Comparisons of Ginsenosides and Anti-inflammatory Effects of White Ginseng and Puffed Red Ginseng (인삼과 팽화홍삼의 Ginsenoside 함량 및 항염효과 비교)

  • In this study, the ginsenoside contents and anti-inflammatory effects of white ginseng (WG) and puffed red ginseng (PRG) were compared. The contents of Rb1, Rg5 and Rk1 were significantly higher in PRG than in WG, whereas the contents of Rg1 and Rb2 were decreased in PRG. The levels of NO production and iNOS expression were suppressed in LPS-stimulated cells by treatment with WG and PRG. Further, the production of cytokines (TNF-$\alpha$ and INF-$\gamma$) and inflammatory proteins (NF-${\kappa}B$ and COX-2) was decreased in cells upon treatment with any of the ginsenosides. The high NO inhibitory activity and cytokine production of PRG is caused by differences in the composition of ginsenosides produced.

Bioavailability and Anti-inflammatory Effect of Fermented Red Ginseng in BALB/c Mouse (BALB/c 마우스에서 발효 홍삼 Ginsenoside의 생체이용율과 항염효과)

  • The fermented red ginseng by microorganism is known to increase pharmacological activity in vivo. To evaluate the bioavailablity of red ginseng fermented by probiotics, we conducted the pharmacokinetic study of ginsenoside Rb1, Rd and total ginsenosides (TG, ginsenosides Rb1 + Rd + Rg1 + F2 + Rg3 + compound K) in BALB/C mice. The AUC value of ginsenoside Rb1 in mice serum administered with 600mg/kg drugs showed 21.93 ± 14.68 ng·h/mL (RGw, water extract), 275.211 ± 110.04 ng·h/mL (RGe, 50% ethanol extract) and 404.91 ± 162.57 ng·h/mL (fRGe, fermented red ginseng extract). Analysis of ginsenoside Rd also showed a higher ACU value in fRGe than in RGw or RGe. And the AUC value of total ginsenosides in mice serum treated with 600 mg/kg were observed 42.12 ± 23.44 ng·h/mL (RGw), 321.44 ± 133.5 ng·h/mL (RGe) and 537.33 ± 229.01 ng·h/mL (fRGe), respectively. Cmax value of ginsenoside Rb1 in mice administered with 600mg/kg were observed 3.67 ± 3.34 ng/mL (RGw), 23.27 ± 8.81 ng/mL (RGe) and 25.52 ± 7.29 ng/mL (fRGe). These results can be considered that the fermented red ginseng has more bioavailability than that of unfermented red ginseng. In quantitative analysis of the inflammation-related cytokines IL-1β and TNF, no significant difference was found between the fermented red ginseng (fRGe) and the red ginseng (RGe).

  • Red ginseng marc is a by-product of Korean red ginseng (panax ginseng CA Meyer) and contains ginsenoside which has pharmacological effects. The Korean red ginseng marc was fermented with Bacillus subtilis (RGMB). This study was carried out to investigate the RGMB effect on swine immunity. The variation of ginsenoside depending on the RGMB fermentation time was analyzed. Swine (Landrace${\times}$Yorkshire) were divided into control group (basic diet) and RGMB group (RGMB 1% diet). One percent RGMB was fed to the RGMB group for 28 days. The biochemical parameters, cytokine and immunoglobulin were analyzed. For 48 hours of fermentation on RGMB, ginsenoside Rb1 had increased 180.94%, Rg3 235.85%. Rg1 wasn't detected before fermentation, but was detected after 48 hours of fermentation. The RGMB had effect of deceasing initial AST concentration $79.33{\pm}12.85U/L$ to $54.00{\pm}14.46U/L$ in final and was significantly lower (P<0.05) than control in final. In final RGMB had significantly lower (P<0.05) ALT concentration of $48.57{\pm}8.26U/L$ comparing with control group of $65.43{\pm}10.31U/L$. RGMB had the effect of significantly decreasing (P<0.05) $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ concentration of $2.44{\pm}1.31ng/mL$, $0.71{\pm}0.36ng/mL$ and $0.51{\pm}0.21ng/mL$. The IgA concentration had significantly increased (P<0.05) in RGMB group of $0.56{\pm}0.06mg/mL$ in final. These results demonstrate that RGMB has effect of increasing immunity and practicable to use as feed additives on swine.

Dexamethasone-induced muscle atrophy and bone loss in six genetically diverse collaborative cross founder strains demonstrates phenotypic variability by Rg3 treatment

  • Background: Osteosarcopenia is a common condition characterized by the loss of both bone and muscle mass, which can lead to an increased risk of fractures and disability in older adults. The study aimed to elucidate the response of various mouse strains to treatment with Rg3, one of the leading ginsenosides, on musculoskeletal traits and immune function, and their correlation. Methods: Six Collaborative Cross (CC) founder strains induced muscle atrophy and bone loss with dexamethasone (15 mg/kg) treatment for 1 month, and half of the mice for each strain were orally administered Rg3 (20 mg/kg). Different responses were observed depending on genetic background and Rg3 treatment. Results: Rg3 significantly increased grip strength, running performance, and expression of muscle and bone health-related genes in a two-way analysis of variance considering the genetic backgrounds and Rg3 treatment. Significant improvements in grip strength, running performance, bone area, and muscle mass, and the increased gene expression were observed in specific strains of PWK/PhJ. For traits related to muscle, bone, and immune functions, significant correlations between traits were confirmed following Rg3 administration compared with control mice. The phenotyping analysis was compiled into a public web resource called Rg3-OsteoSarco. Conclusion: This highlights the complex interplay between genetic determinants, pathogenesis of muscle atrophy and bone loss, and phytochemical bioactivity and the need to move away from single inbred mouse models to improve their translatability to genetically diverse humans. Rg3-OsteoSarco highlights the use of CC founder strains as a valuable tool in the field of personalized nutrition.

Ginsenosides Rb1 and Rg1 Decrease Proliferation but Increase Neuronal Differentiation of Hippocampal Neural Progenitor Cells (진세노사이드 Rb1과 Rg1에 의한 해마 신경전구세포의 분화 증가)

  • Ginseng is the best known and most popular herbal medicine used worldwide. In spite of reported beneficial effects of ginseng on the CNS, there is few scientific evidences established at the cellular level. Among more than 30 ginsenosides, Rb1 and Rg1, the active ingredients of ginseng, are regarded as the main compounds responsible for many pharmaceutical actions of ginseng. Daily treatment with Rb1 or Rg1 for 3 d significantly decreased the number of bromodeoxyuridine(BrdU)(+) cells in primary neural progenitor cells(NPCs) isolated from hippocampi at embryonic day 16.5(E16.5). In contrast, treatment with Rb1 or Rg1 greatly increased the number of microtubule associated protein(MAP2) (+) cells. In addition, the transcription factors, Ngn1 and Hes1, proneural members of the basic helix-loop-helix(bHLH) family, significantly increased in Rb1 or Rg1 treated-NPCs. Based on these results, we suggest for the first time that ginsenosides Rb1 and Rg1 decrease proliferation but promote neuronal differentiation of hippocampal NPCs.

Cosmetic Potential of Enzymatic Treated Ginseng Leaf

  • The objective of this study was to evaluate the potential use of ginseng leaf as a cosmetic material. In this research, we employed enzymatic treated ginseng leaf by using Ultraflo L to improve the recovery of ginsenosides from the ginseng leaf and studied the biological activities and skin safety of the enzymatic treated ginseng leaf for use as a cosmetic material. The total ginsenoside contents of the non-enzymatic treated ginseng leaf (NEGL) and Ultraflo L treated ginseng leaf (UTGL) were 271 and 406 mg/g, respectively. The level of metabolite ginsenosides (sum of Rg2, Rg3, Rg5, Rk1, compound K, Rh1, Rh2, and F2) was higher in UTGL (93.1 mg) compared to NEGL (62.4 mg) in one gram ginseng leaf extract. The increase in amounts of ginsenoside types in UTGL compared to NEGL was generally 140% to 157%. UTGL exhibited relatively higher 2,2-diphenyl-2-picrylhydrazyl hydrate ($IC_{50}$, 2.8 mg/mL) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt ($IC_{50}$, 1.6 mg/mL) radical scavenging activities compared to NEGL (4.8 mg/mL and 2.2 mg/mL). The UTGL group showed normalized hydrogen peroxide, lipid peroxidation and visual wrinkling grade induced-UVB exposure. The UTGL did not induce any adverse reactions such as erythema and edema on intact skin sites; however, some guinea pigs treated with UTGL on abraded skin sites showed very slight erythema. The primary irritation index (PII) score of UTGL was 0.05 and it was classified as a practically non-irritating material (PII, 0 to 0.5). In skin sensitization tests with guinea pigs, UTGL had a positive rate of skin sensitization at 40%, and the mean evaluation score was 0.4.

Studies on the Optimum Light Intensity for Growth of Panax Ginseng II. Effect of Light Intensity on the Contents of Saponin and Free Sugar in the Ginseng Leaf (인삼생육의 최적광량 구명에 관한 연구 제2보 광도가 인삼엽내 Saponin 및 유리당함량에 미치는 영향)

  • This study was conducted to define the effects of light intensity on the amount of saponin and free sugar and the ratio of triol group saponin and diol group saponin (PT/PD) in the leaf of Panax ginseng C.A. Meyer. 4-Year-old ginseng plants were grown under the shadings of different light transmittance rate(LTR) of 5%, 10%, 20% and 30% for 5 months and the leafiets were sampled from 2nd low at late August to determine the amount of saponin and free sugar.-Rd was main ginseuoside in the diol group saponin but in triol group saponin, ginsenoside-Re showed highest value and next was ginsenoside-$Rg_1$ and $Rg_2$ respectively. Up to 20% of light transmittance rate (LTR), the ginseng leaves grown under high light intensity showed an increase in the amount of total saponin and the ratio of PT/PD but the amount of total saponin and the ratio of PT/PD but the amount of total saponin and the ratio of PT/PD was decreased at the ginseng leaves grown under the shading of 30% LTR. The ginseng leaves grown under the shading of 20% LTR showed a significant increase in the amount of glucose and fructose but a significant decrease of sucrose content. A significant positive correlation ($r=0.992$^{**}$) was recognized between the of amount of total saponin and glucose.

Effects of Ginsenosides on $pp60^{c-src}$ Kinase, Intracellular Calcium and Cell Proliferation in NIH 373 Cells

  • In the present study, we examined effects of ginseng saponins (ginsenosides) on pp60c-src protein tyrosine kinase (PTK) activity, intracellular calcium concentration ([$Ca^{2+}$]i), and cell proliferation in NIH3T3 cells. Eight different ginsenosides [ginsenoside-Rb1 (G-$Rb_1$), -$Rb_2$, -Rc, -Rd, -Re, -Rf, -$Rg_1$, -$Rg_2$) and ginseng total saponin (GTS) were used for these experiments. All ginsenosides and GTS tested stimulated the activation of $pp60^{c-src}$ kinase, and especially G-$Rb_1$,-Rd,-$Rg_1$, and -$Rg_1$ showed a higher stimulatory effect than others at 16.7 $\mu\textrm{g}$/ml of ginsenosides with a 18 hr-incubation, increasing the activity by 4.5, 3.5, 3.5, and 3.0-fold, respectively, over that of untreated control. In addition, both G-Rd and -$Rg_2$)Rg2 increased ($Ca^{2+}$), to 202 and 334 nM, respectively, about 2-3-fold above the basal level within 7min at 250 $\mu\textrm{g}$/yml of ginsenosides. The increases of ($Ca^{2+}$), were eliminated by Pretreatment of EGTA, an extracellular calcium chelator, suggtasting that they result from an influx of calcium ion from extracellular medium rather than an efflux from intracellular calcium store, endoplasmic reticulum (ER). All ginsenosides studied enhanced cell proliferation to 1.2-1.4-fold over that of untreated control at 5~250 $\mu\textrm{g}$/ml of concentrations. Interestingly the promotion of cell proliferation by ginsenosides corresponded with the activation of c-src kinase, which is an early step in the mitogenic signaling cascade. Taken together, we suggest that some ginsenosides may lead to cellProliferation via the activation of cellular signal transduction Pathway involving $pp60^{c-src}$ kinase.

Study on the Correlation between The Ginsenoside Contents and Growth Characteristics of Wild-simulated Ginseng with Different Year-Roots (Panax ginseng C. A. Meyer) (산양삼 연근별 생육특성과 진세노사이드 함량 간의 상관관계 연구)

  • 본 연구는 7년, 13년근 산양삼의 생육특성과 진세노사이드(G) 함량 간의 상관관계를 구명하기 위하여 수행되었다. 6개소의 산양삼의 생육특성을 조사한 결과, 뇌두길이, 뿌리길이, 생중량, 단면적, 표면적, 부피에 있어 13년근 산양삼이 7년근 산양삼에 비하여 유의적으로 높은 것을 확인하였다. 진세노사이드11종에 대한 함량은 G-Rb1, Rb2, Rc, Rd, Re, Rf, Rg1, Rg2 함량이 13년근 산양삼이 7년근 산양삼 보다 유의적으로 높은 수치를 확인하였다. 또한 산양삼과 인삼(재배삼) 진세노사이드 함량을 비교한 결과, 13년 산양삼에서 G-Rb1, Rd, Re, Rf, Rg1이 4년, 5년근 인삼(재배삼)에 비해 유의적으로 함량이 높은 것으로 확인되었다. 산양삼 연근별 생육특성과 진세노사이드 함량 간의 상관관계를 분석한 결과, G-Rb1, Rb2, Rc, Rd, Re, Rf, Rg1, Rg2 함량은 뇌두길이, 생중량, 단면적, 표면적, 부피와 유의정인 정의 상관관계를 보였으며, G-Rb1, Re, Rf, Rg2는 줄기직경과 부의 상관관계를 확인하였다. 본 연구는 산양삼의 7년근과 13년근을 대상으로 생육특성과 진세노사이드 함량 상관관계를 구명함으로써 연근에 따른 품질규격 정립에 유용한 정보를 제공 할 것으로 판단된다.

Influence of organic acids and heat treatment on ginsenoside conversion

  • Background: Heat treatments are applied to ginseng products in order to improve physiological activities through the conversion of ginsenosides, which are key bioactive components. During heat treatment, organic acids can affect ginsenoside conversion. Therefore, the influence of organic acids during heat treatment should be considered. Methods: Raw ginseng, crude saponin, and ginsenoside $Rb_1$ standard with different organic acids were treated at $130^{\circ}C$, and the chemical components, including ginsenosides and organic acids, were analyzed. Results: The organic acid content in raw ginseng was 5.55%. Organic acids were not detected in crude saponin that was not subjected to heat treatment, whereas organic acids were found in crude saponin subjected to heat treatment. Major ginsenosides ($Rb_1$, Re, and $Rg_1$) in ginseng and crude saponin were converted to minor ginsenosides at $130^{\circ}C$; the ginsenoside $Rb_1$ standard was very stable in the absence of organic acids and was converted into minor ginsenosides in the presence of organic acids at high temperatures. Conclusion: The major factor affecting ginsenoside conversion was organic acids in ginseng. Therefore, the organic acid content as well as ginsenoside content and processing conditions should be considered important factors affecting the quality of ginseng products.
