Several studies arduously reported that gayal (Bos frontalis) is an independent bovine species. The population size is shrinking across its distribution. In Bangladesh, it is the only wild relative of domestic cattle and also a less cared animal. Their body size is much bigger than Bangladeshi native cattle and has prominent beef type characters along with the ability to adjust in any adverse environmental conditions. Human interactions and manipulation of biodiversity is affecting the habitats of gayals in recent decades. Besides, the only artificial reproduction center for gayals, Bangladesh Livestock Research Institute (BLRI), has few animals and could not carry out its long term conservation scheme due to a lack of an objective based scientific mission as well as financial support. This indicates that the current population is much more susceptible to stochastic events which might be natural catastrophes, environmental changes or mutations. Further reduction of the population size will sharply reduce genetic diversity. In our recent investigation with 80K indicine single nucleotide polymorphism chip, the $F_{IS}$ (within-population inbreeding) value was reported as $0.061{\pm}0.229$ and the observed ($0.153{\pm}0.139$) and expected ($0.148{\pm}0.143$) heterozygosities indicated a highly inbred and less diverse gayal population in Bangladesh. Prompt action is needed to tape the genetic information of this semi-domesticated bovine species with considerable sample size and try to investigate its potentials together with native zebu cattle for understanding the large phenotypic variations, improvement and conservation of this valuable creature.
Indigenous (native) breeds of livestock have higher disease resistance and adaptation to the environment due to high genetic diversity. Even though their extinction rate is accelerated due to the increase of commercial breeds, natural disaster, and civil war, there is a lack of well-established databases for the native breeds. Thus, we constructed the native pig and chicken breed database (NPCDB) which integrates available information on the breeds from around the world. It is a nonprofit public database aimed to provide information on the genetic resources of indigenous pig and chicken breeds for their conservation. The NPCDB (http://npcdb.snu.ac.kr/) provides the phenotypic information and population size of each breed as well as its specific habitat. In addition, it provides information on the distribution of genetic resources across the country. The database will contribute to understanding of the breed's characteristics such as disease resistance and adaptation to environmental changes as well as the conservation of indigenous genetic resources.
Chrysosplenium aureobracteatum Y. I. Kim & Y. D. Kim (Saxifragaceae) is a recently described endemic species growing in the central part of the Korean peninsula. It requires constant monitoring for conservation due to its limited distributions. There is also a need for molecular markers for proper assessments of the genetic differentiation of C. aureobracteatum from species morphologically similar to it. In this study, we developed microsatellite markers that can be used to evaluate the genetic diversity of this species, representing fundamental data with which to conserve the natural populations of the species. A total of 17 expressed sequence tag-simple sequence repeat (EST-SSR) markers were developed by the Illumina pair-end sequencing of the transcriptomes of C. aureobracteatum. These markers were successfully applied to populations of C. aureobracteatum and to its most closely related species, C. barbatum, revealing high polymorphism in both species. The EST-SSR markers developed in this study were proven to be useful not only to monitor the population genetic structure of C. aureobracteatum for conservation purposes but also to study the genetic delimitation of the species from species closely related to it.

In this review, we compared the success rates of DNA amplification and introduced the efficient non-invasive sampling of fecal samples collected from captive and wild Korean long-tailed gorals (Naemorhedus caudatus) by referring to previous non-invasive studies, including three important references (Kim et al., 2008; Kim, 2021; Kim, 2022). A large difference in PCR success rates in the captive and wild populations was observed for mitochondrial (100 and 70.0%), sex-linked (44.4 and 20.8%), and microsatellite markers (73.9 and 34.8%, respectively). Out of the three types of genetic markers, the mitochondrial maker showed the highest success rate, followed by microsatellite and sex-linked markers. In addition, we estimated two factors that affected the PCR success, including the length of the amplified fragments (long, medium, and short) and the type of primer (universal and specific) in fecal samples from a captive population. The length of the PCR fragment was inversely proportional to the PCR success (5.3, 44.4, and 55.6% for long, medium, and short fragments, respectively), and the specific primer set (100%) was more efficient than the universal primer set (60.0%). This review is fundamental but would be greatly helpful for new non-invasive conservation genetic studies, particularly those that use fecal samples from captive and wild populations of rare endangered species. We recommend beginning conservation genetic experiments using mitochondrial markers and then nuclear markers, such as microsatellite and sex-linked markers, to save time, costs, and labor.

For the spring ephemeral and myrmecochorous perennia, Corydalis filistipes Nakai (Fumariaceae), rare and narrow endemic to Ullung Island in Korea, genetic diversity and structure of 4 subpopulations of the species were investigated with allozyme markers. Levels of genetic diversity (A=1.73, $P_{95%}$=61..2%, Ho=0.201, He=0.167) were relatively lower than those of other endemic species with widespread distribution range, but considerably higher than other endemic species with similar life history traits isolated in island. The moderate level of genetic diversity within subpopulations in C. filistipes is characteristic of the species with predominantly outcrossing, myrmecochorous seed dispersal by dual function of the elaisome and mode of sexual and asexual reproduction by the cleistogamy. The analysis of fixation indices showed an overall excess of heterozygotes (mean $F_{IS}=-0.1889,\;F_{IT}=-0.1226$) relative to H-W expectations. About 5.6% of the total genetic variation was found among subpopulations ($F_{ST}$=0.0557). The strategies of reasonable conservation and management, and the maintenance mechanism of genetic diversity of Corydalis filistipes Nakai, endemic plant species in Korea were discussed.

We investigated the genetic diversity of an endemic rare species, Forsythia ovata Nakai by examining 93 ISSR amplicons in 84 individuals distributed among five populations. The overall percentage of polymorphic ISSR amplicons was 54.8% and mean number of amplicons per ISSR primer was 6.6. The amount of genetic diversity was relatively lower than other shrub species. The Mt. Seokbyeong and Mt. Seorak B populations had the highest level of genetic diversity. Although the Seokgae-jae population had the lowest level of genetic diversity, the population was genetically the most distinctive from the other populations. About 30.6% of the total variation was allocated between five populations, which was slightly higher than other shrub species. Such a pattern of genetic variation may have resulted from the limited distribution and small population sizes of F. ovata. The UPGMA dendrogram based on Nei's genetic distance showed some decisive geographic patterns. These results suggest that, in addition to the preservation of the natural stands, the conservation of larger number of populations with small number of individuals per population is more effective for the dynamic ex situ conservation and for maintaining the genetic diversity of F. ovata than smaller number of populations with large number of individuals.
We investigated levels and distribution of genetic variation in Korean populations of Calystegia soldanella and C. japonica, clonally reproducing herbaceous perennials. Calystegia soldanella is one ofecologically important beach plants growing only on sand and beach dunes in Europe, East Asia, the Pacific Islands, and the west coast of North America. In contrast, C. japonica usually grows on small mounds of paddy fields, roadsides, and waste places with patchy distribution. Starch gel electrophoresis was conducted on leaves collected from 13 populations of C. soldanella and eight populations of C. japonica. The levels of genetic variation of the two species are very comparable; means of expected heterozygosity (Hep) were 0.100 and 0.099 for C. soldanella and C. japonica, respectively. These values were also very similar to those for species with similar life-history and ecological traits. However, the proportion of total genetic diversity partitioned among populations (GST) of C. soldanella (0.146) was considerably lower than that of C. japonica (0.383). In addition, means of Nei's genetic identity (Ⅰ) for C. soldanella and C. japonica were 0.985 and 0.900, respectively, which supports a restricted gene flow resulting from obligate clonal reproduction of C. japonica. Significant differences in allele frequency were detected among populations at eight and nine of nine polymorphic loci for C. soldanella and C. japonica (P<0.01), respecitvely. Considering the ecological importance of C. soldanella, the isolated beach populations coupled with present destruction of natural habitats of the species may result in erosion of genetic diversity in the near future. In this respect, conservation efforts should be focused on those populations that currently maintain the most genetic diversity such as those populations in the eastern and southeastern Korean Peninsula and Hamduck Beach, Cheju Island.
The genetic diversity and structure of eight populations of Coreanomecon hylomeconoides Nakai, an endemic Korean plant, were investigated using 50 ISSR loci from eight primers. The average percentage of polymorphic loci was 47.3%. The Shannon's index (SI=0.218) and gene diversity (h=0.142) were relatively lower than those of other long-lived perennials. The Sancheong (SI=0.233, h=0153), Gwangyang (SI=0.263, h=0.171), and Suncheon (SI=0.241, h=0.159) populations showed greater genetic diversity than the Namhae and Gwangju populations, which are on the edge of the distribution. Analysis of molecular variance (AMOVA) showed that 18% of the total variation could be attributed to differences among populations, and 82% to differences within populations, indicating moderate gene flow among adjacent populations. These results were supported by value of Nm (2.184). The UPGMA conducted using the genetic distance and Bayesian cluster analysis showed a remarkable geographic trend structured into east and west regions. Overall, the results indicate that the Sancheong and Gwangyang populations, which had a large population size and higher degree of genetic diversity, should be the focus of in situ conservation.

Taxonomic and phylogenetic studies of firefly in Japan have been reviewed. Fourty-six lampyrid species and one rhagophthalmid are distributed in the Japanese Islands including the Ryukyus. Recently, molecular phylogenetic approaches have been employed in the systematic study of firefly using mitochondrial and luciferase genes. Based on the molecular phylogenetic trees, evolutionary process of flashing patterns related strictly to mating behavior was estimated. Furthermore, genetic diversity studies revealed geographic differentiation patterns within species, and conservation measures of firefly were proposed to protect genetic resources endemic to the localities.
Tissue culture techniques arguably are an important approach for ex situ conservation of rare and endangered plant species. However, there is utmost importance on maintaining the genetic integrity of the introduced plants especially in tree species. To examine the genetic integrity of the micropropagated plants, we randomly screened few hardened plants of Syzygium travancoricum, a critically endangered tree taxon, using Randomly Amplified Polymorphic DNA (RAPD) markers. Twenty-three random. primers were tried and twenty-five polymorphic loci were identified. The dendrogram based on the Unweighted Pair-Group Method Arithmetic Average and Nei's similarity index depicted about 97% homology between the mother plants and micropropagated plants. Further, an attempt was made to reintroduce the micropropagated plants in the wild. Over three hundred small trees could be successfully established.
