A review on the medicinal potentials of ginseng and ginsenosides on cardiovascular diseases

  • Ginseng is widely used for its promising healing and restorative properties as well as for its possible tonic effect in traditional medicine. Nowadays, many studies focus on purified individual ginsenoside, an important constituent in ginseng, and study its specific mechanism of action instead of whole-plant extracts on cardiovascular diseases (CVDs). Of the various ginsenosides, purified ginsenosides such as Rb1, Rg1, Rg3, Rh1, Re, and Rd are the most frequently studied. Although there are many reports on the molecular mechanisms and medical applications of ginsenosides in the treatment of CVDs, many concerns exist in their application. This review discusses current works on the countless pharmacological functions and the potential benefits of ginseng in the area of CVDs. Results: Both in vitro and in vivo results indicate that ginseng has potentially positive effects on heart disease through its various properties including antioxidation, reduced platelet adhesion, vasomotor regulation, improving lipid profiles, and influencing various ion channels. To date, approximately 40 ginsenosides have been identified, and each has a different mechanism of action owing to the differences in chemical structure. This review aims to present comprehensive information on the traditional uses, phytochemistry, and pharmacology of ginseng, especially in the control of hypertension and cardiovascular function. In addition, the review also provides an insight into the opportunities for future research and development on the biological activities of ginseng.

Effect of Ginsenosides from Panax ginseng on Proliferation of Human Osteosarcoma Cell $U_2OS$

  • Object To find out which of the 27 ginsenosides isolated from Panax ginseng C.A. Mey that may inhibit the proliferation of human osteosaocoma cell line $U_2OS$. Methods Effects of each individual ginsenoside on the proliferation of $U_2OS$ cell were studied by determining the viability of cancer cells during culture with or without the presence of the test compound. DNA assay was determined by flow cytometry. Results Ginsonosides -Ro, $-Rh_l,\;-Rh_2,\;-F_1\;and\;-L_8$ at concentrations of 5 ,umol/L could obviously suppress the proliferation of $U_2OS$ cells while ginsenosides $-Rg_1,\;-F_3,$ -Rf, PPT and PT significantly inhibited the cancer cells. Flow cytometry revealed that ginsenosides $-Ro,-Rg_1-Rf,-F_1-Rh_2,PPT$ and PT induced cell cycle arrest at $G_0/G_1$ phase with obvious decrease of cell count at Sand $G_2+M$ phase, Moreover, ginsenosides $-Rf_1,-Rg_1,\;-F_1$ and PPT induced significantly high rates of cell death as compared with the control. Conclusion These data suggested that ginsenosides inhibited $U_2OS$ proliferation Via cell cycle arrest or induction of cell death.

Determination of Ginsenosides Content in Korean Ginseng Seeds and Roots by High Performance Liquid Chromatography

  • A high performance liquid chromatography (HPLC) method has been successfully developed to identify and quantify major ginsenosides in Korean ginseng seeds and roots. Using gradient elution of acetonitrile and water without buffer, the 6 major ginsenosides ($Rb_1,\;Rb_2$, Rc, Rd, Re, and $Rg_1$) were identified. Compared with ginseng roots, the amount of ginsenoside Re and Rd in ginseng seeds were significantly higher than those in ginseng roots (p<0.05). In ginseng seeds, the content of protopanaxtriol (PPT) was higher than that of protopanaxdiol (PPD) and the ratio of PPT and PPD was approximately 2.2 : 1. However, the content of PPT was lower than that of PPD in ginseng roots. It should be mentioned that both content of PPT and PPD in ginseng seeds were much higher than those in ginseng roots.

LC-MS/MS 및 ICP를 이용한 당뇨환의 화학적 특성 분석 (Characteristic Effects of Dangnyo-hwan for Diabetes Control Studied Using LC-MS/MS and ICP)

  • Objectives: The primary aim of this study was to identify bioactive compounds in Dangnyo-hwan, a Korean herbal medicine, through instrumental analysis using LC-MS/MS and ICP, and investigate its potential use in diabetes treatment. Methods: The extract of Dangnyo-hwan has 12 medicinal herbs; these were compared with 18 marker substances selected from literature survey. Results: LC-MS/MS analysis could detect 9 of the 18 bioactive compounds: citruline, catalpol, berberine, ginsenoside Rb1, ginsenoside Rg1, oleanolic acid, β-sitosterol, mangiferin, and schizandrin. While harmful heavy metals such as As, Pb, Cd, Hg, Ni, and Cu were not present in high concentrations, Zn concentration was 4.2 mg in 100 g Dangnyo-hwan. Conclusions: Instrumental analysis such as LC-MS/MS and ICP was successfully used to identify bioactive compounds in Dangnyo-hwan. Detection of 9 bioactive substances and Zn from the herb medicine is a valuable finding, and suggests that Dangnyo-hwan is a candidate medicine for diabetes. Further investigations like in vitro assay, percent GPR 119 activity, and percent human DGAT-1 inhibition are underway.

인삼제품(人蔘製品)의 품질개량(品質改良)에 관(關)한 연구(硏究) (제일보(第一報)) - 인삼근(人蔘根) 및 엽(葉) Saponin의 비교연구(比較硏究) - (Quality Control of Ginseng Products(Part I) - The saponins isolated from ginseng roots and leaves -)

  • 인삼엽(人蔘葉)을 ether, benzene-butanol 혼합용매등으로 색소류(色素類)기타 지용성물질(脂溶性物質)등을 제거하고, methanol과 n-butanol로 연속추출하여 n-butanol soluble saponin을 얻고 계속해서 liquid chromotography 를 행하여 분획별(身劃別)로 인삼엽(人蔘葉) saponin을 분리 정제하였고, 그 화학적(化學的)및 분자구조학적(分子構造學的) 특성(特性)을 인삼근(人蔘根) saponin과 비교 검토한 결과는 다음과 같다. 1. 인삼근(人蔘根)과 엽(葉) saponin의 2차원(차원) TLC pattern은 다소(多少) 차이(差異)차가 있으나, 다수(多數)의 spot들이 일치(一致)하였다. 인삼엽(人蔘葉)에는 ginsenoside Rd가 존재하지 않고, ginsenoside Rc는 엽(葉)에, ginsenoside $Re_1$$Rg_1$는 근부(根部)에 더 많이 분포(分布)되어 있었다. 2. 인삼엽(人蔘葉)에는 인삼근(人蔘根)에 비하여, panaxadiol 계(系) saponin이 panaxatriol 계(系) saponin보다 더높은 함량을 보였다. 3. n-butanol soluble saponin으로 liquid chromatography를 행하여 인삼근(人蔘根)에서는 Fr. 1.과 Fr. 2.를 얻었고, 인삼엽(人襲葉)에서는 Fr. 1, Fr. 2. 및 Fr. 3.의 분획(分劃)을 얻을 수 있었다. 인삼근(人蔘根)의 Fr. 1 및 엽(葉)의 Fr. 1.과 Fr. 2.등 3매의 분획(分劃)은 saponin test반응(反應)에 양성(陽性)을 보이고, 그 IR spectra는 동일(同一)한 것으로 나타났다.

가공방법에 따른 몇가지 백삼 제품의 사포닌 및 유리당 조성 변화 (Composition of Saponin and Free Sugar of Some White Ginsengs with Processing Conditions)

  • 수삼으로 제조된 백삼 제품들의 가공방법의 차이에 따른 성분 변화에 대한 조사로서 원료삼인 수삼과 전통적인 방법으로 제조한 백삼, 태극삼 A, 새로운 방법으로 제조된 태극삼 B의 일반성분, 추출수율, 사포닌 함량 및 조성, 유리당 조성을 비교 분석 하였다. 4가지 시료의 조단백질은 13.34-15.33%, 조지방은 1.04-2.05%, 조회분은 3.13-5.25%, 총 식이섬유는 9.72-15.46%, 당질은 64.75-73.48% 범위의 함량이었다. 추출수율은 열수 추출이 80% 메탄올 추출보다 전체적으로 약 2배 이상 높게 나타났으며, 가공하지 않은 수삼의 열수 추출 수율이 56.4%로 가장 높았다. 사포닌 함량은 열수 추출시 수삼의 총 사포닌이 2.40%로 가장 많이 추출되었으며, 태극삼 A는 태극삼 B보다 비율로 볼 때 약 20% 이상 적은 1.45%의 총 사포닌 함량을 나타내었다. Ginsenoside 조성은 열수 추출시 $ginsenoside-Rg_1$이 수삼은 36.52%이지만 태극삼 A와 B는 68.66, 67.89%고 약 2배의 함량을 나타내었다. $Ginsenoside-Rb_1$은 태극삼 A차 B에 18.41, 17.43%만이 함유되어 있었다. 한편 태극삼 A와 태극삼 B의 총 사포닌 양에는 큰 차이가 있었지만 ginsenosides의 조성비에 있어서는 서로 큰 차이가 없는 것으로 나타났다. 80% 메탄올 추출의 경우에도 가공방법이 다르게 제조된 백삼 제품별로 ginsenoside조성에 큰 차이를 나타내었다. 수삼과 백삼에서는 열수 추출과 80% 메탄올 추출 모두에서 함량비는 다를지라도 fructose, glucose, sucrose, maltose만이 함유되어 있었지만 열 처리된 태극삼 A와 B에는 rhamnose도 각각 2.56, 4.01%씩 함유되어 있었다.

Ginsenosides repair UVB-induced skin barrier damage in BALB/c hairless mice and HaCaT keratinocytes

  • Background: Ginsenosides (GS) have potential value as cosmetic additives for prevention of skin photoaging. However, their protective mechanisms against skin barrier damage and their active monomeric constituents are unknown. Methods: GS monomer types and their relative proportions were identified. A UVB-irradiated BALB/c hairless mouse model was used to assess protective effects of GS components on skin epidermal thickness and transepidermal water loss (TEWL). Skin barrier function, reflected by filaggrin (FLG), involucrin (IVL), claudin-1 (Cldn-1), and aquaporin 3 (AQP3) levels and MAPK phosphorylation patterns, were analyzed in UVB-irradiated hairless mice or HaCaT cells. Results: Total GS monomeric content detected by UPLC was 85.45% and was largely attributed to 17 main monomers that included Re (16.73%), Rd (13.36%), and Rg1 (13.38%). In hairless mice, GS ameliorated UVB-induced epidermal barrier dysfunction manifesting as increased epidermal thickness, increased TEWL, and decreased stratum corneum water content without weight change. Furthermore, GS treatment of UVB-irradiated mice restored protein expression levels and epidermal tissue distributions of FLG, IVL, Cldn-1, and AQP3, with consistent mRNA and protein expression results obtained in UVB-irradiated HaCaT cells (except for unchanging Cldn-1 expression). Mechanistically, GS inhibited JNK, p38, and ERK phosphorylation in UVB-irradiated HaCaT cells, with a mixture of Rg2, Rg3, Rk3, F2, Rd, and Rb3 providing the same protective MAPK pathway inhibition-associated upregulation of IVL and AQP3 expression as provided by intact GS treatment. Conclusion: GS protection against UVB-irradiated skin barrier damage depends on activities of six ginsenoside monomeric constituents that inhibit the MAPK signaling pathway.

pH 및 고온 열처리가 홍삼물추출물의 주종 사포닌 성분변화에 미치는 영향 (Effects of pH and High Temperature Treatment on the Changes of Major Ginsenosides Composition in Korean Red Ginseng Water Extract)

  • This study was carried out to investigate the changes of ginsenosides composition in Korean red ginseng water extract (RGWE) after heated with high temperatures above $100^{\circ}C$. RGWEs were adjusted with pH 3.0, pH 7.0 and pH 10.0, respectively, and then heated at 100,110 and $120^{\circ}C$ for 30 minutes by using autoclave. Total ginsenosides of RGWE treated with heating showed decreasing tendency when compared with control. By TLC analysis, decreasing effect of ginsenosides in RGWE were significantly observed in the acidic condition of pH 3.0, particulary. By HPLC analysis, total ginsenoside of control showed 1.89%, while those of RGWE treated with 100, 110 and $120^{\circ}C$ showed 1.22, 1.05 and 0.97%, respectively. The ratio of protopanaxadiol (PD) to protopanaxatriol (Pr) saponins in control was 1.89, while that of PD/PT in treated RGWEs were level of 1.33 to 1.47. By the result of decreased ratio of PD/PT in RGWE, it was considered that PD type saponin such as ginsenoside$-Rb_{1}$, $-Rb_{1}$, -Rc and -Rd was more unstable than PT type saponin such as ginsenoside-Re and Rg against high temperature heating above $100^{\circ}C$.

저온에서 1차 추출한 홍삼농축액의 기능성분 특성 I - Ginsenoside 위주로 - (Characteristics of Functional Components of Red Ginseng Concentrate First Extracted at Low Temperature I - Focused on Ginsenoside -)

  • 홍삼의 일반적인 가공법인 물과 주정 혼합용매로 추출 여과하여 농축액으로 제조 시 증숙 등의 가열처리에 의해 가수분해되거나 중합반응이 일어나게 되어 현재 기능성이 밝혀진 30여 가지 주요 진세노사이드 외 극미량인 200여종의 진세노사이드가 검출되고 있어서, 오히려 홍삼 본래의 기능성 규명과 효능 구현을 복잡하게 만들고, 추출 횟수를 거듭할수록 떫은맛이 강해지는 등 건강기능식품으로서 부정적인 영향을 배제하기 위하여 저온(40℃ 미만)에서 1차만 추출한 홍삼농축액을 제조한 결과 수율은 고형분 66-77% 정도에서 26-30%를 나타내어 기존의 연구 결과인 40.47±0.47% 보다 현저히 적은 수율을 나타내었고, 동결건조 분말의 진세노사이드 함량분석시험을 진행한 결과 시료 18종 전체에서 진세노사이드 34종이 검출되었고, 특히 홍삼 지표성분 3종인 Rb1, Rg1, Rg3 합계는 42.48 mg/g으로 시판 홍삼제품의 환산량 9.0 mg/g과 8.7 mg/g 보다 4-5배 많았다. 본 시료는 진한 홍삼의 향취와 함께 면역력 증진 등 6가지 기능발현에 9-25%의 소량 복용으로 1일 복용량이 유의성 있게 작아서 복용의 편의성과 함께 제품의 보관, 유통에 장점이 있는 것으로 판단된다.

장뇌삼 첨가 탁주의 이화학적 특성 및 Ginsenosides 함량 (Physicochemical Characteristics and Ginsenosides Compositions of Makgeolli Added with Mountain Ginsengs)

  • Rice wine (makgeolli) containing various amounts of mountain ginsengs (MG) are being prepared with nuruk and yeasts, and the physicochemical characteristics and contents of ginsenosides in MG-makgeolli were analyzed. Average particle size of MG powder is $29.1{\mu}m$. MG slice (20 g) or powder (0~20 g) and rice (3,000 g) were used for 12 days fermentation of makgeolli, makgeolli containing slice of MG (SW-makgeolli), makgeolli containing 2 g (PW1-makgeolli), 10 g (PW2- makgeolli), 20 g (PW3-makgeolli) of powder of MG, respectively. Soluble solids and pH levels show no differences between five kinds of makgeolli groups, whilst the presence of high amounts of MG (PW3-makgeolli) caused decreases in ethanol and acidity. Major free amino acids in MG-makgeolli are glutamic acid and arginine. Total contents of 14 ginsenosides are approximately 2.5 g/100 g of dried MG powder and major ginsenoside were ginsenosides Re, Rb1, Rb2, Rg1, Rc and Rf. During the propagation of makgeolli containing MG, the ginsenosides Rb1, Rb2, Rb3, and Rc decreased, whilst ginsenosides Rg3 and compound K increased highly. It indicates that ginsenosides in MG are metabolized to different forms of ginsenosides by brewing microorganisms.
