To know possibility of raw ginseng production for white- and red-ginseng by direct-sowing culture, seeds were directly sowed or seedlings were transplanted at soil condition of sandy loam in ginseng field. After cultivation, the characters of 5-year-old ginseng were investigated. Number of survived plants was increased with increase the seeding quantity in direct-sowing culture, the survived plants in direct-sowing culture was higher than that of transplanting one. Rate of the numbers of survived plants to numbers of seeds sowed in plots of 134 or 90 seeds sowed per tan, 180 x 90cm area, inspire of high number of survived plant was high compared to that of transplanting culture. Occurrence rate of rusty root of ginseng in direct-sowing culture was low significantly compared to that of transplanting culture. Root yield showed in the order of 134, 268, 90 seeds sowed, the values of yield in direct-sowing culture were high obviously compared to that of transplanting one. Individual root weight was increased with decrease the seeding quantity, however, the root weight in plot of 90 seeds sowed showed almost equal the weight in transplanting culture. The number of usable raw ginseng for white- and red-ginseng was increased with decrease of the seeding quantity; the numbers were higher than that of transplanting culture remarkably. We concluded that optimum seedling rate in direct-sowing culture of ginseng was 90 to 134 seeds per tan considering the yield per area and production rate of large root.

Hairy root culture of ginseng is industrially prospected because the cultivation period of ginseng is relatively long. In this study, the effect of medium concentration and sucrose concentration on hairy root culture of ginseng was evaluated. The optimization of ginseng hairy roots transformed by Agrobacterium rhizogene were performed liquid medium. The MS(Murashinge & Skoog basal medium) concentration was selected with 1/2 strength MS and the optimal sucrose concentration was determined at 2-3%(w/v). At the optimum culture condition, The yield (the ratio of weight of grown hairy root cultures to weight of fresh ginseng hairy roots) and production rate of ginseng root were 19.42 times and 5.73 g/l-day. The major ginsenosides were Rb group, Re and Rg1. The produced total ginsenoside content in the solid medium was 9.87 (mg/g) and increased 1.34 times in the liquid medium (13.23 mg/g). In solid culture, the contents of ginsenosides Rb, Re and Rg1 were 2.14, 3.65 and 1.87 mg/g, respectively. In liquid culture, the contents of ginsenosides Rb, Re and Rg1 were 3.54, 4.12 and 2.63 mg/g, respectively.
To develop the production of ginseng root using plant tissue culture technology, submerged culture conditions were optimized by means of the fractional factorial design with 4 factors and 3 levels by a RSM computer program. The ginseng (Panax ginseng C. A. Meyer) roots induced by plant growth regulators were cultured on SH medium and the effects of various pH of medium, sucrose concentration, nitrogen concentration and phosphate concentration on fresh weight of the ginseng root were investigated. The fresh weight of ginseng root increased with a decrease in nitrogen concentration and fresh weight of ginseng root varied from 1.00 to 2.33g under various conditions. The optimum pH of medium and sucrose concentration determined by a partial differentiation of the model equation, nitrogen and phosphate concentration were pH 5.6, sucrose 3.8%, nitrogen 50 mg/L and phosphate 80.7 mg/L, respectively. Under these conditions, the predicted growth of ginseng root was estimated to be 2.36g.
Effects of media and growth regulators on the growth and saponin accumulation of ginseng root were investigated to develop the ginseng root culture system. When Panax ginseng C. A. Meyer roots were induced and cultured in various liquid media, the maximum root growth and saponin production were obtained in SH medium and an initial doubleing time of ginseng root was approximately 10 days. The patterns and contents of ginsenosides of cultured ginseng root in various media were different from each other. SH and White media resulted in higher total ginsenosides contents than the other media. Among different combinations and concentrations of growth regulators, SH medium containing 4.0mg/ L NAA gave best growth of ginseng roots, while saponin content was highest in SH medium containing 0.5mg/L BAP. These results suggested that the rapid production of ginseng saponin is possible by root culture of Panax ginseng.
The effect of ginseng saponin on chick embryonic dorsal root ganglia organ culture and brain, spinal cord, muscle dissociation cultures was studied. The fiber outgrowth in explanted chick embryonic dorsal root ganglia was markedly induced by water and alcohol extracts of ginseng, total ginseng saponin, protopanaxadiol and protopanaxatriol glycosides as well as ginsenosides R/sub b1/, R/sub d/, R/sub 0/+R/sub a/+R/sub b1/, and R/sub b2/+R/sub c/+R/sub e/ mixtures. The life span of the cultured chick embryonic dorsal root ganglia and potentiation of nerve cell density were also observed with all of these ginseng saponins. The effect of ginseng saponin on chick embryonic dorsal root ganglia organ culture was more marked in the absence of the chick embryonic extract which was known to contain nerve growth factor-like material in the culture media. However, the ginseng saponin did not influence the cultured central nervous system such as brain and spinal cord cells and cultured skeletal muscle cells with respect to the morphological changes, maturation and life span of these cells.
In order to pursue some physiological studies on organogenesis in ginseng tissue culture, ginseng root explants were cultured on a modified MS medium containing NAA and kinetin. The activities of peroxidase and some enzymes were investigated and their isoenzyme patterns were also observed. The activity of peroxidase decreased by 20% in one week's culture and increased thereafter by 80% in culturing for 7 weeks compared with the control group. Glucose-6-phosphate dehydrogenase activity increased by 400% after culturing for 5 weeks and increased during the days preceeding root formation. The activities of glutamate dehydrogenase and acid phosphatase also increased during the culture. After 3 weeks' culture, new peroxidase isozyme (pH 7.6) appeared and 7 weeks' culture, another new peroxidase isozyme (pH unidentified) appeared. These patterns were also identified by using FPLC. After 7 weeks' culture, a new esterase isozyme of pH 8.5 appeared and isozyme patterns of acid phosphatase were quite changed compared with the isozyme patterns of tissue cultured for 5 weeks. In so far as these new isoenzymes appear distinctively after 7 weeks' culture, root differentiation is supposed to be induced after 7 weeks' culture.
In this study, the effects of auxin and casein hydrolysate (CH) on the growth of ginseng hairy root was elucidated. Ginseng hairy root was cultured under light and dark conditions in MS solid and liquid medium with various concentrations of auxin and CH for fifty days. After harvesting the cultures, the fresh and dry weight of cultures were examined, respectively. In the MS solid culture, 1 mg/L of IBA was most effective on the growth of ginseng hairy root under the dark condition, whereas IAA and CH did not affect on the growth of ginseng hairy root. In the MS liquid culture, the growth was maintained regularly by the treatments of IAA and NAA. IBA and CH restrained the growth of ginseng hairy root.
To elucidate the effect of chitin and chitosan on the production of ginsenosides and growth, ginseng hairy root was cultured on the 1/2 MS medium supplemented with chitin and chitosan of various concentrations and culture period. The highest growth was obtained with 1 mg/L of chitin. However, the growth was inhibited by 20 mg/L or above. The contents and productivity of ginsenosides were the highest when ginseng hairy roots were cultured on 40 mg/L chitin and applied of the third-weeks of culture period. Ginseng hairy root culture with 1 mg/L of chitosan resulted in the best growth, but the highest ginsenosides level was appeared in 30 mg/L chitosan. Ginsenosides content was increased when it was treated at the forth-week after culture as 30 mg/L of chitosan.
This study was carried out to obtain the basic information about the large scale propagation of ginseng (Panax ginseng C.A. Meyer). Therefore, the stem cuttings of 1-year old ginseng, treated with various concentrations of plant growth regulators for 5 seconds (quick dipping) and 24 hours (prolonged soaking), were cultured. The root formation of stem cuttings was varied with the concentrations, kinds, and treatment methods of plant growth regulators. Besides normal-looking roots various malformed roots were observed. In the prolonged soaking method, the culture of stem cuttings, treated with 10 ppm of IBA or NAA, resulted in profuse root regeneration. And stem cuttings, in quick dipping method, treated with 2000 ppm of IBA or NAA resulted in more excellent root regeneration. In general, IBA was more vigorous for the root formation than NAA, The treatment with 50 ppm kinetin or 100 ppm BA brought good result for the retardation of senescence of stem cuttings and BA treatment was more effective than kinetin. As for the saponin content of roots derived from stem cutting culture, the roots, formed by non-treatment of growth regulators, were higher in saponin content than those formed by treatment of growth regulators.

To investigate the effect of culture conditions on growth and ginsenosides accumulation, we cultured the ginseng hairy root under three different media, white light or ultra-violet irradiation. The MS/B5 medium containning MS basal salt and B5 vitamin was good for the growth and ginsenoside accumulation. The light during the culture period of ginseng hairy root was irradiated. The growth was abundant in the ginseng hairy root cultured in dark. But the ginsenosides accumulation was higher than in the ginseng hairy root cultured in the light irradiation. When the ginseng hairy root was cultured in 20 L bioreactor, the ginsenosides accumulation was observed at 34% higher than the hairy root cultured in dark. UV irradiated the ginseng hairy root during the culture period. The long time irradiation of UV was caused decreasing the growth of ginseng hairy root, but the accumulation of ginsenosidess was increased as to the irradiated time.
