Plant Growth Promoting Activities of Some Rhizosphere Bacteria and their Effect on Brassica rapa Growth

  • The necessity to develop economical and eco-friendly technologies is steadily increasing. Plant growth promoting rhizomicrobial strains PGPR are a group of microorganisms that actively colonize plant roots and increase plant growth and yield. Pot experiments were used to investigate the potential of some rhizobacterial strains to enhance the Brassica rapa growth. Microbial strains were successfully isolated from the rhizosphere of Panax ginseng and characterized based on its morphological and plant growth promotion characters. Surface disinfected seeds of Wisconsin Fast B. rapa were inoculated with the selected PGPR microorganisms. The different pots treatments were inoculated by its corresponding PGPR ($10^7cfu\;mL^{-1}$) and incubated in the growth chamber at $25^{\circ}C$ and 65% RH, the light period was adjusted to 24 hours (day). NPK chemical fertilizer and trade product (EMRO, USA) of effective microorganisms as well as un-inoculated control were used for comparison. Plants harvested in 40 days were found to have significant increase in leaf chlorophyll units and plant height and also in dry weight of root and shoot in the inoculated seedlings. Root and shoot length and also leaf surface area significantly were increased by bacterial inoculation in sterile soil. The study suggests that Rhodobacter capsulatus and Azotobacter chroococcum are beneficial for B. rapa growth as they enhance growth and induced IAA production and phosphorus solubilization. This study presents some rhizomicrobial strains that significantly promoted growth of Wisconsin Fast Plant B. rapa in pot experiment under different soil conditions.

The Radioprotective Effect of Kuei-Pi-Tang as a prescription of Traditional Chinese Medicine in Mice (생쥐에서 귀비탕 및 구성단미의 방사선 방호효과)

  • In this study, we evaluated the radioprotective effects of Kuei Pi Tang and its ingredients, as a prescription of traditional Oriental medicine. The jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells were investigated in mice irradiated with high and low dose of ${\gamma}$ rays. In the mice administered with Kuei Pi Tang extract before irradiation, the jejunal crypts were protected significantly(p<0.005) and the frequency of radiation induced apoptosis was reduced(p<0.05). In the experiment on the effects of the ingredients of Kuei Pi Tang, the results indicated that the extract of Dangui(Angelica sinensis), Suanzaoren(Zizyphus valgaris), Rensan (Panax ginseng), Fuling(Poria cocos) and Muxiang(Saussurea lappa) might have a major radioprotective effect. Although the mechanisms of this inhibitory effect remain to be elucidated, these results indi cated that Kuei Pi Tang might be a useful radioprotector, especially since it is a relatively nontoxic natural product.

  • The consumption of herbal medicine and related products (herbal products) have increased in South Korea. At the same time the quality, safety, and efficacy of herbal products is being raised. Currently, the herbal products are standardized and controlled according to the requirements of the Korean Pharmacopoeia, the National Institute of Health and the Ministry of Public Health and Social Affairs. The validation of herbal products and their medicinal component is important, since many of these herbal products are composed of two or more medicinal plants. However, there are no tools to support the validation process. Interest in deep learning has exploded over the past decade, for herbal medicine using algorithms to achieve herb recognition, symptom related target prediction, and drug repositioning have been reported. In this study, individual images of four herbs (Panax ginseng C.A. Meyer, Atractylodes macrocephala Koidz, Poria cocos Wolf, Glycyrrhiza uralensis Fischer), actually sold in the market, were achieved. Certain image preprocessing steps such as noise reduction and resize were formatted. After the features are optimized, we applied GoogLeNet_Inception v4 model for herb image recognition. Experimental results show that our method achieved test accuracy of 95%. However, there are two limitations in the current study. Firstly, due to the relatively small data collection (100 images), the training loss is much lower than validation loss which possess overfitting problem. Secondly, herbal products are mostly in a mixture, the applied method cannot be reliable to detect a single herb from a mixture. Thus, further large data collection and improved object detection is needed for better classification.

Fabrication of an ultra-fine ginsenoside particle atomizer for drug delivery through respiratory tract (호흡기를 통한 약액 전달을 위한 진세노사이드 초미세입자 분무장치 제작)

  • Objectives: The purpose of this study is to fabricate an ultra-fine ginsenoside particle atomizer that can provide a new treatment method by delivering ginsenoside components that have a therapeutic effect on respiratory diseases directly to the lungs. Methods: We fabricated the AAO vibrating mesh by using the micromachining process. The starting substrate of an AAO wafer has a 350nm pore diameter with 50㎛ thickness. A photomask having several 5㎛ opening holes with a 100㎛ pitch was used to separate each nanopore nozzle. The photoresist structure was optimized to pattern the nozzle area during the lift-off process precisely. The commercial vibrating mesh was removed from OMRON's NE-U100 product, and the fabricated AAO vibrating mesh was installed. A diluted sample of 20mL with 30% red ginseng concentrate was prepared to atomize from the device. Results: As a result of liquid chromatography analysis before spraying the ginsenoside solution, ginsenoside components such as 20S-Rg3, 20R-Rg3, and Rg5 were detected. After spraying through the AAO vibrating mesh, ginsenosides of the same component could be detected. Conclusion: A nutrient solution containing ginsenosides was successfully sprayed through the AAO vibrating mesh with 350 nm selective pores. In particular, during the atomizing experiment of ginsenoside drug solution having excellent efficacy in respiratory diseases, it was confirmed that atomizing through the AAO vibrating mesh while maintaining most of the active ingredients was carried out.

Studies on Nutrient Components between the Chungjung Chicken Meats and General Chicken Meats (청정닭고기와 일반닭고기의 영양성분에 대한 연구)

  • It was performed to find out difference of the nutritional contents between chungjung chicken meats grown chemicals free and general chicken meats. The chungjung chicken was eat and given feeds of good quality mixed living microbial agent, power of ginseng leaves and by-product of extract liquefied sea foods from fishes producted clean area of the Western Sea, including plentiful calciums, amino acids, docosahexanoic acid(DHA) etc. Addition of they improved chungjung chicken yields and meat quality compared to general chicken given feeds mixed antibiotic etc. Also, chungjung chickens eaten miced type develop resistance to in disease. There were significant trend in crude proteins, calciums, crude lipids contents, and unsaturated fatty acids between the chungjung chicken meats and the general chicken meats. The crude proteins and calciums contents were higher in the chungjung chicken meats than the general chicken meats, but the crude lipids contents were higher in the general chicken meats than that. Also, contents of the n6 fatty acids and n3 fatty acids in chungjung meats were higher than that. Especially, contents of DHA in the chungjung chicken meats were detected higher than the general chicken meats. Therefore, the results from this experiments will provide a information for the nutritional knowledge of the 'white meats', especially the clean chicken meats.

Effect of the Dried-Medicinal-Herbs Mixing Ratio on the Sensory and Quality Characteristics of Samgyetang for Ginseng Chicken Soup (한약재 재료의 혼합비율에 따른 한방 삼계탕의 기호도와 품질특성)

  • The physiochemical characteristics and sensory properties of the Samgyetang herb to which different dried medicinal herbs were added were investigated to find the best formulation for the product. For the Samgyetang herbal product, Schizandra chinensis, Atractylodes lancea, Glycyrrhiza uralensis, and Zizyphus jujube were used at a fixed ratio in the formulation, and Paeonia lactiflora (PL), Codonopsis lanceolata (CL), and Scutellaria baicalensis (SB) were added, with different amounts. The pH of Samgyetang increased along with the SB amount, and the soluble-solid content increased along with the CL amount. S7, which had the highest SB addition level, had the highest L value, and the b values were the highest in S7, which had the highest SB addition level. In the sensory evaluation, S7, which had the highest amount of SB, had the highest score in flavor, taste, and overall acceptability. In conclusion, the best formulation for Samgyetang would include PL 8 g, CL 4 g, SB 12 g, Schizandra chinensis 6 g, Atractylodes lancea 4 g, Glycyrrhiza uralensis 2 g, and Zizyphus jujube 4 g.

The Novel Approach of Gene Detection by Single-neuronal Cell Manipulation (단일 도파민뉴런을 이용한 새로운 유전자발현 검출기법)

  • RT-PCR is an useful method to investigate the expression of target gene as detection tools. Although RT-PCR is the powerful detection method for tissues, it was difficult to amplify the target gene product using the single cell. To clarify the expression level of the genes related to Parkinson's disease (PD), I performed the laser dissection of single cell from Substantia nigra. I examined the mRNA expression level in the dopaminergic neuron isolated from the PD patients by the single cell RT-PCR method. It is known that tyrosine hydroxylase (TH), DOPA decarboxylase (DDC) are involved in biosynthesis of the catecholamine such as dopamine. Little has been known about the gene expression features of these enzymes in single dopaminergic neuron. I could detect the specific gene products in single cell level. The different expression was observed in PD-related gene products from the single neuron of PD patients. Interestingly, TH gene expression was significantly decreased with comparing the ratio of decrease in other PD-related genes. Hence, I represented data that indicate the RT-PCR method described in this report is an effective method in detecting a specific single-cell mRNA level related with diseases.

Antioxidant Effect of Tocopherols and Tocotrienols and cis/trans-, trans/trans-Hydroperoxide Isomer from Linoleic Acid Methylester (토코페롤류의 항산화작용과 Linoleic Acid Methylester에서 생성된 cis/trans-, trans/trans-Hydroperoxide Isomer)

  • Antioxdant effect was studied in model system with linoleic acid methylester and tocochromanols $({\alpha}-,\;{\beta}-,\;{\gamma}-,\;{\delta}-tocopherol\;and \;{\alpha}-,\;{\gamma}-,\;{\delta}-tocotrienol)$ under definite autoxidation condition-temperature $(40,\;60,\;80^{\circ}C),\;O_2\;(0,\;10,\;20%\;O_2\;in\;N_2)$. 13-Hydroperoxy-9-cis-11-trans-, 13-hydroperoxy-9-trans-11-trans-, 9-hydroperoxy-10-trans-12-cis-, 9-hydroperoxy-10-trans-12-trans-octadecadienoic acid methylester as the major oxidation product were produced from linoleic acid methylester by autoxidation, analyzed with HPLC and antioxidant activities were compared by their quantitative changes. Experimental results showed that all added tocochromanols except ${\alpha}-tocotrienol$ had antioxidant effect at $60^{\circ}C$, and also ${\alpha}-tocopherol$, ${\alpha}-tocotrienol$ and ${\delta}-tocotrienol$ had prooxidant effect at $80^{\circ}C$. And cis/trans-hydroperoxide was predominantly produced at $40^{\circ}C$, but trans/trans-hydroperoxide at $80^{\circ}C$. Except no reproductive experimental data in produced hydroperoxides amount, the production ratio of cis/trans-:trans/trans-hydroperoxides in the autoxidation condition of range from $40^{\circ}C/10%,\;O_2\;to\;60^{\circ}C/20%\;O_2$ were as follows: ${\alpha}-T>{\alpha}-T_3>{\gamma}-T>{\beta}-T>{\gamma}-T_3>{\delta}-T>{\delta}-T_3$. This result showed that ${\alpha}-tocopherol$ among tocochromanols had the lowest antioxidant effect.

Detection of Mutations to Zidovudine in the pol Gene of Human Immunodeficiency Virus-1 by Direct Sequencing (인면역결핍 바이러스 pol 유전자 염기서열 결정에 의한 지도부딘 (ZDV) 내성 돌연변이의 탐지)

  • The nested polymerase chain reaction (PCR) assay was used to determine the sequences of reverse transcriptase (RT) codons 41, 67, 70, 210, 215 and 219 of human immunodeficiency virus-1 (HIV-1) pol gene. Template DNA was obtained from uncultured peripheral blood mononuclear cells from 27 Korean HIV-1 infected patients treated with ZDV and Korean red ginseng. The second PCRs were done for 2 separated regions (RT codons $13{\sim}98$ and $152{\sim}259$) with $5\;{\mu}l$ of the first PCR productNucleotide sequences were determined by direct sequencing. In the 27 patients, CD4+ cell count decreased from $230{\pm}117/{\mu}l$ to $152{\pm}162/{\mu}l$ for $46{\pm}26$ months (Mo), and actual duration of ZDV intake was $72{\pm}16$ Mo. In the 16 patients who had been treated with ZDV therapy ${\ge}25$ Mo, the incidences of 70R, 215F/Y, and 41L were 61%, 28% and 22%, respectively and those of 67N, 210W and 219Q were 17%. The incidences of 215F/Y were 6.7% for group ${\le}12$ Mo treatment, 22.7% for group with 13 to 24 Mo, and 27.8% for group ${\ge}25$ Mo. There was no mutation in 9 patients. It might be associated with the interruption of ZDV therapy for more than 6 months in 6 patients. This study shows that the detection of mutation could be useful prognostic marker with other clinical and virological data, and very low mutation rate is dectected compared to overseas reports.
