Purification and antibacterial effect of lysozyme from flounder, Paralichthys olivaceus (양식넙치로 부터의 Lysozyme 정제와 어류병원성 세균에 대한 정균작용)

  • Lysozymes were isolated and purified from various organs of cultured flounder by using chitin-coated cellulose column chromatography. The molecular weights of them were compared with each other in 15% SDS-PAGE gels. The result showed that all lysozymes isolated from various organs of flounder had the same molecular weight of about 14000. To clarify the role of lysoryme as a body defence, the antibacterial activities of flounder lysozyme on seven bacterial pathogens, five Gram-negative and two Gram-positive species, were investigated. The lysozyme had substancial antibacterial activity on four strains, two Gram-negative and two Gram-positive species. These suggest that flounder lysozyme plays a role in body defence against both Gram-negative and Gram-positive bacterial pathogens.

Development of Thixotropic Inorganic-Type Grout and Its Engineering Characteristics (무기계 가소성 그라우트의 개발 및 공학적 특성)

  • A thixotropic grout has been developed for the use of filling a tail void in the shield TBM and as well as various ground voids. The grout developed in this study is a mixture of inorganic substance, cement and some functional additives. Its engineering characteristics was investigated by measuring a viscosity and unconfined compressive strengths. The optimum mix proportion for an effective thixotropic grout was proposed through several repeated laboratory tests. The various physical properties such as thixotropy, unconfined compressive strengths, and durability of the thixotropic grout and the gels produced from the grout were compared with those of the well-known waterglass-type grout such as L.W.. The thixotropic grout developed in the study exhibited an excellent performance for back-filling of tail voids in the shield TBM based on experimental results compared to the existing waterglass grout.

Quality Characteristics of Skate Pipyun prepared with Skate Skin (향신채소와 허브를 첨가한 홍어 피편의 품질특성)

  • The purpose of this study was to provide the basic information of making Skate pipyun made of skate skin. Test for quality characteristics of skate pipyuns seasoned with garlic, ginger, rosemary and basil were carried out. Hardness of GAR(4% of garlic and rosemary) was the highest, while that of GIB(4% of ginger and basil) was the lowest. As for the adhesiveness, springiness and cohesiveness, each specimen did not manifest significant difference. Water contents (82.43~85.56%) were no significant differences. The pipyuns added with spices and herbs were appeared weakly alkali with a range of pH 8.46~8.58. In the sensory evaluation, GIB was the most preferred in overall preference with significantly higher evaluation of flavor, savory taste, and lower fishy odor. The stability of gels decreased during storage at $10^{\circ}C$ for 8 hours, and at $20^{\circ}C$ for 60 min, and the greater was in GIB. Therefore, heating a mixture of 40% skate skin with 60% water for 10 min is recommended as optimum conditions for producing Skate pipyun.

  • In case of glass ceramics and powder preparation from the metalakoxide solutions, metalakoxide solutions with a various species of alkoxy groups have unique characteristics. Therefore, in this study, the mixing ability of homogeneous sol, gel morphology and physical properties of gels were investigated by the changes in terms of the different four alkoxy groups, CH3-, C2H5-, i-C3H7-n-C4H9-, along with the catalyst for the purpose of the observation about the homogenous transition range from sol to gel. As a result, when the fixed condition was mol ratio of H2O/Si(OR)4=2.0 and variables were batch composition and addition amount of catalyst, the characteristics of Tetra-normal-Butoxysilane and Tetra-iso-propoxysilane systems had very narrow sol-gel conversion region than Tetramethoxysilane and Tetraethoxysilane system. And silicon-alkoxide, systems having narrow sol-gel conversion region were enlarged by addition of catalyst. In viewpoint of the weight loss of gel produced by hydrolysis of silicon alkoxide systems with different four alkoxy groups, the amounts of weight loss of gel containing large molecular alkoxy groups were much more than those of small molecular alkoxy group.

Effective Diffusivity of Substrate of an Immobilized Microorganism in Ca- Alginate Gels (고정화 미생물의 기질 유효 확산)

  • The fiffusion characteristics of substrate of varing biomass concentrations into and from Ca- alginate gel beads in well-stirred solutions were investigated. Ca-alginate gel beads were immobilized by Zymomonas mobilis or free from cells. The values of the diffusion coefficient of substrate were calculated by means of the method of Least squares and Random pore model. Reaction rates are expressed by the Michaelis-Menten type equation, and the results are compared with experimental data. Intraparticle effective diffusivity of substrate resistance on reaction by using immobilized Z.mobilis entrapped by Ca-alginated gel seemed to be restricted by cell density. The experimental data also indicated relationship between the effective diffusivity and the cell concentration used in the gel preparation.

Study on Anti-Cancer Effects of Backhapgogumtanggami-bang (백합고금탕가미방의 항종양 효과에 관한 연구)

  • Objectives : The effects of aqueous extract of Backhapgogumtanggami-bang (BGTG, a newly devised herb medicine) on the induction of apoptotic cell death were investigated in human lymphoid origin leukemia cell lines, HL-60. Methods : Cells were treated with various concentrations and $400{\;}\mu\textrm{g}/ml$ BGTG for 12 hr. Genomic DNA was isolated and separated on 1.8% agarose gels. Lysates from the cells were used to measure the activity of caspase-2, -3, -8, and -9 protease by using fluorogenic peptide. Cells were preincubated with SB-203580 for 30 min. Nuclear protein from the cells was incubated with oliginucleotide probe of AP-l and NF-kB. Nuclear extracts from the cells were isolated and reacted with antibodies. Results : The viability of HL-60 cells were markedly decreased by BGTG extract in a dose- and time-dependent manner. BGTG extract induced the apoptotic death of HL-60 cells which was characterized by the DNA fragmentation. The activations of Caspase-2, 3, and 9 were induced by BGTG. However, selective inhibition of the p38 mitogen-activated protein kinase pathways by SB-203580 did not affect the extent of BGTG extract-induced cell death. Furthermore, we observed the transient activations of transcriptional factors such as AP-l and NF-kB. Conclusions : These results suggest that BGTG extract induced apoptotic death of HL-60 cells and caspase activations as well as the modulation of transcriptional factors such as AP-1 and NF-kB.

  • Fluorescence recovery after photobleaching (FRAP) has been widely used for the measurement of molecular diffusion in living cells and tissues. We developed an image-based FRAP (iFRAP) technique using a modified real-time microscope and a 488 nm Ar-ion laser. A fractional intensity curve was obtained from the time-lapse images of fluorescence recovery in the bleached spot to determine the diffusion coefficient of fluorescently labeled macromolecules in porous medium. We validated iFRAP through experiments with agar gels (0.5% and 1.5% w/v) containing FITC-Dextrans (10, 70 and 500 kDa MW). Further validation was performed by a Monte Carlo approach, where we simulated the three-dimensional random walk of macromolecules in agar gel model. Diffusion coefficients were deduced from the mean square displacement curves and showed good agreements with those measured by iFRAP.

  • Background and Objective: Protein expression in colon and rectal cancer (CRC) and paired normal tissues was examined by two-dimensional gel electrophoresis (2-DE) to identify differentially expressed proteins. Materials and Methods: Five fresh colorectal cancer and paired adjacent normal tissues were obtained and differentially expressed protein spots were determined using PDQuest software, with identification on the basis of MALDI-TOF mass spectra. Results: Compared with normal colorectal mucosa, protein abnormal expression of 65 spots varying more than 1.5 times were found in 2-DE gels from colorectal cancer samples (P<0.05); forty-two proteins were up-regulated and 23 were down-regulated; twelve protein spots were identified using mass spectrometry, of which 8 were up-regulated, includimng HSPB1and Annexin A4, while 4 were down-regulated, the results being consistent with Western blot findings. Conclusions: Two-dimensional electrophoresis reference maps for CRC tissues and adjacent normal mucosa (NMC) were established and 12 differentially expressed proteins were identified. Up-regulated HSPB1 and Annexin A4 may play many important roles in the pathogenesis of colorectal cancer.

Preparation and Characterization of Proton Conductive Phosphosilicate Membranes Based on Inorganic-Organic Hybrid Materials

  • A series of proton conductive inorganic-organic hybrid membranes doped with phosphoric acid ($H_3PO_4$) and/or triethylphosphate (PO(OEt)$_3$) have been prepared by sol-gel process with 3-glycidoxypropyltrimethoxysilane (GPTMS), 3-aminopropyltriethoxysilane (APTES) and tetraethoxysilane (TEOS) as precursors. High proton conductivity of 3.0 ${\times}$ $10^{-3}$ S/cm with composition of 50TEOS-30GPTMS-20APTES-50$H_3PO_4$ was obtained at 120 ${^{\circ}C}$ under 50% relative humidity. Thermal stability of membrane was significantly enhanced by the presence of SiO$_2$ framework up to 250 ${^{\circ}C}$. XRD revealed that the gels are amorphous. IR spectra showed a good complexation of $H_3PO_4$ in the matrix. The conductivity under 75% relative humidity was significantly improved by addition of APTES due to the increase in concentration of defected site in hybrid matrix. The effect of PO(OEt)$_3$, humidifying time, and heat-treatment were also investigated. PO(OEt)$_3$ had no improvement on conductivity and conductivity increased with humidifying time, however, decreased with heating temperature.
