Purpose: This study was carried out to determine the protective effects of vitamin C on the hepatotoxicily induced by radiation. Materials and Methods : The Spraque Dawley rats were randomly divided into 3 groups; the control group, the radiation exposed group, aud the radiation and vitamin C-treated group. SOD activity, ca-talase, malondialdehyde and liver enzymes were analyzed to assess the antioxidant effects of vitamin C. Results: The increased level of malondialdehyde and the decreased catalase activity that were induced by radiation were improved after vitamin C but there was no statistical significance among three groups. The superoxide dismutase activity of the liver was increased by vitamin C, but there were no statistically significant differences between the vitamin C-treated group and the non vitamin C-treated group. The level of liver enzymes in sera such as glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, lactate dehyrogenase and alkaline phosphatase were remarkably elevated by radiation. The levels of those enzymes were decreased in the vitamin C-treated group and statistical significance was noted for the GPT level ($p<0.01$). On the electromicrographic findings, the hepatic cell destruction was considerably decreased in the vitamin C-treated group. Conclusion: Vitamin C is thought to be an effective antioxidant against the hepatotoxicity induced by radiation.
A total of fifty Hanwoo steers raised from 14 to 29 months were used to investigated the effects of dietary mugwort on the performance and meat quality during refrigerated storage. A feeding trial was tested for 14.7 months and experimental diets included 0 and 2% dried wild mugwort. Total weight gain and average daily gain in this study were significantly higher in mugwort treatment than control treatment. Feed efficiency was improved by mugwort treatment compared with control treatment. Any difference in GOT (glutamic oxaloacetic transaminase), GPT (glutamic pyrubic transaminase), and BUN (blood urea nitrogen) was not observed between groups. Carcass weight and longissimus muscle area were higher for mugwort treatment than control. As fattening increased, total cholesterol in blood increased. Total cholesterol in blood tended to be lower in mugwort treatment than control treatment (p<0.05). The heating loss, $L^*$, $a^*$, $b^*$, chroma, and hue unit of all treatments were not affected treatment, but showed a significant decrease in all treatments during refrigerated storage. For fatty acid composition in longissimus muscle, dietary mugwort supplementation increased unsaturated fatty acid than saturated fatty acid (p<0.05). Based on these findings, it is obvious that supplementation of mugwort at 2% level will be useful to decrease cholesterol, increase unsaturated fatty acid, and improve growth performance of Hanwoo steers.

Purpose: Paeonia Radix Alba is a traditional herbal medicine used to treat the liver and the spleen. Many studies have reported that Paeonia Radix Alba extract (PR) affects liver injury, but there has been no study on liver injuries induced by thioacetamide (TAA). Therefore, we aimed at evaluating the effect of PR on a TAA-induced acute liver injury (ALI) model. Methods: The antioxidant activity of PR was assayed by the content of total polyphenol, total flavonoid, 1,1-diphenyl-2'-picrylhydrazyl (DPPH), and 2,2'-azino-bis (3-ethylbenzo-thiazoline-6-sulfonicacid) (ABTS) radical scavenging activities in vitro test. ALI was induced via-intraperitoneal injection of TAA (200 mg/kg body weight) for three consecutive days. Also, silymarin (100 mg/kg body weight) and PR (100 or 200 mg/kg body weight) were administered at 1 hours 30 minutes prior to TAA treatment. The levels of ammonia, glutamic oxaloacetic transaminase (GOT), and glutamic pyruvic transaminase (GPT) were analyzed using an assay kit. The expressions of antioxidant proteins including Nrf2, Keap1, HO-1, SOD, catalase, and GPx-1/2 and oxidative stress-related proteins including NOX2, p47phox, and p22phox were evaluated by the western blot analysis. Results: PR showed excellent antioxidant activity in vitro. TAA administration increased the levels of ammonia, GOT, and GPT in the ALI control group compared to the normal group, whereas it was significantly reduced by PR pretreatment. Moreover, NADPH oxidase protein expressions were upregulated after TAA treatment, while the elevated expressions were inhibited by PR pretreatment. The expressions of antioxidant protein were downregulated in the ALI control group, whereas Nrf2 activation in the PR group was accompanied by increased levels of antioxidant enzymes. Conclusion: PR administration increased the antioxidant enzymes via activation of the Keap1/Nrf2 pathway and inhibited the protein levels of NADPH oxidase factors. Taken together, these results showed that PR treatment may be considered to ameliorate acute liver injury induced by TAA.

This study aimed to verify the effect of Citrus unshiu peel water extract (CUP) on a mouse model of acute gastritis (AG) induced by HCl/ethanol. Several studies have found that CUP has anti-inflammatory effects. The AG model was induced by oral administration of 150 mM HCl/60% ethanol (550 µL) to all groups except the control group. Also, for drug treatment, sucralfate (10 mg/kg) and CUP (100 or 200 mg/kg) were orally administered for 90 minutes before induction. The effect of CUP treatment was confirmed by gross gastric mucosal damage measurement, and the levels of Glutamic Oxaloacetic Transaminase (GOT), Glutamic Pyruvic Transaminase (GPT), and myeloperoxidase were reduced as well as the levels of oxidative stress biomarkers and their related proteins. In addition, the levels of inflammatory proteins, mediators, and cytokines were significantly downregulated byPI3K/Akt signaling. Taken together, these results show that CUP treatment alleviates AG by regulating PI3K/Akt signaling.
The present study investigated the survival rate, respiration rate, plasma stress index, and histological changes according to exposure time of cultured red seabream (Pagrus major) and olive flounder (Paralichthys olivaceus) exposed to Cochlodinium polykrikoides red tide. Fish cultured in natural seawater were used as the control group. Cochlodinium polykrikoides density was set to 5,500±200 cells·ml-1 in the experimental groups. All red seabreams died within 1 hour of exposure to red tide, whereas all olive flounders died within 5 hours of exposure. Analysis of physiological response revealed that in red seabream, plasma glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), catalase (CAT), and glutathione peroxidase (GPx) concentrations were increased; plasma glucose and superoxide dismutase (SOD) concentration were decreased. Meanwhile, in olive flounders, plasma cortisol, GOT, and GPT concentrations were increased; plasma glucose concentrations were increased during the first hour of exposure, followed by decrease after 5 hours; and plasma SOD, CAT, and GPx concentrations decreased during the first hour of exposure. Histological analysis revealed structural damage to the gills of both red seabream and olive flounder. In conclusion, the exposure of red seabream and olive flounder to Cochlodinium polykrikoides red tide at the density of 5,500 cells·ml-1 induces oxidative stress, which activates antioxidant defense mechanisms, ultimately leading to liver and gill damage.
The purpose of this study was to determine the effect of external biologger attachment methods on the blood parameters and attachment efficiency of spotted sea bass Lateolabrax maculatus (mean body weight 2630.8 g). The fish were tagged using four different external attachment methods with dummy biologgers: no attachment (control), anchor attachment (AA), monofilament attachment (MA), and silicon tube attachment (SA), each with triplicates. Blood indices and biologger attachment efficiency were assessed on days 1, 7, 14, 28, 56, and 84 after attachment. The concentrations of hematocrit, Na+, Cl-, glutamic pyruvic transaminase and total protein, and the activity of superoxide dismutase in blood were not affected by the external attachment method of biologger. The concentrations of glutamic oxaloacetic transaminase (day 1 of attachment), hemoglobin (day 56) and total cholesterol (day 56 and 84) in AA group, the concentrations of glucose and cortisol (day 14) and total cholesterol (day 84) in MA group showed significantly higher than those of control (p<0.05). During the experiment period, the SA group had no differences from the control in all blood properties. The biologger attachment efficiencies of the AA, MA, and SA groups after 84 days were 0.0%, 33.3%, and 100.0%, respectively. These results indicate that the optimum external biologger attachment method under our experimental conditions is SA type.

This study was conducted to investigate the effects of several additives(macroalgae, wasabi, and herb) in formulated diets on the growth, body composition, blood chemistry and non-specific immune response of juvenile flounder. Three replicates of juveniles (average weight 8.4 g) in flow-through aquarium system were fed one of six isonitrogenous (45%) and isolipidic (8%) diets containing 5 and 10% Undaria powder, 2% wasabi leaf, 2% wasabi stem, and 0.5% herb (Obosan) for 8 weeks. Survival was not affected by the different dietary additives (P>0.05). The highest weight gain and feed efficiency offish fed the diet containing 0.5% herb were significantly higher than those of fish fed the diets containing 10% Undaria powder (P<0.05). No significant differences were found in contents of moisture, crude protein, lipid and ash of whole body (P>0.05). Fish fed the diet containing 10% Undaria powder showed the highest moisture and the lowest crude lipid contents in the liver. Although hematological parameters (red blood cell, hematocrit and hemoglobin) and serum constituents (glucose, total cholesterol and glutamic-oxaloacetic transaminase) contents of fish varied between treatments, no specific trend was observed throughout feeding periods. Lysozyme activity in the serum and nitroblue tetrazolium reduction of macrophage in the head kidney from fish fed the diets containing herb was significantly higher than those of fish fed the control diet. The results of this study suggest that herb as an additive in this formulated diet may improve growth and non-specific immune response of juvenile flounder.
The effect of conjugated linoleic acid (CLA) isomers esterified in diacylglycerol (DAG)-rich oil on lipid metabolism was investigated. Since dietary DAG has been known to induce the regression of atherosclerosis, CLA-DAG and olive-DAG oils containing similar levels of DAG (51.4~54.2%) were synthesized from olive oil. Hyperlipidemic C57BL/6J mice were then fed high-fat high-cholesterol diets supplemented with these oils (5% each) for 7 wk. The CLA-DAG diet containing 2.1% CLA isomers (0.78% c9,t11-CLA; 1.18% t10,c12-CLA) remarkably increased the levels of total plasma cholesterol and glutamic oxaloacetic transaminase (GOT) along with hepatic cholesterol and triacylglycerol (TAG) contents. Furthermore, the CLA-DAG diet inhibited fat uptake into adipose tissue whereas fat deposition (especially in the liver) was increased, resulting in the development of fatty livers. Hepatic fatty acid composition in the CLA-DAG mice was different from that of the olive-DAG mice, showing higher ratios of C16:1/C16:0 and C18:1/C18:0 in the liver. The activity of hepatic acyl-CoA:cholesterol acyltransferase (ACAT) was higher in CLA-DAG mice while plasma lecithin:cholesterol acyltransferase (LCAT) activity and the ferric reducing ability of plasma (FRAP) were lower in CLA-DAG mice compared to the olive-DAG animals. Results of the present study suggest that CLA incorporation into DAG oil could induce atherosclerosis in mice.
The objectives of present study were to investigate the effects of benzo[a]pyrene(BaP) on cytotoxicity, lipid peroxidation and antioxidant enzymes in rat hepatocyte primary culture. Primary cultures of rat hepatocytes were incubated for 24 hr, 48 hr or 72 hr in the presence of various concentrations (0, 10, 20, 30, 50 or 100 $\mu.$ M) of BaP. Cytotoxicity and cell viability were determined by measuring glutamic oxaloacetic transaminase(GOT) activity, lactate dehydrogenase(LDH) activity and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MIT) value. Lipid peroxidation was evaluated using thiobarbituric acid reactive substances(TBARS) assay. Effects on antioxidant system were determined by measuring glutathione peroxidase(GPx) activity, glutathione reductase(GR) activity and glutathione concentration. Activities of GOT and LDH, MTT value as well as TBARS concentration were not affected by up to 100 $\muM$ of BaP for 24 hr incubation. However, BaP at the concentration of 50 $\muM$ for 48 hr incubation or at the concentration of 30 $\muM$ for 72 hr incubation began to increase LDH activity and TBARS concentration but decrease MTT value, representing that BaP caused cytotoxicity and decreased cell viability in dose- and time-dependent manners. GPx activity began to be decreased by BaP at the concentration of 50 $\muM$ for 72 hr incubation. Whereas, GR activity began to be decreased by BaP at the concentration of 20 $\muM$ for 72 hr incubation. Glutathione concentration began to be decreased by BaP at the concentration of 20 $\muM$ for 72 hr incubation and was further reduced to 90% by 100 $\muM$ of BaP. These results demonstrate that BaP caused cytoctoxicity and decreased cell viability by increasing lipid peroxidation and decreasing glutathione concentration as well as activities of GPx and GR.

This study investigated the potential hepatoprotective benefits of Crassostrea gigas oyster hydrolysates. Oysters are known to have many biofunctional properties. In particular, oyster enzymatic hydrolysates produce substances with beneficial functions. The potential hepatoprotective effects of C. gigas hydrolysates against damage induced by tacrine were evaluated in vitro in HepG2 cells. Peptides were generated from C. gigas by enzymatic hydrolysis with Neutrase, Flavourzyme, or Protamex enzyme preparations. Tacrine treatment induced considerable cell damage in HepG2 cells, as shown by significant leakage of glutamic oxaloacetic transaminase (GOT) and lactate dehydrogenase (LDH). Cells treated with C. gigas hydrolysates showed an increased resistance to oxidative challenge compared to control cells, as revealed by higher cell survival against tacrine-induced hepatotoxicity. In addition, treatment with C. gigas hydrolysates reduced the leakage of GOT and LDH. These findings indicate that enzyme hydrolysates derived from C. gigas may be of benefit for developing hepatoprotective foods and drugs.
