Estimation of Genetic Variations and Selection of Superior Lines from Diallel Crosses in Layer Chicken (산란계종의 잡종강세 이용을 위한 유전학적 기초연구와 우량교배조합 선발에 관한 연구)

  • The subject of this study was to obtain some genetic information for developing superior layer chickens. Heterosis and combining ability effects were estimated with 5,759 progenies of full diallel crosses of 6 strains in White Leghorn. Fertility, hatchability, brooder-house viability, rearing- house viability, laying-house viability, age at 1st egg laying, body weight at 1st egg laying, average egg weight, hen-day egg production, hen-housed egg production, and feed conversion were investigated and analyzed into heterosis effect, general combining ability, specific combining ability and reciprocal effect by Grilling's model I. The results obtained were summarized as follows; 1. The general performance of each traits was 94.76% in fertility, 74.05% in hatchability, 97.47% in brooder-house viability, 99.72% in rearing-house viability, 93.81% in laying-house viability, 150 day in the age at 1st egg laying, 1,505g in the body weight at 1st egg laying, 60.08g in average egg weight, 77.11% in hen-day egg production, 269.8 eggs in hen-housed egg Production, and 2.44 in feed conversion. 2. The heterosis effects were estimated to -0.66%, 9.58%, 0.26%, 1.83%, -3.87%, 3.63%, 0.96%, 4.23%, 6.4%, and -0.8%, in fertility, hatchability, brooder-house viability, laying-house viability, the age at 1st egg laying, the body weight at 1st egg laying, average egg weight, hen-day egg Production, hen-housed egg production and feed conversion, respectively. 3. The results obtained from analysis of combining ability were as follows ; 1) Estimates of general combining ability, specific combining ability and reciprocal effects were not high in fertility. It was considered that fertility was mainly affected by environmental factors. In the hatchability, the general combining ability was more important than specific combining ability and reciprocal effects, and the superior strains were K and V which the additive genetic effects were very high. 2) In the brooder-house viability and laying-house viability, specific combining ability and reciprocal effects appeared to be important and the combinations of K${\times}$A and A${\times}$K were very superior. 3) In the feed conversion and average egg weight, general combining ability was more important compared with specific combining ability and reciprocal effects. On the basis of combining ability the superior strains were F, K and B in feed conversion, F and B in the average egg weight. 4) General combining ability, specific combining ability and reciprocal effects were important in the age at 1st egg laying and the combination of V ${\times}$F, F${\times}$K and B${\times}$F were very useful on the basis of these effects. In the body weight at 1st egg laying, general combining ability was more important than specific combining ability and reciprocal effects, relatively. The K, F and E strains were recommended to develop the light strain in the body weight at 1st egg laying. 5) General combining ability, specific combining ability and reciprocal effects were important in the hen-day egg production and hen-housed egg production. The combinations of F${\times}$K, A${\times}$K, and K${\times}$A were proper for developing these traits. 4. In general, high general combining ability effects were estimated for hatchability, body weight at 1st egg laying, average egg weight, hen-day egg production, hen-housed egg production, and feed conversion and high specific combining ability effects for brooder-house viability, laying house viability, age at 1st egg laying, hen-day egg production and hen-housed egg production, and high reciprocal effects for the age at 1st egg laying.

Evaluation of Germplasm and Development of SSR Markers for Marker-assisted Backcross in Tomato (분자마커 이용 여교잡 육종을 위한 토마토 유전자원 평가 및 SSR 마커 개발)

  • This study was conducted to achieve basal information for the development of tomato cultivars with disease resistances through marker-assisted backcross (MAB). Ten inbred lines with TYLCV, late blight, bacterial wilt, or powdery mildew resistance and four adapted inbred lines with superior horticultural traits were collected, which can be useful as the donor parents and recurrent parents in MAB, respectively. Inbred lines collected were evaluated by molecular markers and bioassay for confirming their disease resistances. To develop DNA markers for selecting recurrent parent genome (background selection) in MAB, a total of 108 simple sequence repeat (SSR) primer sets (nine per chromosome at average) were selected from the tomato reference genetic maps posted on SOL Genomics Network. Genetic similarity and relationships among the inbred lines were assessed using a total of 303 polymorphic SSR markers. Similarity coefficient ranged from 0.33 to 0.80; the highest similarity coefficient (0.80) was found between bacterial wilt-resistant donor lines '10BA333' and '10BA424', and the lowest (0.33) between a late blight resistant-wild species L3708 (S. pimpinelliforium L.) and '10BA424'. UPGMA analysis grouped the inbred lines into three clusters based on the similarity coefficient 0.58. Most of the donor lines of the same resistance were closely related, indicating the possibility that these lines were developed using a common resistance source. Parent combinations (donor parent ${\times}$ recurrent parent) showing appropriate levels of genetic distance and SSR marker polymorphism for MAB were selected based on the dendrogram. These combinations included 'TYR1' ${\times}$ 'RPL1' for TYLCV, '10BA333' or '10BA424' ${\times}$ 'RPL2' for bacterial wilt, and 'KNU12' ${\times}$ 'AV107-4' or 'RPL2' for powdery mildew. For late blight, the wild species resistant line 'L3708' was distantly related to all recurrent parental lines, and a suitable parent combination for MAB was 'L3708' ${\times}$ 'AV107-4', which showed a similarity coefficient of 0.41 and 45 polymorphic SSR markers.

QTL Mapping for 6-Year-Old Growths of a Single Open-Pollinated Half-Sib Family of a Selected Clone 7-1037 in Loblolly Pine(Pinus taeda) and Average Effect of QTL Allele Substitution (테다소나무 7-1037 클론의 단일 반형매 풍매가계 6년생 생장에 대한 QTL mapping과 QTL 대립유전자 치환의 평균효과)

  • We conducted QTL mapping for 6-year growths of open-pollinated half-sib progenies from a selected clone 7-1037 in Pinus taeda. With an AFLP marker analysis on haploid DNA samples from the megagametophytes of the open-pollinated seeds, we constructed 20 framework maps spanning a total of 1,869 cM in total length and 18.5 cM in an average interval length between markers. Composite interval mapping reveals that one QTL explains 5.9% of the total phenotypic variation of height, and three QTLs account for 3.9~5.6% of the variation of diameter at breast height (DBH). There are no correlations between the QTLs. The genetic effects of the QTLs are 39.6 cm in height and 7.20~9.41 mm in DBH, respectively, The average effects of gene substitution of the markers closely linked with the QTLs are 44.3 cm in height and 8.38~11.81 m in DBH. Under an assumption that the within-family heritability for the growth traits of loblolly pine is less than 0.2, the QTLs account for 26.8% of the additive genetic variance of the progenies. In terms of relative selection efficiency, the individual selection based on QTL markers could be 5 times as high as phenotypic selection. The results in this study indicate that the QTL mapping method with open-pollinated half-sib family could be more practical and applicable to the conventional seed orchard-based selection work than other mapping methods with a single full-sib family, in particular from the viewpoint that it can provide crucial information for within-family individual selection such as breeding value.

  • This study was carried out to estimate genetic correlations among broiler body weights for the selection of broiler breeders at early age, and to investigate the difference of growth rates between male and female in mixed-sex population of broiler from 2wks to 8 wks of age. A total of 1109 records were used in this study. the heritability estimates from combined variance components for 2-,4-, 6- and 8- weeks body weights were 0.32, 0.31, 0.35 and 0.29 respectively. Genetic correlations were 0.91 between 4wks and 8wks body weights, 0.94 between 6wks and 8wks body weights. Records obtained on 4wks and 8wks might be useful in a selection program to improve traits in 8wks broiler weight. Growth rates of male and female from nonlinear equations from 2wks to 8wks were shown differently in both linear and quadratic terms, Even though both sexes showed closely equal body weights at 2wks of age, they manifested great difference about 300g at 8wks of age. At all. ages roles were havier, grew at a faster than female. From this result it might be profitable either to select the combination of lines which has little sex effect or to rear male and female broiler separately up to the marketing body weights.

  • Garlic (Allium sativum L.) has been used as both food and medicine in many cultures for thousands of years. Garlic cultivars are completely sterile and propagated through vegetative method. Collection of a large number of fertile accessions of these genus is needed to explore genetic variability. In order to investigate genetic variation among Allium species and its possibilities for direct cultivation in Korea, we characterized 12 accessions of A. longicuspis, flowering wild garlic which had collected from Central Asia, the main center of garlic diversity. Most of A. longicuspis accessions showed higher over-wintering and bolting rate, longer scape length and more number of bulbils than Korean landraces cultivar, Danyang and Euiseong, but A. longicuspis accessions exhibited smaller size of bulbs and bulbils. Most accessions of A. longicuspis had more number of cloves per bulb, except K229596 and K248824 than Korean landraces. All the accessions of A. longicuspis from Central Asia had complete bolters having many flowers and topsets in umbel. Further studies of A. longicuspis should focus on securing true seeds through removal of topsets and crosses among accessions to create the genetic variability.

STUDIES ON THE DIMORPHISM AND TRANSITION OF BISEXUALITY OF HETEROSTYLOUS POLYGONACEAE (여뀌과 이형경식물의 Dinorphism과 Bisexuality의 변화)

  • The present experiments were designed in order to clarify the differences between the long and short styled plants and the transgressive gradition in the degree of dimorphism among the three heterostylous species of the Polygonus, P. japonica, F. esculentum, and P. senticosa, based on investigations regarding the floral structure, ecological and physiological traits, the results of which are summarized as follows: (1) P. japonica, although it exhibits typical dimorphism, has undergone so high a differentiation between long and short styled that its long styled individuals behave as if they were female; and short styled individuals as if male. In long-styled individuals, filament, anther, and pollen grains show signs of degeneration, most of the pollen being abortive. On the other hand, in short styled individuals, the filament, anther, and pollen grains have attained remarkable development; the pollen grians are large and fertile. In short-plant the fertilized flowers readily drop off in every stage of their embryo development. This species has completely lost the self-fertile property, which is characteristic of the non-dimorphic Polygonum genus. Although this specsei typically exhibits the physiological characteristics of the non-dimorphic Polygonum genus. Although this specisei typically exhibits the physiological characteristics of dimorphism in controlled pollination, the short-styled individuals bear no seed in nature, thus misleading taxonomists to idenfity the short-styled plant as male. 2) The morphological feature of the flower organ of P. senticosa obviously indicates definite dimorphism. Physiologically, however, no differentiation towards dimorphism was observed, the species still retaining, both in long and short-individuals, the self-fertile property common to the Polygonum genus. Elaborate examinations revealed that regardless of the modes of pollination, both fertiization and seed setting flourish, no differentiation betwen legitimate and illegitimate unions being recognizable. This sort of physiological property has not been observed in the investigations of other heterostylous plants. It is assumed that this species is differentiated structurally into dimorphism, but not yet physiologically. In nature, however, this plant would have more opportunities to be cross-pollinated, i.e., legitimately combined, than self-pollinated because of the development of two forms of flowers. 3) In terms of heterostylism, the F. esculentum just occupies the intermediate position between P. japonica and P. senticosa structurally, ecologically, and physiologically. Doescription of some of the physiological behavior of the plant will suffice to demonstrate the above facts. While P. japonica has completely lost its self-fertile property, P. senticosa still retains it wolly. In F. esculentum 2-6% of self-fertility is the result in illegitimate combination. There occur occasionally hereditary self fertile individuals among some of the F. or 20 min. irradiation plot, when they reach any stage of the same bacterial population. In addition to this increase of total population in the plots with the more dose of UV light irradiation, it seems that the more dose of UV light irradiation is the more shortened the generation time of Azotobacter. Therefore, it is clear that variation of reproductive rate must be, mere or less, due to the genetic effects induced by UV light irradiation. On the other hand, the lag phase or logarithmic growth phase in nonirradiated culture is shortened prominently, and this must be due to the difference in bacterial number of the original inoculm. The generation time of Azotobacter is shortened by exogeneous treatment of nuclei acid derivatives, and the degree is greater in case of DNA derivatives than RNA dervatives. W.H. Price reported that the rate of ribose nucleic acid to protein in Staphylococcus muscae is proportional to the generation time: that is the faster the cell can form ribose nucleic acid, the more rapid its growth. This explains the shortening of generation time by exogeneous RNA derivatives in this work reasonably. On the other hand, it is well known that the desoxyribose nuclic acid content per cell is constant and independent of the generation time. A.D. Laren and W.N. Takahashi reported that the infectious RNA from TMV is 6 times as sensitive to inactivation by UV as it is in the form of intact virus, and that inactivation of infectious TMV involves onlu a local change on RNA chain. But, the effect of exogeneous DNA in this work suggests that irradiated living cell which cotain DNA bring about some change on DNA moleculs as well as RNA molecules. And if the mutagenic effects of UV take into consideration, it is very reasonable. Therefore, it is clear that the variation of the generation time by UV irradiation is, more or less, due to the genetic effects. Therefore, it seems that the shortness of the average lifewpan of Azotobacter by UV irradiation is resulted not only from the influence of the environmental conditions, but also from the variation of genetic factor of the individual.

Current Statues of Phenomics and its Application for Crop Improvement: Imaging Systems for High-throughput Screening (작물육종 효율 극대화를 위한 피노믹스(phenomics) 연구동향: 화상기술을 이용한 식물 표현형 분석을 중심으로)

  • Food security has been a main global issue due to climate changes and growing world population expected to 9 billion by 2050. While biodiversity is becoming more highlight, breeders are confronting shortage of various genetic materials needed for new variety to tackle food shortage challenge. Though biotechnology is still under debate on potential risk to human and environment, it is considered as one of alternative tools to address food supply issue for its potential to create a number of variations in genetic resource. The new technology, phenomics, is developing to improve efficiency of crop improvement. Phenomics is concerned with the measurement of phenomes which are the physical, morphological, physiological and/or biochemical traits of organisms as they change in response to genetic mutation and environmental influences. It can be served to provide better understanding of phenotypes at whole plant. For last decades, high-throughput screening (HTS) systems have been developed to measure phenomes, rapidly and quantitatively. Imaging technology such as thermal and chlorophyll fluorescence imaging systems is an area of HTS which has been used in agriculture. In this article, we review the current statues of high-throughput screening system in phenomics and its application for crop improvement.

Development of HRM Markers Based on SNPs Identified from Next Generation Resequencing of Susceptible and Resistant Parents to Gummy Stem Blight in Watermelon (수박에서 덩굴마름병 감수성 및 저항성 양친에 대한 차세대 염기서열 재분석으로 탐색된 SNP 기반 HRM 분자표지 개발)

  • Watermelon (Citrullus lanatus) is an economically important vegetable crop all over the world, which has functional compounds such as lycopene and citrulline. Gummy stem blight caused by Didymella bryoniae is one of the most devastative diseases in watermelon. Single nucleotide polymorphisms (SNPs), which are genetic variations occurring between individuals with respect to a single base, were often used to construct genetic linkage maps and develop molecular markers linked to a variety of horticultural traits and resistance to several diseases. In this study, we developed high-resolution melting (HRM) markers based on SNPs generated from NGS resequencing of two parents in watermelon. Plant materials were C. lanatus '920533' (female and susceptible parent), C. amarus 'PI 189225' (male and resistant parent), and their $F_1$ and $F_2$ progenies. A total of 13.6 Gbp ('920533') and 13.1 Gbp ('PI 189225') of genomic sequences were obtained using NGS analysis. A total of 6.09 million SNPs between '920533' and 'PI 189225' were detected, and 354,860 SNPs were identified as potential HRM primer sets. From these, a total of 330 primer sets for HRM analysis were designed. As a result, a total of 61 HRM markers that have polymorphic melting curves were developed. These HRM markers can be used for the construction of SNP-based linkage maps and for the analysis of quantitative trait loci (QTLs) related to gummy stem blight resistance.

Studies on Inheritance and Ecological Variation of the Culm Length and Its Related Characters in Short-Statured Rice Varieties (수도단간품종의 간장 및 관련형질의 유전과 생태적 변이에 관한 연구)

  • These studies were aimed at clarification of genetic and ecological variation in culm length, panicle length and plant height of the $\textrm{F}_2$ plants in some selected crosses made between semi-dwarf rice varieties and tall Japonica ones. One Indica semi-dwarf, Taichung Native 1, one Indica $\times$ Japonica hybrid, IE51 and one Japonica semi-dwarf, Tankanbaekmang were used as short-gene donors while two of medium maturity varieties, Jinheung and Kwanok and one late veriety, Palkweng were used as the corresponding counterpart of respective dwarf varieties in a series of crosses. Five different crosses, Kwanok $\times$ Tankanbaekmang, Palkweng $\times$ Tankanbaekmang, Jinheung $\times$ T(N)1, Kwanok $\times$ T(N)1 and Kwanok $\times$ IE51, were made among the above six varieties. The $\textrm{F}_2$ plants of these crosses together with the concerned parental varieties were grown under several different conditions including three levels of each nitrogen and planting space, three planting seasons and three locations in 1968, to investigate variation in length of culm and panicle, and plant height. On the other hand, the F$_3$ progenies which were derived from the shortest 10 percent of the plants of three $\textrm{F}_2$ populations, Kwanok $\times$ T(N)1, Jinheung $\times$ T(N) 1 and Kwanok $\times$ IE51 grown in the previous year, were compared each other on the basis of selection efficiency in culm length. The experimental results could be summarized as follows; 1. Genetic behavior A. It was revealed that Tankanbaekmang, one of Japonica dwarf has a simple recessive gene responsible for short culm expression, showing a typical segregation ratio of three tall to one short culm plants in $\textrm{F}_2$ generation of the crosses either with Kwanok or Palkweng. B. In the both combinations, segregation pattern of the panicle length was exactly same as that of culm length. It seems that the same gene controls both culm length and panicle length. C. No difference between segregation of culm length and plant height in the above crosses was observed. D. T(N)1, one of Indica semi-dwarf did not show such a simple genetic behavior as detected from the crosses with Tankanbaekmang in segregation of culm length but formed a continuous and normal distribution curve. Therefore, some nonallelic genic actions might be involved in expression of culm length of the counterpart varieties of T(N)1. In particular, a transgressive segregation appeared toward the direction of longer culm length in case of Jinheung $\times$ T(N)1. The genetic behavior of panicle length and plant height generally coincided with that of culm length in all the cases. E. IE51 demonstrated exactly the same genetic behavior as that of T(N)1 when this variety was crossed with Kwanok. It was clearly clarified that the simple recessive gene controlling dwarfism from T(N)1 was well incorporated into this variety. 2. Ecological variation A. In general, there was a decreasing tendency in culm length and plant height of rice plant as seeding delayed while it was not so noticeable in panicle length. The decreasing magnitude varied from variety to variety and from cross to cross. Genetic behavior of the culm length and related characters of these materials was not disturbed by the variation of seeding season, nitrogen level, planting space and experimental location. E. The elongation mode of the upper three internodes was very similar to the segregation mode of culm length, panicle length and plant height in $\textrm{F}_2$ populations of . all the crosses investigated in this study. Accordingly, this result confirmed that the roles of the upper three internodes are very important in manifesting plant stature in rice. C. The effect of nitrogen on culm length and the related other two characters seemed to be meager. However, it was true to show an increasing tendency of those characters as nitrogen level got increased from 4 kg to 12kg per l0a, with different magnitude depending upon variety or cross. D. Also, the effect of planting space on culm length, panicle length and plant height was relatively small in all the cases. Those characters varied again depending upon variety or cross. However, a general increasing tendency was detected in manifestation of those traits under denser planting space condition. E. All the parental varieties produced shorter culm, panicle and plant height when they were grown at the lower latitude locations. It might be attributed to the fact that their reproductive growth accelerated with increased temperature prevailing at the lower latitude locations such as Iri and Mi1yang. On the countrary, $\textrm{F}_2$ population reacted differently to the different locations from the parental varieties. All the $\textrm{F}_2$ plants produced the longest culm, panicle and plant at Milyang. 3. Selection efficiency A. The heritability of culm length in Kwanok $\times$ T(N)1, Kwanok $\times$ IE51 and Jinheung$\times$T(N)1 was 92 percent, 74 percent and 55 percent, respectively. B. The actual genetic advance for culm length obtained from the progeny lines of the selected plants(10 precent) from the $\textrm{F}_2$ generation, was comparable to the expected advance calculated from the original $\textrm{F}_2$ populations. As compared with the $\textrm{F}_2$ population, the $\textrm{F}_3$ plants of Kwanok $\times$ T(N)l shortened on the average by 20.8cm, those of Kwanok $\times$ IE51 did 8.7cm and those of Jinheung$\times$T(N)1 20.0cm, respectively. C. Panicle length of the populations was differently affected from one cross to another by the selection based upon culm length in $\textrm{F}_2$ Kwanok $\times$ T(N)1 did not show any noticeable shortening of its culm length due to the selection pressure. On the other hand, both Kwanok $\times$ IE51 and Jinheung $\times$ T(N)1 showed a considerable shortening of their panicles in case of selection for culm length. Based upon the above results, it could be concluded that the ecological variation in culm length, panicle length and plant height was relatively small and fallen within the range of genetic variation. Considering from the fact that the simple recessive gene governing short height of Tankanbaekmang always accompanied with some undesirable characters such as short panicle and extremely small grain, the short gene of T(N)1 seemed to be more useful as dwarf gene source since it did not carry short gene together with such undesirable traits.

  • This study was carried out to estimate the effects of parity, milking time, milking interval and days in milk(DIM) on variation in milk yield between consecutive milkings(am to pm to am), morning and evening milk yield and its components, somatic cell counts(SCS), milking duration, milk flow rate and peak milk flow in Holstein dairy cattle. Records from one hundred and twenty two heads of Holstein cattle at National Livestock Research Institute, Korea were used for this study from July 1 to August 8, 2005. The experimental herd had average 1.6$\pm$0.9 parities, 199.8$\pm$109.1 DIM and 12.26$\pm$4.06kg milk yields at each milking. Milking yield, percent milk fat and SNF, milking duration and average milk flow were significantly varied by parity, milking time and DIM. Percent milk protein and lactose were varied by parity and DIM, however SCS and average milk flow were affected by parity and milking time. Milking interval significantly affected the consecutive, morning and evening milk yield and average milk flow. However, MUN was not affected by parity, milking time, DIM and milking interval. Milk yield was decreased with increasing parity. Milk yield in the morning was higher than that of in the evening. Milk yield between consecutive milking was not affected by parity, however, affected by milking time. Percent milk Fat, SNF and SCS were higher at in evening milk than those of in morning milk. Milk protein, lactose, SNF, SCS, milking duration and peak milk flow rate were influenced by parity. This study suggested that milk yield variation between consecutive milking, milking flow rate, and milking duration could be important traits for enhancing Holstein cattle productivity however, and more study is needed to estimate genetic parameters for such traits.
