Relationship between the mRNA Expression Levels of Sperm Phospholipids Hydroperoxide Glutathione Peroxidase and Heparin-Binding Protein, and in-Vivo Fertility in Boars (돼지 정자내 Phospholipids Hydroperoxide Glutathione Peroxidase와 Heparin-Binding Protein의 발현 수준과 번식 능력의 관계)

  • The objective of the study was to determine the relationship between the mRNA expression levels of sperm phospholipids hydroperoxide glutathione peroxidase (PHGPx) and heparin-binding protein (HBP), and in-vivo fertility in boars. The farrowing rate was not correlated with litter size. Sperm PHGPx mRNA expression level of the larger litter size (over 10) group $(2,414.7{\pm}400.7)$ was high that of smaller litter size (below 8) group $(1,875.8{\pm}311.2)$. Sperm HBP mRNA expression level was also higher in the larger litter size $(2,255.9{\pm}360.8)$ group than the smaller litter size $(2,155.4{\pm}378.0)$. However, significant differences were not observed. Sperm PHGPx mRNA level was correlated positively with litter size (r=0.206). Because the expression levels of PHGPx and HBP are not strongly correlated with in-vivo fertility, PHGPx and HBP can not be considered a predictive measure for fertility in boars.

Effects of Salt and Drought Stresses on Seed Germination and Gene Expression Pattern in Tall Fescue (염과 건조 스트레스 조건에서 톨 페스큐의 종자 발아율과 유전자 발현 변화분석)

  • Salinity and drought stresses are probably the most significant abiotic factor limiting plant's growth, also negatively affect seed germination and early seedling development. To study on effect of NaCl and PEG stress on seed germination and gene expression pattern of tall fescue, the levels of NaCl and PEG-induced water stresses were determined in first experiment. Different concentration of NaCl (0 to 350 mM) and PEG (0 to 30%) were used for seed treatment. Seed Germination percentage reduced with increasing osmotic potential of growth medium either due to NaCl or PEG. Seeds were not germinate at 350 mM NaCl or 30% PEG treatment. On the basis of the results, Kentucky31(E-) had more resistant than Fawn in both stress conditions. Furthermore, we have used an annealing control primer-based differential display reverse transcription-polymerase chain reaction method to identify salt- and drought stress-induced differentially expressed genes (DEGs) in tall fescue leaves. Using 120 annealing control primers, a total of 4 genes were identified and sequenced. The possible roles of the identified DEGs are discussed in the context of their putative role during salinity and drought stresses.

STUDIES OF GINSENG EXTRACT ON AGE-RELATED ENZYMES (고려인삼 추출물이 노화관련효소에 미치는 영향)

  • 노화는 모든 다세포 유기물의 특징이다. 노화가 됨에 따라서 효소활성 및 면역반응의 감소와 과산화지질과 지방갈색물질의 축적, 효소와 염색질을 포함하는 단백질 구조의 변화, 호르몬계의 불균형 등이 일어난다. 그렇지만, 노화가 어떻게 일어나는지에 관하여는 현재까지 확실하지 않다. 본 연구진은 노화와 관련된 효소들에 관하여 연구를 하여 왔으며, 노화가 진행되는 동안의 효소의 활성을 유지시켜주거나, 또는 효소의 활성이 감소되는 것을 지연시켜 주는 물질을 찾고자 노력하였다. 그 가운데 하나로서, 고려인삼 추출물을 흰쥐에 기간별로 투여하여 효소활성의 차이, 열에 대한 안정성, 기질에 대한 친화력, 전기 영동 상의 이동성과 면역적인 반응을 대조군과 비교하였다. Glucose-6-phosphate dehydrogenase, 6-phosphog-luconate dehydrogenase, glutathione redutease, glutathion peroxidase와 같은 노화와 관련된 효소들의 활성을 고려인삼 추출물을 1개월간 흰쥐에 ($60{\~}80$g)투여하여 대조군과 비교 조사하였으나, 별 차이가 없었다. 그러나 고려인삼 추출물을 15개월간 투여하였을 때에는 이러한 노화관련 효소들의 활성이 급격히 증가함이 ($70{\~}200\%$) 관찰되었다. 예견된 바와 같이, 효소의 열에 대한 안정성과 기질에 대한 친화력도 증가함이 관찰되었다. 그러나 glucose-6-phosphate dehydrogenase의 경우에서 전기영동상의 차이 및 면역적인 반응은 대조군과 유사하였다. 이상의 결과는 고려인삼 추출물이 노화와 관련된 효소들의 활성이 감소되는 것을 지연시켜줄 수 있으며, 노화를 어느정도 지연시켜 줄 수 있음을 의미한다. 이와 같은 결과를 포함한 실험자료를 국제 인삼심포지움에서 발표할 것이다.$-tocopherol의 항 산화작용을 더욱 효과적으로 촉진 시킬 것으로 생각된다.-L(독성 호르몬-L)의 작용을 억제함으로서 암환자의 체중 감소를 방지하고, 식욕감퇴를 개선할것으로 생각된다.해되었으며, $Rb_{1}$은 장내의 효소와 tetracycline-resistantant bacteria에 의해 Rd와 2 종류의 미확인 물질로 분해되었다.xA_{2}$ synthetase 억제제인 imidazole의 효과와 유사하였다. 4. G-Re는 $1{\times}10^{-5}g/ml$ 이하의 농도에서는 효과가 없으나 $1{\times}10^{-4}g/ml$ 이상의 농도에서 농도의존적으로 유의성 있는 $PGE_{2},\;PGF_{2}{\alpha},\;TXB_{2}$의 생성억제와 함께 6-keto-$PGF_{1}{\alpha}$ 증가를 보였다. 이는 prostacyclin synthetase를 자극하는 serotonin의 효과와 같은 작용으로서 prostacyclin synthetase 억제제인 tranylcypromine에 대하여 길항효과를 보였다. 5. $TxB_{2}$생성억제 작용을 나타내는 ginsenoside들의 효과를 뒷받침하기 위하여 인삼 saponin 성분을 전처치한 patelet rich plasma에서 혈소판 응집시험 결과, ADP로 유도된 혈소판 응집반응에는 모든 인삼 saponin 성분들이 효과가 없었으나 arachidonic acid로 유도된 혈소판 응집반응에는 $G-Rb_{2}$, G-Rc, G-Re의 순으로 농도 의존적인 억제현상을 보였다. 이상의 결과와 같이 인삼 saponin 성분들은 arachidonic acid로부터 cyclooxygenase를

Effect of Aging on the Liver Damage in Bromobenzene-pretreated Rats (연령이 다른 흰쥐에 Bromobenzene 미치는 영향)

  • To evaluate an effect of growth periods on the bromobenzene-induced liver damage, bromobenzene was administrated to 5-week-old rats and 10-week-old rats pretreated with bromobenzene 5 times every other day for 10 days and then the animals were sacrificed. The results were obtained as follows; The increasing rate of serum levels of alanine aminotransferase, xanthine oxidase activity, hepatic lipid peroxide contents, liver weight per body weight (%) and decreasing rate of hepatic contents of protein to each control group were higher in 10-week-old rats than 5-week-old rats by the pretreatment of bromobenzene. According to the above results, 10-week-old rats indicated more severe liver injury than 5-week-old those in case of bromobenzene pretreatment. On the other hand, hepatic aniline hydroxylase activity was more increased in 10-week-old rats than 5-week-old rats both in control and bromobnezene pretreated rats where as the reverse in hepatic glutathione S-transferase. In case of hepatic GSH determination at the intervals of 2, 4, 8, 24 hours throughout 24 hr after administration of single dose of bromobenzene to 5-week-old and 10-week-old rats both in control and bromobnezene pretreated, the rate of GSH utilization was lower in 10-week-old rats than 5-week-old rats. In conclusion, from the above experimental, it is deduce that the 10-week-old rats showed more severe liver injury than 5-week-old rats by the bromobenzene treatment because the disposal ability of bromobenzene in liver was lower in 10-week-old rats than 5-week-old rats.

Effect of Tota1 Saponin from Red Ginseng on Acvtivities of Antioxidant Enzymes in Pregnant Rats (홍삼 사포닌이 수태중인 흰쥐의 항산화 효소활성에 미치는 영향)

  • Pregnancy is a physiological state accompained by a high energy demand of many bodily functions and an increased oxygen requirement. Because of the increased intake and utilization of oxygen, increased levels of oxidative stress would be expected. So we observed the activities of the hepatic antioxidant enzymes from rat treated with total saponin from the red ginseng against free raicals produced in pregnant rats. The activity of superoxide dismutase (SOD) in the control group was slightly decreased during pregnancy, and SOD activity in total saponin treated group was not observed any siginificant change compared with the control group. The activities of glutathione peroxidase (GPX), glutathione reductase (GRD) and catalase in the control group have shown the decreasing tendency during pregnancy, whereas the activities of GRD and catalase in total saponin treated group showed significant increased tendency compared with the control group. GPX activity in total saponin treated group was slightly decreased tendnency compared with the control group. The activity of glutathione-S-transferase (GST) in the control group was increased to keep the state of homaeostasis tendency in pregnant rats. On the other hand, the activity of GST after total saponin treatment was increased than control group. Activity of all enzymes in the control group and total saponin treated group recovered the normal level after delivery of rats. In spite of the physiological changes in vivo, the inflaunce of total saponin on activaties of hepatic antioxidant enzyme in pregnant rats seems to be regulated the biological homeostatic adaptation mechanism which protects the maternal liver aganist oxygen induced toxicity

Comparison of toxicity and detoxifying enzyme activity in carp (Cyprinus carpio) treated with some synergistic pesticides (농약 상호간의 협력작용에 의한 잉어의 독성과 해독효소 활성의 비교)

  • This study was performed to investigate effects of probable detoxifying enzyme activity and toxicity by pesticides and their combinations in the fresh water fish. Seven pesticides including IBP, isoprothiolane, cartap, ridomil, chlorothalonil, captafol and endosulfan were subjected to investigate for their acute toxicites and synergism possibilities. The $LC_{50}$ value of endosulfan was the lowest at showing 0.0079 ppm and that of metalaxyl was the highest as showing 40 ppm over. The synergism effects of relative pesticides were observed in the combinations of isoprothiolane+IBP and isoprothiolane+cartap. The changes of glycogen contents in fish liver were assayed for 5 pesticides and its highest inhibition effect of glycogen showed in IBP treated fish. The activity of probable detoxifying enzymes including carboxylesterase (CE), glutathion S-transferase (GST) and lactate dehydrogenase (LDH) were assayed in carp liver at dose of sublethal concentrations. Effects of pesticides on changes in each enzyme activities were as follows: carboxylesterase (CE) activities were the highest in IBP and gtutathion S-transferase (GST) activities were the highest in iosoprothiolane+IBP. Both activities of carboxylesterase (CE) and glutahtion S-transferase (GST) were increased by 5 chemicals. The highest LDH activity showed in isoprothiolane treated fish, while the lowest activity was observed in isoprothiolane+cartap. Sublethal exposure to cartap and isoprothiolane+cartap in carp exerted various effects on LDH activity.

Effect of Alcohol Detoxification Beverage that Contained Bulnesia sarmienti on Alcohol-metabolizing Enzymes and Antioxidant Enzyme Activities (Bulnesia sarmienti 를 함유한 숙취해소 음료가 알코올대사 및 항산화 효소활성에 미치는 영향)

  • This study was carried out to investigate the effect of a beverage that contained Bulnesia sarmienti(BSP, 2.5%) on rats to which alcohol was administered. The treatment of the BSP group reduced the serum alcohol concentration to 52%, compared to 47% in the positive control(PC) group. Similar pattell1s were observed in the enhancement of alcohol dehydrogenase(ADH), acetaldehyde dehydrogenase(ALDH), alkaline phosphate(ALP), alanine aminotransferase(ALT), asparate aminotransferase(AST), total cholesterol(CHOL), ${\gamma}$-glutamyltrasferase(GGT), glucose(GLU), total bilirubin, and total protein(TP) in the serum. Also, in the BSP group, the lipoxidase(LPO), glutathion-S-transferase(GST), XO, catalase(CAT), and superoxide dismutase(SOD) were significantly reduced, compared to the CO and PC groups in the liver. The glutathione(GSH) activity increased in the BSP group, though. These results indicate that Bulnesia sarmienti extract can enhance alcohol metabolization activity.

Effects of Laminaria Japonica Extract Supplement on Blood Glucose, Serum Lipids and Antioxidant Systems in Type ll Diabetic Patients (다시마추출물이 제2형 당뇨병 환자의 혈당, 지질 및 항산화 체계에 미치는 영향)

  • We performed a randomized double-blind placebo-controlled trial to determine whether Laminaria japonica extract (LJE) supplement modulates blood glucose, serum lipids and antioxidant systems in type II diabetic patients. We also measured critical parameters assessing safety in liver and kidney functions after LJE supplement. A total of 37 patients (18 males and 19 females) were randomized to either LJE group or placebo group. The treatment group received four 350 mg of LJE capsules (1.4 g, total) per day for 12 weeks. The placebo group received the same dose of cellulose capsules. Baseline characteristics regarding general life style and dietary intake pattern were similar between the two groups. There were no significant influences of LJE supplement except for waist circumference on anthropometric parameters. As the whole, 12 weeks of LJE supplement resulted in a little decrease in fasting blood glucose (FBG) and glycated hemoglobin (HbA1c), but a significant decrease was not observed. Total cholesterol, LDL-cholesterol and triglyceride concentrations were significantly (p<0.05) lowered in LJE group. The antioxidant enzymes, glutathion peroxidase (GSH-px) and superoxide dismutase (SOD) levels were elevated in the LJE group (p<0.05) compared to the placebo. The increase of these enzymes was associated significantly with the decrease of MDA concentration (p<0.05). Furthermore, LJE supplement showed no adverse effects on the functions of liver and kidney. Findings from this study suggest that LJE supplement can help improve serum lipid status in type II diabetic subjects without adverse effects.

Synergistic action of pesticide mixtures using glutathione-s-transferase- and esterase-inhibiting properties in diamondback moth (Plutella xylostella L.) (Glutathione-S-transferase와 esterase 효소 저해특성을 이용한 농약의 혼합 상승효과)

  • In vitro inhibitory activity of 34 insecticides and 31 fungicides to glutathione-S-transferase and esterases extracted from rats was determined. Of tested pesticides, the pesticides with high activity on both detoxifying enzymes were mixed with pesticides that are known to be detoxified by detoxifying enzymes. Glutathione-S-transferase was inhibited by thiodicarb $(I_{50}:1.87\times10^{-4}M)$, thiocyclam $(7.40\times10^{-4}M)$, dithianon $(7.55\times10^{-5}M)$, and tolylfluanide $(8.66\times10^{-5}M)$, while esterases by dichlorvos $(8.95\times10^{-8}M)$, pirimicarb $(2.74\times10^{-6}M)$, pyrazophos $(3.31\times10^{-5}M)$, and benomyl $(4.96\times10^{-5}M)$. After acephate known to be detoxified by glutathione-S-transferase was mixed with glutathione-S-transferase-inhibiting pesticides and phenthoate known to be detoxified by esterases was mixed with esterases-inhibiting pesticides, insecticidal activities of such mixtures were determined against diamondback moth (PlutelLa xylostella L.). Synergistic action was observed in all pesticide combinations. The highest synergistic action was obtained when phenthoate was combined with dichlorvos, showing that co-toxicity coefficients were 1512 and 1877 after 24 and 48 hours of treatment, respectively. Several other combinations of pesticides, such as phenthoate with benomyl, and acephate with dithianon, also showed synergism, showing that their co-toxicity coefficients were about 1,000 and 500, after 24 hours of treatment, respectively. Our results showed that combinations of pesticides inhibited by detoxifying enzymes and ones detoxified by detoxifying enzymes resulted in increased toxicities of pesticides, suggesting that such combinations could be used to develop pesticide mixtures with more broad spectrum and high effectiveness.

Hepatic Oxygen Free Radical Metabolizing Enzyme Activities and Serum Lipid Profile in Rats Fed Diet Supplemented with Monascus Pigment (흰쥐에 있어서 홍국 첨가 식이가 혈청 지질성분 및 간조직의 유해산소 대사효소활성에 미치는 영향)

  • To investigate the hepatic oxygen free radical metabolizing system and changes of serum cholesterol levels in rats fed a diet supplemented with Monascus pigment (MP), Sprague-Dawley rats weighing about 300 g have been fed a diet supplemented with 2% or 4% MP for a month. The rats fed 2% MP supplemented diet gained less body weight than the control rats and those fed 4% W supplemented diet. Those fed 2% or 4% MP supplemented diet had no remarkable changes in liver function on basis of liver weight/body weight, serum levels of xanthine oxidase, alanine amino transferase activity In rats fed 2% and 4% MP supplemented diet, hepatic cytochrome P45O dependent aniline hydroxylase activity significantly (p<0.05) declined about 32%, 37% respectively and showed no significant differences between rats fed 2% and 4% MP supplemented diet whereas those fed 2% MP supplemented diet showed about 29% increased hepatic xanthine oxidase activity. And hepatic glutathione S-transferase and glutathione peroxidase activites in rats fed 2% MP supplemented were more increased by about 17%, 28% respectively than the control rats. There were no significant differences both in between those fed 2% and 4% MP supplemented diet. Especially rats fed 2% or 4% MP supplemented diet showed a significant (p<0.05) increase in hepatic catalase activity by 41%, 25% compared with control rats and those fed 4% MP supplemented diet showed more decrease in tendency of catalase activity than those 2% MP supplemented diet. But hepatic superoxide dismutase activity and glutathione content were appeared to be similar value among three groups. On the other hand, rats fed 2% MP supplement diet showed 17% increased levels of serum HDL-choresterol and 26% decreased value of LDL-cholesterol and serum level of triglyceride. But no different value were appeared between those fed 2% and 4% MP supplemented diet. Especially in those fed 2% and 4% MP supplemented diet, artherogenic index were significantly (p<0.05) declined by 37%, 29% respectively compared with control. In conclusion, it is likely that rats fed a diet supplemented with a proper quantity of MP may have the potential of oxygen free radical detoxication and lowering of artherogenic index.
