커규민의 인도메타신 유도 위점막 손상에 대한 치료 효과 (Curcumin Induces Recovery from Indomethacin-Induced Gastric Mucosal Lesions in Rats)

  • 인도메타신은 nonsteroid anti-inflammatory agent로 심각한 위점막 손상을 야기한다. 본 논문에서는 rat의 위점막 손상에 대한 커규민의 치료효과를 규명하였다. 인도메타신 유도 위점막 손상에 대한 커규민의 치료효과를 검증하기 위하여 인도메타신(25 mg/kg)의 경구투여를 통하여 위점막 손상을 유발한 후 다양한 농도(10, 50, 100 mg/kg)의 커규민을 3일간 경구 투여하였다. 실험 결과, 인도메타신의 처리는 위점막에 궤양부위를 증가시킨 반면, 3일간 커규민의 경구투여는 농도 의존적으로 궤양부위를 유의성있게 감소시켰다. 뿐만 아니라, 커규민은 인도메타신에 의해 유도되는 위점막에서의 지질과산화 증가를 상당히 억제시켰고 radical scavenging enzyme인 superoxide dismutase, catalase, glutathione peroxidase의 활성을 농도 의존적으로 증가시켰다. 이상의 결과들을 종합하여 볼 때, 커규민은 인도메타신 유도 위점막 손상을 지질과산화의 억제와 radical scavenging enzymes의 활성화를 통하여 치료한다.

A Protective Effect for Panax ginseng in the Rat Stomach

  • The effect of ginseng on gastric ulcer and gastric acid secretion was investigated in pylorus-ligated rats. Methods: Sprague-Dawley strain rats were used after 24 hours fast. Pylorus-ligation was performed under light ether anaesthesia, then gastric mucosal damage was evoked in conscious pylorus-ligated rats by the administration of subcutaneous (s.c.) indomethancin (20mg/kg), s.c. histamine (150mg/kg) or by pylorus-ligation (Shay ulcer). Ginseng was given by intragastric (i.g.) or intraperitoneal (i.p.) route simultaneously with the ulcerogens. Rats were killed after 3h (indomethacin) and histamine models) or after 18h (Shay ulcer), when the gastric secretory responses, the number and severity of gastric mucosal lesions and mucosal mucus content deetermined. the effect of i.p. ginseng on basal gastric acid secretion and on gastric acide secretion in indomethacin (20mg/kg, s.c.)-treated rats was also investigated in urethane anesthetized rats. Gastric acid secretion was measured by flushing of the gastric lumen with saline every 15min through an oesophageal cannula. Results: In conscious pylorus-ligated rats, i.g. ginseng(12.5-50mg/$m\ell$; 50-200mg/kg) protected against gastric mucosal lesions evoked by s.c. indomethacin or s.c. histanmine in the d3-h pylorus-lighted rat, withoutmodifying gastric acid secretory responses. Ginseng given i.p. (150 or 200mg/kg) did not reduce the gastric lesions produced by histamine or by ligating the pylorus (Shay ulcer) Ginseng given orally in 50mg/$m\ell$ (200mg/kg) increased gastric mucus secretion in saline- and indomethacin-treated conscious pylorus-ligated rats. In anaesthetized rats ginseng (50 or 200mg/kg) did not modify basal gastric acid secretion or gastric acid secretion in the indomethacin-treated rats. Conclusions: ginseng given orally exerts gastroprotective effects in the rat stomach. Such anti-ulcer effect does not involve changes in gastric acid secretory responses. In addition, ginseng possesses stimulatory effect on gastric mucus secretion, which could be one mechanism by which the compound exerts its antiulcer effect. Our data are in favor for a beneficial effect for topically applied ginseng on the gastric mucosa.

Mouse의 위점막 염증에 대한 사삼맥문동탕의 면역반응 연구 (The Effects of Sasammaickmoondong-tang against Gastric Mucosal Lesions)

  • Objectives: This study was carried out to investigate the effects of Sasammaickmoondong-tang (SME) on gastric mucosal lesions induced by indomethacin in mice. Methods: The normal group was no inflammation-induced mice. The control group was gastro-inflammation-induced mice. The sample group was mice administered SME after gastro-inflammation elicitation. Results: In the common morphology and histochemical change, the control group was observed with various injury-mucous surface cell, micro-villi, paneth cell, surface epithelial cell, goblet cell - by hemorrhagic erosion, while the sample group was as same as the normal group. In the immunohistochemical change, the distributions of COX-1, Bcl-2, and BrdU treated with SME were noticeably higher than in the control group (p<0.05). The distributions of TUNEL, NF-B, COX-2, IL-2R-, NK-1.1, ICAM-1, and CD11b/18 in those treated with SME were noticeably lower than in the control group (p<0.05). Finally, the distribution of SBA was the same as in the normal group. Conclusions: According to the above results, it is supposed that Sasammaickmoondong-tang is applicable to gastric mucosal lesions.

  • Objectives : This study was carried out to investigate the effects of sihogeji-tang on indomethacin-induced gastric mucosal lesions in rats. Methods : Experimental rats were classified into three groups. The normal group had no inflammation elicited. The control group was rats administered water after elicitation of gastro-inflammation. The sample group were administered sihogeji-tang after gastro-inflammation elicitation. Results : In the common morphology and histochemical change, the control group were observed to have various injuries by hemorrhagic erosion, while the sample group had noticeably fewer injuries than the control. In the immunohistochemical change, the distribution of HSP-70, PCNA treated with sihogeji-tang noticeably increased over the control group. The distribution of NF-kB, COX-2 treated with sihogeji-tang noticeably decreased compared to the control group. (p<0.05) Conclusions : Sihogeji-tang had significant effects on indomethacin-induced gastric mucosal lesions in rats.

  • This study was carried out to investigate the effects of Hyangsapyeongwisan(HP) on gastric mucosal lesions induced by indomethacin in mice. The control group consisted of gastro-inflammation elicited mice. The sample group consisted of mice given HP after onset of gastro-inflammation. In common morphological and histochemical change, various cell abnormalities were observed in the control group, such as mucous surface cell, peanut cell, surface epithelial cell, goblet cell abnormalities, all caused hemorrhagic erosion. The sample group was the same as the control group. In the immunohistochemical change, the distributions of COX-I, BrdU treated with HP were notably higher than those of the control group(p$NF-{\kappa}B$, COX-2, PKC, IL-12B in mice treated with HP were notably lower than those of the control group(p

  • Objectives : This study was carried out to investigate the effects of Jaeumgeonbi-tang extract on indomethacin-induced gastric mucosal lesions of mice. Methods : Experimental groups were classified into non-treatment group (CON group), non-administered group (GE group), misoprostol administered group (MA group) and Jaeumgeonbi-tang extract administered group (JG group). This study examined the morphological change, distribution of mast cells, mucous secreted cells and apoptotic cells, BrdU, COX-1, Hsp70, NF-κB p50, PKC, COX-2 and TNF-α of gastric mucosa. Results : 1. The hemorrhagic erosion of gastric mucosa and infiltrated mast cells were reduced in the MA and JG groups. 2. PNA reaction and mucous secreted cells were increased in the MA and JG groups. 3. The distribution of apoptotic cells, Hsp70, NF-κB p50, PKC, COX-2 and TNF-α were increased in the gastro­inflammation elicitated group, but decreased in the MA and JG groups. 4. The MA and JG groups showed increase on COX-1, BrdU. Conclusions : Jaeumgeonbi-tang extract had excellent effects on indomethacin-induced gastric mucosal lesions.

산약이 알콜로 유도된 흰쥐의 위 점막 손상 예방에 미치는 영향 (Protective Effect of Rhizoma Dioscoreae on the Gastric Mucosal Lesions Induced by Alcohol in Rats)

  • The aim of this study was carried out to investigate the protective effect of Rhizoma Dioscoreae on the gastric mucosal lesions induced by alcohol in rats. Experimental groups were treated by oral infusion with Rhizoma Dioscoreae extract at the dose of 0.03465 g/ml(OA-RD1 group), 0.0693 g/ml(OA-RD2 group), and 0.1386 g/ml(OA-RD3 group), while D.W group was administrated with the distilled water and control group did not pretreated. Experimental groups pretreated for 14 days, and given orally 1 ml of 75% alcohol two times(30min interval). The animals were killed 1hr 30min after alcohol treatment, and measured rats body weight, absolute stomach weight, relative stomach weight, SOD activity, glutathione peroxidase(GPx) activity, observed gastric mucosal lesions. The body weight was unremarkable changed. In once as dose intake group's absolute stomach weight was increased and In once as dose intake group, twice as dose intake group's relative stomach weight was increased. SOD activity, glutathione peroxidase activity in twice as dose intake group is remarkably increased. Light microscopy Observations of congestion, hemorrhage, and erosion in gastric mucosal lesions were shown severely in control group than OA-RD1, OA-RD2, OA-RD3. These results suggest that the proposed gastroprotective effect may involve activation of antioxidant effect. And Twice as dose is especially effective.

갈근(葛根)의 항산화작용에 의한 위점막 보호효과와 내인성 Sulfhydryl 화합물의 영향 (Antioxidants Effects and Roles of Endogenous Sulfhydryls in the Gastric Mucosal Protection of Puerariae Radix)

  • Ethanol induces compoundhemorrhagic gastric lesions and causes a dose-dependent decrease in the concentration of endogenous nonprotein sulfhydryls in rat gastric mucosa. Sulfhydryl-containing drugs protect rats from ethanol - induced gastric lesions. Based on this findings, we investigated the involvement of sulfhydryl compounds in the antioxidant effects of Puerariae radix, a traditional herbal medicine, against ethanol - induced gastric lesions in the absence and presence of iodoacetamide(IDA. sulfhydryl blocking agent) in rats. respectively. Because of the known role of sulfhydryls in gastric cytoprotection, its role in gastric antioxidation was of intrest. In vitro, Puerariae radix extract(PRE) reduced linoleic acid autooxidation and exert DPPH radical scavenging effect. In vivo. PRE increased antioxidants(SOD, catalase. GSH) and reduced lipid peroxide level in ethanol-induced gastric mucosal lesions. But treatment with PRE plus IDA significantly inhibit the antioxidant effects such as SOD and GSH but did not affect catalase levels. These results suggest that Puerariae radix may play roles in the gastric cytoprotection through antioxidant effects and increase of SOD activity and GSH level are dependent of endogenous sulfhydryls.

Protective effect of ginsenoside Re on acute gastric mucosal lesion induced by compound 48/80

  • The protective effect of ginsenoside Re, isolated from ginseng berry, against acute gastric mucosal lesions was examined in rats with a single intraperitoneal injection of compound 48/80 (C48/80). Ginsenoside Re (20 mg/kg or 100 mg/kg) was orally administered 0.5 h prior to C48/80 treatment. Ginsenoside Re dose-dependently prevented gastric mucosal lesion development 3 h after C48/80 treatment. Increases in the activities of myeloperoxidase (MPO; an index of neutrophil infiltration) and xanthine oxidase (XO) and the content of thiobarbituric acid reactive substances (TBARS; an index of lipid peroxidation) and decreases in the contents of hexosamine (a marker of gastric mucus) and adherent mucus, which occurred in gastric mucosal tissues after C48/80 treatment, were significantly attenuated by ginsenoside Re. The elevation of Bax expression and the decrease in Bcl2 expression after C48/80 treatment were also attenuated by ginsenoside Re. Ginsenoside Re significantly attenuated all these changes 3 h after C48/80 treatment. These results indicate that orally administered ginsenoside Re protects against C48/80-induced acute gastric mucosal lesions in rats, possibly through its stimulatory action on gastric mucus synthesis and secretion, its inhibitory action on neutrophil infiltration, and enhanced lipid peroxidation in the gastric mucosal tissue.

  • Objective : This study was carried out to investigate the effects of Jisildochehwan extract on indomethacin-induced gastric mucosal lesions of mice. as compared with misoprostol. Methods : Experimental mice were classified into four groups. No gastro-inflammation elicited mice were the normal group(NOR). Gastro-inflammation elicited mice were the control group (CON). Misoprostol-administered mice after gastro-inflammation elicitation were the misoprostol-administered group (MA). Jisildochehwan-administered mice after gastro-inflammation elicitation were the Jisildochehwan-administered group(JA). In this study, we examined superoxide dismutase(SOD) ability, anti-inflammatory ability arrangement of mucous-secreted cells. distribution of mucosal neck cells, mucosal surface cells. neutral mucous-secreted cells. PAS reaction. COX-1, $TNF-{\alpha}$, $NF-{\kappa}B$ p65 and iNOS. Results : The SOD ability of the Jisildochehwan group increased dose-dependently. The hemorrhagic erosion and the damage of arrangement of mucous-secreted cells increased in the CON group. but decreased in the MA and JA groups. The COX-1 positive were decreased in the CON group, but increased in the MA and JA groups. The distribution of mucosal neck cells and mucosal surface cells. PAS positive reaction of surface mucous cells were decreased in CON group, but increased in MA and JA group. $TNF-{\alpha}$, $NF-{\kappa}B$ p65 and iNOS increased in the CON group, but decreased in the MA and JA groups. In all the results, the effects were better in the JA group than in the MA group. Conclusion : Jisildochehwan extract was more effective than Misoprostol. Jisildochehwan has excellent protective effects on indomethacin-induced gastric mucosal lesions.
