Systematic Propagation of High Quality Garlic (Allium sativum L.) Through Shoot Apical Meristem Culture 1. Organogenesis from in Vitro Cultured Shoot-tips (생장점배양에 의한 우량마늘 체계적 증식 1.생장점배 양으로부터 기관형성)

  • Since garlics (Allium sativum L.) are propagated through cloves, infection by virus or other pathogens may become severe problem if not using high quality seed bulbs every year resulting in the reduction of yield and bulb quality, In order to solve this problem, the establishment of virus-free bulb production and its supply system have been required because no chemicals were found to eliminate viruses from seed bulbs. This experiment was conducted to develop an effective production technique of high quality seed bulbs using shoot-tip culture. Over 90% of shoot-tips explanted on January L 1990 were survived at constant temperature of either 20, 24 or 28$^{\circ}C$, wheres 88% at alternate temperature (28/20$^{\circ}C$). The growth of shoot and root was most vigorous at constant 24$^{\circ}C$, and least at alternate temperature (28/20$^{\circ}C$) condition. When shoot-tips were explanted June 21 to August 1,1991, survival and growth of shoot-tips was most vigorous on MS medium supplymented with 0.1 mg/L NAA and 2 mg/L kinetin and least 1 mg/L Gh$_3$. The shoot-tips taken from the seed bulbs stored at 4$^{\circ}C$ for 15 to 60 days were placed on MS medium, shoot growth and in vitro bulblet formation increased slightly as affected by the increase of told treatment period at 4$^{\circ}C$.

On Improvement of Garlic Productivity by Inactivation of Virus in Garlics (마늘 Virus 불활성화에 의한 생산성 향상에 관하여)

  • The effect of heat or chemotherapeutant treatment on the mosaic virus infected garlic(Allium sativum L.) scales and that of chemotherapeutant added to the culture medium were summarized as following. The treatment of the virus infected garlic scales at $37\~57^{\circ}C$ for 35 days to one hour in water or in air shelved no effort of inactivating the virus. Although treatment of the garlic scales at $62\~72^{\circ}C$ for 90 to five minutes reduced the mosaic symptom on the leaves of the garlic plants grown after the heat treatment, it reduced the growth vigor of the plants so greatly that complete inactivation of the virus in garlics was not feasible. The mosaic symptom on the leaves of garlic plant was reduced when the infected garlic scales were grown after 24 hours soaking in $10\~50\;ppm$ Malachite Green, 2,4-Dichlorophnoxy Acetic Acid, or in $20\~100\;ppm$ Quinhydron. These chemotherapeutants, however, inhibited the growth of garlic plant at the high concentration. Garlic scales soaked in $10\~50\;ppm$ Naphthyl Acetic Acid showed the least mosaic symptom without its complete extiction on the garlic leaves. When incorporated into the modified Murashige-Skoog's medium, $0.5\~l.5\;ppm$ Naphthyl Acetic Acid could inactivate the mosaic virus in newy developed garlic plants showing no mosaic symptom on the leaves, no inclusion bodies and intact nuclei in the leaf tissue cells.

Antioxidant Activities of Domestic Garlic (Allium sativum L.) Stems from Different Areas (산지별 국내산 마늘종의 항산화 성분과 항산화 활성 비교)

  • Antioxidant compounds and activities were investigated for both northern (Uiseong) and southern ecotype (Namhae) domestic garlic stems. The contents of chlorophyll, carotenoid, vitamin C, phenolic compounds and allicin in garlic stems were measured. Antioxidant activities of methanol and ethanol extracts of garlic stems were compared determining lipid peroxidation on the linoleic acid system, electronic donating ability (EDA), superoxide dismutase (SOD)-like activity and nitrite scavenging ability (NSA). The amounts of chlorophyll, carotenoid, phenolic compounds and allicin in northern ecotype were significantly higher, but vitamin C content was lower than those of southern ecotype garlic stems. The allicin contents of garlic stems and garlic bulbs were $26.1{\pm}1.0{\sim}28.2{\pm}0.9$ and $33.2{\pm}0.7{\sim}33.5{\pm}0.8\;mg%$, respectively. All antioxidant activities were higher in methanol extracts of garlic stems than in ethanol extracts and were higher with northern ecotype than those with southern ecotype extract. The SOD-like activity of garlic stem extract was higher than that of garlic bulbs while EDA, lipid peroxidation and NDA of garlic stem extracts were lower than those of garlic bulbs. Antioxidant activities of garlic stems were more than 50% of garlic bulbs.

Production of Mother Bulbs with High Quality Using Bulbils of Garlic (Allium sativum L.) (마늘의 주아(珠芽)를 이용(利用)한 우양종구생산(優良種球生産))

  • This study was carried out to determine the promising method for the production of mother bulbs using garlic bulbs at the experimental field of Ange high school, Eusung in Korea from 1996 to 1998. The bulbs and bulbils were taken from a local cultivar 'Eusung' garlic. The growth of bulbil and yield potential were compared with different harvesting times and cultivation methods of bulbil. The harvesting time of bulbils which were capable of producing the mother bulbs was ranged from 15 days to 30 days after the emergence of flower stalks. The best results based on the yield and number of bulbs were obtained from the bulbils sampled at the just before garlic harvesting, 30 days after flower-stalk emergence. The mulching with transparent polyethylene(PE) film in bulbil cultivation was more effective for the plant growth and bulb formation than that with black PE film. The PE mulching from 30 days after seeding of bulbils to 30 days (late-April) before bulb harvesting was the moot effective for the increase of bulb yield. In the cultivation of garlic bulbils, application of lime (150 kg/10a) and compost (3,000 kg/10a) without any chemical fertilizers produced more bulbs in comparison with the standard level of fertilizer (N - $P_2O_5$ - $K_2O$ - compost - lime = 24.8 - 8.0 - 19.2 - 3,000 - 150 kg/10a). It was determined that the involucre seeding and assemble seeding of bulbils was effective and labour-saving methods for the production of mother bulbs from garlic bulbils. Input of labour force for the involucre seeding was decreased up to 61% as compared with the conventional drill seeding of bulbils. Mean yield from the cultivation of bulbil-derived single cloves was remarkably increased up to 95% in comparison with that of the common cloves. The larger size of single clove produced more yields, and the optimum planting density of single clove was $20{\times}10cm$. The cloves differentiated directly from bulbil showed to have higher yield potential than that from common cloves.

Effect of Garlic (Allium sativum L.) Extracts on Formation of N-Nitrosodimethylamine (마늘 추출물이 N-Nitrosodimethylamine의 생성에 미치는 영향)

  • This study was conducted to investigate the effect of garlic extracts on electron donating ability nitrite-scavenging activity and N-nitrosodimethylamine (NDMA) formation. Content of total phenolic compounds was the highest in ethyl acetate extract. Electron donating ability was significantly increased with increasing sample concentration, as $88.0{\pm}1.53%$ from ethyl acetate extract and $77.5{\pm}0.8%$ from butanol extract in concentration of 10 mg/mL. Butanol extract from garlic with highest nitrite scavenging activity and inhibition effect of NDMA formation exhibited $96.9{\pm}0.75%$ and $68.6{\pm}1.15%$ in concentration of 10 mg/mL, respectively. The butanol extract was fractionated into 5 fractions using prep-LC. Nitrite-scavenging activity and inhibition effect of NDMA formation were excellent in fraction III, which further fractioned into 3 subfractions (III-a, III-b and III-c) using prep-LC. Of the 3 subfractions, its activity was the highest in the subfraction III-c. Therefore butanol extract of garlic was suggested to be effective inhibitor of NDMA formation.

Oxidative Stress Inhibitory Effects of Low Temperature-Aged Garlic (Allium sativum L.) Extracts through Free Radical Scavenging Activity (저온숙성마늘의 라디칼 소거 활성을 통한 산화스트레스 억제 효과)

  • Garlic has drawn attention as a food material for its anti-oxidative and anti-inflammatory properties as well as for prevention and treatment of cancer. In order to increase efficiency, various aging methods for garlic have been attempted. In particular, thermally processed garlic is known to have higher biological activities due to its various chemical changes during heat treatment. Therefore, in this study, we investigated the anti-oxidative effects of garlic extracts aged at low temperature ($60{\sim}70^{\circ}C$). In the results, 2,2-diphenyl-1-picrylhydrazyl and 2,2-azino-bis (3-ethylbenzo-thiazoline-6-sulfonate) radical scavenging activities and ferric reducing ability of low temperature-aged garlic (LTAG) were similar to those of raw garlic. LTAG also showed decreased lipopolysaccharide (LPS)-induced production of reactive oxygen species, although there were not significant differences among samples. In addition, xanthine oxidase activity was inhibited by LTAG; the 15 days and $60^{\circ}C$ extract showed outstanding inhibition compared with the others. To understand the molecular mechanisms behind the anti-oxidative activity of LTAG, we performed quantitative real-time PCR analysis. The 30 days and $70^{\circ}C$ extract upregulated mRNA expression of antioxidant enzymes such as Cu/Zn-superoxide dismutase (SOD), Mn-SOD, glutathione peroxidase, and catalase in LPS-stimulated RAW 264.7 cells. This result indicates that LTAG can be a functional food as a nature antioxidant and antioxidant substance.

Effects of Applying Slow-Release Fertilizer on Southern Type Garlic (Allium sativum L.) Cultivation (난지형 마늘 재배 시 완효성 비료 시용효과)

  • This experiment was conducted to evaluate the effect of slow-release fertilizer on warm type garlic cultivation in the Southwestern region of Korea. The slow-release fertilizers used were UF(Urea-Formaldehydes)70, 100, 130, CDU(Crotonylidene Diurea)100, and IBDU(Isobuty- lidene Diurea)100. The slow-release fertilizer treatment slightly increased plant height, leaf sheath diameter, and leaf number compare with conventional fertilizer treatment. Also, large bulb rate, nitrogen and pyruvic acid contents of garlic in slow-release fertilizer plot were higher than other treatments. Application of slow-release fertilizer increased the commercial yields.

  • This study was designed to evaluate the effects of garlic extracts of various concentrations on the growth of various pathogens and human colon cancer cell lines in vitro. For antibacterial effects against microorganisms, minimal inhibition concentrations (MIC) of alliin were from 5,000 to 20,000 ppm. MIC of ethanol extract were from 1,250 to 10,000 ppm. For cytotoxic effect of alliin and ethanol extract against human colon cancer cell lines (HCT-15), the growth rates of the cancer cells in medium containing alliin and ethanol extract were inhibited gradually to a significant degree in proportion to the increase of these concentration. Morphology of HCT-15 cells in medium containing alliin and ethanol extract were seen to be shrinked and fragmented. The results show that the causes of the antibacterial and cytotoxic effect against a wide range are thermostable substances isolated by the ethanol.

Changes in Endogenous Abscisic Acid, Jasmonic Acid and Sucrose Content during Bulb Development in the Cold-type Cultivar of Garlic (Allium sativum L.) of Korea (한지형 마늘의 인경 발육과정에서 식물내생호르몬 Abscisic Acid, Jasmonic Acid 및 당 함량변화)

  • This study was performed to investigate the role of plant growth substances on the bulbing of cold type of garlic (Allium sativum L. cv. Uiseongmaneul) during long and short day conditions. The change in endogenous plant hormones such as abscisic acid (ABA), jasmonic acid (JA) and sugar contents in leaf blade and sheath was examined during the growth stage from bulb differentiation (starting at April 16) to bulbing (April 24 to May 18) in cold type of garlic. In the long day condition, ABA contents were higher than short day condition and ABA contents of leaf sheath were higher than leaf blade. ABA contents of growth stage in garlic were not changed during growth stage from April 16 to May 2, however it rapidly increased during bulbing (May 2 to May 18). On the other hand, endogenous JA contents in short day condition did not change in long day condition, it increased from April 16 to May 2. JA contents in the leaf sheath (33.85-62.04 $ng{\cdot}g^{-1}$ DW) were higher than leaf blade (15.39-30.04 $ng{\cdot}g^{-1}$ DW). These results showed that garlic bulb differentiation and bulbing was induced by JA in leaf sheath. In long day condition, total sugar contents in the leaf blade were increased from bulb differentiation (April 16) to bulbing (May 4) and it was decreased during bulbing (May 4 to May 18) while the total sugar content in leaf sheath were gradually increased from bulb differentiation (April 16) to bulbing (May 18). In conclusion, our results showed that there is a significant correlation between the bulb development of garlic and hormonal content in the leaf sheath.

Antimicrobial Activity and Distilled Components of Garlic(Allium sativum L.) and Ginger(Zingiber officinale Roscoe) (마늘과 생강의 항균성 및 증류성분)

  • In order to investigate the possible use of spices as natural preservatives) antimicrobial activities of garlic and ginger, representative spices, were examinated. Distilled components of garlic and ginger were also analyzed. Each extract of garlic was more suppressive on the growth of Gram negative bacteria than that of Gram positive bacteria. Extracts of garlic were showed effective suppressions on the growth of yeasts. Of the extracts of garlic, ether extract was highest among antimicrobial activities. In the case of ginger, ether extract of ginger was effective for growth inhibition of tested bacteria. Distilled components of garlic and ginger were extracted by simultaneous steam distillation extraction apparatus. The concentrates were analyzed with GC/MSD and Kovat's retention index and identified 13 components and 21 components respectively. Distilled components of garlic and ginger were full of sulfur compounds and hydrocarbons, respectively. di-2-Propenyl disulfide, methyl 2-propenyl disulfide, 2-vinyl-4H-1,3-dithiirane and 3,3'-thiobis-1-propene were major principal aroma compounds in garlic. ${\alpha}-zingibirene$, ${\beta}-phellandrene$, ${\beta}-sesquiphellandrene$ and camphene were major principal compounds in ginger.
