Study on Reaction Behavior of Rigid Polyurethane Foam with Various Types and Contents of Gelling Catalysts (젤화 촉매의 종류 및 함량에 따른 경질 폴리우레탄 폼의 반응거동에 관한 연구)

  • The reaction behavior of rigid polyurethane foams were studied on the effects of gelling catalysts of amine type, such as; dimethylcyclohexyl amine (DMCHA) and of potassium type, such as; potassium octoate (PO). Rigid polyurethane foams were provided with polymeric 4,4'-diphenylmethane diisocyanate, polyester polyol, silicone surfactant, blowing agent and a few gelling catalysts. As the contents of catalyst, DMCHA increased from 0 to 2.0 g, the reaction time decreased from ca. 330 to ca. 35 sec and due to the exothermic reaction, the maximum temperature increased from ca. 217 to ca. $234^{\circ}C$, respectively. As the contents of PO increased from 0 to 2.5 g, the reaction time decreased from ca. 79 to ca. 38 sec and the maximum temperature increased from ca. 182 to ca. $271^{\circ}C$, respectively. The kinetic parameters were calculated and the conversions were based on the temperature rising method of adiabatic process. As the content of DMCHA increased, the rate constant $k_0$ increased. But in the case of PO catalyst, $k_0$ did hardly depend upon its amount, and showed us similar reaction rate constants.

Isolation of the Exopolysaccharide Producing Microorganism and their Cultural Characterisitics (다당류를 생산하는 미생물의 분리와 배양특성)

  • A screening was performed to isolate exoploysaccharide-producing microorganisms, which synthesized specific exopolysaccharide for the substitutive of commercial polysaccharides, from natural sources. Soil bacterium, one of 378 mucoid isolates, was finally selected as potential producer of polysaccharides which made the culture broth very viscous and thus examined in detail for optimal medium composition. Isolated strain was identified as Xanthomonas sp. EPS- 1 from the results of morphological and biochemical characteristics. The composition of optimal medium for exopolysaccharide production was as follows: 50 g sucrose, 1.5 g peptone, 2 g KH$_{2}$PO$_{4}$, 2 g MgSO$_{4}$, -7H$_{2}$O, 3 g NaCl, 0.05 g CaCO$_{3}$, 0.07 g FeSO$_{4}$-7H$_{2}$O and 0.05 g MnSO$_{4}$-7H$_{2}$O in 1 liter of distilled water. From the experiments of temperature and pH dependence, the optimal conditions for exopolysaccharide biosynthesis seemed to be 30$\circ$C and 8.0, respectively. About 14.9 gram of maximum exopolysaccharide per liter was obtained at the initial pH 8.0, 30$\circ$C and 250 rpm in a flask culture. The exopolysaccharide EPS-1 had such potential as an emulsifying agent and a gelling agent in comparision with commercial exopolysaccharide.

  • Oil gelling agent was synthesized with PPG, PTMG and TDI at 7$0^{\circ}C$ for 4hours. PPGs and PTMGs having various average molecular weights (M$_{n}$: 1000, 2000, 3000) were employed to investigate the ratio of oil gelation and water gelation. As M$_{n}$ of PPG, in result, was decreased from 3000 to 1000, the ratio of oil gelation was increased from 130% to 290% for PPG and from 250% to 310% for PTMG. Ratio of oil gelation was increased approximately two times when EG was added. As the amount of hydrophilic compound in the prepolymer was increased, ratio of oil gelation was increased from 290% to 1120% for PPG and increased from 310% to 1310% for PTMG, due to the increased dispersion of prepolymer in the water/oil mixture.ure.

Preparation and Characterization of Gel Food for Elderly (고령자용 겔상식품의 제조 및 특성연구)

  • Model gel food samples of yanggaeng (Y) were prepared for the elderly with various gelling agents, including agar (AG), low acyl gellan (GL), ${\kappa}$-carrageenan (CA), locust bean gum (LB), glucomannan (GM), and xanthan gum (XA) in different combinations as follows (in 1:1 ratio): LB+CA, GM+XA, and GM+CA. The quality characteristics of the different combinations were compared. The results revealed that water loss was highest for Y-GL, whereas there was no significant difference among the other samples. Y-GL showed the highest values for lightness in color, whereas Y-AG showed the lowest. Regarding textural properties, Y-LB+CA had the highest hardness value, whereas Y-GL had the lowest; the hardness of Y was related to the cross-section of the added gel. Finally, Y-GM+XA exhibited the highest score in overall acceptability in the sensory test by elderly, indicating that the preferable texture by elderly is slightly chewy, but not adhesive.

Culture Characteristics of Streptomyces spp. on Improved Polyacrylamide Gel and Agar Media

  • Application of polyacrylamide gel (PAG) instead of agar to solid cultures of Streptomyces spp. was studied. The improved media were prepared by 1) gelling 20 ml of 5% acrylamide in a glass petri dish at room temperature, 2) washing by running water for more than 8 hr to remove residual reaction reagents, 3) drying at 5$0^{\circ}C$ for 12 hr to make a gel film, 4) autoclaving at 121$^{\circ}C$ for 15 min, and 5) swelling gel for about 4 hr by adding sterile liquid medium. In PAG media there were no differences from the observation of morphological characteristics showing during the cellular differentiation on agar media, whereas the ability to utilize carbohydrates differed somewhat from agar media. Agar media thus were little favorable for biochemical tests which the growth was determined depending on the formation of colony, but washed PAG was superior to serve as a solidifying agent.

  • The gel electrolytes were prepared with sulfuric acid and phosphoric acid, where hydrophilic fumed silica was used as a gelling agent. The influences of gel electrolyte on performance of the valve regulated lead acid (VRLA) batteries were investigated employing capacity tests, electrochemical impedance spectroscopy and scanning electron microscopy. The initial capacities of the sulfuric gel VRLA batteries were higher than that of phosphoric gel VRLA batteries. The sulfuric gel VRLA battery using 1.210 specific gravity of sulfuric acid with hydrophilic fumed silica exhibited the highest capacity of 0.828Ah. In the impedance measurements, the ohmic and charge transfer resistances for the phosphoric gel VRLA batteries were higher thanthat of sulfuric gel batteries. The morphology of electrodes of phosphoric gel VRLA batteries were more deteriorated in the SEM image.

  • The hyphal tip phenoloxidases of Coprinus congregatus were localized by the protoplast-concanavalin A method. Protoplast were generated from cultures grown on a solid medium which was solidified with a new gelling agent, Pluronid Polyol F127, instead of agar. the enzymes were associated with the cell membrane which might work as a transducer in the light recepter complex.

A Study of Textural Properties and Preferences of Fruit Pectin Jelly (펙틴을 이용한 과즙젤리의 질감특성과 기호도 연구)

  • This study was done to evaluate the effects of grape variety, pH and sugar contents on textural and sensory properties of grape pectin jelly. As a gelling agent, pectin 1% was selected due to highest sensory scores in preparatory experiments and also nutritional and economical reasons. To get the proximate composition of grape extract, moisture, pH, reducing sugar, and total titratable acidity was measured. Grape pectin jellies, made by 12 different recipies, were tested by sensory and instrumental evaluatoin. The results were as follows; 1) Hardness and chewiness were lowered in grape jelly which has lower pH and lower sugar contents. 2) People preferred grape jelly which has lower hardness, brittleness and sweetness, and higher sourness. 3) Comparing overall quality, Recipe C1 in Jelly 1 and recipe B1 in Jelly 2 were chosen as best. 4) Sensory hardness showed positive correlation with instrumental hardness, brittleness and chewiness.

  • This study was designed to establish Korean food culture by considering traditional fruit preserve. A historical approach of traditional fruit preserve were reviewed by the cooking book published from 1670 to 1988 in Korea. The results obtained from this study are as follows. Cooking method of fruit preserve was firstly published to Omshikdimibang in 1670 and published moderate cooking book in 1988 but fruit preserve has not been cooked yet. Cooking method of fruit juice and starch had been started in Samkuk dynasty and was completed in Lee Dynasty. Compared with the western fruit jelly, mung bean starch was used as a gel forming material in Korean traditional fruit preserve while gelatin was used as a gelling agent in western fruit jelly. Western fruit jelly was succeed in innovation, but traditional fruit jelly was failed to innovation.

  • Effects of media and plant growth regulators on the germination of somatic embryos of Angelica tree(Aralia elata Seem.) was studied for the mass production of Angelica tree through tissue culture. MS medium was found to be the most effective for the germination of somatic embryos(65% germination rate), Among the MS medium, the medium containing 25% less inorganic salts and 1% less sucrose was found to be the most effective. Gelling agent with 0.2-0.3% gelrite promoted the germination of somatic embryo$(65{\sim}70%)$ and caused good growth of shoots and roots. 0.1 mg/l of BA and kinetin treatment caused $65{\sim}70%$ germination rate of somatic embryos and good growth of shoots and roots, and resulted in high percentage of dry matter. 1mg/l or 5 mg/l treatment of putrescine, and 10 mg/l treatment of spermidine caused 90% germination rate of somatic embryos and good growth of plant organs, and inhibited vitrification of regenerated plants.
