Effects of Dietary Copper Exposure on Accumulation and Histopathological Change in Liver of Juvenile Rockfish, Sebastes schlegeli

  • Experiments were carried out to investigate the accumulation and the histopathological changes in liver of juvenile rockfish, S. schlegeli, after sub-chronic dietary Cu (0, 50, 125, 250 and 500mg/kg) exposure for 60 days. Cu accumulation in liver was significantly increased with dietary exposure period and concentration for 60 days, and has a linear relation with dietary exposure days. After 60 days of Cu dietary exposure, the Cu concentration in the liver was $75.9\pm12.05,\;126.29\pm22.11\;and\;360.44\pm45.26\;{\mu}g/g$ dry weight and was approximately 11-fold, 18-fold and 51-fold higher than in the control diet group at 125, 250 and 500 mg/kg Cu diet group. The accumulation factors were increased with the dietary exposure period in liver of rockfish. In the primary exposed stage, the effect of hepatic tissue in the rockfish exposed to dietary Cu observed enlargement of hepatocytes nuclei, activity of hepatic cells and the swelling of hepatic cells. While exposed time and concentration were increased, the distinct granulation, irregular shape and necrosis of hepatic cells were observed. It was observed that granule degeneration and necrosis showed a part of cells in hepatic tissue after 60 days at 500 mg/kg.

Epigallocatechin-3-gallate rescues LPS-impaired adult hippocampal neurogenesis through suppressing the TLR4-NF-κB signaling pathway in mice

  • Adult hippocampal dentate granule neurons are generated from neural stem cells (NSCs) in the mammalian brain, and the fate specification of adult NSCs is precisely controlled by the local niches and environment, such as the subventricular zone (SVZ), dentate gyrus (DG), and Toll-like receptors (TLRs). Epigallocatechin-3-gallate (EGCG) is the main polyphenolic flavonoid in green tea that has neuroprotective activities, but there is no clear understanding of the role of EGCG in adult neurogenesis in the DG after neuroinflammation. Here, we investigate the effect and the mechanism of EGCG on adult neurogenesis impaired by lipopolysaccharides (LPS). LPS-induced neuroinflammation inhibited adult neurogenesis by suppressing the proliferation and differentiation of neural stem cells in the DG, which was indicated by the decreased number of Bromodeoxyuridine (BrdU)-, Doublecortin (DCX)- and Neuronal Nuclei (NeuN)-positive cells. In addition, microglia were recruited with activating TLR4-NF-${\kappa}B$ signaling in the adult hippocampus by LPS injection. Treating LPS-injured mice with EGCG restored the proliferation and differentiation of NSCs in the DG, which were decreased by LPS, and EGCG treatment also ameliorated the apoptosis of NSCs. Moreover, pro-inflammatory cytokine production induced by LPS was attenuated by EGCG treatment through modulating the TLR4-NF-${\kappa}B$ pathway. These results illustrate that EGCG has a beneficial effect on impaired adult neurogenesis caused by LPS-induced neuroinflammation, and it may be applicable as a therapeutic agent against neurodegenerative disorders caused by inflammation.

Fucoidan Increases Porcine Neutrophil Extracellular Trap Formation through TNF-α from Peripheral Blood Mononuclear Cells

  • Fucoidan extracted from brown seaweed has a variety of biological activities. Neutrophil extracellular traps (NETs) formation is an immune response for the invasion of pathogens. Neutrophils release granule protein and chromatin that form extracellular fibers that bind microbes. These NETs degrade virulence factors and kill bacteria. The aim of this study was to investigate the effect of fucoidan on NET formation of porcine peripheral blood polymorphonuclear cells (PMNs). The NET formation was determined by fluorescence emission of propidium iodide (PI) in PMNs by a fluorescence microplate reader. The production of tumor necrosis factor (TNF)-α from peripheral blood mononuclear cells (PBMCs) was measured by ELISA method. Fucoidan itself did not show any direct effect on NET formation. However, NET formation of PMNs was increased by the culture supernatant from PBMCs treated with fucoidan. The NET formation of PMNs were also enhanced by treatment with recombinant porcine (rp) TNF-α. The ability of culture supernatant from PBMCs treated with fucoidan to increase the NET formation of PMNs was inhibited by addition of goat anti-rp TNF-α polyclonal antibody (pAb) (IgG) prior to the culture. The increase of NET formation by rp TNF-α was also inhibited by goat anti-rp TNF-α pAb (IgG). The level of TNF-α in culture supernatant from PBMCs was increased by treatment with fucoidan. These results suggest that fucoidan increases porcine NET formation, which is mediated by TNF-α produced from PBMCs.

Effects of Macrophage on Biodegradation of β-tricalcium Phosphate Bone Graft Substitute (대식세포가 β-tricalcium Phosphate 뼈이식제의 생분해에 미치는 영향)

  • Various calcium phosphate bioceramics are distinguished by their excellent biocompatibility and osteoconductivity. Especially, the exceptional biodegradability of $\beta$-TCP makes it a bone graft substitute of choice in many clinical applications. The activation of osteoclasts, differentiated from macrophage precursor cells, trigger a cell-mediated resorption mechanism that renders $\beta$-TCP biodegradable. Based on this evidence, we studied the biodegradation process of granular-type $\beta$-TCP bone graft substitute through in vitro and in vivo studies. Raw 264.7 cells treated with RANKL and M-CSF differentiated into osteoclasts with macrophage-like properties, as observed with TRAP stain. These osteoclasts were cultured with $\beta$-TCP nano powders synthesized by microwave-assisted process. We confirmed the phagocytosis of osteoclasts by observing $\beta$-TCP particles in their phagosomes via electron microscopy. No damage to the osteoclasts during phagocytosis was observed, nor did the $\beta$-TCP powders show any sign of cytotoxicity. We also observed the histological changes in subcutaneous tissues of rats implanted with granule-type $\beta$-TCP synthesized by fibrous monolithic process. The $\beta$-TCP bone graft substitute was well surrounded with fibrous tissue, and 4 months after implantation, 60% of its mass had been biodegraded. Also, histological findings via H&E stain showed a higher level of infiltration of lymphocytes as well as macrophages around the granule-type $\beta$-TCP. From the results, we have concluded that macrophages play an important role in the biodegradation process of $\beta$-TCP bone graft substitutes.

ELECTRON MICROSCOPIC AND IMMUNOHISTOCHEMICAL STUDY ON THE PROLIFERATION OF RABBIT SUBMANDIBULAR GLANDULAR CELL AFTER DUCT LIGATION AND CUT (가토 악하선 도관 결찰과 절단 후 악하선 세포의 증식에 관한 전자 현미경 및 면역조직화학적 연구)

  • Obstructive sialadenitis of major salivary glands is a common entity that occurs either in sialolithiasis or in foreign-body obstruction of the excretory ducts. This is characterized histologically by the presence of duct-like structural groups in a highly fibrotic stroma. Although the pathologic features are well recognized, the various cell types involved in the atrophy and subsequent regeneration of the obstructed salivary gland have been controversial. For this reason, an animal model of obstructive sialadenitis that induced atrophy in the salivary gland was used. Experimental study was performed to observe changes of submandibular gland in rabbit and apply the results to clinical activity. Forty-five rabbits each weighing about 3Kg were used and divided into control and experimental group. In the experimental group, ducts of submandibular gland was ligated and cutted divided into each twenty rabbits. Rabbits were serially sacrificed on the 3rd, 5th, 14th, 30th day of experiment. The submandibular glands were dissected out at sacrifice and stained with H&E, MT, immunohistochemical stain and the histological examinations were carried out under the light and transmission electron microscope. After examination and comparison of all specimens, the results of this study were as follows: 1. In the features of H&E stain, moderate infiltration of inflammatory cell were present at 3rd day of experiment. The features of ductal metaplasia was observed after 7th day in the ligation group and destructive changes was continued. In the cutting group, atrophic changes were less severe than ligation group but the small ductule were separated from stroma after 7th day. 2. In the feature of MT stain, apposition of connective tissue was increased in all group, more active in ligation group. 3. In the features of immunohistochemical stain, ligation group showed increased PCNA positive response at 7th day and the higher activity of duct cells was observed. Severance group showed more PCNA positive response than ligation group at 30th day. 4. In TEM features, ductal metaplasia was started at 7th day and degenerative change with margination of nucleus had been severe. Although ductal metaplasia was seen in the severance group, more numerous granule in different size was founded than ligation group. From above results, degenerative change was identified with ductal metaplasia, apically apposition of granule, r-ER destruction in ligation group. Severance of duct elicit degenerative change of grandular cells but the change was less severe than ligation group and more PCNA positive cell was founded at acinar cell.

Screening of genes differentially expressed in cultured human periodontal ligament cells and human gingival fibroblasts (배양된 치주인대세포와 치은섬유아세포에서 상이하게 발현된 유전자들의 검토 양상)

  • Periodontal ligament(PDL) cells and human gingival fibroblasts(HGFs) play important roles in development, regeneration, normal function, and pathologic alteration. PDL cells and HGFs have the similarity related with general characteristics of fibroblast such as spindle shaped morphology, the presence of vimentin intermediate filament and the synthesis of interstitial collagens and fibronectin. There were many studies about the differences between PDL cells and HGFs, but they were not about whole gene level. In this study, we tried to explain the differences of gene expression profiles between PDL cells and HGFs, and the differences among three individuals by screening gene expression patterns of PDL cells and HGFs, using cDNA microarray. Although there were some variants among three experiments, a set of genes were consistentely and differentially expressed in one cell type. Among 3,063 genes, 49 genes were more highly expressed in PDL cells and 12 genes were more highly expressed in HGFs. The genes related with cell structure and motility were expressed more highly in PDL cells. These are cofilin 1, proteoglycan 1 secretory granule, collagen type I(${\alpha}$ 1), adducin gamma subunit, collagen type III(${\alpha}$ 1), fibronectin, lumican(keratan sulfate proteoglycan), and ${\alpha}$ -smooth muscle actin. Tissue inhibitor of metalloproteinase known as the enzyme controlling extracellular matrix with matrix metalloproteinase is more highly expressed in PDL cells, osteoprotegerin known as osteoclastogenesis inhibitory factor is more highly expressed in HGFs. We performed northern blot to verify cDNA microarray results on selected genes such as tissue inhibitor of metalloproteinase, fibronectin, osteoprogeterin. The result of northern blot analysis showed that each cell expressed the genes in similar pattern with cDNA microarray result. This result indicates that cDNA microarray is a reliable method in screening of gene expression profiles.

  • The nervous tissue in the cerebral ganglion of Korean planaria was observed using electron microscope. The obtained results are as follows: A cerebral ganglion is composed of the nerve cells, neurosecretory cells, neuroglial cells and neuropils. The nerve cells are round or ovoidal-shaped cells (diameter, $5{\mu}m$), which has a large ellipsoidal nucleus containing the evenly developed heterochromatin. Their cytoplasms were found to be relatively simple, because of their undeveloped cell organelles. The neurosecretory cells are long and ellipsoid or spindle-shaped cells, where there were found a large ellipsoidal nucleus and cytoplasm filled with secretory granules (diameter, 60 nm). The neuroglial cells were seldom observed. They are spindle-shaped cells (size, $6\times0.8{\mu}m$), which were observed mainly among the nerve fibers. The neuropils are formed by the nerve fibers and nerve endings which are filled with mitochondria, neurotubules and secretory granules of four kinds (high electron dense granules of sizes 75 nm, 50 nm and 37 nm, and electron lucent granule of size 30 nm etc.). These granular vesicles are divided into single vesicle type and compound vesicle type in the nerve terminals, and neuronal synapses were observed to be the axo-dendritic and dendro-dendritic synapse type.

  • The changes of rough surfaced endoplasmic reticulum of plasma cells in the regional lymph node after injection of Bovine Gamma Globulin summarized as follow: The rough ER of plasma cells before antigenic stimulation shows flattened sac but changed to vesicular at 3rd day and vacuolar at 7th to 10th day. lntradisternal granules are appeared within the lumina or rough ER at 7th to 10th day. The rough ER is changed to distended sac or flattened sac at 20th day. The results suggest that normally flattened sac of rough ER is changed to vesicular and then to vacuolar after antigenic stimulation. But they are returned to distended sac or flattened sac with disappearance of antigenic stimuli.

  • In the goby Micropercops swinhonis, the follicular layer of full-grown oocytes consists of an outer layer (theca cell) and an inner layer (granulosa cell). As the oocyte grows, columnar cells of inner granulosa layer secrete mucin to their cytoplasm and then surround the oocyte. Such granulosa cells appear to be cuboidal cells in the early vitellogenesis, yolk vesicle stage, to be replaced by columnar cell secreting mucins (adhesive materials) in the middle vitellogenesis, yolk granule stage. The enveloping layer of the oocyte has a muco- follicle layer filled with mucins. The mucins are an amorphous and electron-dense substance. Interestingly, the oocyte enveloping layer becomes thickened towards the animal pole as vitelogenesis proceeds. A zona radiata of about $7.8{\sim}11.5\;{\mu}m$ thick is present below the muco-follicle layer. The zona radiata is composed of an one-layered electron-dense externa and a three to five-layered electron-less interna.

Structure of the Ovary and Ultrastructural Study of Vitellogenesis in the Oocytes in Female Gomphina veneriformis (Bivalvia: Veneridae) in Eastern Korea

  • The structure of the ovary, ultrastructure of oocytes and morphological characteristics of vitellogenesis during oogenesis in female Gomphina veneriformis were investigated in clams collected from coastal waters of Samchok, Gangwon-do, Kore. In the previtellogenic oocytes, the Golgi complex was involved in the formation of a number of vacuoles. In the early vitellogenic oocytes, lipid droplets appeared among the Golgi complex, endoplasmic reticulum, and mitochondria in the cytoplasm of the oocyte were involved in the formation of lipid droplets. Coated vesicles, resulting from endocytosis appeared at the basal region of the early vitellogenic oocyte. The uptake of nutritive materials in the coated vesicles formed by receptor-mediated endocytosis appeared through the formation of coated endocytotic pits on the oolemma. In the late vitellogenic oocytes, large yolk granules were formed by a combination of small yolk granules. In the mature oocyte, a mature yolk granule in composed of three components: crystaline core, electron lucent cortex, and a limiting membrane. According to cytological and histological observations, vitellogenesis occurred by way of endogenous autosynthesis and exogenous heterosynthesis. Autosynthesis involved the conbined activities of the Golgi complex, mitochondria, rough endoplasmic reticulum, whereas heterosynthesis involved endocytotic incorporation of extraovarian precursors at the basal region of the early vitellogenic oocyte. The follicle cells which was attached to oocytes, were involved in the development of the previtellogenic and early vitellogenic oocytes as a kind of nutritive cells containing a number of glycogen particles and lipid droplets in the cytoplasm.
