New Record of a Limacodid Moth, Hampsonella takemurai (Lepidoptera: Limacodidae) from Korea

  • We report a limacodid moth, Hampsonella takemurai (Inoue) for the first time in Korea based on four males from the southwestern islands of Shinan-gun, Jeollanam-do. The genus Hampsonella Dyar was designated with the type species Parasa dentata Hampson and comprises six species, H. acatharta (Hampson, 1897), H. albidula Wu and Fang, 2009, H arizana (Wileman, 1916), H. dentata (Hampson, 1893), H. membra Solovyev and Witt, 2009, and H. takemurai (Inoue, 1986). Hampsonella takemurai can be diagnosed by the bipectinate male antennae, the blackish forewing with a large, dark blackish postmedial marking, and the blackish hindwing. The male genitalia can be diagnosed by the short triangular uncus, the long digitate gnathos, and the simple membranous valva. We provided the diagnosis and photographs of adult and male genitalia and COI gene sequence.

Phylogenetic analysis of Locusta migratoria (Orthoptera: Acridae) in Haenam-gun, Jeollanam-do, Korea using Two Mitochondrial Genes (마이토콘드리아 유전자 2개를 이용한 대한민국 전라남도 해남군 발생 풀무치 Locusta migratoria (메뚜기목: 메뚜기과)의 계통분석)

  • An outbreak of the migratory locust, Locusta migratoria, in the environment-friendly reclaimed plantations of forage crops in Sanyimyeon, Haenam-gun, Jellanam-do, Korea in August 2014 caused severe damages to various crops. Owing to its first occurrence in the Korean history, the causes underlying the outbreak and phase-transition of the migratory locust were not known. It is critical to establish the genetic relationship of the migratory locust in Sanyimyeon, Haenam-gun with the other previously reported strains in the world in order to understand the mechanisms responsible for its outbreak. The gene sequences of the 16S ribosomal RNA (rRNA) and displacement-loop (D-loop) of the mitochondria of various regional species of the migratory locust were used to perform the phylogenetic analysis. Our results suggested that the migratory locusts in Sanyimyeon, Haenam-gun are closely related with the Eurasian strains of the northern lineage. In future, these two mitochondrial genes can be used for elucidating the genetic population structures in migratory locusts in various regions. In addition, the sequence information of these genes can be used to enhance our understanding of the genetic basis of the outbreak of migratory locusts.

Soil Bacterial Community in Red Pine Forest of Mt. Janggunbong, Bonghwa-Gun, Gyeongbuk, Korea, Using Next Generation Sequencing (차세대염기서열방법을 이용한 경북 봉화군 장군봉 소나무림의 토양 박테리아 군집 구성)

  • The soil microbiome plays important roles in material cycling and plant growth in forest ecosystem. Although a lot of researches on forest soil fungi in Korea have been performed, the studies on forest soil bacterial communities have been limited. In this study, we conducted next generation sequencing (NGS) targeting 16S rRNA gene to investigate the soil bacterial communities from natural red pine (Pinus densiflora) forest in Mt. Janggunbong, Bonghwa-gun, Gyeongbuk, Korea. Our results showed that the entire bacterial communities in the study sites include the phyla Proteobacteria, Acidobacteria, Actinobacteria, Planctomycetes, which have been typically observed in forest soils. The composition ratio of Proteobacteria was the highest in the soil bacteria community. The results reflect that Proteobacteria is copiotroph, which generally favors relatively nutrient-rich conditions with abundant organic matter. Some rhizobia species such as Burkholderia, Bradyrhizobium, Rhizobium, which are known to contribute to soil nitrogen-fixation, exist in the study sites. As a result of correlation analysis between soil physicochemical characteristics and bacteria communities, the soil pH was significantly correlated with the soil bacteria compositions.

Genotype analysis of Cryptosporidium spp. prevalent in a rural village in Hwasun-gun, Republic of Korea

  • Two species of Cryptosporidium are known to infect man; C. hominis which shows anthroponotic transmission between humans, and C. parvum which shows zoonotic transmission between animals or between animals and man. In this study, we focused on identifying genotypes of Cryptosporidium prevalent among inhabitants and domestic animals (cattle and goats), to elucidate transmittal routes in a known endemic area in Hwasun-gun, Jeollanam-do, Republic of Korea. The existence of Cryptosporidium oocysts was confirmed using a modified ZiehlNeelsen stain. Human infections were found in 7 $(25.9\%)$ of 27 people examined. Cattle cryptosporidiosis cases constituted 7 $(41.2\%)$ of 17 examined, and goat cases 3 $(42.9\%)$ of 7 examined. Species characterizations were performed on the small subunit of the rRNA gene using both PCR-RFLP and sequence analysis. Most of the human isolates were mixtures of C. hominis and C. parvum genotypes and similar PCR-RFLP patterns were observed in cattle and goat isolates. However, sequence analyses identified only C. hominis in all isolates examined. The natural infection of cattle and goats with C. hominis is a new and unique finding in the present study. It is suggested that human cryptosporidiosis in the studied area is caused by mixtures of C. hominis and C. parvum oocysts originating from both inhabitants and domestic animals.

  • As an effort to explore fungal diversity, fungal survey was undertaken in 2017 in the vicinity of demilitarized zone (DMZ) located in Yeoncheon-gun, Gyeonggi-do, Korea. For the survey, wild plants and soils were sampled and subjected to fungal isolation. A total of 18 genera and 23 species including five unrecorded fungal species, Mortierella sclerotiella, M. sossauensis, M. verticillate, Eupenicillium saturniforme, and Trichoderma hispanicum, were obtained from the survey. This study described their morphological characteristics including colony features formed on media, light microscopic images and molecular characteristics of nucleotide sequences of the internal transcribed spacer (ITS) region, 18S and 28S rDNA, ${\beta}-tubulin$ gene, calmodulin, and translation elongation factor $1{\alpha}$ ($tef1{\alpha}$) nDNA genes.

Peltaster fructicola: Undescribed Sooty Blotch and Flyspeck Species on Apple Fruit in Korea

  • While investigation of the fungal diseases on apples collected from Cheongsong-gun and Bonghwa-gun in Gyeongbuk province, Korea, between August and September 2023 isolated five fungal strains from fruits with sooty blotch and flyspeck (SBFS) disease. The strains were designated as KNUF-23-CS02, KNUF-23-CS-06, KNUF-23-CS12, KNUF-23-BH01, and KNUF-23-BH03. When grown on potato dextrose agar and 2% water agar, the cultural characteristics of the strains were similar to those previously reported characteristics of Peltaster fructicola Pf001. The strains produced monoblastic, hyaline conidiogenous cells; the conidia were hyaline, unicellular, cylindrical to ovoidal, and 3.5-7×1.7-3.9 and 4.0-6.6×1.8-3.2 μm in size on synthetic nutrient-poor agar or water agars, respectively. Secondary conidia production by microcyclic conidiation and budding was observed. The KNUF-23-BH03 strain was shown to cause SBFS symptoms similar to those observed on the apples in the pathogenicity test. Molecular phylogenetic analyses were conducted based on the isolated species sequences of the internal transcribed spacer region, nuclear large ribosomal DNA subunit, and mitochondrial small ribosomal RNA subunit gene. The five strains were clustered with Peltaster fructicola Pf001. Based on the cultural and morphological characteristics and phylogenetic analysis, the five strains were identified as Peltaster fructicola, which has not been previously reported in Korea.

In vitro Study of Anti-inflammatory Effects of Salvia Miltiorrhiza Extracts Using Luciferase Reporter Gene Assay (Luciferase Reporter Gene Assay를 이용하는 단삼추물문의 소염 및 진통작용에 대한 in vitro 연구)

  • In order to identify the anti-inflammatory and analgesic properties of natural herbal extracts, widely used in the Korean traditional medicine, an in vitro screening system was designed using pGL3, a luciferase reporter vector, and the tumor necrosis factor (TNF)-α and cyclooxygenase (COX)-II as target genes. The promoter regions of each gene was generated by PCR using the human chromosome as template DNA, and inserted into pGL3 vector with Kpnl and Hindlll. The final construct was transfected into human myleomonocytic leukemia cells (U937) that could be differentiated and activated by phorbol 12-myristate 13-acetate (PMA) or lipopolysaccharide (LPS). Using this system, we tested the anti-inflammatory and analgesic effects of several herbal extracts being regarded to have the medicinal effects of diminishing the body heat and complementing Qi. The well-known chemicals of PD98059 and berberine chloride were used as controls of the transcriptional inhibitors of TNF-α and COX-II, respectively. Among them, Salvia miltiorrhiza (Dan-Sam) was found to exhibit the significant medicinal properties of anti-inflammatory and analgesic effects.

Gene Expression Analyses in Hypothalami of Immobilization-stressed and BoshimgeonbiTang-treated Mice Using cDNA Microarray (구속 스트레스 (immobilization stress)를 가한 rat의 hypothalamus에서의 유전자 발현 및 포심건비탕의 항스트레스 효과에 관한 cDNA microarray 분석)

  • The genetic effects of restraint stress challenge on HPA axis and the therapeutic effect of Boshimgeonbi-Tang on the stress were studied with cDNA microarray analyses on hypothalamus using an immobilization-stress mouse as stress model. Male CD-1 mice were restrained in a tightly fitted and ventilated vinyl holder for 2hours once a day, and this challenge was repeated for seven consecutive days. The body weights of the immobilization-stress mice were diminished about 25 percent degree as compared to normal ones. Seven days later, total RNA was extracted from the organs of the mouse, body-labeled with CyDye/sup TM/ fluorescence dyes (Amersham Bioscience Co., NJ), and then hybridized to cDNA microarray chip. Scanning and analyzing the array slides were carried out using GenePix 4000 series scanner and GenePix Pro/sup TM/ analyzing program, respectively. The expression profiles of 109 genes out of 6000 genes on the chip were significantly modulated in hypothalamus by the immobilization stress. Energy metabolism-, lipid metabolism-, apoptosis- and signal transduction-related genes were transcriptionally activated whereas DNA repair-, protein biosynthesis-, and structure integrity-related genes were down-regulated in hypothalamus. The 58 genes were up-regulated by the mRNA expression folds of 1.5 to 7.9. and the 51 genes were down-regulated by 1.5 - 3.5 fold. The 20 genes among them were selected to confirm the expression profiles by RT-PCR. The mRNA expression levels of Tnfrsf1a (apoptosis), Calm2 (cell cycle), Bag3 (apoptosis), Hspe1 (protein folding), Aatk (apoptosis), Dffa (apoptosis), Itgb1 (cell adhesion), Vcam1 (cell adhesion), Fkbp5 (protein folding), BDNF (neuron survival) were restored to the normal one by the treatment of Boshimgeonbi-Tang.

Analysis of p53-Dependency of Differentially Expressed Genes by Capsaicin in Human Colorectal Cancer Cell (인간 대장암 세포주에서 capsaicin 처리에 의한 차별적인 유전자 발현의 p53 의존성 분석)

  • In the present study, we investigated anti-proliferative activities of capsaicin and gene expression changes in response to capsaicin treatment in human colorectal HCT116 cells. The results showed that capsaicin decreased cell viabilities in a dose dependent manner and induced global gene expression changes. We found that 103 genes were up-regulated more than twofold, whereas 153 genes were down-regulated more than twofold by $100\;{\mu}M$ capsaicin treatment. Among the up-regulated genes, we selected 4 genes (NAG-1, DDIT3, GADD45A and PCK2) and performed RT-PCR to confirm the microarray data. We found that $100\;{\mu}M$ of capsaicin increased tumor suppressor p53 gene expression. In addition, the results showed that NAG-1, DDIT3 and GADD45A expressions were not dependent on p53 presence, whereas PCK2 expression. The results of this study may help to increase our understandings of the molecular mechanism of anti-proliferative activity mediated by capsaicin in human colorectal cancer cells.

Effects of osmoticum treatments and shooting chances on the improvement of particle gun-mediated transformation in Phalaenopsis (유전자총을 이용한 팔레놉시스 형질전환 효율향상에 삼투압 조절제 및 발사횟수차이가 미치는 영향)

  • This study was carried out to develop an efficient transformation protocol via particle bombardment with PLBs (protocorm-like bodies) in Phalaenopsis. To achieve this aim, osmoticum treatment and an increasing shooting chances in particle bombardment process were applied for this study. In addition, pCAMBIA3301: ORE7 vector which contains a herbicide-resistance bar gene as a selectable marker and ORE7 gene as a gene of interests were employed. With regard to the increasing chances of shooting in particle bombardment, double shooting was the best results with 1.5 ~ 2.5 times higher than those of a single or triple shooting treatment in the productioon of PPT (D-L-phosphinothricin)-resistant PLBs. However, regeneration rate of shoots in double shooting was not high as a single shooting. Further, double shooting showed 35 ~ 40% higher than that of a single shooting in the frequency of browning. Regarding effects of different osmotic treatments, combination of 0.2 M sorbitol with 0.2 M mannitol showed the best results in transformation efficiency, regeneration of transformants and reduction of browning. Putative transgenic Phalaenopsis plants were analyzed by PCR analysis and confirmed the presence of bar and ORE 7 gene. Also, real-time PCR was conducted by using 21 transgenic plants and showed only 4 plants had one copy of transgene; whereas, the other 17 plants had more than 2 copies of transgene. Transgenic phalaenopsis plants produced in this study were transferred to pots and flowered normally without morphological variations in flower and leaf.
