길항성 근원 세균이 딸기 시설재배에서 발생하는 잿빛곰팡이병의 생물학적 제어에 미치는 영향 (Effects of Antagonistic Rhizobacteria on the Biological Control of Gray Mold in Greenhouse Grown Strawberry Plants)

  • This study was carried out to clarify the effects of antifungal bacterial strains isolated from the greenhouse soil grown strawberry plants on the growth inhibition of plant pathogen, gray mold (Botrytis cinerea) infected in strawberry plants in Damyang and Jangheung districts. Antagonistic bacterial strains were isolated and investigated into the antagonistic activity against gray mold. Screened ten bacterial strains which strongly inhibited Botrytis cinerea were isolated from the greenhouse grown strawberry plants, and the best antifungal microorganism designated as SB 143 was finally selected. Antifungal bacterial strain SB 143 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. Bacillus sp. SB 143 showed 59.4% of antifungal activity against Botrytis cinerea. By the bacterialization of culture broth and heated filtrates of culture broth, Bacillus sp. SB 143 showed 93.1% and 32.1% of antagonistic activity against Botrytis cinerea.

카네이션의 시설재배에서 길항성 세균을 이용한 Fusarium Wilt 의 생물학적 방제 (Biological Control of Fusarium Wilt of Carnation Plants by Antagonistic Microorganism in Greenhouse)

  • This study was carried out to screen and select the effects of antifungal bacterial strains which inhibit the growth of plant pathogen, Fusarium oxysporum(fusarium wilt) occurred in carnation plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it's identification. Twenty bacterial strains which strongly inhibited Fusarium oxysporum were isolated from roots of carnation plants and the soil in greenhouse, and the best antifungal bacteria designated as C121, was finally selected. Antagonistic bacterial strain, C121 was identified to be the genus Bacillus sp. based on the morphological, biochemical and cultural characterizations. The Bacillus sp. C121 showed 58.1% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the cultural broth and the heat bacterialization culture filtrate of it, Bacillus sp. C121 was shown 92.1% and 21.0% of antifungal activity, respectively.

온실재배 토마토에서 발생하는 위조병의 미생물학적 제어 (Biological Control of Fusarium Wilt of Tomato Plants by Antagonistic Microorganism in Greenhouse)

  • This study was conducted to screen the antagonistic bacteria which inhibit the growth of plant pathogen, fusarium wilt(Fusarium oxysporum) occurred in tomato plants in greenhouse. We isolated an effective bacterial strains and investigated into the antifungal activity of the antagonistic microorganism and it’s identification. Ten bacterial strains which strongly inhibited Fusarium oxysporum were isolated from the nature, and the best antagonistic bacterial strain designated as KC175, was selected. The antagonistic strain KC175 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. The Bacillus sp. KC175 showed 58.2% of antifungal activity against the growth of Fusarium oxysporum. By the bacterialization of the culture broth and the heat bacterialization culture filtrate of it, Bacillus sp. KC175 showed 91% and 18% of antifungal activity, respectively.

제주 전통 발효식품 쉰다리에서 분리한 세균의 군집 조사 및 어류질병세균과 인체유해세균에 대한 항균활성효과 (Phylogenetic Diversity and Antibacterial Activity of Bacteria from Shindari of Jeju Traditional Fermented Food)

  • 옛날부터 제주도는 지형적인 특징으로 인하여 보리를 주식으로 사용하였다. 식은 보리밥을 누룩과 함께 단기간 발효시켜 그대로 음용 또는 단맛을 첨가하여 단술 또는 쉰다리라 불리는 제주 발효음료를 만들어 먹었다. 막걸리와 비슷한 원료로 짧은 시간 발효하여 만들어지는 쉰다리는 막걸리의 연구에 비해 미비한 실정이다. 이에 본 연구에서는 제주 전통 막걸리인 쉰다리에 군집되어있는 세균을 파악하였다. 또한, 분리 균주로부터 어류와 인체 유해 세균에 대한 항균 활성을 확인하였다. 분리 균주에서 Firmicutes 73%와 Proteobacteria 27%로 Firmicutes문이 우점문으로 나타났다. 또한, Pediococcus속, Bacillus속이 각각 25%로 가장 우세하였고, 다음으로 Cronobacter속 22%, Enterococcus속 16%, Aneurinibacillus속 5%, Klebsiella속 4%, Paenibacillus속이 2% 순으로 나타났다. 항균 활성 결과에서는 어류질병 세균 P. Damselae와 인체 유해세균 S.mutans를 제외한 모든 균에 대해서 생육저해 환을 나타냈다.

제주 연안의 해수로부터 분리한 Cellulase 생산균 Bacillus sp. GC-1과 GC-4의 동정 (Identification of a Cellulase Producing Marine Bacillus sp. GC-1 and GC-4 Isolated from Coastal Seawater of Jeju Island)

  • GC-1과 GC-4로 명명된 두 종의 그람 양성 박테리아가 제주도 연안해수로부터 동정되었다. 이 두 균주는 16S rRNA 유전자 염기서열 분석과 생리적 특성 분석결과를 토대로 Bacillus 속의 박테리아로 규명되었다. 균주 GC-1의 16S rRNA 유전자 염기서열은 B. tequiliensis와 B. subtilis subsp. inaquosorum의 16S rRNA 유전자 염기서열과 99.91%의 상동성을 보였고, 균주 GC-4의 16S rRNA 유전자 염기서열은 B. altitudinis, B. stratosphericus 및 B. aerophilus의 16S rRNA 유전자 염기서열과 100%의 상동성을 보였다. 그러나 두 균주의 생리학적-유전학적 특성 분석 결과, 이들이 계통적 유연관계를 갖는 다른 Bacillus 속의 균주들과 상당한 차이가 있었고, 따라서 조사된 Bacillus 속과는 다른 속에 속할 가능성이 높았다. 이러한 결과는 Bacillus 속이 진화과정 중에 다양한 변종으로 진화되었음을 암시한다.

메주 발효 관련 Bacillus, Enterococcus, Staphylococcus 속 우점종 확인 (Identification of the Predominant Bacillus, Enterococcus, and Staphylococcus Species in Meju, a Spontaneously Fermented Soybean Product)

  • 국내 5개 지역에서 수집한 12개 메주로부터 Bacillus, Enterococcus, Staphylococcus 속 bacteria를 선택배지를 이용하여 분리하고, 16S ribosomal RNA 유전자 및 gmk(guanylate kinase) 유전자 염기서열분석을 통해 동정하여, 이들 속의 우점종을 검토하였다. Bacillus 속과 Enterococcus 속은 모든 시료로부터 검출되었고, Bacillus 속은 11개 시료에서 총균수 대비 15% 이상 검출되어 메주 발효에서 가장 우점하는 bacteria로 확인되었다. Staphylococcus 속은 6개 시료에서만 검출되었다. Bacillus 속으로 동정된 151개 분리주는 B. velezensis, B. sonorensis 순으로 우점하였고, B. subtilis, B. licheniformis가 그 뒤를 이었다. Enterococcus 속으로 동정된 165개 분리주 중, 163균주가 E. faecium으로 확인되었다. Staphylococcus 속으로 동정된 82개 분리주에는 6종의 coagulase-negative Staphylococcus 존재가 확인되었고, S. xylosus가 우점종으로 확인되었다.

Xylanase 생산균 Bacillus sp. MX47의 분리 및 동정 (Isolation and Characterization of a Xylanolytic Bacterium, Bacillus sp. MX47)

  • A xylanolytic bacterial strain, MX47, was isolated from rotting plant matter in soil. The strain was aerobic and gram positive, and grew between pH 6.0 and 11.0. Cells were susceptible to thiostrepton and chloramphenicol. The major fatty acids (>3%) comprised 64.55% of iso-$C_{15:0}$, 22.76% of anteiso-$C_{15:0}$, and 3.92% of iso-$C_{17:0}$. The G/C content of the DNA was 44.15 mol%. The predominant respiratory quinone was menaquinone 7 (MK-7). Searches for 16S rRNA gene sequence similarity as well as phylogenetic analyses strongly suggested that the strain should be classified to the genus Bacillus. However, its biochemical characteristics, including acid production and enzyme activities, are different from those of other Bacillus strains in the same clade, and therefore, we propose the name Bacillus sp. MX47.

Isolation and Characterization of Mannanase-Producing Bacillus amyloliquefaciens YJ17 from Spent Mushroom (Flammulina velutipes) Substrates

  • The mannanase-producing bacteria, designated YJ17, was isolated from spent mushroom (Flammulina velutipes) substrates. The isolate YJ17 was a facultative anaerobic and was grown at temperatures ranging from $20^{\circ}C$ to $50^{\circ}C$ with an optimal temperature of $40^{\circ}C$. The DNA G+C content of the YJ17 was 44 mol%. The major fatty acids were anteiso-15:0 (38.9%), 17:0 (7.6%), and iso-15:0 (36.5%). The 16S rRNA gene sequence similarity between the isolate YJ17 and other Bacillus strains was from 98% to 99%. In the phylogenetic analysis based on these sequences, the isolate YJ17 and Bacillus amyloliquefaciens clustered within a group together and separated from other species of Bacillus. Based on the physiological and molecular properties, the isolate YJ17 was classified within the genus Bacillus as B. amyloliquefaciens YJ17. The optimal pH and temperature for mannanase activity of B. amyloliquefaciens YJ17 were pH 7.0 and $50^{\circ}C$, respectively.

호알칼리성, 고온성 Bacillus sp. TA-11의 분리와 $\beta$-galactosidase의 생산 (Isolation of Alkalophilic, Thermophilic Bacillus sp. TA-11 and Production of $\beta$-Galactosidase)

  • $\beta$-galactosidase를 강력하게 생산하는 호알칼리성, 고온성 세균 TA-11를 부엽토에서 분리하여 Bacillus sp. TA-11로 동정 하였다. Bacillus sp. TA-11에 의한 $\beta$-galactosidase 생산은 lactose 1.5%, peptone과 yeast ext. 각각 0.4%, $MgSO_4$ 0.2%, $NH_4$CL 0.05% 및 NaCl 0.2% 등을 함유한 배지의 pH를 10.0으로 하여 시험균을 접종한후 $50^{\circ}C$에서 2일간 진탕배양 하였을 때 가장 좋았다.

Isolation of Bacillus sp. SHL-3 from the Dry Soil and Evaluation of Plant Growth Promoting Ability

  • Excess use of chemical fertilizer causes soil acidification and accumulation of salt, and thus might bring to desertification of soil. To overcome this problem, it needs limited usage of chemical fertilizer and increased usage of natural fertilizer as an alternative. In this study, dry soil-borne Bacillus sp. SHL-3, which was isolated from arid and barren soil, with plant growth promoting activity was isolated for identification and to determine optimal culture condition. A bacterial strain SHL-3 had the IAA productivity ($5.16{\pm}0.10mg\;L^{-1}$), ACC deaminase activity ($0.36{\pm}0.09$ at 51 hours) and siderophore synthesis. It was identified as genus Bacillus sp.. Also, optimal culture condition of SHL-3 were $20^{\circ}C$ and pH 7 in LB medium. Bacillus sp. SHL-3 had up to 4% salt tolerance in the medium. We evaluated the plant growth promotion ability of SHL-3 using yam (Dioscorea japonica Thunb.). As a result, Bacillus sp. SHL-3 was effective on the increase of the shoot length (202.4% increase for 91 days). These results indicate that Bacillus sp. SHL-3 can serve as a promising microbial resource for the biofertilizers of soil.
