하는 것이다.
