Effects of Folium Artemisiae Argyi and Moxa Tar' Herbal Acupuncture on Transient Forebrain Ischemic Injury in Rats (애엽(艾葉) 및 구진(灸津) 약침(藥鍼)이 일과성(一過性) 전뇌(前腦) 허혈(虛血) 손상(損傷)에 미치는 효과(效果))

  • Objectives : Acupuncture and herbal medicine have been used to prevent and treat the cerebrovascular accident, such as a stroke, and many studies of acupuncture and moxibustion concerning to the stroke have been undertaken in the human and various animals. Recently, the new therapeutic tool, that is herbal acupuncture, has been developed since the 1950' and applied to various diseases including the cerebrovascular accident. The main characteristics of herbal acupuncture are a combination of acupuncture and herbal medicine. It was not well known the therapeutic effect and the mechanism of herbal acupuncture on transient forebrain ischemic injury, although it has been used frequently in clinics. Methods : In this study, effects of folium Artemisiae Argyi and moxa tar' herbal acupuncture on the $GV_{20}$, named Baek-Hue, on neuroprotection after the transient forebrain ischemia were investigated in Sprague-Dawely rats. Expressions of cFos, FosB and BDNF protein in the hippocampus and cortex were observed at 2 hrs and 48 hrs after transient forebrain ischemia by immunohis- tochemistry and ELISA technique. Results : Expression of cFos protein was increased slightly in the hippocampus and cortex at 2 hrs after transient forebrain ischemia, but FosB protein was increased highly comparing to cFos protein. However, pretreatment with folium Artemisiae Argyi or moxa tar' herbal acupuncture on $GV_{20}$ significantly increased expression of cFos protein and significantly decreased expression of FosB protein compared to control group, respectively. These features were observed in the motor cortex and retrosplenial granular cortex as well as the hippocampus. Also, pretreatment with folium Artemisiae Argyi and moxa tar' herbal acupuncture on$GV_{20}$ significantly increased the expression of BDNF protein in the hippocampus and the cortex compared to control group at 48 hrs after transient forebrain ischemia, respectively. Conclusions : These results suggest that pretreatment with folium Artemisiae Argyi or moxa tar' herbal acupuncture on $GV_{20}$ has neuroprotective effect on transient forebrain ischemia and theherbal acupuncture on $GV_{20}$ may be related to antioxidative function.

  • A series of trapdoor model tests was systematically performed in order to investigate soil arching. The mobilized soil arching was clearly observed by change of the vertical earth pressure loaded on trapdoor of soil container box. A slow decent of the loading plate at the trapdoor results in loosening zone over the trapdoor and the stress in this loosening zone was transferred to the stationary zone in the vicinity of the trapdoor. In particular, it was observed that the vertical earth pressure rapidly decreased in the loosening zone and increased in the stationary zone at the trapdoor. Both the maximum decreasing rate of the vertical earth pressure in the loosening zone and the increasing rate of the vertical earth pressure in the stationary zone were not influenced by the ground density, but affected by the size of the trapdoor. The loosening zone could be defined by the elliptical configuration, in which the major axis was twice as long as the height of the loosening zone at the center of trapdoor and the minor axis was the same as the width of trapdoor. The height of loosening zone at the center of trapdoor was one and a half times as long as the width of trapdoor loading plate.

  • Idiopathic CD4+ T-lymphocytopenia is defined as a depletion of CD4+ lymphocytes below $300/mm^3$ in the absence of HIV infection or other known causes of immunodeficiency. Many infectious diseases have been reported to be associated with idiopathic CD4+ T-lymphocytopenia, and there have also been a few cases of mycobacterial infection in idiopathic CD4+ T-lymphocytopenia. Until now, it has been unclear as to whether CD4+ T-lymphocytopenia is a predisposing factor for or a consequence of the mycobacterial infection. Pulmonary alveolar proteinosis is an uncommon disease characterized by the intraalveolar deposition of amorphous granular material that stains positive with PAS, and its association with mycobacterial infection has rarely been reported. Recently, we experienced a previously healthy young man who had been diagnosed as idiopathic CD4+ T-lymphocytopenia with disseminated mycobacterium kansasii infection and pulmonary alveolar proteinosis, and report this case.

  • The adsorption and desorption behavior of biofilter-medium was investigated on the performance of an adsorption column. Continuous flow-isothermal adsorption experiments were performed to treat waste air containing such a VOC as ethanol under the same condition of > 90% relative humidity as the condition of the feed to a biofilter process. In case of feeding waste air containing ethanol of 1,000 ppmv (or 2,050 mg ethanol/$m^3$) to the adsorption system at the rate of 2 L/min, the onsets of its breakthrough and reaching the state of dynamic equilibrium at the exit had been delayed 10 and 3 times, respectively, later than those at the 1st stage sampling port. Moreover, in case of 2,000 ppmv (or 4,100 mg ethanol/$m^3$), they had been delayed 9 and 3 times, respectively. Thus, regardless of feeding concentration, the ratios of delaying period were observed to be quite consistent each other at the exit of the adsorption column. With regard to the period of desorption, the ratios of delaying period were consistent each other to be 1.5 for both cases. In addition, the effect of microbial activity and sterilization-process was studied on adsorption equilibrium. The ethanol concentration in the vapor phase of vials packed with sterilized granular activated carbon (GAC) was quite consistent to that with unsterilized GAC. However, the ethanol concentrations in the vapor phase of vials packed with unsterilized compost and the unsterilized mixture of GAC and compost were higher than those with sterilized compost and the sterilized mixture of GAC and compost, respectively.

  • The morphological and fine structural changes during the oogensis of Clonorchis sinensis were studied on the developing ovums in the ovary and ootype with electron microscope. Adult worms were removed from the hepar of the which and previously infected with metacercariae of Clonorchis sinensis. The ovary including the Mehlis' glands and an ootype from adult worm was prefixed for 1-2 hours in 1.25% glutaraldehyde buffered with 0.2M cacodylate at PH 7.2, secondarily fixed for 30 minutes in potassium bicromate and postfixed for an hour in 1% osmic acid buffered with 0.4M cacodylate at PH 7.2. After fixation tissues were dehydrated in an alcohol series, embedded in Epon 812 from propylene oxide and stained with saturated uranyl acetate and $Pn(NO_3)_2$ solution. Material was examined with a Hitachi HS-7S electron microscope. The periphery of the ovary, except for the posterior region, is made up of oogonia. As the oogonia divide they proliferate primary oocytes toward the central part of the ovary. After a period of growth the primary oocyte leaves the ovary and is penetrated by a sperm in the ootype. Sperm penetration immediately activates the primary oocyte to resume its meiotic activity. After the oocytes meiotic activity is completed, the pronuclei fuse to form a single cleavage nucleus which possesses two nucleoli. As the oocytes develop their cytoplamic materials are abundant; small mitochondria are abundant and often their profiles are more unmerous in one part of the cytoplasm than elsewhere; the granular endoplasmic reticulum becomes alveolar-sac form after it leaves the ovary it becomes stratified form. The reticulate Golgi apparatus is apparent in the developed oocyte. A little of cortical granules are distributed inside of the plasma membrane I oogonia and large quantity of cortical granules are arranged just inside of the plasma membrane of the primary oocyte and after fertilization they are disappeared with broken out.

Ultrastructure of the Submandibular Gland in the Lesser White-Toothed Shrew, Crocidura suaveolens (작은땃쥐 Crocidura suaveolens 악하선의 미세구조)

  • The ultrastructure of submandibular gland was examined in the lesser white-toothed shrew, Crocidura suaveolens. The submandibular gland of C. suaveolens was a mixed gland composed of serous and mucous acinar cells. Secretory granules from the acini were discharged through the salivary ducts into the oral cavity. Serous and mucous acinar cells had well developed rough endoplasmic reticulum and mitochondria and large amount of granules. In case of serous acinar granules, an immature granule was formless and had only dense specks, and a matured granule was a complete round type delimiting by a single membrane and had a homogeneous dense center with dense specks on the border. In case of mucous acinar granules, while an immature granule was a round type and had an only homogeneous matrix and an indistinct limiting membrane, a mature granule was an even round type having a variety of pattern with several dense bands into the homogeneous matrix and had a distinct membrane. Therefore, a mature mucous acinar granule of C. suaveolens was not only distinct from those of the other mammalian species to have a variety of pattern but also from those of C. lasiura to have an even round type. A great serous-like secretory granules and Myelin-like body were observed in the cytoplasm and lumen of granular duct cells. Myelin-like body, a characteristic structure only reported in salivary gland of three shrews, was discharged from secretory cell into lumen by the manner of exocytosis which has little differences from discharging manner of secretory granules.

  • Germ cell development and cyclic changes in the epithelial cells of the seminal vesicle of the male rapa whelk, Rapana venosa, were investigated by cytological and histological observations. The process of germ cell development can be classified into five stages: (1) spermatogonial, (2) primary spermatocyte, (3) secodary spermatocyte, (4) spermatid, and (5) spermatozoon. In particular, four atypical cells (Type IA, IB, IIA and IIB cells) occur among normal germ cells in the acini during spermatogenesis. Presumably, the atypical cells, which have lysosome-like vacuoles or lysosome-like bodies in the cells, are involved in breakdown and absorption themselves in the acini. However, atypical cells were not found in the epithelial cells of the inner layer of the seminal vesicle. A considerable amount of spermatozoa are transported from the testis towards the the seminal vesicles until late July. The main coupulation period is between June and July. The process of the cyclical changes of the seminal vesicles can be classified into three phases: (1) resting, (2) accumulating, and (3) spent. Yellow granular bodies are involved in resorption or digestion of residual spermatozoa.

Characteristics of the Small Scale Leucocratic Granites in the Eastern Parts of the Taebaegsan Region, Korea (태백산 지역 동부에 분포하는 소규모 우백질 화강암체의 특징)

  • Precambrian granitic gneisses and Cambrian meta-sedimentary rocks are prevalently distributed in the eastern part of the Taebaegsan region, and biotite granitic batholith of the Jurassic period (?) is found in the southern part of Uljin-si. But small scale leucocratic granitic stocks which commonly found here and there have been rather neglected in the previous studies. The presence of leucocratic granites could be differentiated from the older granitic rocks and biotite granite through the outcrop characteristics, mineral species and geochemical compositions. For the effective comparison between the older granitic rocks and leucocratic ones, pale gray to gray coloured Hongjesa granitic gneiss with granular texture was selectively chosen. The Hongjesa granitic gneiss and biotite granite usually have rather plenty of coloured minerals such as biotite and chlorites. But the leucocratic granites often show sericitic alteration due to the albitization and greisenisation during the post-magmatic alteration, and shows rather bright appearance because of poor amount of coloured minerals. Since all of granitic rocks passed rather high degrees of magmatic differentiation, they belong to calc-alkalic and peraluminous in their characters. Among the alkali elements of the leucocratic rocks $K_2O$ shows higher increase than those of the other granitic rocks, and $Na_2O$ only represents slight decrease than those of the Hongjesa granitic gneiss and Uljin granite. On the other hand, CaO and total Fe content are clearly decreased than those of the Hongjesa granitic gneiss and Uljin granite.

  • The development of trachea in fetuses on 60, 90 and 120 days of gestation and neonates of Korean native goats was investigated by microscopy and scanning and transmission electron microscopy. The results were summarized as follows; Light microscopic studies: 1. In the 60-day-old fetuses, the tracheal walls were differentiated and divided into four layers of the mucosa, submucosa, muscle and cartilage, and adventitia. The tracheal epithelium is composed of stratified ciliated columnar epithelium at 60- and 90-day-old fetuses while the epithelium observed at 120-day-old fetuses was pseudostraified ciliated colummar epithelium. 2. In the 90-day-old fetuses, tracheal glands extended into the submucosa and peripheral area of the tracheal cartilage. The blood vessels were observed in the submucosa and adventitia. The elastic and collagenous fibers were observed in the tracheal walls. 3. In the neonates, the tracheal walls consisted of mucosa with well-developed folds, submucosa, tracheal glands, muscle and cartilage, collagenous and elastic fibers, and adventitia, which were more developed than those of 120-day-old fetuses. The tracheal epithelium was developed as that in adults. Scanning electron microscopic studies: 4. In the 60-day-old fetuses, most of tracheal epithelial cells were nonciliated but short microvilli were sporadically observed on the luminal surface. On rare occasions, a few cells have solitary cilium. 5. In the 90-day-old fetuses, the ciliated cells appeared increasingly and cilia elongated longer than those of 60-day-old fetuses. 6. In the 120-day-old fetuses, the nonciliated cells covered with microvilli in dome-shape were barriered by thick carpet of cilia. The nonciliated cells also have many papillary projectons on the apical surface. 7. In the neonates, the nonciliated cells in tracheal epithelium were covered compactly with numerous cilia, and many secretory droplets were found on the cilia. Transmission electron microscopic studies: 8. In the 60-day-old fetuses, nonciliated cells of the tracheal epithelium contain large amounts of glycogen granules in the supernuclear and subnuclear areas meanwhile a few cell organelles were formed. Cilia were well formed along the apical cell membranes of the ciliated cells. Also found in the ciliated cells were basal corpuscles, mitochondria and short chains in granular endoplasmic reticulum(GER). Between the epithelial cells presented were well-defined junctional complex with zonula occludens and desmosomes. The nuclei were variable in size and shape. The more developed nucleoli were observed conspicuosly. 9. In the 90-day-old fetuses, nonciliated cells contained large glycogen granules. Accumulated glycogen granules were observed in the subnuclear and supranuclear portion of the cytoplasm. A few short microvilli were covered with glycocalyx. Ciliated cells contained numerous mitochondria and short chains of GER. 10. In the 120-day-old fetuses, the ciliated cells contained numerous mitochondria, abundant short chains of GER and nucleoli. Nonciliated cells contained some Golgi complex and mitochondria. The cell borders were well-defined and distinct junctional complex with zonula occludens, desmosomes, and interdigitorum. 11. In the neonates, well-developed goblet cells were observed in the tracheal epithelium. Ultrastructures of ciliated and nonciliated cells on the tracheal epithelia were similar in pattern as those in adults.

Also, pretreatment with folium artemisiae argyi' herbal acupuncture on $LR_3$ significantly increased the expression of BDNF protein in the hippocampus ($263.26{\pm}44.56pg/ml$ extracted by water, $275.35{\pm}51.47pg/ml$ extracted by moxa tar)and the cortex ($102.33{\pm}13.65pg/ml$ extracted by water, $109.54{\pm}9.37pg/ml$ extracted by moxa tar) compared to the hippocampus $(134.07{\pm}2.96pg/ml)$ and the cortex $(61.16{\pm}4.11pg/ml)$ in control group at 48 hrs after transient forebrain ischemia. These results suggest that pretreatment with folium artemisiae argyi'herbal acupuncture on $LR_3$ has neuroprotective effect on transient forebrain ischemia and the herbal acupunture on $LR_3$ may be related to antioxidative function of folium artemisiae argyi.
