Manufacturing Sunsik Smoothie with Lactic Acid Bacteria and Germinated Grain Enzyme and Its Characteristics (유산균 및 발아효소를 첨가한 선식 스무디의 제조와 특성)

  • Sunsik has been popular as well-being and healthy food to some Asian people, but it still has a limit to other foreigners because of its taste and appearance. This study tried to modify Sunsik into smoothie type for foreigners and investigate its physicochemical characteristics. Germinated black and brown rice was prepared. The germination condition of two cereals was steeping for 24 hr at room temperature, and then germinating for 24 hr at $30^{\circ}C$. After germination, the ${\alpha}$-amylase activity of germinated grains was 13~15 times higher than before germination. The enzyme activity of brown rice was 9.16 CU/g, but germinated brown rice was 152.63 CU/g. In case of black rice, enzyme activity before germination was 7.47 CU/g, and enzyme activity after germination was 97.96 CU/g. The lactic acid bacteria was grown in 50 g germinated brown rice powder with 100 ml malt solution, 30 g tomato juice, and 1.5 g rice bran. After manufacturing beverage using milk and Sunsik and the cell count of lactic acid bacteria was $1.3{\times}10^5CFU/ml$ enough to use starter. According to sensory test, the optimal concentration of Sunsik smoothie was 30 g Sunsik in 200 ml of milk. The viscosity was $5.97{\pm}1.2$ centipoise. The color of Sunsik beverage was evaluated as L value : $63.50{\pm}0.41$, a value: $-0.35{\pm}0.06$, and b value: $8.85{\pm}0.19$.

Inhibitory Effects of Water-soluble Extracts of Barley, Malt, and Germinated Barley on Melanogenesis in Melan-a Cells

  • In recent times, the demand for edible medication for the treatment of hyperpigmentation has increased significantly. Therefore, the discovery of a stable, safe and inexpansive antimelanogenic component from natural substances, such as grains, is of particular interest. The levels and activities of some metabolites and/or enzymes can be increased. In the present study, we investigated the antimelanogenic effects of water-soluble extracts from barley (BE), malt (ME) and germinated barley (GBE) in melan-a cells. The inhibitory effects of ME and GBE on melanin production were significantly greater than that of BE. Interestingly, the content of ferulic acid, the proposed active component of barley, was also higher in ME and GBE than in BE by HPLC analysis. Western blot analysis of the expression of melanogenic enzymes in melan-a cells treated with BE, ME or GBE indicated the expression of both tyrosinase and tyrosinase-related protein 2 (TRP-2) significantly decreased after treatment with BE, ME or GBE. These results suggest that besides BE, ME and GBE also inhibit melanin production most likely through suppression of tyrosinase and TRP-2 expression. ME and GBE were more efficacious at inhibiting melanin production than BE was and may also represent potential skin-whitening agents.

Quality and physicochemical characteristics of newly developed Sunsik products with germinated brown rice (발아현미 선식의 품질 및 이화학적 특성)

  • The quality and physicochemical characteristics of newly developed Sunsik products were investigated. Sunsik A, B, and C were prepared under different roasting temperatures and times (A: $220^{\circ}C$, 15 min; B: $220^{\circ}C$, 25 min; C, D, E: $230^{\circ}C$, 15 min). Sunsik D was prepared using grain flours passed through a 100 mesh screen, whereas the other products were passed through a 140 mesh screen. Sunsik E was prepared by the addition of germinated grains. The moisture content of Sunsik products ranged from 3.24-7.10%. The viscosity and sugar content ranged from 57-74 cP and $1.5-1.7^{\circ}Bx$, respectively. The pH values were similar among the samples. Sunsik D had the highest gelatinization enthalpy, whereas Sunsik C had the lowest. The viable cell counts were in the range from 3-4 log CFU/g and coliform groups were not detected. Sunsik B and E had higher scores for savory flavor and sweet taste than other Sunsik products.

Amylolytic Activity of Brown Rice and Black Rice during Germination (현미와 흑미의 발아과정 중 amylolytic activity)

  • Enzymatic activity in germinated cereal grains is important for the saccharification of starch materials. This study was conducted to investigate the amylolytic activities of germinating brown rice and black rice that have different amylose contents. Brown rice and black rice were steeped at room temperature for 24 h and germinated at 20 and $30^{\circ}C$ for 1, 2, and 3 days. ${\alpha}$- and ${\beta}$-Amylase activities in normal brown rice increased very slightly during the 3-day germination period, but the enzymatic activities were slightly higher in low-amylose (waxy type) brown rice. Diastatic power (DP), a measure of starch-saccharifying enzyme, was higher in the germinating brown rice with low amylose than in those with normal amylose content. ${\alpha}$- and ${\beta}$-Amylase activities in black rice increased gradually during germination, and DP of low-amylose black rice appeared to be higher than that of normal brown rice. Amylase activities in brown rice and black rice germinated at $30^{\circ}C$ were higher than those germinated at $20^{\circ}C$. Compared to brown rice, the overall amylolytic activity of germinated black rice was observed to be higher than that of brown rice.

Pasting Properties and Antioxidant Characteristics of Germinated Foxtail Millet and Proso Millet with Added Cooked Rice (발아 조와 기장 첨가 밥의 호화 및 항산화 특성)

  • This study was carried out to analyze the pasting and antioxidant characteristics of cooked mixed grains consisting of germinated millet and rice with varying addition rates and cooking methods. In this study, we chose two foxtail millets: glutinous ('Samdachal') and non-glutinous ('Samdame'), along with a glutinous proso millet ('Ibaekchal'). Cooked mixed grain rice-added germinated millet was cooked by general and high pressure cooking methods, with and without fermented alcohol. The peak and trough viscosity of germinated millet were decreased significantly with increasing amounts of germinated millet. The phenolic compounds and radical scavenging activity of cooked rice-added germinated millet increased significantly with the addition of germinated millet (p<0.05). The total polyphenol contents of cooked rice-added germinated Samdachal, Samdame, and Ibaekchal were 127.74~194.11, 128.20~192.56, and $128.88{\sim}171.28{\mu}g\;GAE/g$, respectively. The total flavonoid contents were 13.79~30.23, 14.77~25.99, and $15.28{\sim}29.56{\mu}g\;CE/g$, respectively. DPPH and ABTS radical scavenging activities were 3.31~5.73 and 6.31~9.69 mg TE/100 g for cooked rice-added germinated Samdachal; 3.14~8.58 and 6.05~10.19 mg TE/100 g for Samdame; and 3.37~7.45 and 6.27~9.27 mg TE/100 g for Ibaekchal, respectively. In this study, phenolic compounds and radical scavenging activity of cooked mixed grain rice-added germinated millet with different addition rates and cooking methods can be used as basic data for manufacturing processed products.

Characteristics of Malt Prepared with Covered Barley, Naked Barley and Wheat (쌀보리, 겉보리 및 밀을 이용한 엿기름의 특성)

  • Quality of Sikhe, the Korean conventional rice beverage depended on the characteristics of saccharifying activities of various amylases, intrinsic flavour, budding rate and so on. To improve the quality of Sikhe, characteristics of malt produced with wheat, covered barley and naked barley were evaluated. The germination rate of wheat was 82%, but those of naked and covered barley were 69% and 56% for 6 days, respectively. Malt prepared from germinated grains with 1.5~2.0 times length of buds had the highest saccharifying power. when the extraction of enzyme and reducing sugar was carried out at 5$0^{\circ}C$ for 4 hr, saccarifying power and reducing sugar contents were the highest. Malt of wheat had the highest saccharifying power. Malt of naked barley had higher saccharifying power than that of covered barley. The amylase types of wheat, covered barley and naked barley were similar to $\beta$-amylase.

Effect of Anaerobic Treatment on Carbohydrate-Hydrolytic Enzyme Activities and Free Amino Acid Contents in Barley Malt

  • Effects of acute anoxia on carbohydrate hydrolytic enzyme activities and free amino acid contents in malt were examined. Malts were prepared with barley grains germinated for 7 days which contained the highest levels of amylolytic and(1-3,1-4)-$\beta$-glucanase activities. $\alpha$-Amylase and $\beta$-amylase activities in malts were not significantly affected by anoxia for 5 or 10 h.(1-3,1-4)-$\beta$-Glucanase activity, however, decreased about 7 to 10% by anoxia for 5 or 10 h. Alanine and $\gamma$-aminobutyric acid content changed drastically. Alanine contents in malts increased by 2.2- and 2-fold, and $\gamma$-aminobutyric acid contents by 1.4- and 1.9-fold under anoxia for 5 and 10 h, respectively.

Changes in Nutritional Components throughout Germination in Paddy Rice and Brown Rice

  • The aim of this study was to investigate changes in 7 nutritional components (fatty acid, protein, fat, ash, total dietary fiber (TDF), $\gamma$-aminobutyric acid (GABA), and $\gamma$-oryzanol) of paddy rice (PR) and brown rice (BR) throughout the germination process, as measured at different shoot lengths (10 mm, 20 mm, and 30 mm). With the increase of shoot length, the nutritional components' concentrations increased, as compared to the concentrations measured before germination. Moreover, BR exhibited higher GABA, $\gamma$-oryzanol, and protein than PR. Among the components, TDF, GABA, and $\gamma$-oryzanol showed significant concentration differences throughout germination, while the others exhibited only slight variations. In particular, GABA and $\gamma$-oryzanol were predominantly increased in grains of 10 mm shoot length. These compounds might prove to be important factors from germinated rice. Additionally, the germinated cultivar 'Keunnun' might also prove to be a very important food source, owing to its high GABA and $\gamma$-oryzanol contents. These results suggest that variations in nutritional components related to the increase of shoot length may prove to be important when considering the beneficial aspects of rice on human health.

  • In order to get information on the ecology of rice false smut, germination ability and pathogenicity of sclerotia and chlamydospores of the pathogen, environmental conditions affecting the disease outbreak and varietal resistance have been investigated. 1. The degree of outbreak of rice false smut was higher in the upland rice in comparison with the paddy field rice in respect to the number of affected grains per ear, the size and weight of smut balls formed on affected grains as well as the ratio of sclerotial formation produced on smut balls. 2. Germination percentage and days required for germination of overwintered sclerotia placed on the soil surface in July were 81% and 19 days, respectively, while those of overwintered sclerotia treated in May were 60-70% and 41 days. Sclerotia placed on the soil surface or under 1 cm depth of the soil surface and incubated at $25-30^{\circ}C$ were germinated well, whereas those placed under 3 cm or 5 cm depth of the soil surface were not germinated at all. Germinability and stroma productivity of sclerotia were reduced when the sclerotia were cutted into small pieces. 3. The average number of stroma formed on a sclerotium was six and that of perithecia formed in a stroma was about 50 to 140. 4. Percentage of germination of chlamydospores on the yellow balls was very high and was decreased as the color of the balls being darken with maturation. 5. Panicle of rice plants were successfully infected by injection inoculation with suspention of ascospores and chlamydospores of the pathogen to the sheaths at the booting stages, while seeding infection by spraying with suspensions of chlamydospores was unsuccessful. 6. More number of infected grains was distributed on basal parts of an affected ear than that of infected ones distributed upper parts of the ear, when the affected ear was divided into five parts from its basal portion to the apical of the ear. 7. The occurrence of the disease was more severe in the late maturing varieties of rice in comparison with the early maturing varieties. 8. When the level of nitrogen applied was increased, the incidence of disease increased, and the infection percentage of the disease was increased as the transplanting date was delayed. 9. The weight of panicles and 1000 kernels and the ratio of ripenness were reduced, and the contamination degree of grains with chlamydospores were increased as the number of smut balls per panicle were increased.

Studies on Incompatibility in Interspecific Hybrid Between Panax ginseng C. A. Meyer and Panax quinquefolium L. (고려인삼과 미국삼의 종간 잡종식물체 불화합성에 관한 연구)

  • This study was carried out to clarify the cause of incompatibility in interspecific hybrid plant between Panax ginseng and p. quinquefolium. The floral structure of F,(p.g. x p.q.) hybrid was normal because the redundant anther was 0.2 mm longer than pistil in Fl hybrid and the size and structure of redundant carpel in F, hybrid were similar to P. ginseng and p. quiquefolium Pollens of $F_1$ hybrid did not germinate on stigma of P-quinquefolium but germinated well on stigma of P. ginseng. Pollen tube was able to penetrate styles completely and seed harvest rate was 16.8% in field. However on stigma of $F_1$ hybrid, Pollen did not germinate when P. ginseng was used as male Parent. In addition, the growth of pollen tube was halted on style and seed was not set when P qlfinquefoEi2a was used as male Parent. These suggest that the inhibitor of pollen germination present on stigma caused $F_1$ hybrid sterility. It took 5 hours for pollen grains to germinate, 12 hours to arrive at in trance of ovule, 16 hours to penetrate micropyles in Panax ginseng.
