Expression of Maturation-Related Genes and Leptin during Sexual Maturation in the Female Goldfish: Effects of Exogenous Kisspeptin

  • Kisspeptin (Kiss) and its cognate receptor, kisspeptin receptor (KissR; G protein coupled receptor 54, GPR54), have recently been recognized as potent regulators of reproduction in teleosts. Additionally, leptin plays an important role in energy homeostasis and reproductive function in teleosts. The purpose of this study was to examine differences in the concentration of the hormones of the Kiss/KissR system and leptin and the expression of their underlying genes, all of which are involved in the sexual maturation of female goldfish, Carassius auratus, following treatment with Kiss. The expression levels of KissR increased after the Kiss injection. Furthermore, the peptide hormone leptin also increased after the injection (in vivo and in vitro). Additionally, the expression of GnRH and GTHs (GTHα, FSHβ, and LHβ) increased in the brain and pituitary (in vitro and in vitro). These results support the hypothesis that Kiss plays important roles in the direct regulation of the hypothalamus-pituitary-gonad axis and leptin in goldfish. Therefore, we suggest that Kiss system gene expression is correlated with energy balance and reproduction.

Contractile Activity of Goldfish Intestine Exposed to 18F Isotope (18F 동위원소 피폭에 의한 금붕어 장관 평활근의 수축활성)

  • Goldfish were exposed to radioactive isotope $^{18}F$, which is used for positron emission tomography (PET). $^{18}F$ is created when the oxygen in water combines with hydrogen on exposure to positrons accelerated by a cyclotron. The temporal change in intestine contractility was measured by physiograph after the exposure to $^{18}F$. The distance between the goldfish and 580 mCi of $^{18}F$ was approximately 4 cm and the exposure was for 4 hrs. The absorption level calculated from the distance, exposure time, and half-life of $^{18}F$ was approximately 2 Gy. The contractile activity of goldfish intestine was lowest on the first day and increased gradually to 100% of the control level by a period of between 5 and 36 days.

Determination of Short-term Bioconcentration Factor on Carbofuran in Carassius auratus (goldfish) (Carassius auratus(goldfish)를 이용한 Carbofuran의 단기간 생물농축계수의 측정)

  • The Bioconcentration factor(BCF) is used as an important criterion in the risk assessment of environmental contaminants. Also it can be used as indicator of biomagnification of environmentally hazardous chemicals through food-chain as well as a tool for ranking the bioconcentration potential of the chemicals in the environment. This paper reports the measured BCF value on carbofuran in Carassius auratus(goldfish), under steady state, and examined corelation between the BCF value and the depuration rate constant. Carassius auratus(goldfish) was chosen as test organism and test periods were 1-day, 3-day and 5-day. Experimental concentrations were 0.05, 0.10 and 0.50 ppm. Carbofuran in fish tissue and in test water was extracted with n-hexane and acetonitril. GC-ECD was used to detect and quantitate carbofuran. The depuration rate of carbofuran from the whole body of goldfish is determined over the 24-h period after treatment. The obtained results were as follows: 1. It was possible to determine short term BCFs of carbofuran through relatively simple procedure in environmental concentrations. 2. $BCF_1$ of carbofuran in concentration of 0.05, 0.10 and 0.50 ppm were 1.66, 1.64 0.61, $BCF_3$ were 2.08, 2.14, 0.66 and $BCF_5$ were 2.21, 2.57, 0.86, respectively. 3. Carbofuran concentration in fish extract was increased as increasing test concentration and prolonging test period, but $BCF_s$ in concentration of 0.50 ppm was greately decreased. 4. Determined deputation rate constants of carbofuran in concentration of 0.05, 0.10, 0.50 ppm were 0.076, 0.082 and 0.089, respectively. 5. It is considered that great decrease of $BCF_s$ in concentration of 0.50 ppm is due to high water solubility and stability of carbofuran in testwater. 6. It is suggested that low BCF of carbofuran is due to its relatively high water solubility and depuration rate, compared to BPMC, carbaryl and chlorothalonil.

Combined Effect of Lidocaine Hydrochloride with Some Surfactants ( I ) (유화제(乳化劑)와 Lidocaine.HCl의 병용효과에 관(關)한 연구(硏究)( I ))

  • The effect of some surfactants, such as tween 40, tween 80, span 80, and triethanolamine, on the absorption of lidocaine was studied using goldfish as a model of absorption kinetics. This model was believed that reciprocal overturn time and death time were the occurrence of biological effect versus drug absorption. The results were as follows; The threshold concentration of lidocaine was significantly reduced by the surfactants. Overturn time and death time of goldfish in the lidocaine solution were reduced by such surfactants as tween 40, tween 80, span 80 and triethanolamine, and more reduced with increasing concentration of surfactants except tween 40 and tween 80. The plots of reciprocal death time versus lidocaine concentration were linear relationship with a positive concentration intercept such as minimum effective concentration (0.2-0.8%). As results it is believed that these surfactants may form complexes with lidocaine or affect the goldfish membrane to make more permeable to lidocaine, which is adsorbed more rapidly.

  • Goldfish retina has been well studied to a great extent. In spite of that, electrical characteristics of dissociated horizontal cells(HCs) have not been identified in detail. Thus the cone-and the rod- HCs dissociated from goldfish retina were investigated electrophysiologically using whole-cell patch-clamping recording. To explore the basic electrical property, We examined voltage-dependent channels in all types of HCs. For the futher understanding of GABAergic pathway, the localization and distribution of GABA receptors was examined in cone- HCs including HC axon terminals(ATs).

Two Types of Voltage-activated Calcium Currents in Goldfish Horizontal Cells

  • In horizontal cells (HCs) that were freshly dissociated from goldfish retina, two types of voltagedependent calcium currents ($I_{Ca}$) were recorded using a patch-clamping configuration: a transient type current and a sustained type current. The cell was held at -40 mV, and the prepulse step of -90 mV was applied before command pulse between -65 and +55 mV. The transient $Ca^{2+}$ current was activated by depolarization to around -50 mV from a prepulse voltage of -90 mV lasting at least 400 ms and reached a maximal value near -25 mV. On the other hand, the sustained $Ca^{2+}$ current was induced by pre-inactivation for less than 10 ms duration. Its activation started near -10 mV and peaked at +20 mV. $Co^{2+}$ (2 mM) suppressed both of these two components, but nifedipine ($20{\mu}M$), L-type $Ca^{2+}$ channel antagonist, blocked only the sustained current. Based on the activation voltage and the pharmacolog$I_{Ca}$l specificity, the sustained current appears to be similar to L-type $I_{Ca}$ and the transient type to T-type $I_{Ca}$. This study is the first to confirm that transient type $I_{Ca}$ together with the sustained one is present in HCs dissociated from goldfish retina.

Physiological Response and Hematological Characteristics of Goldfish (Carassius auratus) to Water Temperature Shock (수온충격에 따른 금붕어(Carassius auratus)의 혈액 성상 및 생리적 반응)

  • The effects of sudden changes of water temperature (WT) on the physiological response and hematological characteristics of the goldfish (Carassius auratus) were examined by manipulating WT in a flow through freshwater culture system with tanks. The WT was dropped from $15^{\circ}C$ to $10^{\circ}C$ within 1 hour and then returned to the original water temperature within 12 hours and maintained for 12 hours at the normal WT. The WT stress give continued for 3 days. Plasma levels of cortisol, glucose and lactic acid were higher in stress group than that of non-stressed group until 72 hours. However, The $Na^+,\;Cl^-$, osmolality and aspartate aminotransferase (AST) levels showed no significant differences in two groups. This results in stress group showed that goldfish exhibit "typical" physiological responses (in cortisol, glucose, lactic acid, hematocrit, red blood cell and hemoglobin) to the stress induced by WT changes.

The Effects of Different Wavelengths of Light-Emitting Diodes on the Expression of Reproduction-Related Genes in Goldfish Carassius auratus

  • We investigated the differences in the expression of the neurohormones kisspeptin (Kiss) and gonadotropin-inhibitory hormone (GnIH) and cytochrome P450 aromatase (P450arom), gonadotropin hormones (GTHs), and sex steroids in the goldfish Carassius auratus exposed to light-emitting diodes (LEDs). The expression levels of Kiss1, Kiss2, G-protein-coupled receptor 54 (GPR54), GTHs, GnIH, and P450arom were compared between the control (white light) and LED-treated goldfish. Furthermore, we measured the plasma levels of follicle-stimulating hormone (FSH) and luteinizing hormone (LH). The levels of Kiss1 mRNA and protein; Kiss2, GPR54, and $GTH{\alpha}$ protein; GTH mRNA; and plasma FSH and LH in the hypothalamus and cultured hypothalamus cells were significantly higher in the green and purple LED treatment groups than in the other groups. These results suggested that red LEDs inhibit the sex maturation hormones, Kiss, GPR54, GTHs, and P450arom, and that GnIH plays a role in the negative regulation of reproductive function in goldfish.

The Optomotor Response of Crusian Carp and Goldfish in Accordance with the Colors of the Wall and the Stripes of the Visual Screen (시각막의 바탕과 줄무늬의 색상에 따른 붕어와 금붕어의 시각운동 반응)

  • The author carried out an experiment to find on the optomotor response of crusian carp, Carassius carassius and goldfish, Carassius auratus in accordance with the colors of the wall and the stripes labeled on the revolving visual screen. The results are as follows; 1. The optomotor response rate of crusian carp was 80% or more in case of B-Y, R-Bl and W-Bl pair of colors, and 50% or less in case of Bl-B, B-Bl, R-W, R-Y, W-Y and Y-W pair. 2. The optomotor response rate of goldfish was 80% or more in all except B-Bl, W-R and W-Y pair. 3. Mean round number of crusian carp was 7.5-9.5 revolutions in case of W-Bl, Y-Bl, W-B and Y-B pair, and 1.8-3.9 revolutions in case of Bl-B, Bl-Y, B-Bl, B-R, R-All and Y-W pair for 3 minutes. 4. Mean rounded number of goldfish was only 4.1-4.9 revolutions in case of W-Bl, W-B and Y-B pair for 3 minutes.

Oxidative Stress and Apoptosis in Goldfish (Carassius auratus) Caused by Exposure to Different Concentrations of Micro-polystyrene

  • Microplastic contamination in waterbodies is a growing source of concern for researchers and other stakeholders. We investigated oxidative stress and toxicity in goldfish (Carassius auratus) in response to exposure to 1-㎛ diameter micro-polystyrene (MP) at concentrations of 0, 10, 100, and 1000 beads/mL (MP 0, MP 10, MP 100, and MP 1000 groups) for 7 d (at day 0, 1, 3, 5, and 7). We analyzed the survival rates; superoxide dismutase (SOD) and catalase (CAT) mRNA expression levels in the liver; SOD and CAT activity in the plasma; caspase-3 mRNA expression in the liver; and the levels of hydrogen peroxide (H2O2) in plasma. Terminal transferase dUTP nick end labeling (TUNEL) assays were also conducted to determine apoptosis levels in the liver. All fish in the MP 1000 group died by day 7 and the MP 100 group had a lower survival rate than the MP 10 and MP 0 groups. The mRNA expression as well as SOD, CAT, and caspase-3 activity levels were increased significantly with increases in MP concentration and exposure time. Finally, according to the TUNEL assay, more apoptosis was observed in the MP 1000 group at day 5 than in other groups. In summary, MP concentrations above 100 beads/mL caused death and oxidative stress to goldfish. We conclude that MP can cause oxidative stress and apoptosis in goldfish, which leads to death.
