Anthraquinones with Antibacterial Activities from Crucianella maritima L. Growing in Egypt

  • From the extracts of Crucianella maritima L. (Rubiaceae), five new anthraquinones namely; 1-hydroxy-2-methyl-6-methoxy anthraquinone, 6-methoxy-2-methyl quinizarin, 6-methyl-anthragallol-2,3-dimethyl ether, 6-methyl-anthragallol-2-methyl ether, and 1-hydroxy-2-carbomethoxyanthraquinone were isolated and identified. In addition, deacetyl asperulosidic acid 6'-glucoside sodium salt, a new iridoid diglucoside, along with twelve known anthraquinones, three flavonols, three sterols, and one triterpene were also isolated and identified for the first time from this plant. Their chemical structures were established by physical, chemical and spectroscopic data, including UV, MS, ID- and 2D-NMR analyses. The antimicrobial, cytotoxic activities and a preliminary clinical trial of the crude extracts and some isolates are also presented. Chemotaxonomical aspects are briefly discussed.

  • A chemical examination of the aerial parts of Euphorbia pekinensis $R_{UPRECHT}$. (Euphorbiaceae) has led to the isolation of seven hydrolyzable tannins and ten fl avonoid glycosides. The former ones have been identified as gallic acid, methylgallate, 3-O-galloyl shikimic acid, 1,3,4,6-tetra-O-galloyl-${\beta}-_D$-glucose, 1,2,3,4,6-penta-O-galloyl-${\beta}-_D$-glucose, corilagin, geraniin and the latter ones as isoquercitrin, quercitrin, astragalin, afzelin, prunin, rutin, kaempferol-3-O-rutinoside, quercetin-3-O-(2"-O-galloyl)-${\beta}-_D$-glucoside and quercetin-3-O-(2"-O-galloyl)-${\alpha}-_L$-rhamnoside on the basis of chemical and spectroscopic evidence.

The Inhibitory Effects of Lactose-${\beta}$-sitosterol on the Inflammatory Responses of HMC-1 Cells and EoL-1 Cells

  • ${\beta}$-sitosterol glucoside exists in a variety of plants and have anti-tumor, anti-microbial, and immunomodulatory activities. Mast cells and eosinophils play important roles in a variety of inflammatory diseases, specifically asthma and atopic dermatitis. In the present study, we used lactose-${\beta}$-sitosterol (L-BS) and investigated the effect of L-BS on inflammatory responses of the human mast cell line, HMC-1 and the human eosinophilic leukemia cell line, EoL-1. In HMC-1 cells, L-BS significantly inhibited cell migration in response to stem cell factor without cytotoxicity. However, the mRNA expression of CC chemokine receptors (CCRs), including CCR1-5, were not altered after L-BS treatment in HMC-1 cells. LPS-induced IL-4 production was also suppressed by L-BS in a dose-dependent manner. In EoL-1 cells, the concentration ranging from 0.1 ${\mu}M$ to 10 ${\mu}M$ of L-BS had no cytotoxicity and had no effect on mRNA expression of major protein-mediators derived from activated eosinophils. However, 100 ${\mu}M$ of L-BS induced the apoptosis of EoL-1 cells in a time-dependent manner. This finding indicates the possibility of L-BS as a potential therapeutic molecule in inflammatory diseases and may contribute to the need to improve current therapeutic drugs.

Flavonoids of Gomphocarpus sinaicus and Evaluation of Some Pharmacological Activities

  • The aerial parts of Gomphocarpus sinaicus Boiss. yielded four flavonoids that were identified as isorhamnetin 3-O-rhamnoglucoside (1), luteolin-7-O-glucoside-3-O-rhamnoside (2), rutin (3) and rutin-7-O-rhamnoside (4). All of the isolated flavonoids were identified by spectroscopic methods (UV, FAB-MS, $^1H-NMR\;&\;^{13}C-NMR$) and in comparison with the literature data. The isolated flavonoids 1, 2 and 4 are reported here for the first time from Gomphocarpus sinaicus Boiss. Three sets of experiments were carried out using the defatted alcoholic extract of Gomphocarpus sinaicus Boiss: the $1^{st}$ experiment indicated that the $LD_{50}$ was 49.82 mg/100 g b.wt. of intraperitoneally (i.p.) injected mice. The toxic signs were recorded within the first 24 hr post-injection. The $2^{nd}$ experiment revealed that the extract of the plant exhibited significant anti-inflammatory effects in normal rats. The $3^{rd}$ experiment was found that the tested doses of the extract in diabetic rats induced a significant decrease in serum glucose, AST, ALT, triglycerides, cholesterol and LDL, while HDL caused a significant increase.

Inhibitory Effects of Methanol Extract, Phenolic Acids and Flavonoids from the Leaves of Eucalyptus darylmpleana against 1,1-Diphenyl-2-picrylhydrazyl Radical

  • The inhibitory effects of the leaves of Eucalyptus darylmpleana (Myrtaceae) on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical was examined. The scavenging effect of the ethyl acetate fraction of Eucalyptus darylmpleana leaves on DPPH radical was stronger than the other fractions, and further purified by silica gel and Sephadex LH-20 column chromatography. 3,4-Dihydroxybenzoic acid, gallic acid, quercetin, quercetin $3-O-{\alpha}-_L-rhamnoside$, quercetin $3-O-{\beta}-_D-glucoside$ and quercetin 3-O-rutinoside were isolated and elucidated by spectroscopic data. Among these components, gallic acid and quercetin $3-O-{\alpha}-_L-rhamnoside$ exhibited potent scavenging activities on DPPH radical with $IC_{50}$ values of 6.02 and $5.54\;{\mu}M$, respectively.

Aldose Reductase Inhibition by Luteolin Derivatives from Parasenecio pseudotaimingasa

  • Effects of the extract and fractions from Parasenecio pseudotaimingasa on rat lens aldose reductase (AR) inhibition have been investigated. Among them, the n-BuOH fraction was exhibited good inhibitory potencies ($IC_{50}$ value 1.42 ${\mu}g/ml$). Phytochemical constituents were isolated from the n-BuOH fraction by open column chromatography. Their structures were elucidated as luteolin-7-O-rutinoside (1) and luteolin-7-Oglucoside (2) on the basis of spectroscopic analysis. Compounds 1 and 2 exhibited strong AR inhibitory activity, with $IC_{50}$ values of 2.37 and 1.05 ${\mu}M$, respectively. This is the first report on the isolation of compounds 1 and 2 from P. pseudotaimingasa. These results suggest that P. pseudotaimingasa could be a useful material in the development of a novel AR inhibitory agent against diabetic complications.

  • Fourteen compounds were isolated from the 70% ethanol extract of Lonicera Caulis (Caprifoliaceae) and their structures were identified as seven aliphatic compounds [long-chain alcohols (1, 2), trilinolein (3), hexacosanol (4), fatty acids (6), 9,12,13-trihydroxyoctadeca-10(E),15(Z)-dienoic acid (10), and pinellic acid (11)] and seven phenolics [bis(2-ethylhexyl) phthalate (dioctylphthalate, DOP) (5), coniferaldehyde (7), caffeic acid docosanoyl ester (8), caffeic acid (9), coniferyl aldehyde 4-O-glucoside (12), linarin (13), and coniferin (14)]. The chemical structures of these compounds were identified on the basis of spectroscopic methods and comparison with literature values. All the compounds except for caffeic acid (9) were isolated from this plant parts for the first time.

  • Twelve compounds were isolated from the 70% ethanol extract of Lonicera Caulis (Caprifoliaceae) and their structures were identified as six triterpenoids [(24S)-cycloart-25-en-$3{\beta}$,24-diol (1), pomolic acid (7), ursolic acid (8), euscaphic acid (9), hederagenin (10), and 23-hydroxytormentic acid (12)] and six sterols [obtusifoliol (2), gramisterol (3), citrostadienol (4), ${\beta}$-sitosterol (5), ergosterol peroxide (6) and ${\beta}$-sitosterol glucoside (11)]. The chemical structures of these compounds were identified on the basis of spectroscopic methods and comparison with literature values. All the compounds were isolated from this plant parts for the first time.

  • Phytochemical investigation of the MeOH extract of the aerial parts of Bistorta manshuriensis resulted in the isolation of two cerebrosides, two lactams, six phenolic compounds and seven flavonoids. Their chemical structures were characterized by spectroscopic methods to be pinelloside (1), soyacerebroside I (2), pterolactam (3), 5-hydroxypyrrolidine-2-one (4), vanillic acid (5), caffeic acid methyl ester (6), protocatechuic acid (7), caffeic acid (8), 3,5-di-O-caffeoyl quinic acid methyl ester (9), chlorogenic acid methyl ester (10), avicularin (11), afzelin (12), quercetin (13), isoorientin (14), quercetin 3-O-${\beta}$-D-glucoside (15), quercitrin (16), and luteolin (17). The isolated compounds (1 - 4, 7, 12, 14) were isolated for the first time from this plant source and the compounds 1 - 4, 9 and 10 were first reported from the genus Bistorta. Compound 17 exhibited moderate cytotoxicity and compound 6 exhibited weak cytotoxicity against four human cancer cell lines in vitro using an SRB bioassay.

Cytotoxic Compounds from the Roots of Pulsatilla koreana

  • Seven compounds including hederagenin 3-[(O-${\alpha}$-L-rhamnopyranosyl-($1{\rightarrow}2$)-${\alpha}$-L-arabinopyranosyl) (1), $3{\beta}$-[(O-${\alpha}$-L-rhamnopyranosyl-($1{\rightarrow}2$)-${\alpha}$-L-arabinopyranosyl)oxy]olean-12-en-28-oic acid (2), caffeic acid methyl ester (3), ferulic acid (4), orebiusin A (5), latifonicinin C (6) and 5-(hydroxymethyl)-2-furfuraldehyde (7) were isolated from the ethyl acetate fraction of the roots of Pulsatilla koreana. Their chemical structures were established based on physicochemical and spectroscopic data analyses. All isolates were investigated for their cytotoxic activity against cancer cell lines. Among them, compound 1 showed inhibitory activity against A549, COLO 205, and L1210 cancer cell lines with $IC_{50}$ values of 15.8, 36.5, and 22.8 ${\mu}g$/mL, respectively.
