Physicochemical Properties of Starch Granules from Endosperm Mutants in Rice (배유돌연변이체 쌀전분의 이화학적 특성 비교)

  • Starches from the eight varieties of rice were analyzed using scanning electron microscope(SEM), differential scanning calorimetry(DSC) and X-ray diffractometry, and tested on the starch-granule susceptibility to 15% $H_2SO_4$ and glucoamylase. The shape of starch granules from normal rice varieties and low-amylose mutants were polygonal while shrunken and floury mutants were globular. According to DSC, starches from Nampung CB243 showed higher onset temperature$(T_o)$, completion temperature$(T_c)$ and Punchilmi, Nampung EM90 showed higher enthalpy$({\triangle}H)$ of gelatinization than others. shr showed the highest hydrolysis rate to 15% $H_2SO_4$ while Nampung CB243 showed the lowest one. Eight varieties of rice starch granules showed A-type pattern on X-ray diffractograms. The lower amylose content rice varieties showed the higher hydrolysis rate treated with glucoamylase.

Physicochemical Properties of Starches from Flavored Glutinous Rice Varieties (향미찹쌀전분의 이화학적 특성비교)

  • Starches of flavored glutinous rice were analyzed by using scanning electron microscope (SEM) and differential scanning calorimetry (DSC) and tested on the starch granule susceptibility to 15% H$_2$SO$_4$, glucoamylase and $\alpha$-amylase. The shape of starch granules form flavored glutinous rice varieties was polygonal and the size was 4~6 $\mu$m in diameter. According to DSC, glutinous rice starch showed onset temperature (T$_{o}$) range 59.8~62.5$^{\circ}C$ and KR92021-B-B-42-3-B and KR92021-B-B-165-1-B showed higher enthalpy ($\Delta$H) on gelatinization than others. The starches from KR92021-B-B-5-2-B and KR92021-B-B-42-3-B showed lower hydrolysis rate using 15% H$_2$SO$_4$ than KR92021-B-B-165-1-B. KR92021-B-B-5-2-B showed higher degree of hydrolysis of glucoamylase and $\alpha$-amylase than the others.

Effect of Chestnut Flour on the Rheology of Dough and Processing Adaptability of White Pan Bread (밤가루 첨가가 밀가루 반죽의 물성과 제빵 적성에 미치는 영향)

  • The effects of replacement of wheat flour with 10, 20, and 30% chestnut flour on the bread-making properties and quality characteristics of bread were evaluated. Among the physical characteristics of the dough, the development time in the farinogram decreased with increasing amounts of chestnut flour, and the dough stability and durability were reduced. In the amylogram, there was a slight increase in the gelatinization starting temperature when compared to the control flour, but the maximum viscosity gradually decreased in response to increasing amounts of chestnut flour. The expansion ratio did not increase in the dough containing 20 or 30% chestnut flour. Moreover, the loaf volume and specific volume of bread containing 20 and 30% chestnut flour were smaller than those of the control and the 10% treatment. The hardness of bread containing 10% chestnut flour($700g/cm^2$) was lower than that of the other treatments($1413-1,627g/cm^2$). Furthermore, bread containing 10% chestnut flour had denser porous structures than the other treatments. The sensory evaluation tests revealed that the 10% treatment had higher sensory scores for the internal and external qualities of bread than the other treatments.

Effects of Panicle Position and Planting Density on the Physicochemical Properties of Starch in Panicle Number Type Rice

  • The tillering potential of panicle number type (PNT) rice greatly varies with planting density. Moreover, grain filling and ripening differ depending on the panicle position, which may further affect rice grain quality. The present study evaluated the grain quality of PNT rice sparsely planted to reduce production costs. The physicochemical characteristics of starch from the grains of PNT type rice 'Ilpum' planted at different densities (37, 50, 60, and 80 plants/3.3 m2) and at different positions of panicles (upper or lower on the culm) were determined. Overall, as the planting density decreased, the number of panicles increased but the starch content decreased, which further reduced the 1,000-grain weight. In particular, at the lowest density (37 plants/3.3 m2), protein content increased but particle size, enthalpy, and relative crystallinity decreased. The effects were more pronounced at lower than at upper panicle positions. These findings indicate that tillering potential differs with planting density, ultimately affecting the palatability of rice grains. Based on these findings, we propose restricting rice transplantation to a planting density of ≤37 plants/3.3 m2 to achieve the best quality of grains at lower costs and with less labor.

Rheological Properties of Dough and Quality Characteristics of Bread Added with Pumpkin Powder (단호박 분말을 첨가한 반죽의 물성 및 빵의 품질특성)

  • This study aims to investigate the quality characteristics of breads with the addition of sweet pumpkin powder. Farinogram showed that the absorption rate of the dough decreased, the development time and stability became shortened, and the degree of attenuation tended to be grown along with increasing the amount of pumpkin powder. From the amylogram, it was found that the gelatinization starting temperature and the maximum viscosity of pumpkin powder added dough seemed to be decreased as the amount of pumpkin powder was increased. Extensogram showed that there was a decrease in the degree of extension of the dough added with increasing the amount of pumpkin powder, while an increase in the degree of resistance and resistance/extensibility. The lightness (L value) and redness (a value) of bread was found to be decreased with increasing the amount of pumpkin powder, while the yellowness (b value) increased. In terms of the dough texture, for which the hardness, cohesiveness and gumminess of the dough were measured, there were no significant differences between groups, while the dough added with 8.0% pumpkin powder showed a significant decrease in the elasticity. The results of sensory evaluation showed that the highest score of color (p<0.05) and taste (p<0.05) were obtained from the bread added with 6.0% and 8.0% pumpkin powder, while the highest score of texture in control and 3.0% pumpkin bread. In addition, the highest score of flavor (p<0.05) and overall acceptance (p<0.05) were observed in bread added with 6.0% pumpkin powder. Upon the results of this study, it was assumed that the development of food products using pumpkin are prospective in response to health-oriented consumers.

Strategic Use of QTL Mapping to Improve the Palatability of Rice

  • The properties of starch play an important role in determining the palatability of rice. In addition, the gelatinization temperature (GT) of rice starch is an important factor in determining the quality of rice because it is related to the cooking time and texture of rice. For the development of high-quality rice, it is important to understand the genetic basis of palatability-related traits, and QTL analysis is an effective method to explain the genetic basis of variation in complex traits. QTL mapping related to alkali digestion value (ADV) of brown and milled rice was performed using the 120 Cheongcheong/Nagdong double haploid (CNDH) line. As a result, 12 QTLs related to ADV were detected, and 20 candidate genes were selected from the RM588-RM1163 region of chromosome 6 through screening by gene function analysis. The comparison of the relative expression level of candidate genes showed that OsSS1q6 is highly expressed in CNDH lines with high ADV in both brown rice and milled rice. In addition, OsSS1q6 has high homology with starch synthase 1 protein, and interact with various starch biosynthesis-related proteins, such as GBSSII, SBE, and APL. Therefore, we suggest that OsSS1q6 identified through QTL mapping could be one of the various genes involved in the GT of rice by regulating starch biosynthesis. This study can be used as basic data for breeding high-quality rice and provides a new genetic resource that can increase the palatability of rice.

Isolation and Physicochemical Properties of Rice Starch from Rice Flour using Protease (단백질분해효소에 의한 쌀가루로부터 쌀전분의 분리 및 물리화학적 특성)

  • This study aimed to investigate the impact of protease treatments on the yield of rice starch (RST) from frozen rice flours, and to compare the physicochemical properties of RST by alkaline steeping (control) and enzymatic isolation (E-RST) methods. Although the yield of E-RST, prepared according to conditions designed by the modified 23 complete factorial design, was lower than the control, the opposite trends were observed in its purity. E-RST (RST1, isolated for 8 h at 15℃ with 0.5% protease; RST2, isolated for 24 h at 15℃ with 1.5% protease; RST3, isolated for 24 h at 15℃ with 0.5% protease) with the yields above 50% were selected. Amylose contents did not significantly differ for the control and RST2. Relative to the control, solubilities were higher for all E-RST, but swelling power did not significantly differ for E-RST except for RST1. Although all E-RST revealed higher gelatinization temperatures than the control, the reversed trends were found in the gelatinization enthalpy. The pasting viscosities of all E-RST were lower than those of the control. Consequently, the enzymatic isolation method using protease would be a more time-saving and eco-friendly way of preparing RST than the alkaline steeping method, even though its characteristics are different.

Quality characteristics of dumpling shell with sea tangle powder (다시마 분말을 첨가한 만두피의 품질 특성)

  • This study investigated the effects of sea tangle powder (SP) on the physicochemical and sensory characteristics of dumpling shells. Various dumpling shell samples were prepared with wheat flour containing different amounts of SP. The characteristics of the dumpling shell samples were investigated based on their viscosity, cooking properties, color values, and texture properties. Their sensory characteristics were evaluated by using seven scores. According to the amylograph data, the composite SP-wheat flour samples showed an increased gelatinization temperature with an increasing SP content, whereas the initial viscosity at $95^{\circ}C$ was decreased after 15 minutes. The increase of SP amount decreased the values of L and b whereas the a value increased. With regard to the textural characteristics, the dumpling shells with an increased amount of SP showed increased hardness, cohesiveness, and adhesiveness, whereas the SP addition reduced the springiness of the dumpling shell. The dumpling shell with the addition of 3% SP showed the highest overall acceptability in the sensory evaluation.

Physicochemical Properties of Several Sweet Potato Starches (품종별 고구마 전분의 이화학적 특성)

  • The physicochemical properties and characteristics of sweet potato starches which were isolated from the six varieties were investigated. The shapes of starch granules which observed through photomicroscope and scanning electron microscope lucre round and polygonal, but those of the Shinmi were most polygonal, and the average diameters were in the range of $10.4{\sim}14.2$ microns. The amylose contents were between 25% and 28%, and blue values and alkali numbers were in the range of $0.29{\sim}0.36$, $7.0{\sim}12$, respectively. The swelling power and solubility patterns of the starches were negligible until $50^{\circ}C$, thereafter it increased rapidly and the Eunmi showed highest water binding capacity of 211.6%. Amylogram pattern of 6% starch solutions were similar to no peak viscosity, but maximum viscosity varied widely with varieties. A significant positive correlation was observed between amylose content and average gelatinization temperature. Taste and texture of the steam cooked sweet potatoes were negatively and positively correlated with moisture and amylose contents, respectively, while those of the microwave cooked sweet potatoes were only positively correlated with amylose contents.

  • The physicochemical properties of starch isolated from two kinds of waxy barley were investigated. The average diameters of starch granules of Changyoung and Smire were 13.6 and 16.6 microns, respectively, and most of starch granules were round shape. X-ray diffraction patterns of two samples were A-types and amylose contents of two waxy barley starches were 4%, blue values and alkali numbers of Changyoung and Smire were 0.22 and 0.18, 5.2 and 4.0, raising powers of them were 167 and 173, respectively. The optical transmitance of 0.1% suspension of waxy barley starches increased rapidly from $51^{\circ}C\;to\;75^{\circ}C$. Amylogram data on 5% of Changyoung and Smire starch solutions showed the gelatinization temperature of 63 and $62^{\circ}C$, maximum peak hight of 920 and 900 B.U., respectively. Waxy barley starches had higher swelling powers than non waxy barley starch. The starch pastes prepared from waxy barleys retrograded much slower than that of non waxy barley.
