Development of an ISSR-Derived SCAR Marker in Korean Ginseng Cultivars (Panax ginseng C. A. Meyer)

  • Recently, new ginseng cultivars having superior agricultural traits have been developed in Korea. For newly developed plant cultivars, the identification of distinctiveness is very important factors not only in plant cultivar management but also in breeding programs. Thus, eighty-five inter simple sequence repeat (ISSR) primers were applied to detect polymorphisms among six major Korean ginseng cultivars and two foreign ginsengs. A total of 197 polymorphic bands with an average 5.8 polymorphic bands and 2.9 banding patterns per assay unit across six Korean ginseng cultivars and foreign ginsengs from 236 amplified ISSR loci with an average 6.9 loci per assay unit were generated by 34 out of 85 ISSR primers. Three species of Panax ginseng including the Korean ginseng cultivars, P. quinquefolius, and P. notoginseng, could be readily discriminated using most tested primers. UBC-821, UBC-868, and UBC-878 generated polymorphic bands among the six Korean ginseng cultivars, and could distinguish them from foreign ginsengs. Sequence characterized amplified region (SCAR) marker system was introduced in order to increase the reproducibility of the polymorphism. One SCAR marker, PgI821C650, was successfully converted from the randomly amplified polymorphism by UBC-821. It showed the expected dominant polymorphism among ginseng samples. In addition, the specific polymorphism for Sunwon was generated by treating Taq I restriction enzyme to polymerase chain reaction products of PgI821C650. These results will serve as useful DNA markers for identification of Korean ginseng, especially Sunwon cultivar, seed management, and molecular breeding program supplemented with marker-assisted selection.

고려인삼(Panax ginseng C.A. Meyer)의 년차간 형질상관 및 경로계수 분석 (The Correlation of Agronomic Characters and Path Coefficient Analysis in Panax ginseng C.A. Meyer)

  • This study was carried out to investigate the correlation of agronomic characters, their path coefficients in 2, 3 and 4-year old ginseng plants, and to provide a useful information for ginseng breeding. Correlation coefficients between stem fen변h, number of leaves and number of Iraflets in 2-year age, and stem diameter and leaf length in 3-year age showed highly significant correlation with number of fruits and root weight in 4-year age. The path coefficient analysis indicated that stem length and number of leaflets might give indirect effects on root weight regardless of plant age. On the other hand, stem length and number of leaflets in 2-year age and, stem diameter and leaf length in 3-year age showed direct effects on root weight in 4-year old ginseng. These results may be used for selection of high-yielding ginseng plants. Key words Selection information, correlation and path coefficient analysis.

Genetic Diversity of Wild and Cultivated Populations of American Ginseng (Panax Quinquefolium) from Eastern North America Analyzed by RAPD Markers

  • The objective of this study was to assess genetic diversity among 6 different wild ginseng populations from New York, Kentucky, North Carolina, Pennsylvania, Tennessee and Virginia, and to compare these wild populations to one cultivated population. RAPD markers were used to estimate the genetic difference among samples from the 7 populations. The 64 random primers were screened, and 15 primers were selected which exhibited the 124 highly reproducible polymorphic markers. The ratio of discordant bands to total bands scored was used to estimate the genetic distance within and among populations. Multidimensional scaling (MDS) of the relation matrix showed distinctive separation between wild and cultivated populations. The MDS result was confirmed using pooled chi-square tests for fragment homogeneity. This study suggests that RAPD markers can be used as population-specific markers for American ginseng.

Analysis of the chloroplast genome and SNP detection in a salt tolerant breeding line in Korean ginseng

  • The complete chloroplast genome sequence of Panax ginseng breeding line 'G07006', showing higher salt tolerance, was confirmed by de novo assembly using whole genome next-generation sequences. The complete chloroplast (CP) genome size is 156,356 bp, including two inverted repeats (IRs) of 52,060 bp, separated by the large single-copy (LSC 86,174 bp) and the small single-copy (SSC 18,122 bp) regions. One hundred fourteen genes were annotated, including 80 protein-coding genes, 30 tRNA genes, and 4 rRNA genes. Among them, 18 sites were duplicated in the inverted repeat regions. By comparative analyses of the previously identified CP genome sequences of nine cultivars of P. ginseng and that of G07006, five useful SNPs were defined in this study. Since three of the five SNPs were cultivar-specific to Chunpoong and Sunhyang, they could be easily used for distinguishing from other ginseng accessions. However, on arranging SNPs according to their gene location, the G07006 genotype was 'GTGGA', which was distinct from other accessions. This complete chloroplast DNA sequence could be conducive to discrimination of the line G07006 (salt-tolerant) and further enhancement of the genetic improvement program for this important medicinal plant.

RAPD Pattern of Ginseng(Panax ginseng C.A. Meyer) Lines Containing High Level of Ginsenoside

  • The important component for medical effect in ginseng is ginsenoside. Korea Ginseng & Tobacco Research Institute contains approximately 200 lines produced by inbred selection. It is assumed that ginseng lines containing high level of ginsenoside should be included in those lines. Besides, new breeding methods such as cell line selection in vitro and hairy root were recently developed. Therefore, this study was carried out to detect genes related to ginsenoside, and to use it for selection marker to select and distribute lines containing high level of ginsenoside. DNA was extracted from both ginseng roots and hairy roots, and the difference between the line containing high ginsenoside(KG101) and normal ginsenoside(KG103) were analysed. As a result, 28 out of 36 primers showed bands, and many primers showed band difference between ginseng lines. It is considered that the bands should be analysed using DNA sequence comparison to check if those are related to ginsenoside. In case of hairy roots of ginseng, almost no differences were found between two lines.

Gibberellins enhance plant growth and ginsenoside content in Panax ginseng

  • The roots of Korean ginseng (Panax ginseng) have a long history of usage as a medicinal drug. Ginsenosides, a group of triterpenioid saponins in ginseng, have been reported to show important pharmacological effects. Many studies have attempted to identify the ginsenoside synthesis pathways of P. ginseng and to increase crop productivity. Recent studies have shown that exogenous gibberellin (GA) treatments promote storage root secondary growth by integration of the modulating cambium stem cell homeostasis with a secondary cell wall-related gene network. However, the dynamic regulation of ginsenoside synthesis-related genes and their contents by external signaling cues has been rarely evaluated. In this study, we confirmed that GA treatment not only enhanced the secondary growth of P. ginseng storage roots, but also significantly enriched the terpenoid biosynthesis process in RNA-seq analysis. Consistently, we also found that the expression of most genes involved in the ginsenoside synthesis pathways, including those encoding methylerythritol-4-phosphate (MEP) and mevalonate (MVA), and the saponin content in both leaves and roots was increased by exogenous GA application. These results can be used in future development of biotechnology for ginseng breeding and enhancement of saponin content.

Thermotolerant Transgenic Ginseng (Panax ginseng C.A. Meyer) by Introducing Isoprene Synthase Gene through Agrobacterium tumefaciens-mediated Transformation

  • The cost of conventional cultivation of ginseng (Panax ginseng C.A. Meyer) is very expensive, because shadow condition should be maintained during cultivation periods owing to inherently weak plant for high-temperature. Therefore, application of plant biotechnology may be possible to overcome these difficulties caused by conventional breeding of ginseng. Transgenic plants were produced via Agrobacterium tumefaciens Gv3101, both carrying the binary plasmid pBI121 mLPISO with nptII and Iso (isoprene synthase) gene. Integration of the transgenes into the P. ginseng nuclear genome was confirmed by PCR analysis using nptII primers and Iso primers. RT-PCR result also demonstrated the foreign isoprene synthase gene in three transgenic plant lines (T1, T3, and T5) which was expressed at the transcriptional level. When whole plants of transgenic ginseng were exposed to high temperature at $46^{\circ}C$ for 1 h, a non-transformed plant was wilted from heat shock, whereas a transgenic plant appeared to remain healthy. We suggest that the introduction of exogenous isoprene synthase is considered as alternative methods far generating thermotolerance ginseng.

인삼(Panax ginseng C.A. Meyer) 신품종 고풍의 육성경과 및 생육특성 (Breeding Process and Characteristics of Gopoong, a New Variety of Panax ginseng C.A. Meyer.)

  • KT&G 중앙연구원 원료연구소에서 육성한 신품종 고풍에 대해서 육성경과, 생산력 검정 및 지역 적응시험을 수행하였다. 1968년에 선발하여 20년 이상 육성된 고풍은 줄기, 잎자루, 꽃대 및 잎맥부위까지도 진한 자색을 띤다. 열매송이 모양은 역삼각형이며 진한 붉은 색으로 재래혼계종인 자경종이나 기개발된 천풍 및 연풍과 지상부 표현형에서의 차이를 나타내었 다. 1981-1984년에 실시한 생산력 검정시험이나 1985-1990년에 실시한 지역적응력시험에서 대비구인 재래혼계종인 자경종과의 지상부 생육 제반 특성에서는 차이가 없었으나 유전적 특성의 발현 재현성이 있었다. 지하부 형질에서는 고풍의 생존율이 대비구인 자경종보다 2.6% 높았고, 고풍의 칸당 수량은 2.3 kg으로 자경종 2.2 kg보다 0.1 kg이 높았으며, 고급홍삼인 천지삼율은 고풍은 16.6%로 대비구인 자경종 9.4%보다 7.2%가 더 높았다. 따라서 고풍은 고급 홍삼제조용 원료삼 품종으로 가능성이 매우 높은 것으로 판단되었다.
