Infection of Clinostomum complanatum(Rudolphi, 1814) (Trematoda : Digenia) metacercaria in goldfish (Carassius auratus) cultured in Korea (금붕어에 기생한 Clinostomum complanatum(Rud., 1814)의 피낭유충에 관하여)

  • Many encysted metacercariae of Clinostomum complanatum(Rudolphi, 1814) were found in the flesh near the skin of goldfish(Carassius auratus) sultured at a fish farm in Chonbuk Province, Korea, in the summer of 1995. The mortality reached about 7% from July 10 to August 20. Encysted and excysted metacercariae were$700\sim877{\times}700\sim775{\mu}m$ and $2.4\sim5.5{\times}0.7\sim1.37{\mu}m$ in size, respectively. The parasites looked yellowish due to the yellow particulate matter in the intestine. Of 276 fish examined, 234(84.8%) were infected with a mean intensity of 216.6(range 1~1.214) parasites. Fish of 2.0~2.5 cm in body length were most frequently(94.3%) infected but those of 3.1~3.5 cm were the least infected(75.27%). The isthmus was most heavily infected. There was no difference in prevalence between two forms of goldfish, Gyariko an Ryugum. The sites of infection were swollen and recognized as yellow spots of 0.4~0.8 mm in diameter. Fish infected with more than 200 metacercariae became moribund with listless swimming and finally died.

  • The regeneration and differentiation of the cutaneous pigment system in the goldfish, Carassius auratus during the wound healing process were studied with high magnification electron microscope. The cutaneous pigment cells of the normal tissues were composed of three kinds of dermal chromatophores-xanthophores, leucoiphores and melanophores. While xanthophores contain two kinds of pigment granules-pterinosomes and carotenoid vesicles, leucophores and melanophores contain amorphous pigment granules (leucosomes) and oval shaped electron dense melanin pigment granules (melanosomes) respectively. After injury, primary wound healing responses being carried out by migration of epidermal cells and hemocytes spreading over the wound surface at the day of wounding. And at the time of primary wound closure, 5 to 7 days after wounding, rER rich cells-presumably common precursors of dermal chromatophores-immigrated into the wound area. First redifferentiated chromatophores appeared 3 weeks after wounding. Pigment granules of the chromatophores were emerged from the cytoplasmic Golgi complex via rough endoplasmic reticulum. Pinocytotic vesicles which associated with accumulation of pigment material, appeared only at the inner surface of the chromatophores adhering to the rER rich cells, characteristically. The differentiation of each chromatophore in addition to integumental wound repair were accomplished within 4 weeks after wounding at most cases, however the total numbers and densities of these repaired chromatophores still primitive state. Moreover, It has been revealed that complete repair of chromatophores at wounded tissues from burns requirs more than 3 months in normal environment.

Effect of Co-existence of Carbaryl and Chlorothalonil on the Short-term Bioconcentration Factor in Carassius auratus(goldfish) (Carbaryl과 Chlorothalonil의 공존이 Carassius auratus(goldfish)를 이용한 생물농축계수에 미치는 영향)

  • This study was performed to investigate the effect of co-existence of carbaryl and chlorothalonil on the short-term bioconcentration factor in Carassius auratus(goldfish). The fishes were exposed to the combined treatment of carbaryl and chlorothalonil(0.05 ppm+0.005 ppm, 0.05 ppm+0.010 ppm, 0.10 ppm+0.005 ppm) for 1, 3 and 5 days, respectively. Carbaryl and chlorothalonil in fish and in test water were extracted with n-hexane and acetonitrile. GC-ECD was used to detect and quantitate carbaryl and chlorothalonil. 1-day, 3-day and 5-day bioconcentration factors($BCF_1, BCF_3$ and $BCF_5$) of each pesticide were calculated from the quantitation results. The depuration rate of each pesticide from the whole body of fish was determined over the 72-h period after combined treatment. The results were as follows: $BCF_1$ values of carbaryl were 3.521, 3.802 and 3.587, respectively, when the concentration of carbaryl and chlorothalonil in combined treatment were 0.05+0.005, 0.05+0.010 and 0.10+0.005 ppm. BCF3 values of carbaryl were 4.825, 4.556 and 3.828, respectively, and $BCF_5$ values of carbaryl were 3.974, 3.921 and 4.186, respectively, under the conditions. While $BCF_1$ of chlorothalonil were 0.829, 0.829 and 1.540, respectively, under the same condition of pesticide concentrations $BCF_3$ of chlorothalonil were 2.040, 2.208 and 3.633, respectively, and $BCF_5$ of chlorothalonil were 6.222, 6.667 and 7.095, respectively, under the conditions. Depuration rate constants of carbaryl were 0.022, 0.022 and 0.152, respectively, when the concentration of carbaryl and chlorothalonil in combined treatment were 0.05+0.005, 0.05+0.010 and 0.10+0.005 ppm. While depuration rate constants of chlorothalonil were 0.004, 0.004 and 0.006, respectively, under the same condition of pesticide concentrations. It was observed that no significant differences of carbaryl and chlorothalonil concentration in fish extracts, test water and $BCF_s$ of carbaryl and chlorothalonil between combined treatment and single treatment. It was considered that no appreciable interaction at experimental concentrations was due to low concentrations, 0.005~0.1 ppm. Co-existence of carbaryl and chlorothalonil had no effect on excretion of each pesticide and depuration rate of chlorothalonil was investigated 1/8 slower than that of carbaryl in combined treatment. Therefore, it is considered that the persistence of chlorothalonil in fish body would be higher than that of carbaryl.

  • Five strains of gram-negative and yellow-pigmented bacteria were recently isolated from diseased freshwater fishes in Korea. All isolates were confirmed as a known fish pathogen of columnaris disease, Cytophaga columnaris based on their colonial and cellular morphology, and on physiological, biochemical and antigenic characteristics. Although the isolates were from different fish species, their characteristics of them were very similar to those of the reference strains of C columnaris (NCMB $2248^T$ and EK 28). Also, profiles of OMPs of Korean isolates were similar to those of the reference strain, C columnaris NCMB $2248^T$ when analyzed by SDS-PAGE.

  • Cyprinids are popular species for aquaculture worldwide, with Asia being a significant contributor to their production. In Korea, common carp (Cyprinus carpio), koi carp (Cyprinus rubrofuscus), and goldfish (Carassius auratus) are cultivated domestically and imported for ornamental or human consumption purposes. Among the viruses that infect cyprinids, cyprinid herpesvirus-2 (CyHV-2), koi herpesvirus (KHV, also known as cyprinid herpesvirus-3), and carp edema virus (CEV) are of particular concern as they cause substantial economic losses to the aquaculture industry. In this study, we investigated these viruses in both of domestic and imported cyprinids. Our results revealed that CyHV-2 was only detected in imported goldfish from Thailand. To further investigate the genetic characteristics of them, the marker A region was analyzed. Despite belonging to the same cluster with isolates from China, France, Poland, and Israel, CyHV-2 detected in this study showed distinct differences in their repetitive sequence sizes. Furthermore, two different forms of KHV/CEV coinfection were identified from domestic koi carp, both of which exhibited typical symptoms. Phylogenetic analysis showed that one KHV isolate (ScKc-2105-K) was of the Asian type and closely related to isolates from Japan, Indonesia, Belgium, Taiwan, and China. Two CEV isolates (ScKc-2105-CE and GhKc-2207-CE) be- longed to the IIa type and showed high similarity with isolates from the USA, France, and Korea. Notably, koi carp injected with cultured KHV (ScKc-2105-K) showed 78.0% cumulative mortality within 14 days post-injection (dpi). Our findings support the importance of regular surveillance of viral diseases in cyprinids.

The Biodegradability and Toxicity of Polyoxyethylene-Alkyl Citric Diester-Triethanolamine Detergent (Polyoxyethylene-Alkyl Citric Diester-Triethanolamine의 생분해 및 독성에 관한 조사 연구)

  • The authors investigated the biodegradability and toxicity of polyoxyethylene-alkyl citric diester-triethanolamine(PAT), which is one of new non-ionic detergents. 1) The PAT is very biodegradable, which 50% is biodegraded for one day and 100% for eight days by the activated sludge treatment and 40% for one day and 93% for eight days by the aerobic domestic sewage treatment. 2) $LD_{50}$ of PAT for chicken, mouse, and rat by oral adminstration are 7.1g/kg, 8.4g/kg and 14.0g/kg reactively. 3) $TL_{m}$ of PAT for goldfish is 64.0mg/l in 24bours. 4) No reaction for humun skin is determined by PAT.

Studies on the absorption of molecular compounds of aminopyrine (Aminopyrine 분자화합물의 흡수에 관한 연구)

  • The absorptions of aminopyrine, molecular compounds of aminopyrine and mixed compounds of aminopyrine were studies in the small intestine of rats (in situ). The molecular compounds of aminopyrine were more absorbed than aminopyrine and mixed compounds of aminopyrine were less absorbed than aminopyrine in small intestine of rats. The appearent permeability coefficients and the absorption velocity constants of the molecular compound of aminopyrine-barbital were highest and the orders of decreasing in values of permeability coefficients and the absorption velocity constants of the other molecular compounds are as follows: Aminopyrine-secobarbital, Aminopyrine-phenobarbital Aminopyrine-amobarbital, Aminopyrine-cyclobarbital Aminopyrine-allobarbital. The orders of decreasing in values of the appearent permeability coefficients and the absorption velocity constants of the mixed compounds of aminopyrine are as follows: Aminopyrine secobarbital, Aminopyrine allobarbital Aminopyrine cyclobarbital, Aminopyrine amobarbital Aminopyrine phenobarbital, Aminopyrine barbital. The relative absorption rates of aminopyrine, molecular compounds of aminopyrine and the mixed compounds of aminopyrine by the goldfish method and the partition coefficients were correlative to the values of circulation method.

  • In inland aquaculture, a lot of fishes often died cause of touch of L. carteri which is attached on the net cage. In this works, the experiment were carried out on classification of species, toxin test, and extermination with some chemicals. A bryozoa from net cage in lake Okjeong is classified into the Phylactolaemata, Lophopodella carteri Hyatt. A bryozoa broke out firstly on the 30th of June with water temperature up to $31^{\circ}C$ and it reattached on the net with water temperature below $30^{\circ}C$. Size of bryozoa colony is about $0.8{\times}0.6{\sim}1.7{\times}1.5cm$. Water column of attachment was 0~5m and the peak is 2~3m. In toxin test on the israeli carp, goldfish and catfish touched with bryozoa for 1 minute at $25^{\circ}C$ of water temperature, a dead fish appeared at 20 minutes after touched. Mortality was 90% for israeli carp and 100% for goldfish at 100 minuties after touched, but catfish was not died at all. It was supposed that the toxin is from nematocyst being around tentacles and this toxin act a deadly poison on a israeli carp and goldfish. In extermination test, the bryozoa treated with 300ppm of formalin, 5% of sodium chloride, 5ppm of malachite green, 200ppm of potassium permanganate, 1000ppm of potassium iodide, 10ppm of DDVP for minute respectively, all of them were not effected.

The Effects of Temperature on Maintaining the Stability of Water Quality in Biofloc-based Zero-water Exchange Culture Tanks (Biofloc을 기반으로 한 무 환수 사육 시스템의 수질 안정 유지에 미치는 수온의 영향)

  • This study explored adequate water temperature ranges for maintaining stable water quality in a biofloc- based zero-water exchange culture system. Five experimental tanks with the following temperatures were set up: 10℃, 15℃, 20℃, 25℃, and 30℃. First, a biofloc-based culture system was developed in the experimental tanks; then, the tanks were stocked with goldfish and went without a water exchange for 60 days. Conditions for developing a biofloc-based culture system and stable water quality in low concentrations of inorganic nitrogen compounds at 10℃, 15℃, 20℃, 25℃, and 30℃ were maintained after 17, 26, 43, 68, and 78 days, respectively. Beginning from when the goldfish were stocked in the biofloc-based culture tanks, concentrations of $NH_4{^+}-N$ remained constant and at low levels at 10℃ and 15℃, but they showed a gradual increase at 20℃, 25℃, and 30℃. Concentrations of $NO_2{^-}-N$ and $NO_3{^-}-N$ at 10℃ and 15℃ did not remain at low levels and immediately increased. While $NO_2{^-}-N$ concentrations at above 20℃ remained constant and stable at relatively low levels, $NO_3{^-}-N$ concentrations showed a gradual increase. Conditions of 15℃ and below could not maintain low and stable concentrations of $NO_2{^-}-N$. In the pH range of 4.0 to 6.0, $NH_4{^+}-N$ concentration decreased as the pH rose. However, there was no correlation between pH and $NH_4{^+}-N$ concentration in the pH range of 6.0 to 8.0. These results indicate that pH levels should be kept at pH 6.0 and above to maintain a low and stable concentration of $NH_4{^+}-N$ at above 20℃.
