Development of Alginate-Celite Immobilization Technique for the Improvement of Ethanol Productivity (에탄올 생산성 향상을 위한 Alginate-Celite 고정화 방법의 개발)

  • The optimal initial pH for the ethanol production by Saccharomyces K35 was found to be 5.0, and about 80% of yield was obtained when 200g/$\ell$ of glucose was used as a substrate, which showed sugar tolerant. As the additives and cross-linking agent, the addition of 1.67%(w/v) Celite R-634 together with 0.33%(v/v) of glutaraldehyde(ACG bead) resulted in better stability, ethanol productivity and cell viability than Ca-alginate bead. Also, ACG bead seemed to be more resistant to phosphate ion than Ca-alginate bead, considering outgrowing cell concentration in the media. Scanning electron microscopic observation depicted that the surface of ACG bead was almost similar to the original state but not for Ca-alginate bead. When repealpd-batch culture was performed with Ca-alginate bead for 60 days in a 500m1 Erlenmeyer flask, ethanol and cell concentration were maintained about 138g/$\ell$-gel and 29~30g/$\ell$-gel, respectively, up to 40 days(7th run number), and then both were rapidly decreased. In the case of ACG bead, ethanol and cell concentration were maintained about 130~150g/$\ell$-gel and 32~35g/$\ell$-gel, respectively, up to 60days(10th run number). Cell viability was maintained about 70%, and outgrowing cell concentration was below 5.8% of total cell concentration.

  • Two types of alginate gel beads capable of floating in the gastric cavity were prepared. The first, alginate gell bead containing olive oil(Al-Oil), is a hydrogel bead and its buoyancy is attributable to olive oil held in the alginate gel matrix. The model drug, metronidazole(MZ), contained in Al-Oil was released gradually into artificial gastric fluid. The profiles of MZ release from Al-Oil shown initial burst and after 90 min they were about 100%. The second, alginate gel bead containing curdlan microsphere(Al-C), is a gel bead with curdlan-MZ microsphere in the matrix. To sustained release rate of drug, alginate bead were prepared curdlan microsphere containing MZ. Results demonstrated that sustained delivery of MZ over 2h can be easily achieved while the bead remained float. The release properties of prepared alginate beads are applicable not only for sustained release of drugs but also for targeting the gastric mucosa.

Optimal Conditions of Co-Immobilized Mixed Culture System with Aspergillus awamori and Zymomonas mobilis (Aspergillus awamori와 Zymomonas mobilis로 구성된 혼합고정화 배양계의 최적 조건)

  • Co-immobilized mixed culture system(A-Z system) composed of two different oxygen-demanding strains, aerobic(Aspergillus awamori) and anaerobic(Zymomonas mobilis) strains, in a Ca-alginate gel beads was developed to increase ethanol production from raw starch as a carbon source. Optimal mixture ratio of A. awamori and Z. mobilis was $1.25{\times}10^{9}\;spores/L-gel$ and 0.5g cells/L-gel, respectively. After 120 hours of cultivation, gel beads distinguished oxygen-rich surface for A. awamori from oxygen-deficient central part for Z. mobilis. At A-Z culture system, yield of ethanol on glucose, $Y_{p/s}=0.18$, was very low and there was high leakage of cells from surface of gel beads. At A-Z 36 cultrue system with changing silicon check valve for cotton plug at 36 hours in A-Z culture system, there was no cell leakage from gel beads, pH was maintained at around 4.3 during cultivation, and yield of ethanol on glucose, $Y_{p/s}=0.36$, showed 2 times higher than that of control culture system(cotton plug culture).

Characteristics of Immobilized Rhodopseudomonas sp. for Wastewater Treatment (폐수처리를 위한 고정화 Rhodopseudomonas sp.균의 특성)

  • Rhodopseudomonas sp. was immobilized in three supports(agar, k-carrageenan, and PVA) in order to remove nitrate in wastewater coming from fish farm. Among them 3% agar was the most suitable support when denitrification rate and bead durability were tested. Optimum bead size was 4mm-diameter when the substrate transfer into the bead and shear stress for bead were considered, and optimum cell loading was 25mg dry $cells/cm^2$gel gel. Ethanol was the best as a carbon source, and optimum C:N ratio, temperature and pH were 1.5:1, $31^{\circ}C$,, and 6, respectively. Under these conditions the maximum denitrification rate in synthetic wastewater was $$345{\MU}{\ell};N_2/Cm^3 gel{\cdot}hr;and that in modified MYC medium was 450{\MU}{\ell}};N_2/Cm^3 gel{\cdot}hr $$.

Ethanol Production by a New Method of Alginate-Immobilization (새로운 Alginate 고정화 방법에 의한 에탄올 생산)

  • When the cells of yeast K35 were immobilized in Ca-alginate gel, cell concentration and viability decreased as alginate concentration increased. Considering the results, 2% (w/v) Ca-alginate concentration would be suitable. Among various concentrations of additives and cross-lin-king agent, the addition of 1.67% (w/v) of bentonite together with 0.33% (v/v) of glutaraldehyde (ABG bead) resulted in the highest ethanol production of 1.8%(w/v), using YPD medium containing 2% glucose. ABG bead seemed to be more resistant to phosphate ion than Ca-alginate bead. 0.33%(w/v) of phosphate was a proper concentration for the ethanol production by ABG bead. Scanning electron microscopic observation depicted that the immobilized cells on the bead surface were coated by alginate gel and that the cells in the internal bead were cross-linked with alginate matrix. When repeated-batch culture was performed with ABG bead for 40 days in a packed-bed reactor, ethanol concentration of about 90~110 g/l-gel was maintained. Cell viability was maintained around 70%, and outgrowing cell concentration was below 6.3% of total cell concentration. Consequently, the results showed that ABG head was a potential carrier for continuous production of ethanol compared to conventional Ca-alginate bead.

Eletron Microscopic Observation of Calcium-Acetylated Seaweed Alginate Gel

  • Seaweed alginate was acetylated by activated carbon immobilized Pseudomonas syringae in a fluidized bed, up-flow reactor. The acetylation degree of seaweed alginate was about 30%. Calcium-acetylated seaweed alginate gel bead was made and compared to calcium-seaweed alginate gel bead by the scanning electron microscopy (SEM). Structural difference of two gel beads may results from increased viscosity and decreased affinity of acetylated seaweed alginate for calcium ion. On the basis of interior and exterior structure of calcium-acetylated seaweed alginate gels and property of acetylated seaweed alginate, it seems that acetylated seaweed alginate is used for the supporter for electrophoresis and packing materials for liquid chromatography and gel filtration.

Sphericity Optimization of Calcium Alginate Gel Beads and the Effects of Processing Conditions on Their Physical Properties

  • In this study, the sphericity of calcium alginate gel beads was optimized using response surface methodology. The optimum conditions for bead sphericity were a concentration of 2.24% sodium alginate, a flow rate of 0.059 mL/sec for the sodium alginate solution, and a 459 rpm rotation for the calcium chloride solution. The predicted and experimental bead sphericities under the optimum conditions were 94.5 and 96.7%, respectively, showing close agreement. We also investigated the processing condition effects for the physical properties of the optimized calcium alginate gel beads. Immersion in hot water slightly decreased bead size and rupture strength. NaCl treatment increased bead size and decreased rupture strength. While the pH of the calcium chloride solution had little effect on bead sphericity, the bead sizes and gel strengths decreased with longer times in each pH solution. The beads coated with pectin and glucomannan showed no significant changes in sphericity, but their sizes decreased with time. The coated beads showed higher rupture strengths than the uncoated beads.

Nitrogen removal performance of anammox process with PVA-SA gel bead crosslinked with sodium sulfate as a biomass carrier

  • In this study,the result shows that polyvinyl alcohol-sodium alginate (PVA-SA) gel bead crosslinked with sodium sulfate are better among the different methods by comparing the relative mechanical strength, mechanical strength swelling and expansion coefficient of beads in water. Subsequently, anammox biomass entrapment by PVA-SA gel was introduced into continuous stirred tank reactor (CSTR). After 24 operation days, the nitrogen removal efficiency achieved 60%, while the nitrogen loading rate (NLR) was $0.14kgN/m^3/d$ and the experiment data indicated that PVA-SA gel bead crosslinked with sodium sulfate can be used to initiate anammox process. Furthermore, it is an alternative for culturing anammox in a long-term operation.

Continuous Nitrate Reduction by Gel and Foam Matrix (GFM) Immobilized Cells (겔-스폰지 혼용 시스템에 고정화된 세포를 통한 질산염의 연속적 분해)

  • In this study the anaerobic degradation of nitrate by in GFM (gel and foam matrix) and bead gel immobilized Paracoccus denitrificans DSM 65 in continous culture was conducted. A novel GFM immobilization system was developed in order to improve conventional system (bead). With increasing nitrate concentration in water, the nitrate reduction rate was increased. The observed maximum denitrification rate by in GFM immobilized cells was 177 mg/L h in buffered water, while that was 33 mg/L h in tap water. In comparison with bead system the reduction activity by GFM system showed $1.2{\sim}2.1$ times better. The denitrification activity was not changed after 16 days storage at $5^{\circ}C$ and also showed better activity than that of free cells or even bead immobilized cells.

Citric Acid Production Using Immobilized Yeast Activated with $CaCl_2$ - containing Medium (고정화효모를 사용한 시트르산 생성에 있어서의 $CaCl_2$ 함유배지에 의한 활성화 효과)

  • Immobilized Candida lipolytica cells were prepared by entrapping the whole cells in calcium alginate gel. To enhance citric acid productivity, immobilized cells were Incubated with activation medium in fluidized-bed reactors. When the activation was done in batch operation, maximum citric acid productivity appeared in a much shorter time than in continuous operation. Activated immobilized cells were enhanced about 10-fold in citric acid production relative to non-activated immobilized cells. The productivity of citric acid was also influenced by bead size. When Immobilized cells were reacted in a fluidized-bed reactor with the same quantity of cells, the citric acid productivity was increased as the bead size was decreased.
