Improving Germination and Seedling Growth of Rigitaeda Pine Tree by Coating and Prime Treatment (리기테다 소나무 종자(種子)의 피복(被覆)과 전처리(前處理)에 의한 발아(發芽) 및 유묘(幼苗) 생장(生長) 촉진(促進))

  • Seeds of Rigitaeda pine tree(Pinus rigida${\times}$P. taeda) was primed with polyethylene glycol(PEG-6000) under different PEG concentrations, treatment period, and temperatures to test uniformity of germination. Coated seeds and PEG treated seeds were sown to compare germination, emergence, establishment, and seedling growth. The results obtained are summarized as follows : 1. As the concentration of PEG increased, as total germination percentage of pine tree was increased. Total germination percentage of pine tree seeds primed with PEG at 15 or $20^{\circ}C$ was better than the seeds treated at $10^{\circ}C$ compared to untreated seeds. 2. Germination rate was improved as primed period long and level of PEG concentration high. 3. Maximum germination rate was high with long PEG treatment period, and markedly improved when the seeds were primed at $20^{\circ}C$ 4. Priming with PEG at $20^{\circ}C$ for 12 days reduced time taken for germination. 5. Osmoconditioned seeds accelerated germination under drought condition with injurious effect of coated seeds by some chemicals. 6. There were much differences in establishment and dry matter production between drill and oversowing method, and primed seeds showed better performances than the coated seeds. Coating effects to seeds were better under drought soil condition rather than moderate moisture condition.

Effects of Seed Pre-treatment and Germination Environments on Germination Characteristics of Ligularia fischeri Seeds (종자 전처리 및 발아환경에 따른 곰취 종자의 발아특성)

  • This study was carried out in order to examine the germination characteristics of Ligularia fischeri seeds, and it was to develop the more efficient pre-treatment and production system of the seeds. It was performed by two ways - temperature control (10, 15, 20 and $25^{\circ}C$) and shading treatment (Full sunlight, 35%, 50%, 75% and 95% of full sunlight). Seed pre-treatment before the each experiment was carried out by temperature (with low temperature and wetting treatment (LTW) for 0, 15, 30, 45 and 60days) and shading treatment (with drying at room temperature (DRT), drying at low temperature (DLT) and water soaking (WS) for 2 days). Seeds of L. fischeri were, regardless of seed pre-treatment, germinated well at $10^{\circ}C$ and the more temperature went up, the more germination rate went down. As a result of surveying shading treatment, 75% shading with DRT was the highest germination rate (68.1%) and 95% shading with WS was the lowest germination rate (48.6%). It was showed over-growth under 95% shading treatment experiment and withered in the full sunlight. As a result of surveying the whole experiment, L. fischeri seeds pre-treated with LTW for 15 days germinated well at $10^{\circ}C$ and under 50~75% shading treatment.

Seed Germination and Effect of Water Depths on Seedling Establishment of Iron-coated Rice Seeds (벼 철분코팅종자의 발아 특성 및 담수깊이에 따른 초기입모 영향)

  • Germination characteristics, seedling emergence, and early seedling growth of iron-coated rice seeds, cultivars Daebo and Samdeokbyeo, under different water depths were compared with those of non-coated seeds (control) and the results evaluated to obtain basic information for establishing stable seedlings in direct water seeding. The total germination percentage of the two seed treatments was similar, but iron-coated seeds had slightly faster germination and shorter mean germination time than non-coated seeds. Water absorption rates of iron-coated seeds were lower than that of non-coated seeds during seed germination. The germination percentage of the two iron-coated rice seed cultivars showed a significant decline of 15-22% after one year of storage under natural conditions. The seedling emergence percentage and uniformity of the two rice cultivars were significantly higher in the iron-coated seeds at 1-13 cm water depths but the percentage of floating seedlings was lower in iron-coated seeds than in non-coated seeds. The iron-coated seeds had a high seedling emergence percentage of 91.3-93.3% at all flooding depths whereas the non-coated seeds had a significantly low seedling emergence percentage of 57.7-71.7% at a water depth of 13 cm. Moreover, the shoot dry weight and seedling health score of iron-coated seeds were significantly higher than those of non-coated seeds, while root dry weights were similar in iron-coated and non-coated seeds, regardless of water depth. These results suggest that iron-coated seeds are more appropriate for stable seedling establishment in direct water seeding than are non-coated seeds.

Effect of ultrasonication, salt solution and liquid smoke treatment on germination of Setaria italica seeds

  • The preference for domestic cereal crop of Setaria Italica has been increased with the recent interest in healthy foods. However, the productivity of Korean domestic produce, which has been decreasing due to the lack of cultivation technology and the low rate of mechanization during cultivation. Increase of germination ability will have a positive effect on the cultivation by reducing the labor of the manpower consuming and weeding work. Therefore, red light, ultrasonication, liquid priming and liquid smoke treatment that are effective for the germination of the seeds evaluated. The seeds of 1.4mm or more were used for the experiment. The priming solution used in the experiment was 1% $KH_2PO_4$ (74mM). During the priming, the light treated seeds at 2000 lux for 15, 30, 60 and 120 minutes. Ultrasonicationd treatment was performed for 5, 10, and 20 minutes at exposures of 60%, 80%, and 100% of ultrasound up to 21.6 kHz during priming. Light or ultrasound treated seeds transferred to priming treatment at $15^{\circ}C$ for 24 hours. The treatment of the liquid smoke was divided into the treatment of the liquid smoke alone and the treatment of the liquid smoke with the priming. The liquid smoke alone was diluted with distilled water without priming solution and the treatment of the liquid smoke was diluted with the salt priming solution. Both treatments were performed at 0.0%, 0.5%, 1.0%, 5.0%, and 10.0% of the liquid smoke (pH7) concentration at $15^{\circ}C$ for 6 hours. After each treatment, the seeds were dried to moisture content ranged 5-8% at $25^{\circ}C$ for 24 hours. All treatments showed better results than the non-treated control. Light treatment for 120 minutes improved for germination percentage (GP), Germination uniformity (GU) and heath seed percentage (HS). Ultrasonication treatment was most effective when treated with ultrasound at 21.6 kHz for 5 minutes in all germination characteristics. Ten % of the liquid smoke increase in 92% GP, 1.8 days MGT, $54%{\cdot}day^{-1}GR$, 0.76 GU and 88% HS comparing to non-treated control (72% GP, 2.3 days MGT, $45%{\cdot}day^{-1}GR$, 1.48 GU, and 63 % HS). This study showed that it is possible to obtain high germination by adding liquid smoke treatment to the seeds supplied to the farmers. The efficacy of light, ultrasonication, inorganic salt priming, and liquid smoke treatment on the seeds found in the experiment will be a positive alternative to labor force problems in the cultivation by improving germination.

Quality and Physiological Characteristics of Pinus densiflora Seeds Under Different Storage Periods and Temperatures (장기 저장과 저장 온도에 따른 소나무 종자 품질과 생리적 특성)

  • Our aim was to evaluate the quality of pine seeds after long-term storage and to analyze the correlation between germination and physiological characteristics. Therefore, we investigated the germination and physiological characteristics of seeds stored for different periods and storage temperatures. Pine seeds lost little viability and vigor after 12 years of storage at 4℃; it is even possible to prolong the storage period at -18℃. The deterioration process operating through long-term storage was characterized by a loss of seed vigor before a loss in seed viability. Electrical conductivity of the seed leachate was significantly higher in seeds that completely lost their viability, and was also found to be significantly correlated with the germination percentage, T50, mean germination time, and germination speed. Among the minerals leaked to the leachate, K had the highest concentration, followed by Na, Ca, Cu, Mg, and Fe, while Mn and Zn were not detected. Concentrations of K, Ca, Cu, Mg, and Fe in the leachate differed significantly by treatment, and in the cases of K, Ca, Cu, Na, and Mg, we observed large differences in the leachate between the seeds that were collected in 2003 and those collected in other years.. Germination percentage was significantly negatively correlated to the concentrations of Ca, Cu, K, Mg, and Na. The coefficient of uniformity of germination was significantly negatively correlated to Cu concentration. The other germination characteristics were not significantly correlated with the mineral concentration. Therefore, the mineral concentrations of pine seed leachate were not reliable indicators of seed deterioration when the differences in quality between seed lots were small. However, electrical conductivity could be an indicator of seed viability and vigor of pine seeds under long-term storage.

Effect of Light, $GA_3$, $KNO_3$ Treatments on the Germination of Chilean Wild Flower (칠레 야생화의 발아율에 미치는 광조건 그리고 $GA_3$, $KNO_3$ 처리 효과)

  • Chilean wild flower seeds showed low germination rate and lacked germination uniformity. This experiment was conducted to find out the effects of light conditions, $GA_3$ (50 mg/L, 100 mg/L, 200 mg/L) and $KNO_3$ (0.1%, 0.2%, 0.4%) treatments on germination of five species of Chilean wild flower seeds. The seeds of $Schizanthus$ $candidus$, $Schizanthus$ $hookerii$, and $Schizanthus$ $litoralis$ did not germinated in light condition. The others did not show a significant different germination rate under light and dark conditions. The germination rate of $Mimulus$ $luteus$, $Schizanthus$ $candidus$, $Schizanthus$ $hookerii$ increased 2.3, 5.6, and 3.2 times compared with control, respectively in $GA_3$-200 mg/L treatment, but that of $Rhodophiala$ $ananuca$ enhanced slightly in $GA_3$-50 mg/L treatment. Those results suggested that the germination rate of Chilean wild flower seeds enhanced by $GA_3$ treatment that is very efficient in breaking seed dormancy.

Coating Method and Germination Test of Coated Tobacco (Nicotiana tabacum L.) Seeds (담배종자의 피복대형화방법과 발아에 미치는 영향)

  • Experiments were conducted to develop a pelletization method of minute tobacco seeds for easy handling at seeding by hand or for mechanical seeding. Serpentine, zeolite, and talc were tested as coating materials and the resulting pills were compared in size uniformity, hardness, length of time needed for coating, percentage of pills containing a single seed, and speed of disintegration when submerged in water. Talc was the poorest and zeolite was good only in hardness. On the other hand serpentine was good in most characters above, except for brittleness. This defect, however, was overcome by double coating, first with serpentine and followed by zeolite. This new pelletizing method results in optimum hardness, uniform size, and high ratio of pellets containing one tobacco seed. When compared to bare tobacco seeds, the double-coated seeds did not differ in germination test.

Seed Treatment to Improve Germinability of Gourd (Lagenaria siceraria Standl.) (종자처리가 수박대목용 박 종자의 발아성에 미치는 영향)

  • Various methods for seed treatments have been devised to improve the rate and uniformity of seed germination of gourd (Lagenaria siceraria Standl.). Hydropriming and priming, the imbibition of seeds in distilled water and osmotic solutions respectively, are useful techniques for early establishment of uniform seedlings. In this study, the optimum conditions for hydropriming and priming were determined in gourd (Lagenaria siceraria Standl.) seeds. Effect of hydropriming was compared with those of osmotic priming. Seeds, hydroprimed or primed with 50 mM solution of $KH_2PO_4$ and $KNO_3$ at $30^{\circ}C$ for 2 days, showed shorter $T_{50}$ (number of days to 50% of the final germination percentage) and MDG (mean number of days to germination) than those unprimed. The relative effectiveness of hydropriming and priming in improving the germinability of gourd seeds was more evident when germinated at $20^{\circ}C$ than at $25^{\circ}C$ and $35^{\circ}C$.

Effects of Seed Specific Gravity by Germination and Seeding Emergence in Proso millet (Panicum miliaceum L.) (기장의 염수선 종자 정선법에 따른 발아 및 출현율 특성)

  • The experiment was implemented to gain basic data for stable proso millet (Panicum miliaceum L.) production through germination viability and establishment uniformity based on seed refining under seed selection with specific gravity of proso millet seed. Seeds of three proso millet cultivars were carefully selected by adjusting NaCl into 9 levels (1.000, 1.005, 1.010, 1.015, 1.020, 1.025, 1.030, 1.035, 1.04 g·L-1), and their standard germination rate, seeding quality and field emergence rate were investigated. Proso millets showed heavier thousand seed weight as specific gravity high. In standard germination, all of the 3 cultivars of selected seeding showed at least 91% germination rate. With respect to the 1.040 g·L-1 selection by specific gravity, 'Hwanggeum' had 98.9%, 'Ibaekchal', 99.4%, and 'Hwangsilchal', 98.9%, recording the highest. Seeding quality was investigated and it was found that, compared with the non-selected seeds, selected seeds had longer plant heights and averaged 2.0, 2.2 and 1.9 cm, respectably. In terms of leaf length, only 'Hwanggeum' and 'Ibaekchal' showed distinctive differences. No significance was found in stem diameter and SPAD. The seeds selected using specific gravity all had longer or heavier average root length, root weight and stem weight. Compared with non-selected seeds, the specific gravity of 1.040 g·L-1 had longer root lengths of 2.9, 3.0 and 2.9 cm. 1.040 g·L-1 was also found to have the heaviest root weight, and stem weight. The field emergence viability investigation found significance solely in the mean emergence time (MET) by the cultivar. High significance was found in percent emergence (PE) and emergence rate index (ERI) in the specific gravity for the selection of seeds. All of the three cultivars showed higher PE and ERI as specific gravity selection high. The 1.040 g·L-1 specific gravity selection had the highest PE of 90.7, 85.3 and 92.8%. For productivity increase based on improved proso millet germination rate and enhanced establishment uniformity, it seems favorable to employ 1.040 g·L-1 specific gravity selection using salt solution.

Characteristics of Seed and Germination of Rhododendron mucronulatum by Collection Dates and Germination Temperatures (채취 시기와 발아 온도에 따른 진달래의 종자 및 발아 특성)

  • This study was carried out in order to secure basic data of seedling mass propagation technique of Rhododendron mucronulatum which is the native tree species of Korea by surveying the characteristics of its fruit and seed. The fruits were collected at Mt. Goryeo in Ganghwa-gun on different dates in 2013; August 26th, September 5th, September 12th, October 4th. The seed germination test was carried out at 5, 10, 15, 20, 25 and $30^{\circ}C$. Moisture content of the fruit was highest (54.5%) in the fruit collected on September 5th. Number of the seeds in a fruit was 91.3 to 116.3, regardless of the collection date. Seed length was highest ($1947.4{\mu}m$) in the seeds collected on October 4th and seed width was highest ($727.3{\mu}m$) in the seeds collected on September 12th. Germination rate of the seeds was highest at $25^{\circ}C$ regardless of the seed collection date, which showed the highest value(27.3%) in the seeds collected September 12th. Meanwhile, the seeds were not germinated not at all at 5, 10 and $30^{\circ}C$. $T_{50}$ and mean germination time of the seeds got shorter at the higher temperature. Germination uniformity got lower when the collection date got later. Germination speed of the seeds was fastest at $25^{\circ}C$. According to the results of this study, it seems that the appropriate time to collect fruit and seed is between September 12th and October 4th, and the appropriate temperature for the seed germination is $25^{\circ}C$.
