이리도이드 배당체(配糖體) (II) -속단(續斷)의 이리도이드 배당체(配糖體) 및 진형과(唇形科) 식물(植物)의 제암효과(制癌效果)- (Iridoid Glucoside from Phlomis umbrosa $T_{URCZ.}$ and Antitumor Activity of Labiatae Plants)

  • A novel iridoid glucoside was isolated from the root of Phlomis umbrosa Turcz. (Labiatae) 'Sok-dan' and named as Umbroside. It was obtained as amorphous powder and its molecular formula is C_{19}H_{18}O_{12}$. By enzyme hydrolysis it produced a stable aglucone and ${\beta}-D-glucose$. The structure of Umbroside was determined as 8-O-acetyl shanzhiside methyl ester. Antitumor activity of Labiatae plants was also tested.

미나리에서 분리한 플라보노이드 화합물의 아플라톡신 B$_1$에 대한 항돌연변이 효과 (Antimutagenic Effect of Flavonoids Isolated from Oenanthe jauanica)

  • 미나리의 MeOH추출물에서 분획한 ethyl acetate 가용부를 silica gel column chromatography를 이용하여 isorhamnetin, hyperoside 및 persicarin의 플라보노이드 화합물을 분리하였으며, in vitro에서 Salmonella typhimurin TA 100균주를 이용하여 항돌연변이 활성을 관찰하였다. 이들 화합물 중 isorhamnetin은 0.5mg/plate에서 33%, 1.0mg/plate에서는 59%, persicarin은 0.5mg/plate에서 40%, 1.0mg/plate에서 47%의 돌연변이 저해효과를 나타내었다. 이와 유사한 플라보노이드 화합물로서 엉겅퀴 지상부에서 분리한 acacetin 7-rutinoside, cirsimaritin 4'-glucoside 및 hispidulin 7-neohesperidoside에 대해서도 1mg/plate 농도에서 acacetin 7-rutinoside와 cirsimaritin 4'-glucoside가 40% 및 49%의 돌연변이 억제활성이 관찰되었다.

  • Plants are known as important source in the search for new drugs. The twelve compounds from Viscum album (Loranthaceae), lupeol (1), betulonic acid (2), betulinic acid (3), terminic acid (4), ursolic acid (5), $\beta$-sitosterol (6), $\alpha$-spinasterol (7), oleanolic acid (8), 5-hydroxy-1-(4′-hydoxyphenyl)-7-(4"-hydroxyphenyl)-hepta-1-en-3-on (9), 2′-hydroxy-4′,6′-dimethoxychalcone-4-O-glucoside (10), 2′-hydroxy-4′,6′-dimethoxychalcone-4-O-[apiosyl(l$\longrightarrow$2)]glucoside (11) and syringin (12) were evaluated for their immunomodulatory properties. Compounds 6 and 11 induced the macrophage tumoricidal activity and the lymphocyte blastogenesis. In addition, these compounds stimulated the macrophages to induce the production of TNF-$\alpha$ and NO. These findings suggest that compounds 6 and 11 are modulating various elements of the host immune response.

  • Two soyasaponins were isolated from the aerial parts of Astragalus tribuloides Del. Their structures were established on the basis of spectroscopic and chemical methods. In addition, ursolic acid, ${\beta}-sitosterol\;{\beta}-D-glucoside$ and isorhamnetin 3-O-glucoside were isolated and identified by comparing their mp, spectral and chromatographic data with those of authentic samples. This is the first report of screening and isolation of the chemical constituents of this species of genus Astragalus.

  • 우리나라에서 채소(菜蔬)로 또 조미품(調味品)으로 많이 이용(利用)되고 있는 재래종(在來種)갓의 anthocyanin 색소(色素)의 구명(究明)을 위(爲)한 실험(實驗)에서 얻어진 결과(結果)를 요약(要約)하면 다음과 같다. 1. T.L.C.에 의(依)해 이종(二種)의 anthocyanin을 분리(分離)하였다. 2. 이들 anthocyanin의 구조구명(構造究明)을 위(爲)하여 Robinson 시험(試驗), Spectrophotometry, T.L. chromatography, 부분가수분해등(部分加水分解等)의 방법(方法)으로 각각 peonidin-3-glucoside와 peonidin-3-galctoside 임을 확인(確認)하였다.

  • An ${\alpha}-{\text\tiny{D}}-glucoside$ of sesterterpene, 11-epiterpestacin, was isolated from the culture of a filamentous fungus Bipolaris sorokiniana NSDR-011. The structure was elucidated by chemical studies and spectroscopic methods including NMR and ESI-MS. 11-ETG (1) named arbitrarily did not inhibit the root growth of Italian ryegrass seedlings even at the level of 200 ppm, while its aglycone 11-ET (2) completely inhibited root growth at level of 100 ppm.

  • The single-vessel multienzyme UDP-${\alpha}$-$\text\tiny{D}$-glucose recycling system was coupled with a forward glucosylation reaction to produce novel glucose moiety-conjugated derivatives of different tetracycline antibiotic analogs. Among five tetracycline analogs used for the reaction, four molecules (chlorotetracycline, doxytetracycline, meclotetracycline, and minotetracycline) were accepted by a glycosyltransferase enzyme, YjiC, from Bacillus licheniformis to produce glucoside derivatives. However, the enzyme was unable to conjugate sugar units to rolitetracycline. All glucosides of tetracycline derivatives were characterized by ultraviolet absorbance maxima, ultra-pressure liquid chromatography coupled with photodiode array, and high-resolution quadruple time-of-flight electrospray mass spectrometry analyses. These synthesized glucosides are novel tetracycline derivatives.

배당화 아크릴레이트와 메타크릴레이트의 효소적 합성 (Enzymatic Synthesis of Glycosyl Acrylate and Methacrylate)

  • Glycosyl acrylate and methacrylate were synthesized by lipase-catalyzed esterification of vinyl acrylate and vinyl methacrylate with ${\beta}$-methyl glucoside in t-butanol as a reaction medium. At the optimum conditions of initial concentration of 150g/l ${\beta}$-methyl glucoside, molar ratio of 1 : 3, 5%(w/v) lipase(Novozym 435) and $50^{\circ}C$, we attained up to 100% conversion for enzymatic glycosylation of vinyl acrylate and vinyl methacrylate by supersaturated solvent process. The polymerizable glycosyl acrylates and methacrylate are expected to have biomedical application as hydrophilic monomers and hydration modifiers to be used for biocdmpatible hydrogel.

배풍등의 페놀성 배당체 및 스테로이드 사포게닌 (A Phenolic Glucoside and Steroidal Sapogenins of Solanum Iyratum)

  • A phenolic glucoside and two steroidal sapogenins were isolated from the aerial part of Solanum Iyratum (Solanaceae). They were identified as 2-hydroxy-3-methoxybenzoic acid glucose ester (1), ${\Delta}^{3,5}$-deoxytigogenin ((25R)-spirosta-3,5-diene) (2), diosgenin (($3{\beta}$, 25R)-spirost-5-en-3-ol) (3), respectively by several spectroscopic methods including IR, MS and NMR. Compound 1 has not been previously isolated from Solanaceae plants. Compounds 2 and 3 are isolated from S. Iyratum for the first time in the present study.

백화사설초(百花蛇舌草)의 성분(成分)에 관한 연구(硏究) (Studies on the Constituents of Oldenlandia diffusa)

  • 국내(國內)에서 생산(生産)된 백화사설초(百花蛇舌草) Oldenlandia diffusa(WiIId.) Roxb. 의 전초로 부터 얻은 MeOH extract를 $CHCl_3, EtOAc$ 및 BuOH로 분획하여 얻은 fraction으로 부터 column chromatography를 실시하여 3종의 화합물을 분리하였다. 이들의 이화학적(理化學的) 및 분광학적(分光學的) data를 종합하여 화합물 I의 화학구조는 ${\beta}-sitosterol$에 소양(小量)의 stigmasterol이 함유된 sterol이며, 화합물 II는 ursolic acid이고, 화합물 III은 ${\beta}-sitostero\;3-0-{\beta}-D-glucoside$임을 밝혔다. 이 식물은 다양한 생리활성(生理活性)이 보고(報告)된 ursolic acid의 중요한 자원식물(資源植物)이 될 수 있음을 본실험을 통하여 밝혔으며 문헌(文獻)에 보고(報告)된 oleanolic acid는 함유(含有)되어 있지 않음을 알았다.
